Individual circadian clocks entrain differently to environmental cycles (zeitgebers, e.g., light and darkness), earlier or later within the day, leading to different chronotypes. In human populations, the distribution of chronotypes forms a bell-shaped curve, with the extreme early and late types _ larks and owls, respectively _ at its ends. Human chronotype, which can be assessed by the timing of an individual's sleep-wake cycle, is partly influenced by genetic factors - known from animal experimentation. Here, we review population genetic studies which have used a questionnaire probing individual daily timing preference for associations with polymorphisms in clock genes. We discuss their inherent limitations and suggest an alternative approach combining a short questionnaire (Munich ChronoType Questionnaire, MCTQ), which assesses chronotype in a quantitative manner, with a genome-wide analysis (GWA). The advantages of these methods in comparison to assessing time-of-day preferences and single nucleotide polymorphism genotyping are discussed. In the future, global studies of chronotype using the MCTQ and GWA may also contribute to understanding the influence of seasons, latitude (e.g., different photoperiods), and climate on allele frequencies and chronotype distribution in different populations.

The paper presents a compact planar Ultra Wide Band ¯lter employing folded stepped impedance resonators with series capacitors and dumb bell shaped defected ground structures. An interdigital quarter wavelength coupled line is used for achieving the band pass characteristics. The transmission zeros are produced by stepped impedance resonators. The ¯lter has steep roll o® rate and good attenuation in its lower and upper stop bands, contributed by the series capacitor and defected ground structures respectively.

There is growing interest in the potential beneficial effects of flavonoids in the aging and diseased brain. We have investigated the potential of the flavanone hesperetin and two of its metabolites, hesperetin-7-O-beta-D-glucuronide and 5-nitro-hesperetin, to inhibit oxidative stress-induced neuronal apoptosis. Exposure of cortical neurons to hydrogen peroxide led to the activation of apoptosis signal-regulating kinase 1 via its de-phosphorylation at Ser963, the phosphorylation of c-jun N-terminal kinase and c-Jun (Ser73) and the activation of caspase 3 and caspase 9. Whilst hesperetin glucuronide failed to exert protection, both hesperetin and 5-nitro-hesperetin were effective at preventing neuronal apoptosis via a mechanism involving the activation/phosphorylation of both Akt/protein kinase B and extracellular signal-regulated kinase 1 and 2 (ERK1/2). Protection against oxidative injury and the activation of Akt and ERK1/2 followed a bell-shaped response and was most apparent at 100 nmol/L concentrations. The activation of ERK1/2 and Akt by flavanones led to the inhibition of the pro-apoptotic proteins, apoptosis signal-regulating kinase 1, by phosphorylation at Ser83 and Bad, by phosphorylation at both Ser136 and Ser112 and to the inhibition of peroxide-induced caspase 9 and caspase 3 activation. Thus, flavanones may protect neurons against oxidative insults via the modulation of neuronal apoptotic machinery.

Emerging evidence suggests that the cellular actions of flavonoids relate not simply to their antioxidant potential but also to the modulation of protein kinase signalling pathways. We investigated in primary cortical neurons, the ability of the flavan-3-ol, (-)epicatechin, and its human metabolites at physiologically relevant concentrations, to stimulate phosphorylation of the transcription factor cAMP-response element binding protein (CREB), a regulator of neuronal viability and synaptic plasticity. (-)Epicatechin at 100-300 nmol/L stimulated a rapid, extracellular signal-regulated kinase (ERK)- and PI3K-dependent, increase in CREB phosphorylation. At micromolar concentrations, stimulation was no longer apparent and at the highest concentration tested (30 mu mol/L) (-)epicatechin was inhibitory. (-)Epicatechin also stimulated ERK and Akt phosphorylation with similar bell-shaped concentration-response characteristics. The human metabolite 3 '-O-methyl-(-)epicatechin was as effective as (-)epicatechin at stimulating ERK phosphorylation, but (-)epicatechin glucuronide was inactive. (-)Epicatechin and 3 '-O-methyl-(-)epicatechin treatments (100 nmol/L) increased CRE-luciferase activity in cortical neurons in a partially ERK-dependent manner, suggesting the potential to increase CREB-mediated gene expression. mRNA levels of the glutamate receptor subunit GluR2 increased by 60%, measured 18 h after a 15 min exposure to (-)epicatechin and this translated into an increase in GluR2 protein. Thus, (-)epicatechin has the potential to increase CREB-regulated gene expression and increase GluR2 levels and thus modulate neurotransmission, plasticity and synaptogenesis.

Diacetyl, like other alpha-dicarbonyl compounds, is reportedly cytotoxic and genotoxic. A food and cigarette contaminant, it is related with alcohol hepatotoxicity and lung disease. Peroxynitrite is a potent oxidant formed in vivo by the diffusion-controlled reaction of the superoxide radical anion with nitric oxide, which is able to form adducts with carbon dioxide and carbonyl compounds. Here, we investigate the nucleophilic addition of peroxynitrite to diacetyl forming acetyl radicals, whose reaction with molecular oxygen leads to acetate. Peroxynitrite is shown to react with diacetyl in phosphate buffer (bell-shaped pH profile with maximum at 7.2) at a very high rate constant (k(2) = 1.0 X 10(4) M-1 s(-1)) when compared with monocarbonyl substrates (k(2) < 10(3) M-1 s(-1)). Phosphate ions (100-500 MM) do not affect the rate of spontaneous peroxynitrite decay, but the H2PO4- anion catalyzes the nucleophilic addition of the peroxynitrite anion to diacetyl. The intermediacy of acetyl radicals is suggested by a three-line spectrum (a(N) = a(H) = 0.83 mT) obtained by EPR spin trapping of the reaction mixture with 2-methyl-2-nitrosopropane. The peroxynitrite reaction is accompanied by concentration-dependent oxygen uptake. Stoichiometric amounts of acetate from millimolar amounts of peroxynitrite and diacetyl were obtained under nonlimiting conditions of dissolved oxygen. In the presence of either L-histidine or 2`-deoxyguanosine, the peroxynitrite/diacetyl system afforded the corresponding acetylated molecules identified by HPLC-MS"". These studies provide evidence that the peroxynitrite/diacetyl reaction yields acetyl radicals and raise the hypothesis that protein and DNA nonenzymatic acetylation may occur in cells and be implicated in aging and metabolic disorders in which oxygen and nitrogen reactive species are putatively involved.

Foram analisadas 72 amostras da água de dessedentação de frangos de corte, 18 da entrada e 54 dos bebedouros pendulares. As amostras foram submetidas às mensurações de coliformes totais, coliformes fecais, estreptococos fecais, Escherichia coli e organismos mesófilos, além da detecção da presença de Salmonella sp e da taxa de demanda de cloro. Os resultados evidenciaram uma elevada contaminação bacteriológica, já na primeira semana, com os indicadores na ordem de 10³ a 10(5). Não houve isolamento de nenhuma cepa de Salmonella sp, provavelmente por não haver ave infectada no plantel. A taxa de demanda de cloro mostrou-se elevada, devido ao maior acúmulo de matéria orgânica nos bebedouros, com o ápice na quinta semana (98,7%), seguida de uma leve diminuição na sexta semana (98,5%). As análises dos resultados evidenciam o bebedouro pendular como um instrumento depreciativo e ineficiente em relação às qualidades higiênico-sanitárias das águas fornecidas aos frangos de corte, contribuindo para um alto risco de contaminação por patógenos de veiculação hídrica. Preconizam-se a utilização de modelos de bebedouros menos deletérios e o monitoramento semanal de cloro residual nos bebedouros pendulares, a fim de se obter uma desinfecção satisfatória da água.

The embryology and seed structure of Blastocaulon scirpeum (Mart.) Giul. and Paepalanthus scleranthus Ruhland were studied in order to contribute to the embryology of Eriocaulaceae and supply data for future taxonomic studies. Both species present: anther with 4-layered wall; conspicuous endothecium with fibrous thickenings; secretory tapetum with uninucleate cells; successive microsporogenesis forming isobilateral microspore tetrads; bicellular pollen grains; orthotropous, bitegmic and tenuinucellate ovule; micropyle formed by the inner integument alone; megagametophyte of the Polygonum type, with a conspicuous antipodal cyst; nuclear and starchy endosperm; reduced, undifferentiated, and bell-shaped embryo; operculate and endotestal seed; seed coat derived from the two ovule integuments; and tanniniferous endotegmen. In addition, Blastocaulon scirpeum shows a bisporangiate anther and a 3-layered ovary wall, while P. scleranthus presents a tetrasporangiate anther that becomes bisporangiate at maturity, and a 2-layered ovary wall. This investigation shows that the bisporangiate condition does not suffice to separate Blastocaulon from Paepalanthus, since it is common to both. It also indicates, based on several embryological aspects, the proximity of Eriocaulaceae and Xyridaceae, which comply mainly with the features presented by the other commelinid families. These results may be used in future cladistic analysis of the family, and contribute to a better understanding of its phylogeny.

The embryology and the seed development of Syngonanthus caulescens are presented. This species possesses: a bithecous and tetrasporangiate anther, with a four-layered wall, a conspicuous endothecium of the baseplate type, a secretory tapetum formed by uninucleate cells, successive microsporogenesis resulting in isobilateral microspore tetrads, spiraperturate and binucleate pollen grains, an orthotropous, pendulous, bitegmic and terminucellate ovule, with a micropyle formed only by the inner integument, a megagametophyte of the Polygonum type, with formation of an antipodal cyst, free-nuclear and starchy endosperm, a broad and bell-shaped embryo, operculate and endotestal seeds, a seed coat derived from the inner layers of both integuments, and tanniniferous endotegmen. These embryological aspects are characteristic not only for Syngonanthus, but for the whole family, with few differences between genera. Furthermore, the pollen grain of the spiraperturate type and the cystic arrangement of the antipodals in the megagametophyte are peculiar and very distinctive features of Eriocaulaceae within the other Poales (commelinids). The population dynamics and reproduction of the hermit crab Clibanarius vittatus were evaluated on Pescadores Beach, located on the estuarine channel of Sao Vicente (São Paulo), Brazil. The hermit crabs were captured by hand during low tide,from May 2001 to April 2003. A total of 2554 hermit crabs were captured, of which 701 were males, 1741 non-ovigerous females, 48 ovigerous females and 64 intersex individuals. The size-frequency distribution of the males was represented by a platykurtic bell-shaped curve, which differed from the leptokurtic bell-shaped curve of the females. The smaller and intermediate classes were composed mainly of females (modal size 6.5-7.5 mm carapace shield length (CSL)), and the larger classes only by males (modal size 9.5-10.5 mm CSL). The overall sex-ratio was skewed toward females (0.39:1/M:F), differing significantly from the expected 1:1. A seasonal reproductive pattern was recorded for C. vittatus in this location, with more intensive reproductive activity in the warmer months. The absence of juveniles suggests that their recruitment area is different than the area inhabited by adults, possibly another area with more protection and specialized or different resources for young.

O mercúrio pode ser encontrado em diversas formas, sendo a orgânica como metilmercúrio (MeHg), considerada a mais tóxica. Facilmente absorvido por via oral, se acumula na cadeia trófica e se amplifica em carnívoros aquáticos, principalmente em peixes, daí o risco maior para as populações que deles se alimentam preferencialmente, como os ribeirinhos Amazônidas. O efeito neurotóxico dessa forma de mercúrio tem sido amplamente demonstrado através de estudos epidemiológicos e experimentais. Alguns desses estudos também mostraram que hormônios e substâncias antioxidantes podem agir protegendo o organismo contra a ação deletéria do mercúrio. A prolactina é um destes hormônios que apresenta ação protetora, mas age também como citocina pró-inflamatória. Desde que o MeHg pode também agir como uma substância imunotóxica, procuramos neste trabalho estudar a ação citoprotetora da PRL em cultivos contínuos de linhagem B95-A de linfócitos de primata afim de avaliar sua fragilidade ao MeHg e sua reatividade a ação da PRL. Com o objetivo de avaliar a integridade funcional dos linfócitos expostos ao MeHg utilizou-se teste de reação colorimétrica para 3-(4,5-dimetiltiazol-2-yl)-2,5-difenil tetrazólio bromide (MTT), o qual detecta atividade metabólica mitocondrial. Para avaliar a resposta imune do linfócito, medidas da concentração do fator de necrose tumoral alfa (TNF α) no sobrenadante do cultivo, foram realizadas por ELISA. É uma citocina pró-inflamatória liberada em resposta a agressão celular de diferentes causas, incluindo estresse oxidativo, um dos efeitos agudos mais evidentes do MeHg, além disso, esta citocina também poder responder a regulação prolactinérgica em linfócitos humanos. Após 18 horas de exposição do cultivo a crescentes concentrações do metal (0,1; 1, 5, 10 e 50 μM) verificou-se significativa diminuição do tipo dose-dependente da viabilidade celular a partir de 1 μM (35%) e progressivamente até 50 μM (80%), quando poucas células íntegras foram encontradas nos cultivos. Um efeito bifásico em forma de “sino” ocorreu na liberação de TNF α, onde concentrações mais baixas de MeHg inibiram (0,1 e 1 μM), a intermediária estimulou (5 μM) e as duas maiores (10 e 50 μM) voltaram a inibir. A prolactina também diminuiu a viabilidade celular, em cerca de 30%, somente na dose mais elevada (10 nM). Por outro lado, na dose de 1 nM a PRL preveniu a diminuição de 40% da viabilidade celular resultante a exposição ao MeHg a 5 μM. Esta dose de 1 nM de PRL foi a única a estimular a liberação de TNF α, mas curiosamente, reverteu a liberação desta citocina quando associada a 5 μM de MeHg, concentração que igualmente estimulou a secreção de TNF α. Os resultados confirmaram a toxidade do MeHg para linfócitos de primatas (linhagem B95-A) e sua reversão por uma possível ação protetora da PRL. Um efeito bifásico na secreção de TNF α resultou da exposição ao MeHg, sugerindo a presença de diferentes mecanismos citotóxicos resultantes a ação mercurial. Por outro lado, a PRL foi pouco efetiva em estimular a secreção daquela citocina, invertendo esta resposta quando associada ao MeHg. No entanto, estes resultados são preliminares e carecem de um estudo mais acurado para sua completa elucidação.

O primeiro estágio de processamento da informação do estímulo visual consiste na contagem de fótons pelas células fotorreceptoras. Nos estágios pós-receptorais a informação de intensidade absoluta do estímulo é transformada em comparações de informações provindas de áreas adjacentes da retina e momentos sucessivos. Essa métrica implementada pelo sistema visual para quantificar o estímulo é chamada de contraste - contraste espacial ou simultâneo e contraste temporal ou sucessivo. A presença de contraste é essencial para a geração de percepção visual consciente no domínio do espaço e do tempo e em três dimensões ortogonais de cores - branca e preta; azul e amarela; verde e vermelha. Uma curva em forma de sinodelimita os limiares de detecção de contraste em função da frequência especial ou temporal do estímulo. Ela é chamada função de sensibilidade ao contraste e é afetada por uma série de fatores ópticos e neurais. Neurônios de diferentes classes contribuem para regiões diferentes da função de sensibilidade ao contraste e suas atividades representam as ações de vias de processamento visual que se estendem da retina ao córtex visual. Investigações básicas e clínicas têm dado suporte à importância do estudo da sensibilidade ao contraste espacial de luminância (branco e preta) como uma ferramenta indicadora da função visual em sujeitos normais e afetados por disfunções neuro-oftalmológicas.

Laccases (benzendiol:oxygen oxidoreductases; EC 1.10.3.2) catalyze the oxidation of a broad range of substrates, such as polyphenols, dyes and pollutants, and thus these enzymes are widely applied in industrial, biotechnological and environmental fields. In order to improve their biotechnological applications, a deep knowledge of structural factors involved in controlling their activity, in various experimental conditions and on different substrates, is required. In the present study, a laccase from the mushroom Rigidoporus lignosus was kinetically characterized. In particular, the stability, the effects of pH, ionic strength and fluoride ion concentration on the kinetic parameters were investigated, using three di-hydroxy-benzene isomers (1,2-dihydroxy-benzene, 1,3-dihydroxy-benzene and 1,4-dihydroxy-benzene) as substrates. The catalytic constant values of the laccase showed a bell-shaped pH profile, with the same optimum pH and pK(a) values for all tested substrates. This behavior appears to be due to the presence of an ionizable residue in the enzyme active site. To identify this residue, the enzyme was derivatized with diethylpyrocarbonate to modify accessible histidine residues, which, according to structural data, are present in the active site of this enzyme. The kinetic behavior of the derivatized laccase was compared with that of the native enzyme and the derivatized residues were identified by mass spectrometry. Mass spectrometry and kinetic results suggest the main role of His-457 in the control of the catalytic activity of laccase from R. lignosus. (C) 2013 Elsevier B.V. All rights reserved.

Uridine adenosine tetraphosphate (Up(4)A) has been recently identified as a novel and potent endothelium-derived contracting factor and contains both purine and pyrimidine moieties, which activate purinergic P2X and P2Y receptors. The present study was designed to compare contractile responses to Up(4)A and other nucleotides such as ATP (P2X/P2Y agonist), UTP (P2Y(2)/P2Y(4) agonist), UDP (P2Y(6) agonist), and alpha,beta-methylene ATP (P2X(1) agonist) in different vascular regions [thoracic aorta, basilar, small mesenteric, and femoral arteries] from deoxycorticosterone acetate-salt (DOCA-salt) and control rats. In DOCA-salt rats [vs. control uninephrectomized (Uni) rats]: (1) in thoracic aorta, Up(4)A-, ATP-, and UP-induced contractions were unchanged; (2) in basilar artery, Up(4)A-, ATP-, UTP- and UDP-induced contractions were increased, and expression for P2X(1), but not P2Y(2) or P2Y(6) was decreased; (3) in small mesenteric artery, Up(4)A-induced contraction was decreased and UDP-induced contraction was increased; expression of P2Y(2) and P2X(1) was decreased whereas P2Y(6) expression was increased; (4) in femoral artery, Up(4)A-. UTP-, and UDP-induced contractions were increased, but expression of P2Y(2), P2Y(6) and P2X(1) was unchanged. The alpha,beta-methylene ATP-induced contraction was bell-shaped and the maximal contraction was reached at a lower concentration in basilar and mesenteric arteries from Uni rats, compared to arteries from DOCA-salt rats. These results suggest that Up(4)A-induced contraction is heterogenously affected among various vascular beds in arterial hypertension. P2Y receptor activation may contribute to enhancement of Up(4)A-induced contraction in basilar and femoral arteries. These changes in vascular reactivity to Up(4)A may be adaptive to the vascular alterations produced by hypertension. (C) 2011 Elsevier Ltd. All rights reserved.

The rhea (Rhea americana americana) is an american bird belonging to Ratite's family. Studies related to its morphology are still scarce. This study aims to describe the macroscopic structures of the oropharyngeal cavity. Five heads (2 to 6 months old) formalin preserved were anatomically dissected to expose the oropharynx. The oropharynx of the rhea was "bell-shaped" composed by the maxillary and mandibular rhamphotheca. The roof and floor presented two distinct regions different in colour of the mucosa. The rostral region was pale pink contrasting to grey coloured caudal region. The median longitudinal ridge extended rostrally from the apex of the choana to the tip of the beak in the roof and it is clearly more prominent and rigid than the homolog in the floor that appeared thin and stretched merely along the rostral portion of the regio interramalis. The floor was formed by the interramal region, (regio interramalis) tongue and laryngeal mound containing glove-shaped glottis. This study confirmed the basic morphology of the oropharinx of the rhea. However, important morphological information not previously described is highlighted and contradictory information present in the literature is clarified.

Benzyltris(trimethylsilyl)silan konnte durch Reaktion mit Lithiummethanid oder Alkalimetall-tert-butanolaten (M-OtBu, M = Na, K, Rb, Cs) in die entsprechenden Alkalimetall-benzylbis(trimethylsilyl)silanide (M-Bnz, Bnz = Si(SiMe3)2CH2Ph) überführt werden. Die Strukturen dieser Verbindungen weisen viele Gemeinsamkeiten mit den entsprechenden Hypersilylverbindungen (Hypersilyl bzw. Hyp = Si(SiMe3)3) auf. Mit Ausnahme der Lithiumverbindung weisen die Natrium-, Kalium- und Rubidiumverbindungen jedoch noch intramolekulare Wechselwirkungen des Benzylrestes mit den Alkalimetallkationen auf. Die Benzylbis(trimethylsilyl)silanide der Alkalimetalle eignen sich zur Darstellung von Kupfersilylverbindungen. So konnten heteroleptische Cuprate (Bnz-Cu-OtBu-, Bnz-Cu-Hyp-) und monoleptische Cuprate (CuBnz2-), aber auch das ungeladene Kupfer-benzyl-bis(trimethylsilyl)silanid hergestellt werden. Auf analoge Weise wurde versucht, Alkalimetall- und Kupferverbindungen des Phenylbis(trimethylsilyl)silanides herzustellen. Da aber bereits bei der Synthese der Alkalimetallverbindungen die entsprechenden Hypersilanide als schwer abtrennbare Nebenprodukte entstanden, konnten keine ausführlichen Untersuchungen angestellt werden. Es konnten die Natrium- und Kaliumsalze des Dihypersilylcuprates hergestellt und strukturell charakterisiert werden. Die Natriumverbindung wurde nur als Nebenprodukt einer Reaktion von Kupfer(I)-cyanid mit Natriumhypersilanid erhalten, die entsprechende Kaliumverbindung ließ sich jedoch mit guten Ausbeuten gezielt aus Kupfer(I)-tert-butanolat herstellen. Beide Verbindungen weisen ein hantelförmiges Hyp-Cu-Hyp-Anion auf, die THF- bzw. THP-koordinierten Kationen liegen isoliert vom Anion vor. Eine neue Klasse von Kupfersilylverbindungen konnten durch Reaktion von Kupfer(I)-cyanid mit Alkalimetallhypersilaniden erzeugt werden. Dabei entstehen heteroleptische Cyano-hypersilyl-cuprate. Großen Einfluss auf die Strukturen üben die Alkalimetallkationen und Lösungsmittelmoleküle aus. So findet man bei den Lithiumverbindungen zweikernige Kupferkomplexe, die je nach Lösungsmittelgehalt unterschiedliche Anordnungen der Cyanid- und Hypersilanidanionen aufweisen. Lediglich die Kaliumverbindung weist lineare Cyanid-Kupfer-Hypersilanid-Einheiten auf, wie man sie analog von Organyl-cyano-cupraten kennt. Trotz vieler Gemeinsamkeiten vieler Silyl- und entsprechender Stannylverbindungen bezüglich Struktur und Reaktivität, erwies sich die Darstellung von Kupferstannyl-verbindungen schwieriger als erwartet. Zwar konnte in einer mehrstufigen Reaktion Kupfer-tris(trimethylsilyl)stannanid erhalten werden, auftretende Zwischen- und Nebenprodukte weisen jedoch auf das Auftreten von Umlagerungen und Redoxprozesse hin, die bei entsprechenden Kupfersilylverbindungen nicht beobachtet wurden.
