© 2016 by the Midwest Political Science Association.Recent research has cast doubt on the potential for various electoral reforms to increase voter turnout. In this article, we examine the effectiveness of preregistration laws, which allow young citizens to register before being eligible to vote. We use two empirical approaches to evaluate the impact of preregistration on youth turnout. First, we implement difference-in-difference and lag models to bracket the causal effect of preregistration implementation using the 2000-2012 Current Population Survey. Second, focusing on the state of Florida, we leverage a discontinuity based on date of birth to estimate the effect of increased preregistration exposure on the turnout of young registrants. In both approaches, we find preregistration increases voter turnout, with equal effectiveness for various subgroups in the electorate. More broadly, observed patterns suggest that campaign context and supporting institutions may help to determine when and if electoral reforms are effective.

Cet article synthétise la théologie protestante des images en montrant qu’elle vise à prévenir l’idolâtrie. Il rappelle que l’iconoclasme de la Réforme a été sélectif. Il souligne l’existence de peintres protestants, de Hans Holbein le Jeune à André Bieler. Il explique comment les principaux réformateurs ont traité la question des images. Il distingue les positions luthérienne et calviniste, notamment quant à leur interprétation du commandement biblique qui interdit les idoles. Il précise enfin à quelles conditions une image peut transmettre l’Évangile.

Cette thèse commence comme une simple question en réponse au modèle du « parfait flâneur » que Baudelaire a élaboré dans Le peintre de la vie moderne (1853): un flâneur peut-il être imparfait? Je suggère trois interprétations possibles du mot « imparfait ». Il permet d’abord de sortir le flâneur du strict contexte du Paris du dix-neuvième siècle et permet des traductions imparfaites de personnages dans d’autres contextes. Ensuite, le flâneur déambule dans la dimension « imparfaite » de l’imagination fictionnelle – une dimension comparable à l’image anamorphique du crâne dans la peinture Les ambassadeurs de Holbein. Enfin, il réfère à l’imparfait conjugué, « l’imparfait flâneur » peut rappeler le personnage antihéroïque de l’humain dont l’existence est banale et inachevée, comme la phrase « il y avait ». Ces trois visions contribuent à la réinterprétation du flâneur dans le contexte de la fin du vingtième siècle. Mon hypothèse est que l’expérience urbaine du flâneur et la flânerie ne sont possibles que si l’on admet être imparfait(e), qu’on accepte ses imperfections et qu’elles ne nous surprennent pas. Quatre études de romans contemporains et de leurs villes respectives forment les principaux chapitres. Le premier étudie Montréal dans City of forgetting de Robert Majzels. J’examine les façons par lesquelles les personnages itinérants peuvent être considérés comme occupant (ou en échec d’occupation) du Montréal contemporain alors qu’ils sont eux-mêmes délogés. Quant au deuxième chapitre, il se concentre sur le Bombay de Rohinton Mistry dans A fine balance. Mon étude portera ici sur la question de l’hospitalité en relation à l’hébergement et au « dé-hébergement » des étrangers dans la ville. Le troisième chapitre nous amène à Hong-Kong avec la série Feituzhen de XiXi. Dans celle-ci, j’estime que la méthode spéciale de la marelle apparait comme une forme unique de flânerie imparfaite. Le quatrième chapitre étudie Istanbul à travers The black book d’Orhan Pamuk. Inspiré par les notions de « commencement » d’Edward Saïd, mon argumentaire est construit à partir de l’interrogation suivante : comment et quand commence une narration? En lieu de conclusion, j’ai imaginé une conversation entre l’auteur de cette thèse et les personnages de flâneurs imparfaits présents dans les différents chapitres.

Cordycepin (3′ deoxyadenosine) is a biologically active compound that, when incorporated during RNA synthesis in vitro, provokes chain termination due to the absence of a 3′ hydroxyl moiety. We were interested in the effects mediated by this drug in vivo and analyzed its impact on RNA metabolism of yeast. Our results support the view that cordycepin-triphosphate (CoTP) is the toxic component that is limiting cell growth through inhibition of RNA synthesis. Unexpectedly, cordycepin treatment modulated 3′ end heterogeneity of ACT1 and ASC1 mRNAs and rapidly induced extended transcripts derived from CYH2 and NEL025c loci. Moreover, cordycepin ameliorated the growth defects of poly(A) polymerase mutants and the pap1-1 mutation neutralized the effects of the drug on gene expression. Our observations are consistent with an epistatic relationship between poly(A) polymerase function and cordycepin action and suggest that a major mode of cordycepin activity reduces 3′ end formation efficiency independently of its potential to terminate RNA chain elongation. Finally, chemical-genetic profiling revealed genome-wide pathways linked to cordycepin activity and identified novel genes involved in poly(A) homeostasis.

To identify genes involved in poly(A) metabolism, we screened the yeast gene deletion collection for growth defects in the presence of cordycepin (3′-deoxyadenosine), a precursor to the RNA chain terminating ATP analog cordycepin triphosphate. Δpho80 and Δpho85 strains, which have a constitutively active phosphate-response pathway, were identified as cordycepin hypersensitive. We show that inorganic polyphosphate (poly P) accumulated in these strains and that poly P is a potent inhibitor of poly(A) polymerase activity in vitro. Binding analyses of poly P and yeast Pap1p revealed an interaction with a kD in the low nanomolar range. Poly P also bound mammalian poly(A) polymerase, however, with a 10-fold higher kD compared to yeast Pap1p. Genetic tests with double mutants of Δpho80 and other genes involved in phosphate homeostasis and poly P accumulation suggest that poly P contributed to cordycepin hypersensitivity. Synergistic inhibition of mRNA synthesis through poly P-mediated inhibition of Pap1p and through cordycepin-mediated RNA chain termination may thus account for hypersensitive growth of Δpho80 and Δpho85 strains in the presence of the chain terminator. Consistent with this, a mutation in the 3′-end formation component rna14 was synthetic lethal in combination with Δpho80. Based on these observations, we suggest that binding of poly P to poly(A) polymerase negatively regulates its activity.

 Cleavage factor IA (CF IA), cleavage and polyadenylation factor (CPF), constitute major protein complexes required for pre-mRNA 3' end formation in yeast. The Clp1 protein associates with Pcf11, Rna15 and Rna14 in CF IA but its functional role remained unclear. Clp1 carries an evolutionarily conserved P-loop motif that was previously shown to bind ATP. Interestingly, human and archaean Clp1 homologues, but not the yeast protein, carry 5' RNA kinase activity. We show that depletion of Clp1 in yeast promoted defective 3' end formation and RNA polymerase II termination; however, cells expressing Clp1 with mutant P-loops displayed only minor defects in gene expression. Similarly, purified and reconstituted mutant CF IA factors that interfered with ATP binding complemented CF IA depleted extracts in coupled in vitro transcription/3' end processing reactions. We found that Clp1 was required to assemble recombinant CF IA and that certain P-loop mutants failed to interact with the CF IA subunit Pcf11. In contrast, mutations in Clp1 enhanced binding to the 3' endonuclease Ysh1 that is a component of CPF. Our results support a structural role for the Clp1 P-loop motif. ATP binding by Clp1 likely contributes to CF IA formation and cross-factor interactions during the dynamic process of 3' end formation.

A presente investigação foi utilizada como suporte ao desenvolvimento das práticas pedagógicas desempenhadas pelo Mestrando Estagiário e comporta como problemática central, no que se refere ao corpo da investigação científica, Anamorfoses e Anamorfismos: da perceção à perceção visual. Nesta investigação, pretendeu-se analisar e fundamentar a componente histórica que enquadra o aparecimento e desenvolvimento da anamorfose, analisando-a de um ponto de vista crítico seletivo, desde os primórdios da sua origem, seguindo as fontes encontradas, até aos trabalhos anamórficos nas práticas artísticas contemporâneas. A primeira parte da dissertação resulta da abordagem de um conjunto de artistas, em particular de Leonardo Da Vinci, Vexierbild Dérchd Schon, Hans Holbein e, nas práticas artísticas contemporâneas, Julian Beever, Felice Varine, Helly Houle, Kurt Wenner, Eduardo Kobra, François Abélanet. Numa segunda etapa, ensaia-se o desenvolvimento de uma reconceptualização do conceito Anamorfismo na investigação, sendo o mesmo utilizado de modo a dar a conhecer ao leitor a verdadeira natureza das criações neste âmbito. O Anamorfismo, foi consequentemente aplicado à poesia, à literatura, à pintura, à arquitetura, às artes do corpo e a outras realidades artísticas. Estas aplicações surgem da necessidade de se incentivar um fruidor perspicaz e capaz de descodificar, analisar, compreender e interagir com o meio envolvente através de uma prática simultaneamente consciente e inconsciente do Anamorfismo. A prática consciente é cumprida pelo fruidor conhecedor dos meandros do processo anamórfico, levando-o à descodificação do meio que o envolve de forma sistemática, informada e cultivada, enquanto a prática inconsciente fundamenta-se na intuição que convoca toda envolvência do Anamorfismo, efetuando um conjunto de leituras do objeto criado partindo da profusão de associações essencialmente automáticas, logo muito ricas de sugestões criativas. Posteriormente, numa etapa mais prática da investigação, passaremos à aplicabilidade destes conceitos no contexto da prática pedagógica inicial, onde procuraremos trabalhar com os alunos de um ponto de vista teórico e seguidamente introduzindo os conteúdos desta investigação num conjunto de várias atividades práticas.

In The Middle Ground (1980), Margaret Drabble uses an intertextual web to depict the feminine search for identity and psychological integrity in middle age. This paper attempts to identify and analyze the absorption and integration of other texts in The Middle Ground. The dialogue, both in theme and style, with Virginia Woolf's Mrs Dalloway is initially considered, mostly from the perspective of parody. Other confluences are also studied: with Russian fairy tales, with paintings by Hans Holbein, J. B.Vanmour, Van Dyck, Claude Lorrain and Peter de Hooch, and also its references to the whole of Drabble's literary work.

Il lavoro consiste nella traduzione dell’adagio 2001, Herculei labores con commento delle righe 1-116, che comprendono il racconto della fatica di Ercole contro l’idra di Lerna e le interpretazioni che Erasmo ne fornisce per introdurre la filologia come impresa erculea in chiave autobiografica. L’introduzione ha lo scopo di presentare una sintesi degli elementi notevoli del commento e alcune osservazioni sull’autorappresentazione di sé dell’umanista. Erasmo fa dell’identificazione con Ercole un topos della propria descrizione in chiave ironica, ma si propone anche come emulo di Girolamo, di cui cura l’edizione delle lettere. Questo lavoro prende in considerazione infine il ritratto di Erasmo dipinto da Holbein e custodito a Longford Castle in relazione al testo dell’adagio, al quale allude con la scritta in primo piano, ΗΡΑΚΛΕΙΟΙ ΠΟΝΟΙ.

INTRODUCTION: Maintaining arterial blood glucose within tight limits is beneficial in critically ill patients. Upper and lower limits of detrimental blood glucose levels must be determined. METHODS: In 69 patients with severe traumatic brain injury (TBI), cerebral metabolism was monitored by assessing changes in arterial and jugular venous blood at normocarbia (partial arterial pressure of carbon dioxide (paCO2) 4.4 to 5.6 kPa), normoxia (partial arterial pressure of oxygen (paO2) 9 to 20 kPa), stable haematocrit (27 to 36%), brain temperature 35 to 38 degrees C, and cerebral perfusion pressure (CPP) 70 to 90 mmHg. This resulted in a total of 43,896 values for glucose uptake, lactate release, oxygen extraction ratio (OER), carbon dioxide (CO2) and bicarbonate (HCO3) production, jugular venous oxygen saturation (SjvO2), oxygen-glucose index (OGI), lactate-glucose index (LGI) and lactate-oxygen index (LOI). Arterial blood glucose concentration-dependent influence was determined retrospectively by assessing changes in these parameters within pre-defined blood glucose clusters, ranging from less than 4 to more than 9 mmol/l. RESULTS: Arterial blood glucose significantly influenced signs of cerebral metabolism reflected by increased cerebral glucose uptake, decreased cerebral lactate production, reduced oxygen consumption, negative LGI and decreased cerebral CO2/HCO3 production at arterial blood glucose levels above 6 to 7 mmol/l compared with lower arterial blood glucose concentrations. At blood glucose levels more than 8 mmol/l signs of increased anaerobic glycolysis (OGI less than 6) supervened. CONCLUSIONS: Maintaining arterial blood glucose levels between 6 and 8 mmol/l appears superior compared with lower and higher blood glucose concentrations in terms of stabilised cerebral metabolism. It appears that arterial blood glucose values below 6 and above 8 mmol/l should be avoided. Prospective analysis is required to determine the optimal arterial blood glucose target in patients suffering from severe TBI.
