The blow flies Chrysomya putoria and C. megacephala have 2n=12 chromosomes, five metacentric pairs of autosomes and an XX/XY sex chromosome pair. There are no substantial differences in the karyotype morphology of these two species, except for the X chromosome which is subtelocentric in C. megacephala and metacentric in C. putoria and is about 1.4 times longer in C. putoria. All autosomes were characterized by the presence of a C band in the pericentromeric region; C. putoria also has an interstitial band in pair III. The sex chromosomes of both species were heterochromatic, except for a small region at the end of the long arm of the X chromosome. Ribosomal genes were detected in meiotic chromosomes by FISH and in both species the NOR was located on the sex chromosomes. These results confirm that C. putoria was the species introduced into Brazil in 1970s, and not C. chloropyga as formerly described.

Rhodnius pallescens is the main vector of Trypanosoma cruzi in Panama and one of the most relevant secondary vectors in Colombia. Despite the importance of this species, there is limited knowledge about the genetic variability along its geographical distribution. In order to evaluate the degree of karyotype variability we analyzed the meiotic behavior and banding pattern of the chromosomes of 112 males of R. pallescens coming from different regions of Colombia and Panama. Using the C-banding technique we identified two chromosomal patterns or cytotypes characterized by differences in the amount, size and distribution of constitutive heterochromatic regions in the chromosome complement (2n = 20 autosomes plus XY in males). The individuals can be easily classified in each cytotype by the analysis of the chromosomes during first meiotic prophase. The frequencies of the cytotypes are variable according to the geographic origin of the populations. This chromosomal divergence together with morphological data supports the existence of three genetically different populations of R. pallescens and provides new information to understand the distribution dynamics of this species.

This region contains extensive euchromatic-like sequence and includes on average one transcript every 100 kb. These transcripts show multiple inter- and intrachromosomal copies, and extensive copy number and sequence variability. The sequencing of the "heterochromatic" regions of the human genome is likely to reveal many additional functional elements and provide important evolutionary information.

The banding pattern (G-, C-, AgNOR-staining) was described in karyotypes of Apodemus alpicola Heinrich, 1952 and A. microps Kratochvil et Rosicky, 1952 collected from the Alps and central Europe, Distinct differences between the two species were revealed in the distribution of C-heterochromatic regions in autosomes and the sex chromosomes, and the distribution of nucleolar organizer regions (NORs). Extensive variation in the distribution pattern of C-heterochromatin and NORs obviously exists among the wood mice of the subgenus Sylvaemus, and individual species can be distinguished according to a specific variation pattern. However, it seems premature to designate individual karyotypic forms as separate species, because the extent of overall geographical interpopulation variation is still not sufficiently known.

Structural variation has played an important role in the evolutionary restructuring of human and great ape genomes. Recent analyses have suggested that the genomes of chimpanzee and human have been particularly enriched for this form of genetic variation. Here, we set out to assess the extent of structural variation in the gorilla lineage by generating 10-fold genomic sequence coverage from a western lowland gorilla and integrating these data into a physical and cytogenetic framework of structural variation. We discovered and validated over 7665 structural changes within the gorilla lineage, including sequence resolution of inversions, deletions, duplications, and mobile element insertions. A comparison with human and other ape genomes shows that the gorilla genome has been subjected to the highest rate of segmental duplication. We show that both the gorilla and chimpanzee genomes have experienced independent yet convergent patterns of structural mutation that have not occurred in humans, including the formation of subtelomeric heterochromatic caps, the hyperexpansion of segmental duplications, and bursts of retroviral integrations. Our analysis suggests that the chimpanzee and gorilla genomes are structurally more derived than either orangutan or human genomes.

The variability of the lengths of the heterochromatic and euchromatic segments of the human Y chromosome was studied by a quantitative method of densitometric measurement in 60 normal and unrelated black individuals (30 with and 30 without devotional surnames), living in Salvador, Bahia, northeastern Brazil. Thirty normal and unrelated Caucasian individuals of European origin, living in Curitiba, Paraná, south Brazil, were included as controls. The heterochromatic segment and total Y chromosome lengths were greater in caucasians than in blacks without devotional surnames, and these were greater than in blacks with devotional surnames. These findings are in agreement with previous reports of a higher percentage of black ancestry in blacks carrying devotional surnames than those carrying non-devotional ones.

Thirty-six specimens of Schizodon nasutus (Anostomidae-Characiformes) from the middle Paraná River (Posadas, Argentina) were analyzed cytogenetically. The karyotype of this species was similar to those described for this species in the literature. C-banding technique showed a rich heterochromatic pattern relative to other Anostomidae species. The NORs were located on one chromosome pair in terminal position and showed a very marked size heteromorphism. A microchromosome was observed with a frequency of about 20% in the sample studied. This additional element was punctiform, negative C-band, and constant in all metaphase plates of the seven carriers. The present study is the first karyotypic approach to Schizodon nasutus from Argentina and the first description of microchromosome in Anostomidae

Peripheral glial cells consist of satellite, enteric glial, and Schwann cells. In dorsal root ganglia, besides pseudo-unipolar neurons, myelinated and nonmyelinated fibers, macrophages, and fibroblasts, satellite cells also constitute the resident components. Information on satellite cells is not abundant; however, they appear to provide mechanical and metabolic support for neurons by forming an envelope surrounding their cell bodies. Although there is a heterogeneous population of neurons in the dorsal root ganglia, satellite cells have been described to be a homogeneous group of perineuronal cells. Our objective was to characterize the ultrastructure, immunohistochemistry, and histochemistry of the satellite cells of the dorsal root ganglia of 17 adult 3-4-month-old Wistar rats of both genders. Ultrastructurally, the nuclei of some satellite cells are heterochromatic, whereas others are euchromatic, which may result from different amounts of nuclear activity. We observed positive immunoreactivity for S-100 and vimentin in the cytoplasm of satellite cells. The intensity of S-100 protein varied according to the size of the enveloped neuron. We also noted that vimentin expression assumed a ring-like pattern and was preferentially located in the cytoplasm around the areas stained for S-100. In addition, we observed nitric oxide synthase-positive small-sized neurons and negative large-sized neurons equal to that described in the literature. Satellite cells were also positive for NADPH-diaphorase, particularly those associated with small-sized neurons. We conclude that all satellite cells are not identical as previously thought because they have different patterns of glial marker expression and these differences may be correlated with the size and function of the neuron they envelope.

Pancreatic deoxyribonuclease preferentially digests active genes during all phases of the cell cycle including mitosis. Recently, a DNAse I-directed in ~ nick translation technique has been used to demonstrate differences in the DNAse I sensitivity of euchromatic and heterochromatic regions of mitotic chromosomes. This ill ~ technique has been used in this study to ask whether facultative heterochromatin of the inactive X chromosome can be distinguished from the active X chromosome in mouse and human tissues. In addition to this, in ~ nick translation has been used to distinguish constitutive heterochromatin in mouse and human mitotic chromosomes. Based on relative levels of DNAse I sensitivity, the inactive X chromosome could not be distinguished from the active X chromosome in either mouse or human tissues but regions of constitutive heterochromatin could be distinguished by their relative DNAse I insensitivity. The use of !D situ nick translation was also applied to tissue sections of 7.5 day mouse embryos to ask whether differing levels of DNAse I sensitivity could be detected between different tissue types. Differences in DNAse I sensitivities were detected in three tissues examined; embryonic ectoderm, an embryo-derived tissue, and two extraembryonic tissues, extraembryonic ectoderm and ectoplacental cone. Embryonic ectoderm and extraembryonic ectoderm nuclei possessed comparable levels of DNAse I sensitivity while ectoplacental cone was significantly less DNAse I sensitive. This suggests that tissue-specific mechanisms such as chromatin structure may be involved in the regulation of gene activity in certain tissue types. This may also shed some light on possible tissue specific mechanisms regulating X chromosome activity in the developing mouse embryo.

Chez la levure Saccharomyces cerevisiae, l'acétylation de l'histone H3 sur la lysine 56 (H3K56ac) est présente sur les histones néo-synthétisées déposées derrière les fourches de réplication et est essentielle pour préserver la viabilité cellulaire en réponse au dommage à l'ADN. La désacétylation d'H3K56 sur l'ensemble du génome catalysée par Hst3 et Hst4 et a lieu en phase G2 ou M. H3K56ac est une lame à double tranchant. L'absence d'H3K56ac rend les cellules sensibles aux dommages à l'ADN. En revanche, un excès d'acétylation d'H3K56 dans un mutant hst3Δ hst4Δ a des conséquences encore plus sévères tels que la thermo-sensibilité, l'hypersensibilité aux agents génotoxiques, l'instabilité génomique ainsi qu'une courte durée de vie réplicative. Les désacétylases Hst3 et Hst4 sont étroitement régulées au cours du cycle cellulaire afin de permettre à l'H3K56ac d'exercer son rôle en réponse aux dommages à l'ADN tout en évitant les conséquences néfastes de l'hyperacétylation d'H3K56. Dans cette thèse, nous avons identifié la machinerie moléculaire responsable de la dégradation de Hst3. De plus, nous avons exploré les raisons pour lesquelles l'absence de désacétylation donne lieu aux phénotypes du mutant hst3Δ hst4Δ. Au chapitre 2, nous démontrons que la dégradation d'Hst3 peut être complétée avant l'anaphase. Ceci suggère que la désacétylation de H3K56 a lieu durant une courte fenêtre du cycle cellulaire se situant entre la complétion de la phase S et la métaphase. De plus, nous avons identifié deux sites de phosphorylation d'Hst3 par la kinase cycline-dépendante 1 (Cdk1) et démontré que ces évènements de phosphorylation conduisent à la dégradation d'Hst3 in vivo. Nous avons aussi démontré que l'ubiquityltransférase Cdc34 et l'ubiquitine ligase SCFCdc4 sont requises pour la dégradation d'Hst3. Finalement, nous avons montré que la phosphorylation d'Hst3 par la kinase mitotique Clb2-Cdk1 peut directement entraîner l'ubiquitylation d'Hst3 par SCFCdc4 in vitro. Au chapitre 3, nous avons étudié les mécanismes moléculaires sous-jacents à la sensibilité extrême du mutant hst3Δ hst4Δ aux agents qui endommagent l'ADN. Nous avons établi qu'en raison de la présence anormale d'H3K56ac devant les fourches de réplication, le mutant hst3Δ hst4Δ exhibe une forte perte de viabilité lorsqu'exposé au méthyl méthanesulfonate (MMS) durant un seul passage à travers la phase S. Nous avons aussi découvert que, malgré le fait que le point de contrôle de réponse aux dommages à l'ADN est activé normalement dans le mutant hst3Δ hst4Δ, ce mutant est incapable de compléter la réplication de l'ADN et d'inactiver le point de contrôle pour une longue période de temps après exposition transitoire au MMS. L'ensemble de nos résultats suggère que les lésions à l'ADN induites par le MMS dans le mutant hst3Δ hst4Δ causent une forte perte de viabilité parce que ce mutant est incapable de compléter la réplication de l'ADN après une exposition transitoire au MMS. Dans la deuxième section du chapitre 3, nous avons employé une approche génétique afin d'identifier de nouveaux mécanismes de suppression de deux phénotypes prononcés du mutant hst3Δ hst4Δ. Nous avons découvert que la délétion de plusieurs gènes impliqués dans la formation de frontières entre l'hétérochromatine et de l'euchromatine atténue les phénotypes du mutant hst3Δ hst4Δ sans réduire l'hyperacétylation d'H3K56. Nos résultats indiquent aussi que l'abondante acétylation de l'histone H4 sur la lysine 16 (H4K16ac) est néfaste au mutant hst3Δ hst4Δ. Ce résultat suggère un lien génétique intriguant entre l'acétylation d'H3K56 et celle d'H4K16. L'existence de ce lien était jusqu'à présent inconnu. Nous avons identifié un groupe de suppresseurs spontanés où H3K56ac est indétectable, mais la majorité de nos suppresseurs ne montrent aucune réduction flagrante d'H3K56ac ou d'H4 K16ac par rapport aux niveaux observés dans le mutant hst3Δ hst4Δ. Une étude plus approfondie de ce groupe de suppresseurs est susceptible de mener à la découverte de nouveaux mécanismes génétiques ou épigénétiques permettant d'éviter les conséquences catastrophiques de l'hyperacétylation d'H3K56 chez le mutant hst3Δ hst4Δ. En résumé, cette thèse identifie la machinerie moléculaire responsable de la dégradation d'Hst3 (une désacétylase d'H3K56) durant une fenêtre de temps situées entre la fin de la phase S et la métaphase. Nos résultats permettent aussi d'expliquer pourquoi la dégradation d'Hst3 précède le début de la phase S durant laquelle l'acétylation d'H3K56 s'accumule derrière les fourches de réplication afin d'exercer son rôle de mécanisme de défense contre le dommage à l'ADN. De plus, nous avons identifié plusieurs suppresseurs qui permettent de contourner le rôle important d'Hst3 et Hst4 en réponse au dommage à l'ADN. Plusieurs suppresseurs révèlent un lien génétique inattendu entre deux formes abondantes d'acétylation des histones chez Saccharomyces cerevisiae, soit H3K56ac et H4K16ac.

L’influence de la couleur dans les mécanismes perceptuels et attentionnels a été étudiée. Quatre couleurs (le rouge, le vert, le bleu et le jaune) ont été calibrées individuellement à travers la technique heterochromatic flicker photometry. Suivant cela, les participants ont déployé leur attention à une cible (un cercle de couleur avec une ligne orientée). Les données électrophysiologiques ont été enregistrées pendant que les sujets performaient la tâche de recherche visuelle, et les analyses ont été basées sur les potentiels évoqués (PÉs). Trois composantes des PÉs ont été examinées : la posterior contralateral positivity (Ppc); la N2pc, reflétant le déploiement de l’attention visuo-spatiale et la temporal and contralateral positivity (Ptc). Des conditions dans lesquelles la cible était bleu ou rouge, lorsque comparées à des conditions avec une cible jaune ou verte suscitaient une N2pc plus précoce. Une amplitude plus élevée est aussi observée pour les cibles rouges pour les composantes Ppc et Ptc, reflétant une sélectivité pré-attentionelle. Ces résultats suggèrent de la prudence dans l’interprétation de données comparant des cibles de différentes couleurs dans des tâches de PÉs, et ce même lorsque les couleurs sont équiluminantes.

Introducción: Lucilia sericata es una especie de importancia médica y forense, utilizada en terapialarval para curar heridas crónicas y en estudios médico-legales empleada en la estimacióndel intervalo post mórtem y el traslado de cadáveres. No existen registros de las característicascitogenéticas de esta mosca en el neotrópico. El objetivo principal de este trabajo fue identificarlas características morfométricas cromosómicas y las estructuras primarias del cariotipo, a partirde especímenes de L. sericata de la cepa Bogotá, Colombia. Materiales y métodos: Se tomaronhuevos embrionados, que fueron previamente esterilizados en su superficie, se maceraron y luegofueron sembrados en el medio de cultivo L-15, suplementado con 20% de sfb, e incubados auna temperatura de 28 ºC, sin atmosfera de C02. La preparación de los cromosomas se obtuvo demonocapas celulares semiconfluentes, empleando diversas soluciones: antimitótica (Colchicina),hipotónica (KCl 0,075 M) y fijadora (Carnoy: metanol y ácido acético; 3:1). Se llevó a cabo la técnicade bandeo C para la identificación de regiones cromosómicas de heterocromatina constitutiva.Resultados: Se obtuvieron parámetros morfométricos de cada par cromosómico. El número diploidedel cariotipo obtenido de los cultivos celulares fue 2n = 12; éstos se clasificaron morfológicamente,de acuerdo con patrones previamente establecidos, así: los pares I, II, IV y V fueronmetacéntricos, y el par III fue submetacéntrico. A su vez, el par sexual fue heteromórfico, siendoel cromosoma X metacéntrico y el cromosoma Y submetacéntrico. El bandeo C fue positivo paratodos los pares cromosómicos. Conclusiones: Se establecieron las características citogenéticas deL. sericata, cepa Bogotá, Colombia, relacionadas con número, forma, tamaño, posición del centrómeroy regiones heterocromáticas de los cromosomas.

Mitotic chromosomes of Metynnis maculatus (KNER 1860) (Teleostei, Characiformes), a fish species that occurs in the Amazon and Parana-Paraguay river basins, were analyzed for the first time by Giemsa and Ag-NOR staining, C-banding and fluorescence in situ hybridization (FISH) with 18S and 5S rDNA sequences. The basic chromosome number of the species is 2n=62 (32M+22SM+4ST+4A) and, in addition to the 62 regular chromosomes, one small acrocentric supernumerary B chromosome was found in part of the specimens analyzed. Four active NORs were present, and constitutive heterochromatin blocks were found in the pericentromeric region of several chromosomes. A heterochromatic block was also present in the interstitial portion of the submetacentric NOR-bearing pair and the B chromosome was entirely heterochromatic. FISH using an 18S rDNA probe confirmed the results obtained with AgNO(3) staining, and an additional signal was also present on the B chromosomes. 5S rDNA sequences mapped only to the largest acrocentric pair. This is the first description of supernumerary B chromosomes in Serrasalminae, and this karyotype characterization may be useful in further studies about chromosome evolution in this fish group.

Kayotypes of four neotropical teiid lizard species (Tupinambinae) were herein studied after conventional as well as silver staining and CBG-banding: Crocodilurus amazonicus (2n = 34), Tupinambis teguixin (2n = 36), Tupinambis merianae and Tupinambis quadrilineatus (2n = 38). The karyological data for T. quadrilineatus as well as those obtained using differential staining for all species were unknown until now. The karyotypes of all species presented 12 macrochromosomes identical in morphology, but differed in the number of microchromosomes: 22 in C. amazonicus, 24 in T. teguixin and 26 in T. quadrilineatus and T. merianae. The Ag-NOR located at the secondary constriction at the distal end of pair 2 is shared by all species, contrasting with the variability observed for this character in species of the related Teiinae. CBG-banding revealed a species-specific pattern in T. quadrilineatus with conspicuous interstitial C-blocks at the proximal region of the long arm of pair 4 and the whole heterochromatic short arm of pair 6. The karyological data reported here corroborates the relationship hypothesis obtained for Tupinambis based on molecular characters. T. teguixin presents the putative ancestral karyotype for the genus with 2n = 36 whereas T. merianae and T. quadrilineatus exhibit 2n = 38, due to an additional pair of microchromosomes.
