The mechanism of hydroxylation reactions catalyzed by m-hydroxybenzoate-4-hydroxylase and anthranilate hydroxylase from Aspergillus niger was investigated using superoxide dismutase from ovine erythrocytes. Inclusion of superoxide dismutase in the assay mixtures of the two enzymes resulted in complete inhibition of the hydroxylation reaction, indicating the possible involvement of superoxide anions (O2−) in these reactions.

Considerações ambientais têm aumentado a pesquisa e o desenvolvimento de sistemas poliméricos aquosos para diversos tipos de aplicações, principalmente como revestimentos. Nesta dissertação, foram sintetizadas formulações não-poluentes à base de poliuretanos dispersos em água (PUDs), com 40% de teor de sólidos, na ausência de solventes orgânicos. Os monômeros empregados foram copolímeros em bloco à base de poli(glicol etilênico) e poli(glicol propilênico) (EG-b-PG), com teor de 25% de segmento hidrofílico EG, poli (glicol propilênico) (PPG), ácido dimetilolpropiônico (DMPA), diisocianato de isoforona (IPDI) e hidrazina (HYD), como extensor de cadeia. Foram variadas as razões entre o número de equivalente-grama de grupamentos isocianato e hidroxila (NCO/OH) e a proporção em equivalente-grama de PPG e dos copolímeros em bloco (EG-b-PG). Foi observado que a incorporação de altas quantidades de copolímero dificultou a síntese dos poliuretanos dispersos em água, levando à formação de géis. O tamanho médio de partícula e a viscosidade das dispersões foram determinados. Os filmes vazados a partir dessas dispersões foram avaliados quanto à capacidade de absorção de água, resistência mecânica, termogravimetria (TG), e caracterizados por espectroscopia na região do infravermelho (FTIR). As dispersões poliuretânicas produzidas se mostraram satisfatórias quando aplicadas como revestimento para madeira, metais e vidro

A área de pesquisa de materiais nanoestruturados tem recebido destacada atenção nos últimos anos por parte da comunidade científica, ressaltando-se os nanocompósitos à base de polímero e argila. Nesta Dissertação foram sintetizados nanocompósitos à base de poliuretano (NWPUs) com argilas do tipo montmorilonita (MMT), hidrofóbicas (organofílicas) e hidrofílica. Os monômeros empregados na síntese foram poli(glicol propilênico) (PPG); polibutadieno líquido hidroxilado (HTPB); ácido dimetilolpropiônico (DMPA), diisocianato de isoforona (IPDI) e hidrazina (HYD), como extensor de cadeia. Nas formulações, foram variadas as proporções de HTPB e o teor das argilas. As argilas organofílica Cloisite 30B e hidrofílica Cloisite Na+ foram incorporadas em 1, 3 e 5 %, enquanto que a organofílica Cloisite 15A foi incorporada no teor de 1%. As dispersões foram caracterizadas quanto ao teor de sólidos totais, tamanho médio de partículas e viscosidade. Os filmes vazados a partir das dispersões foram caracterizados por espectroscopia na região do infravermelho (FTIR), difração de raios-x (XDR), microscopia eletrônica de varredura (SEM) e microscopia de força atômica (AFM). A resistência térmica e a transição vítrea dos materiais foram determinadas por termogravimetria (TG) e calorimetria diferencial de varredura (DSC), respectivamente. O teor de absorção de água e o comportamento mecânico dos filmes foram avaliados. Foi verificada, por XRD, a ausência do pico de cristalinidade, característico das argilas puras e não-deslaminadas. As micrografias obtidas por SEM confirmam uma dispersão homogênea das argilas na matriz poliuretânica. Os filmes à base de nanocompósitos (NWPUs) apresentaram propriedades mecânicas superiores às apresentadas por aqueles obtidos a partir das dispersões sem a presença de argila (WPUs). A adesão dos revestimentos formados, pela aplicação das dispersões em diversos substratos, também foi verificada visualmente. Os resultados das análises mostraram que houve a formação de nanocompósitos à base de água, formados tanto a partir da argila hidrofílica quanto das organofílicas

Foram produzidas dispersões aquosas não-poluentes e inovadoras de poli(uretano-uréia) à base de poli(glicol propilênico) (PPG) e de copolímeros em bloco, poli(glicol etilênicob-glicol propilênico), (EG-b-PG) com teor de 7% de unidades de glicol etilênico. Os poli(uretano-uréia)s foram preparados, em duas etapas, pelo método do prepolímero. Na primeira etapa, foram obtidos anionômeros, sintetizados em massa, à base do copolímero EG-bPG, PPG, ácido dimetilol propiônico (DMPA) (gerador dos sítios aniônicos) e diisocianato de isoforona (IPDI). Ainda na primeira etapa, os grupos carboxílicos do DMPA foram submetidos à neutralização com trietilamina (TEA). Na segunda etapa, foi realizada a dispersão do prepolímero anionômero e, em seguida, foi conduzida a reação de extensão de cadeia com a hidrazina (HYD). Nas diferentes formulações, foram variadas a razão NCO/OH e as proporções de PPG e do copolímero em bloco EG-b-PG. Além disso, foram obtidas dispersões aquosas programadas para teores de sólidos de 40 e 50%. As dispersões foram avaliadas quanto ao teor de sólidos totais, tamanho médio de partícula e viscosidade. Os filmes, obtidos por vazamento das dispersões, foram avaliados quanto à absorção de água, termogravimetria (TG) e propriedades mecânicas (ensaio de tração). Os filmes vazados e os monômeros foram caracterizados por espectrometria na região de infravermelho (FTIR) As dispersões obtidas se mostraram adequadas para serem aplicadas como revestimento para madeira, metais e vidro

Tetralin hydrogenation (HYD) and thiophene hydrodesulfurization (HDS) were studied for the supported MoS2 and WS2 sulfides, either non-promoted or promoted with Co and Ni. The supports used were ZrO2, alumina-stabilized TiO2 and pure alumina. Preparation of catalysts included presulfidation of non-promoted system with subsequent addition of promoter and resulfidation. It has been found that the nature of promoter plays determining role for the catalytic performance. The most active in both HYD and HDS reactions are Ni-promoted Mo and W catalysts, supported on ZrO2. (C) 2003 Published by Elsevier B.V.

A series of unsupported dimolybdenum nitride (gamma-Mo(2)N) catalysts differing in surface area were prepared by temperature programmed reduction of MoO(3) with a mixture of NH(3):N(2) (90:10). Characterization of catalysts by BET, XRD, TPR and XPS techniques was carried out. The samples were used as catalysts in hydrotreating reactions (simultaneous hydrodesulfurization of thiophene and hydrogenation of cyclohexene). Low surface area gamma-Mo(2)N materials show much higher specific conversions than those with higher surface area. These results indicate that HDS and HYD reactions over gamma-Mo(2)N seem to be structure-sensitive. The relative exposure extent of crystalline planes (111) and (200) over the different catalysts can be associated with their hydrogen adsorption capacities and with their catalytic performances. The catalytic activities are significantly affected by the catalyst pretreatment conditions. (C) 1999 Elsevier Science B.V. All rights reserved.

The primary purpose of the current investigation was to develop an elevated muscle fluid level using a human in-vivo model. The secondary purpose was to determine if an increased muscle fluid content could alter the acute muscle damage response following a bout of eccentric exercise. Eight healthy, recreationally active males participated in a cross-over design involving two randomly assigned trials. A hydration trial (HYD) consisting of a two hour infusion of a hypotonic (0.45%) saline at a rate of 20mL/minVl .73m"^ and a control trial (CON), separated by four weeks. Following the infusion (HYD) or rest period (CON), participants completed a single leg isokinetic eccentric exercise protocol of the quadriceps, consisting of 10 sets of 10 repetitions with a one minute rest between each set. Muscle biopsies were collected prior to the exercise, immediately following and at three hours post exercise. Muscle analysis included determination of wet-dry ratios and quantification of muscle damage using toluidine blue staining and light microscopy. Blood samples were collected prior to, immediately post, three and 24 hours post exercise to determine changes in creatine kinase (CK), lactate dehydrogenase (LD), interleukin-6 (IL-6) and Creactive protein (CRP) levels. Results demonstrated an increased muscle fluid volume in the HYD condition following the infusion when compared to the CON condition. Isometric peak torque was significantly reduced following the exercise in both the HYD and CON conditions. There were no significant differences in the number of areas of muscle damage at any of the time points in either condition, with no differences between conditions. CK levels were significantly greater 24hour post exercise compared to pre, immediately and three hours post similarly in both conditions. LD in the HYD condition followed a similar trend as CK with 24 hour levels higher than pre, immediately post and three hours post and LD levels were significantly greater 24 hours post compared to pre levels in the CON condition, with no differences between conditions. A significant main effect for time was observed for CRP (p<0.05) for time, such that CRP levels increased consistently at each subsequent time point. However, CRP and IL-6 levels were not different at any of the measured time points when comparing the two conditions. Although the current investigation was able to successfully increase muscle fluid volume and an increased CK, LD and CRP were observed, no muscle damage was observed following the eccentric exercise protocol in the CON or HYD conditions. Therefore, the hypotonic infusion used in the HYD condition proved to be a viable method to acutely increase muscle fluid content in in-vivo human skeletal muscle.

Solutions of a two-dimensional dam break problem are presented for two tailwater/reservoir height ratios. The numerical scheme used is an extension of one previously given by the author [J. Hyd. Res. 26(3), 293–306 (1988)], and is based on numerical characteristic decomposition. Thus approximate solutions are obtained via linearised problems, and the method of upwind differencing is used for the resulting scalar problems, together with a flux limiter for obtaining a second order scheme which avoids non-physical, spurious oscillations.

Acanthamoeba species are frequently isolated from soil and water collections. In the environment, the organisms multiply as phagotrophic trophozoites and encyst under adverse conditions. Several species are known to infect man, causing keratitis and opportunistic diseases. The mechanisms underlying tissue damage and invasion by the amoebae are being elucidated and the involvement of secreted peptidases, particularly serine peptidases, has been demonstrated. Here, elastase activity was examined in Acanthamoeba-conditioned medium (ACM), making use of elastin-Congo red (ECR) and synthetic peptide p-nitroanilide substrates. ACM hydrolysed ECR over a broad pH range and optimally at a pH of 7.5 and above. Indicating the activity of serine and metallopeptidases, Congo red release was potently inhibited by PMSF, antipain, chymostatin and 1,10-phenanthroline, partially reduced by elastatinal and EDTA, and unaffected by 1,7-phenanthroline and E-64. Screening with synthetic substrates mainly showed the activity of serine peptidases. ACM efficiently hydrolysed Suc-Ala(2)-Pro-Leu-pNA and Suc-Ala(2)-Pro-Phe-pNA over a broad pH range (7.0-9.5) and was weakly active against Suc-Ala(3)-pNA, a substrate found to be optimally hydrolysed at a pH around 7.0. Following ammonium sulfate precipitation of ACM proteins and FPLC analysis, the majority of the ECR-splitting activity, characterised as serine peptidases, bound to CM-sepharose and co-eluted with part of the Suc-Ala(2)-Pro-Phe-pNA-hyd to lysing activity in a gradient of 0-0.6 M NaCl. In the corresponding FPLC fractions, serine peptidases resolving in the region of 70-130 kDa were detected in gelatin gels. Overall, the results demonstrate that trophozoites secrete elastases, and additionally suggest the high molecular weight serine peptidases as possible elastase candidates. (C) 2009 Elsevier B.V.
