In chronic myeloid leukemia and Philadelphia-positive acute lymphoblastic leukemia patients resistant to tyrosine kinase inhibitors (TKIs), BCR-ABL kinase domain mutation status is an essential component of the therapeutic decision algorithm. The recent development of Ultra-Deep Sequencing approach (UDS) has opened the way to a more accurate characterization of the mutant clones surviving TKIs conjugating assay sensitivity and throughput. We decided to set-up and validated an UDS-based for BCR-ABL KD mutation screening in order to i) resolve qualitatively and quantitatively the complexity and the clonal structure of mutated populations surviving TKIs, ii) study the dynamic of expansion of mutated clones in relation to TKIs therapy, iii) assess whether UDS may allow more sensitive detection of emerging clones, harboring critical 2GTKIs-resistant mutations predicting for an impending relapse, earlier than SS. UDS was performed on a Roche GS Junior instrument, according to an amplicon sequencing design and protocol set up and validated in the framework of the IRON-II (Interlaboratory Robustness of Next-Generation Sequencing) International consortium.Samples from CML and Ph+ ALL patients who had developed resistance to one or multiple TKIs and collected at regular time-points during treatment were selected for this study. Our results indicate the technical feasibility, accuracy and robustness of our UDS-based BCR-ABL KD mutation screening approach. UDS was found to provide a more accurate picture of BCR-ABL KD mutation status, both in terms of presence/absence of mutations and in terms of clonal complexity and showed that BCR-ABL KD mutations detected by SS are only the “tip of iceberg”. In addition UDS may reliably pick 2GTKIs-resistant mutations earlier than SS in a significantly greater proportion of patients.The enhanced sensitivity as well as the possibility to identify low level mutations point the UDS-based approach as an ideal alternative to conventional sequencing for BCR-ABL KD mutation screening in TKIs-resistant Ph+ leukemia patients

Convection in the tropics is observed to involve a wide-ranging hierarchy of scales from a few kilometers to the planetary scales and also has a profound impact on short-term climate. The mechanisms responsible for this behavior present a major unsolved problem. A promising emerging approach to address these issues is cloud-resolving modeling. Here a family of numerical models is introduced specifically to model the feedback of small-scale deep convection on tropical planetary waves and tropical circulation in a highly efficient manner compatible with the approach through cloud-resolving modeling. Such a procedure is also useful for theoretical purposes. The basic idea in the approach is to use low-order truncation in the meriodonal direction through Gauss–Hermite quadrature projected onto a simple discrete radiation condition. In this fashion, the cloud-resolving modeling of equatorially trapped planetary waves reduces to the solution of a small number of purely zonal two-dimensional wave systems along a few judiciously chosen meriodonal layers that are coupled only by some additional source terms. The approach is analyzed in detail with full mathematical rigor for linearized equatorial primitive equations with source terms.

In the last fifteen years a fundamental shift in the strategic balance in Asia – Pacific has taking place. The article aims at discussing the difficulties the Japan-US military Alliance has adaptating to this new regional strategic environment. It also explains the new strategy that President Obama´s Administration is trying to implement, initially called “Pivot to Asia” and later “Rebalance”. The article tries to show the challenges that this new strategy implies for both Japan and the United States, enumerating different scenarios some of them not very likely and highlighting on the contrary the possibility of a more assertive and military independent Japan.

This study focuses on the European Economic Area (EEA), which is a very ‘deep’ instance of market integration between the European Union, Iceland, Liechtenstein and Norway. The EEA in fact is just the ‘internal-market-minus’, i.e. minus agriculture and fisheries, for its three non-EU members. As an extension to the single market of a mere 1% in GDP, the EEA has attracted little attention, not least because it functions quite well. Even less known, however, is the role of Liechtenstein within the EEA, which this book also attempts to clarify.

In their March 2015 Consultation paper on the review of the European Neighbourhood Policy (ENP), the European Commission and the High Representative raised the question whether the Deep and Comprehensive Free Trade Areas (DCFTAs) “are the right objective for all or should more tailor-made alternatives be developed, to reflect differing interests and ambitions of some partners?” Such ambitious but complex trade agreements have now finally been signed with Ukraine, Moldova and Georgia, but they are still on the table for several Mediterranean ENP countries. Although these Mediterranean partners have a completely different political, economic and legal relationship with the EU, some important lessons can be drawn from the ‘Eastern DCFTA experience’. In particular, the DCFTA negotiators should avoid overly ambitious and ill-defined legislative approximation commitments and develop a comprehensive implementation strategy.

Purpose – This paper aims to focus on developing critical understanding in human resource management (HRM) students in Aston Business School, UK. The paper reveals that innovative teaching methods encourage deep approaches to study, an indicator of students reaching their own understanding of material and ideas. This improves student employability and satisfies employer need. Design/methodology/approach – Student response to two second year business modules, matched for high student approval rating, was collected through focus group discussion. One module was taught using EBL and the story method, whilst the other used traditional teaching methods. Transcripts were analysed and compared using the structure of the ASSIST measure. Findings – Critical understanding and transformative learning can be developed through the innovative teaching methods of enquiry-based learning (EBL) and the story method. Research limitations/implications – The limitation is that this is a single case study comparing and contrasting two business modules. The implication is that the study should be replicated and developed in different learning settings, so that there are multiple data sets to confirm the research finding. Practical implications – Future curriculum development, especially in terms of HE, still needs to encourage students and lecturers to understand more about the nature of knowledge and how to learn. The application of EBL and the story method is described in a module case study – “Strategy for Future Leaders”. Originality/value – This is a systematic and comparative study to improve understanding of how students and lecturers learn and of the context in which the learning takes place.

This document is summarizing a major part of the work performed by the FP7-JERICO consortium, including 27 partner institutions, during 4 years (2011-2015). Its objective is to propose a strategy for the European coastal observation and monitoring. To do so we give an overview of the main achievements of the FP7-JERICO project. From this overview, gaps are analysed to draw some recommendations for the future. Overview, gaps and recommendation are addressed at both Hardware and Software levels of the JERICO Research Infrastructure. The main part of the document is built upon this analysis to outcome a general strategy for the future, giving priorities to be targeted and some possible funding mechanisms, but also upon discussions held in dedicated JERICO strategy workshops. This document was initiated in 2014 by the coordination team but considering the fact that an overview of the entire project and its achievement were needed to feed this strategy deliverable it couldn’t ended before the end of FP7-JERICO, April 2015. The preparation of the JERICO-NEXT proposal in summer 2014 to answer an H2020 call for proposals pushed the consortium ahead, fed deep thoughts about this strategy but the intention was to not propose a strategy only bounded by the JERICO-NEXT answer. Authors are conscious that writing JERICO-NEXT is even drawing a bias in the thoughts and they tried to be opened. Nevertheless, comments are always welcome to go farther ahead. Structure of the document The Chapter 3 introduces the need of sustained coastal observatories, from different point of view including a short description of the FP7-JERICO project. In Chapter 4, an analysis of the JERICO coastal observatory Hardware (platforms and sensors) in terms of Status at the end of JERICO, identified gaps and recommendations for further development is provided region by region. The main challenges that remain to be overcome is also summarized. Chapter 5 is dedicated the JERICO infrastructure Software (calibration, operation, quality assessment, data management) and the progress made through JERICO on harmonization of procedures and definition of best practices. Chapter 6 provides elements of a strategy towards sustainable and integrated coastal observations for Europe, drawing a roadmap for cost-effective scientific-based consolidation of the present infrastructure while maximizing the potential arising from JERICO in terms of innovation, wealth-creation, and business development. After reading the chapter 3, for who doesn’t know JERICO, any chapter can be read independently. More details are available in the JERICO final reports and its intermediate reports; all are available on the JERICO web site (www.jerico-FP7.eu) as well as any deliverable. Each chapter will list referring JERICO documents. A small bibliographic list is available at the end of this deliverable.

This paper investigates three decision problems with potential to optimize operation and maintenance and logistics strategies for offshore wind farms: the timing of pre-determined jack-up vessel campaigns; selection of crew transfer vessel fleet; and timing of annual services. These problems are compared both in terms of potential cost reduction and the stochastic variability and associated uncertainty of the outcome. Pre-determined jack-up vessel campaigns appear to have a high cost reduction potential but also a higher stochastic variability than the other decision problems. The paper also demonstrates the benefits and difficulties of considering problems together rather than solving them in isolation.

An unstructured mesh �nite volume discretisation method for simulating di�usion in anisotropic media in two-dimensional space is discussed. This technique is considered as an extension of the fully implicit hybrid control-volume �nite-element method and it retains the local continuity of the ux at the control volume faces. A least squares function recon- struction technique together with a new ux decomposition strategy is used to obtain an accurate ux approximation at the control volume face, ensuring that the overall accuracy of the spatial discretisation maintains second order. This paper highlights that the new technique coincides with the traditional shape function technique when the correction term is neglected and that it signi�cantly increases the accuracy of the previous linear scheme on coarse meshes when applied to media that exhibit very strong to extreme anisotropy ratios. It is concluded that the method can be used on both regular and irregular meshes, and appears independent of the mesh quality.
