Several lines of evidence indicate that the adapter molecule p130CAS (crk-associated substrate (CAS)) is required for src-mediated cellular transformation. CAS has been shown to be heavily tyrosine-phosphorylated in src-transformed cells, and genetic variants of src that are deficient in CAS binding are also unable to mediate cellular transformation. In this report, we investigated whether CAS phosphorylation and/or its association with src are required elements of the transformation process. Expression of the carboxy-terminal src binding domain of CAS in Rat 1 fibroblasts expressing a temperature-sensitive allele of v-src inhibited the formation of src-CAS complexes and also inhibited tyrosine phosphorylation of CAS. However, expression of this protein had no effect on morphological transformation, src-mediated actin rearrangements, or anchorage-independent growth of these cells when grown at the src-permissive temperature. Thus, the ability of activated src to mediate cellular transformation is either largely independent of endogenous CAS phosphorylation and/or its association with CAS or, alternatively, the carboxy-terminus of CAS may substitute for endogenous CAS in the process of src-mediated transformation.

p130(Cas) (crk associated substrate) has the structural characteristics of an adapter protein, containing multiple consensus SH2 binding sites, an SH3 domain, and a proline-rich domain. The structure of p130(Cas) suggests that it may act to provide a framework for protein-protein interactions; however, as yet, its functional role in cells is unknown. In this report we show that p130(Cas) is localized to focal adhesions. We demonstrate that p130(Cas) associates both in vitro and in vivo with pp125(FAK) (focal adhesion kinase), a kinase implicated in signaling by the integrin family of cell adhesion receptors. p130(Cas) also associates with pp41/43(FRNK) (pp125(FAK)-related, non-kinase), an autonomously expressed form of pp125(FAK) composed of only the C-terminal noncatalytic domain. We show that the association of p130(Cas) with pp125(Fak) and pp41/43(FRNK) is direct, and is mediated by the binding of the SH3 domain of p130(Cas) to a proline-rich sequence present in both the C terminus of pp125(FAK) and in pp41/43(FRNK). In agreement with recent studies we show that p130(Cas) is tyrosine-phosphorylated upon integrin mediated cell adhesion. The association of p130(Cas) with pp125(FAK), a kinase which is activated upon cell adhesion, is likely to be functionally important in integrin mediated signal transduction.

Adaptor proteins play an important role in signal transduction by regulating the establishment and maintenance of functionally important protein complexes. A recently described member of this group of proteins is p130cas (CAS), which contains numerous sequence motifs predicted to be involved in mediating protein-protein interactions. We propose that adaptor molecules like CAS may help determine the response of a cell to a particular signal by interacting with specific subsets of cellular proteins. To test this hypothesis, we have identified potential binding partners of CAS that may play a rote in cellular transformation by the oncoproteins v-SRC and/or v-CRK. We show that individual domains of CAS associate with specific subsets of proteins in vitro, and that many of these interactions are dependent on the state of tyrosine-phosphorylation of CAS. Sequences necessary for interacting with the focal adhesion kinase pp125FAK (FAK), v-SRC and v-CRK have been mapped to distinct regions of CAS. In addition, the identification of a number of putative CAS-binding partners that are present in crk-transformed cell extracts but undetectable in normal and src-transformed cell extracts supports a model in which unique protein complexes are formed in response to different signals.

International exhibitions were greatly responsible for the modernization of western society. The motive for these events was based on the possibility of enhancing the country’s international status abroad. The genesis of world exhibitions came from the conviction that humanity as a whole would improve the continual flow of new practical applications, the development of modern communication techniques and the social need for a medium that could acquaint the general public with changes in technology, economy and society .
Since the first national industrial exhibitions in Paris during the eighteenth century and especially starting from the first Great Exhibition in London’s Hyde Park in 1851 these international events spread steadily all over Europe and the United States, to reach Latin America in the beginnings of the twentieth century . The work of professionals such as Daniel Burnham, Werner Hegemann and Elbert Peets made the relation between exhibitions and urban transformation a much more connected one, setting a precedent for subsequent exhibitions.
In Buenos Aires, the celebration of the centennial of independence from Spain in 1910 had many meanings and repercussions. A series of factors allowed for a moment of change in the city. Official optimism, economical progress, inequality and social conflict made of this a suitable time for transformation. With the organization of the Exposición Internacional the government had, among others, one specific aim: to achieve a network of visual tools to set the feeling of belonging and provide an identity for the mixture of cultures that populated the city of Buenos Aires at the time. Another important objective of the government was to put Buenos Aires at the level of European cities.
Foreign professionals had a great influence in the conceptual and factual shaping of the exhibition and in the subsequent changes caused in the urban condition. The exhibition had an important role in the ways of thinking the city and in the leisure ideas it introduced. The exhibition, as a didactic tool, worked as a precedent for conceiving leisure spaces in the future. Urban and landscape planners such as Joseph Bouvard and Charles Thays were instrumental in great part of the design of the Exhibition, but it was not only the architects and designers who shaped the identity of the fair. Other visitors such as Jules Huret or Georges Clemenceau were responsible for giving the city an international image it did not previously have.
This paper will explore on the one hand the significance of the exhibition of 1910 for the shaping of the city and its image; and on the other hand, the role of foreign professionals and the reach these influences had.

RATIONALE: Cigarette smoke exposure is associated with an increased risk of the acute respiratory distress syndrome (ARDS); however, the mechanisms underlying this relationship remain largely unknown.

OBJECTIVE: To assess pathways of lung injury and inflammation in smokers and non-smokers with and without lipopolysaccharide (LPS) inhalation using established biomarkers.

METHODS: We measured plasma and bronchoalveolar lavage (BAL) biomarkers of inflammation and lung injury in smokers and non-smokers in two distinct cohorts of healthy volunteers, one unstimulated (n=20) and one undergoing 50 μg LPS inhalation (n=30).

MEASUREMENTS AND MAIN RESULTS: After LPS inhalation, cigarette smokers had increased alveolar-capillary membrane permeability as measured by BAL total protein, compared with non-smokers (median 274 vs 208 μg/mL, p=0.04). Smokers had exaggerated inflammation compared with non-smokers, with increased BAL interleukin-1β (p=0.002), neutrophils (p=0.02), plasma interleukin-8 (p=0.003), and plasma matrix metalloproteinase-8 (p=0.006). Alveolar epithelial injury after LPS was more severe in smokers than non-smokers, with increased plasma (p=0.04) and decreased BAL (p=0.02) surfactant protein D. Finally, smokers had decreased BAL vascular endothelial growth factor (VEGF) (p<0.0001) with increased soluble VEGF receptor-1 (p=0.0001).

CONCLUSIONS: Cigarette smoke exposure may predispose to ARDS through an abnormal response to a 'second hit,' with increased alveolar-capillary membrane permeability, exaggerated inflammation, increased epithelial injury and endothelial dysfunction. LPS inhalation may serve as a useful experimental model for evaluation of the acute pulmonary effects of existing and new tobacco products.

US presidents have expanded executive power in times of war and emergency,sometimes aggressively so. This article builds on the application of punctuated equilibria theory by Burnham (1999 and Ackerman (1999). Underpinning this theory is the notion that rapid changes in - or external shocks to - domestic and international society impose new and insistent demands on the state. In so doing, they produce important and decisive moments of institutional mobilization and creativity, disrupt a pre-existing, relatively stable, equilibrium between the Congress and the president, and precipitate decisions or nondecisions by the electorate and political leaders that define the contours for action when the next crisis or external shock occurs. The article suggests that the combination of President George W. Bush's presidentialist doctrine, 9/11 and the 'war' on terror has consolidated a new, constitutional equilibrium. While some members of Congress contest the new order, the Congress collectively has acquiesced in its own marginalization. The article surveys a wide range of executive power assertions and legislative retreats. It argues that power assertions generally draw on precedent: on, for example, a tradition of wartime presidential extraconstitutional leadership extending to presidents, such as John Adams and Abraham Lincoln,as well as to Cold War and post-Cold War presidentialism.

Le présent mémoire de maîtrise constitue une recherche qualitative qui porte sur le rôle des agents d’intervention œuvrant au sein des centres jeunesse, dans le processus de réadaptation des jeunes. Peu des études et chercheurs se sont consacrés au rôle des agents d’intervention. Pourtant ils travaillent quotidiennement auprès des jeunes placés en CJM-IU et collaborent avec les éducateurs. Les pages suivantes s’attardent au rôle de l’agent d’intervention ainsi qu’à la manière dont ce rôle est situé dans un contexte de centre de réadaptation comme le Centre jeunesse de Montréal – Institut universitaire (CJM-IU). Le rôle et les pratiques de l’agent d’intervention sont expliqués selon la perception des agents eux-mêmes, de jeunes et de chefs de permanence rencontrés. Ainsi, à la fin de ce mémoire de maîtrise, un portrait plus détaillé de l’agent d’intervention qui travaille auprès des adolescents de la Cité-Des-Prairies et du Mont St-Antoine est dressé. L’évolution du rôle et les pratiques du milieu sont également relatées afin de répondre aux objectifs de recherche.

El siguiente trabajo tiene como objetivo el Estado del arte acerca de la discusión teórica de la repercusión de la unión monetaria en el principio de soberanía nacional, específicamente el caso de Gran Bretaña, ya que éste es el único país que expresa abiertamente su incertidumbre referente a algún tipo de amenaza a su soberanía. Se pretende precisar si existen criterios concluyentes, o por el contrario determinar si no hay claridad con respecto al futuro de Gran Bretaña como miembro de la unión monetaria.A partir de lo anterior, se plantean como sus propósitos particulares construir un marco conceptual acerca de la soberanía. Este estado del arte se inicia con citas de los principales autores de finales de siglo XVII y XVIII y finaliza con las posiciones conceptuales aportadas por los teóricos modernos motivados por el nacimiento de la Unión Europea como un nuevo orden político. Entre las que se citan: modelo centrico o intergubernamentalista, modelo de gobernabilidad multi-nivel, modelo neo-funcionalista y modelo federalista. Igualmente, el trabajo, busca desarrollar un marco conceptual sobre Unión Monetaria y su evolución hasta el logro de una unión fiscal europea en el periodo de 1950-2010, establecer las relaciones teóricas entre Soberanía del Estado y Unión Monetaria y por último realizar un análisis histórico hermenéutico de la Unión Europea y el caso del Reino Unido de acuerdo a las relaciones evidenciadas en los apartados teóricos entre soberanía del Estado y Unión Monetaria.

Este trabajo lleva a cabo una compilación del material escrito acerca del liderazgo educativo, sus implicaciones sociales, educativas y culturales. Así mismo identifica las corrientes que actualmente predominan en el liderazgo educativo como es el Liderazgo pedagógico y el liderazgo administrativo. También brinda un acercamiento a los esfuerzos y la situación actual del liderazgo educativo en Colombia, con el fin de determinar si existe en efecto y liderazgo educativo efectivo en nuestro país. Este trabajo se lleva a cabo recopilando la información de la situación actual y literatura de varios países y encontrando puntos en comuna lo largo de los mismos.

El propósito de esta disertación es rescatar las interpretaciones de Tierra y Mar en lo que se refiere al surgimiento del Orden Internacional Moderno, con el objetivo de explicar la percepción de los acontecimientos por parte de Carl Schmitt y su aporte filosófico-jurídico a la comprensión de la historia y teoría de las Relaciones Internacionales. La particularidad de esta obra de Schmitt es su estilo profundo: la variedad y amplitud de temas que abarca, que contrasta con la brevedad del texto. El presente trabajo ha encontrado vacíos en el estudio de Tierra y Mar, así como interpretaciones distintas a las propuestas por el autor, en lo concerniente al surgimiento del Orden Internacional Moderno. La originalidad de la obra de Schmitt consiste en proponer, en realidad, dos órdenes internacionales distintos que surgen en el mundo moderno.

El emprendimiento como campo de estudio ha cobrado una creciente relevancia en las agendas académicas y gubernamentales del último quinquenio por su capacidad para empujar el desarrollo económico y los procesos de innovación en los países. Un ejemplo de ello es Colombia, donde el nivel de impacto ha sido tan elevado que se han formalizado desde marcos regulatorios como la ley 1014 del 2006 para el fomento a la cultura empresarial, hasta instituciones de todo tipo para el fomento al emprendimiento empresarial. No obstante, los esfuerzos por estructurar espacios para estimular el emprendimiento deben trascender más allá del simple asistencialismo, para centrarse en la definición de planes estratégicos que permitan la formación de emprendedores de una manera articula y congruente con objetivos y políticas claramente identificadas. Por tal razón, desde el centro de emprendimiento de la Universidad de Rosario se impulsó el desarrollo de un estudio que permitiese la caracterización del perfil emprendedor de los estudiantes, a fin de identificar sus fortalezas y debilidades. Este estudio toma como base la metodología de características del comportamiento emprendedor o CCE para estudiar los alumnos de pregrado de la Facultad de Administración en cada uno de sus tres programas. Al final con estos resultados el Centro Emprendimiento podrá formular cada uno de sus planes, programas y proyectos, así como también políticas asociadas al desarrollo de las cualidades necesarias para formar mejores emprendedores (Pulgarín, S. 2011).

Este texto comienza con un ensayo autobiográfico que explora los retos y beneficios de lo que implica ser un académico marxista en este tiempo. A este ensayo le sigue una discusión sobre varios temas del análisis de clase, con particular énfasis en dos temas: las clases y la desigualdad, y la relación entre clase y poder. La segunda sección aborda el tema del socialismo como posible futuro del capitalismo. Su autor procura clarificar el estatus conceptual del socialismo y discute las razones por las que ciertas reformas, tales como los subsidios básicos universales, en últimas no podrían realizarse por completo sin la introducción de alguna forma de socialismo. Preguntas a la desigualdad concluye con un examen del problema general del marxismo, en tanto que tradición radical de la teoría social. Allí se discuten tres temas en particular: los principios fundamentales del marxismo analítico como estrategia para reconstruir el marxismo como teoría social científica; la relación entre el marxismo y el feminismo como teorías sociales emancipadoras y las perspectivas del marxismo tras el colapso de los regímenes comunistas.

1. Distance sampling is a widely used technique for estimating the size or density of biological populations. Many distance sampling designs and most analyses use the software Distance. 2. We briefly review distance sampling and its assumptions, outline the history, structure and capabilities of Distance, and provide hints on its use. 3. Good survey design is a crucial prerequisite for obtaining reliable results. Distance has a survey design engine, with a built-in geographic information system, that allows properties of different proposed designs to be examined via simulation, and survey plans to be generated. 4. A first step in analysis of distance sampling data is modeling the probability of detection. Distance contains three increasingly sophisticated analysis engines for this: conventional distance sampling, which models detection probability as a function of distance from the transect and assumes all objects at zero distance are detected; multiple-covariate distance sampling, which allows covariates in addition to distance; and mark–recapture distance sampling, which relaxes the assumption of certain detection at zero distance. 5. All three engines allow estimation of density or abundance, stratified if required, with associated measures of precision calculated either analytically or via the bootstrap. 6. Advanced analysis topics covered include the use of multipliers to allow analysis of indirect surveys (such as dung or nest surveys), the density surface modeling analysis engine for spatial and habitat-modeling, and information about accessing the analysis engines directly from other software. 7. Synthesis and applications. Distance sampling is a key method for producing abundance and density estimates in challenging field conditions. The theory underlying the methods continues to expand to cope with realistic estimation situations. In step with theoretical developments, state-of- the-art software that implements these methods is described that makes the methods accessible to practicing ecologists.
