Aims/hypothesis. This study was designed to determine whether inhibition of formation of AGE and advanced lipoxidation end-products (ALE) is a mechanism of action common to a diverse group of therapeutic agents that limit the progress of diabetic nephropathy. We compared the effects of the ACE inhibitor enalapril, the antioxidant vitamin E, the thiol compound lipoic acid, and the AGE/ALE inhibitor pyridoxamine on the formation of AGE/ALE and protection against nephropathy in streptozotocin diabetic rats.

S-(2-Succinyl)cysteine (2SC) has been identified as a chemical modification in plasma proteins, in the non-mercaptalbumin fraction of human plasma albumin, in human skin collagen, and in rat skeletal muscle proteins and urine. 2SC increases in human skin collagen with age and is increased in muscle protein of diabetic vs. control rats. The concentration of 2SC in skin collagen and muscle protein correlated strongly with that of the advanced glycation/lipoxidation end-product (AGE/ALE), N(epsilon)-(carboxymethyl)lysine (CML). 2SC is formed by a Michael addition reaction of cysteine sulfhydryl groups with fumarate at physiological pH. Fumarate, but not succinate, inactivates the sulfhydryl enzyme, glyceraldehyde-3-phosphate dehydrogenase in vitro, in concert with formation of 2SC. 2SC is the first example of spontaneous chemical modification of protein by a metabolic intermediate in the Krebs cycle. These observations identify fumarate as an endogenous electrophile and suggest a role for fumarate in regulation of metabolism.

Carboxyl-functionalized multiwalled carbon nanotubes (MWCNTs) have been successfully radiolabelled with cobalt-57 (57Co) (T1/2 = 270 days) via the attachment of the bifunctional caged ligand MeAMN3S3sar. In this study MeAMN3S3sar has been synthesized and coupled to MWCNTs to form the conjugate MWCNT–MeAMN3S3sar. Synthesis was confirmed with nuclear magnetic resonance. X-ray photoelectron spectroscopy (XPS) confirmed the conjugation. Non-radioactive labelling of this conjugate was completed with Cu(II) ions to confirm the stability of the MeAMN3S3sar after coupling with the MWCNTs. The complexation of the Cu(II) was also confirmed with XPS. Transmission electron microscopy was used to demonstrate that the coupling reaction had a negligible effect on the size and shape of the MWCNTs. Radiolabelling of the MWCNT–MeAMN3S3sar conjugate and pristine (untreated) MWCNTs (non-specific) with the gamma-emitting radioactive isotope 57Co were compared. The radiolabelling efficiency of the MWCNT–MeAMN3S3sar conjugate was significantly higher (95% vs. 0.1%) (P ⩽ 0.001) than for the unconjugated pristine MWCNTs. This will allow for the potential tracking of nanoparticle movement in vitro and in vivo.

The effects of modifying blends of poly(vinyl chloride) (PVC) with linear low density polyethylene (LLDPE) by means of acrylic acid, maleic anhydride, phenolic resins and p-phenylene diamine were investigated. Modification by acrylic acid and maleic anhydride in the presence of dicumyl peroxide was found to be the most useful procedure for improving the mechanical behaviour and adhesion properties of the blend. The improvement was found to be due mainly to the grafting of the carboxylic acid to the polymer chains; grafting was found to be more effective in LLDPE/PVC blends than in pure LLDPE.

A novel biosensor for glucose was prepared by adsorption of 1,1`-bis(4-carboxybenzyl)-4,4`-bipyridinium di-bromide compound (H(2)BpybcBr(2)) onto the surface of a nanocrystalline TiO(2) film deposited onto FTO glasses, which was used as a platform to assemble the enzyme glucose oxidase to the electrode surface. The H(2)BpybcBr(2)/TiO(2)/FTO modified electrode was characterized by scanning electron microscopy, X-ray fluorescence image, cyclic voltammograms and spectroelectrochemical measurements. The immobilization of GOD on functionalized TiO(2) film led to stable amperometric biosensing for glucose with a linear range from 153 mu mol L(-1) to 1.30 mmol L(-1) and a detection limit of 51 mu mol L(-1). The apparent Michaelis-Menten constant (K(m)) was estimated to be 3.76 mmol L(-1), which suggested a high enzyme-substrate affinity. The maximum electrode sensitivity was 1.25 mu A mmol L(-1). The study proved that the combination of viologen mediators with TiO(2) film retains the electrocatalytic activity of the enzyme, and also enhances the electron transfer process, and hence regenerating the enzyme in the reaction with glucose. The criteria for accepting or rejecting a technology extend beyond the intrinsic properties of the finished product such as physical performance. There are also extrinsic factors such as the history of the product and trust in the manufacturers and suppliers, as well as the perceptions and risk management strategies of various stakeholder groups. A methodology was trialled to take into account the extended supply chain of the product, while simultaneously engaging stakeholders to determine and to understand their perceptual frameworks. Three pine decking products manufactured using different amounts and types of chemical modification were compared using life cycle assessment and the comments of 114 respondents from six stakeholder groups in New Zealand. The results of the perceptual research include a quadrant diagram which allows a visual comparison of the responses of different stakeholders to actual or hypothetical products, aiding the identification of when and why certain technologies may be disqualified from acceptability or become the topic of public debate.

To help avoid conflict and even market rejection, the design and introduction of new technologies can be aided by consideration of the potential concerns of different stakeholder groups. An assessment of their desired goals for technological change may be useful for pre-empting changes in the  parameters of acceptability for technologies. As part of a research  programme evaluating the parameters of acceptability for bio-based  technologies using life cycle assessment of products and an analysis of the  perceptual frameworks of stakeholders, respondents from four stakeholder  groups in New Zealand were interviewed about the desirable and  undesirable trajectories for chemical modification technologies. Three  examples of pine decking products derived using different amounts and  types of chemical modification served to help explore the contemporary  criteria and rationale for acceptability. The responses of the 70 respondents  in those four groups indicated that new technologies need to be able to  prove their worth with regard to both tangible and intangible qualities if they  are to be accepted into the market in the place of either the existing product  or an inorganic competitor.

Anatase nanoparticles were obtained through a modified sol-gel route from titanium isopropoxide modified with acetic acid in order to control hydrolysis and condensation reactions. The modification of Ti(O(i)Pr)(4) with acetic acid reduces the availability of groups that hydrolyze and condense easily through the formation of a stable complex whose structure was determined to be Ti(OCOCH(3))(O(i)Pr)(2) by means of FTIR and (13)C NMR. The presence of this complex was confirmed with FTIR in the early stages of the process. A doublet in 1542 and 1440 cm(-1) stands for the asymmetric and symmetric stretching vibrations of the carboxylic group coordinated to Ti as a bidentate ligand. The gap of 102 cm(-1) between these signals suggests that acetate acts preferentially as a bidentate rather than as a bridging ligand between two titanium atoms. The use of acetic acid as modifier allows the control of both the degree of condensation and oligomerization of the precursor and leads to the preferential crystallization of TiO(2) in the anatase phase. A possible reaction pathway toward the formation of anatase is proposed on the basis of the intermediate species present in a 1:1 Ti(O(i)Pr)(4):CH(3)COOH molar system in which esterification reactions that introduce H(2)O into the reaction mixture were seen to be negligible. The Rietveld refinement and TEM analysis revealed that the powder is composed of isotropic anatase nanocrystallites.
