Background The malignant B cells in chronic lymphocytic leukemia receive signals from the bone marrow and lymph node microenvironments which regulate their survival and proliferation. Characterization of these signals and the pathways that propagate them to the interior of the cell is important for the identification of novel potential targets for therapeutic intervention. Design and Methods We compared the gene expression profiles of chronic lymphocytic leukemia B cells purified from bone marrow and peripheral blood to identify genes that are induced by the bone marrow microenvironment. Two of the differentially expressed genes were further studied in cell culture experiments and in an animal model to determine whether they could represent appropriate therapeutic targets in chronic lymphocytic leukemia. Results Functional classification analysis revealed that the majority of differentially expressed genes belong to gene ontology categories related to cell cycle and mitosis. Significantly up-regulated genes in bone marrow-derived tumor cells included important cell cycle regulators, such as Aurora A and B, survivin and CDK6. Down-regulation of Aurora A and B by RNA interference inhibited proliferation of chronic lymphocytic leukemia-derived cell lines and induced low levels of apoptosis. A similar effect was observed with the Aurora kinase inhibitor VX-680 in primary chronic lymphocytic leukemia cells that were induced to proliferate by CpG-oligonucleotides and interleukin-2. Moreover, VX-680 significantly blocked leukemia growth in a mouse model of chronic lymphocytic leukemia. Conclusions Aurora A and B are up-regulated in proliferating chronic lymphocytic leukemia cells and represent potential therapeutic targets in this disease.

A presente pesquisa teve por objetivo estudar o desenvolvimento inicial no campo de pessegueiros 'Aurora-1' enxertados nos Clones 05; 10 e 15 de umezeiro (Prunus mume Sieb. et Zucc.) e 'Okinawa' [Prunus persica (L.) Batsch] propagados por estacas herbáceas. O experimento foi conduzido em propriedade rural no município de Taiaçu-SP, em Argissolo Vermelho-Amarelo sob espaçamento de 1,5m x 6,0m, com uso de irrigação por microaspersores subcopa. Foi adotado o delineamento em blocos ao acaso, com quatro tratamentos (porta-enxertos) e cinco repetições, sendo que cada parcela foi constituída de três plantas. Após 23 meses da implantação, foi possível concluir que: a) os porta-enxertos Clones 05; 10 e 15 de umezeiro e 'Okinawa' não influenciaram de maneira diferenciada no vigor de pessegueiros 'Aurora-1'; b) o Clone 05 de umezeiro antecipou o pleno florescimento do 'Aurora-1', em relação às plantas enxertadas sobre 'Okinawa'; c) os porta-enxertos estudados não influenciaram no número de frutos por planta e na brotação do 'Aurora-1'; d) não foram observados sintomas de incompatibilidade entre os Clones 05; 10 e 15 de umezeiro e 'Okinawa' propagados por estacas herbáceas com a cv. Aurora-1 de pessegueiro.

O presente trabalho teve por objetivo estudar o diâmetro do tronco, a fenologia e a produção de gemas em pessegueiros 'Aurora-1', enxertados no porta-enxerto 'Okinawa' propagado por sementes e por estacas herbáceas, em três espaçamentos (6 x 2 m, 6 x 3 m e 6 x 4 m). No 2º e 3º anos após o plantio das mudas (2005 e 2006, respectivamente), foram estudadas 13 variáveis na cultivar-copa 'Aurora-1', além de sete avaliações trimestrais de diâmetro do tronco, mensuradas a 5 cm acima e abaixo do ponto de enxertia. Nas condições experimentais adotadas, conclui-se que: a) o diâmetro do tronco de pessegueiros 'Aurora-1' não é influenciado pelo método de propagação do porta-enxerto 'Okinawa' nem pelos diferentes espaçamentos entre plantas; b) não há diferença de diâmetro do tronco entre as medições feitas acima e abaixo do ponto de enxertia e não foram constatados sintomas visíveis de incompatibilidade com a cultivar-copa 'Aurora-1', em ambas as formas de propagação do porta-enxerto; c) os métodos de propagação do porta-enxerto 'Okinawa' estudados não exercem nenhum efeito diferenciado na fenologia, no comprimento de ramos mistos, na produção de gemas floríferas e vegetativas e em sua relação, avaliadas na cv. Aurora-1; d) os diferentes espaçamentos estudados não influenciaram na fenologia, no comprimento de ramos mistos e no número de gemas floríferas por ramo da cv. Aurora-1.

While the Cluster spacecraft were located near the high-latitude magnetopause, between 10:10 and 10:40 UT on 16 January 2004, three typical flux transfer event (FTE) signatures were observed. During this interval, simultaneous and conjugated all-sky camera measurements, recorded at Yellow River Station, Svalbard, are available at 630.0 and 557.7nm that show poleward-moving auroral forms (PMAFs), consistent with magnetic reconnection at dayside magnetopause. Simultaneous FTEs seen at the magnetopause mainly move northward, but having duskward (eastward) and tailward velocity components, roughly consistent with the observed direction of motion of the PMAFs in all-sky images. Between the PMAFs meridional keograms, extracted from the all-sky images, show intervals of lower intensity aurora which migrate equatorward just before the PMAFs intensify. This is strong evidence for an equatorward eroding and poleward moving open-closed boundary (OCB) associated with a variable magnetopause reconnection rate under variable IMF conditions. From the durations of the PMAFs we infer that the evolution time of FTEs is 5-11 minutes from its origin on magnetopause to its addition to the polar cap.

We apply a numerical model of time-dependent ionospheric convection to two directly driven reconnection pulses during a 15-min interval of southward IMF on 26 November 2000. The model requires an input magnetopause reconnection rate variation, which is here derived from the observed variation in the upstream IMF clock angle, q. The reconnection rate is mapped to an ionospheric merging gap, the MLT extent of which is inferred from the Doppler-shifted Lyman-a emission on newly opened field lines, as observed by the FUV instrument on the IMAGE spacecraft. The model is used to reproduce a variety of features observed during this event: SuperDARN observations of the ionospheric convection pattern and transpolar voltage; FUV observations of the growth of patches of newly opened flux; FUVand in situ observations of the location of the Open-Closed field line Boundary (OCB) and a cusp ion step. We adopt a clock angle dependence of the magnetopause reconnection electric field, mapped to the ionosphere, of the form Enosin4(q/2) and estimate the peak value, Eno, by matching observed and modeled variations of both the latitude, LOCB, of the dayside OCB (as inferred from the equatorward edge of cusp proton emissions seen by FUV) and the transpolar voltage FPC (as derived using the mapped potential technique from SuperDARN HF radar data). This analysis also yields the time constant tOCB with which the open-closed boundary relaxes back toward its equilibrium configuration. For the case studied here, we find tOCB = 9.7 ± 1.3 min, consistent with previous inferences from the observed response of ionospheric flow to southward turnings of the IMF. The analysis confirms quantitatively the concepts of ionospheric flow excitation on which the model is based and explains some otherwise anomalous features of the cusp precipitation morphology.

We employ a numerical model of cusp ion precipitation and proton aurora emission to fit variations of the peak Doppler-shifted Lyman-a intensity observed on 26 November 2000 by the SI-12 channel of the FUV instrument on the IMAGE satellite. The major features of this event appeared in response to two brief swings of the interplanetary magnetic field (IMF) toward a southward orientation. We reproduce the observed spatial distributions of this emission on newly opened field lines by combining the proton emission model with a model of the response of ionospheric convection. The simulations are based on the observed variations of the solar wind proton temperature and concentration and the interplanetary magnetic field clock angle. They also allow for the efficiency, sampling rate, integration time and spatial resolution of the FUV instrument. The good match (correlation coefficient 0.91, significant at the 98% level) between observed and modeled variations confirms the time constant (about 4 min) for the rise and decay of the proton emissions predicted by the model for southward IMF conditions. The implications for the detection of pulsed magnetopause reconnection using proton aurora are discussed for a range of interplanetary conditions.

The main objective of this research was to evaluate chemical and physical changes in 'Aurora-1' peach harvested at two maturity stages, packed in different types of packaging and kept under refrigeration. Fruit were harvested at the mature green and ripe stages, packed in four different types of packaging (control, PD-900 (TM), PVC and PET) and stored at 6 degrees C. The following variables were evaluated every eight days: coloration, accumulated fresh mass loss, firmness, appearance, acidity, total soluble solids contents, soluble sugars, and percentage of pectin solubilization. We observed that the postharvest life was influenced by packaging and the mature green fruits showed lower disease occurrence. Fresh mass loss was lower in packed fruits. The peel of mature green fruits developed a characteristic ripe peach color at the end of storage, but PD-900 (TM) provided a delay in color change. Packaging also influenced the firmness, allowing for more firmness retention than for the control fruits at both harvest stages. The organic acid content decreased in the packaged fruits and increased in the control fruits. In the packaged fruit, the amount of sugar increased until the eighth day and then decreased until the end of the storage period. The 'Aurora-1' peaches did not show compromised quality by packaging use and exhibited an increase in harvest life to 24 days (compared to 16 days for the control).

The production of minimally processed fruit has increased significantly in the last few years due to the high nutritional value, convenience and safety of minimally processed fruit. The objective of the present study was to evaluate the effects of the conservation temperature and processing conditions on the quality and shelf-life of minimally processed peaches. The processing procedure consisted of washing, sanitization, peeling and stone removal. To remove the stone, the fruit was cut longitudinally, and three types of products were obtained, including halves, quarters and one-eighth segments. The fruit pieces were immersed in 2% ascorbic acid for 3 min and were packed in rigid polystyrene trays (Meiwa (c) M-54) coated with 14 mu m PVC film (Omnifilm (TM)) (about 200 g per pack). The pieces were stored at 65% RH at 3, 6 or 9 degrees C for 12 days, and were evaluated every four days. The appearance, fresh mass loss, color, O-2 and CO2 concentration, acidity, total soluble solids, total and soluble pectin content and ascorbic acid concentration were measured. Minimally processed peaches stored at 3 degrees C maintained higher quality due to control of ripening and senescence Moreover, an interaction effect between one-eighth segments and a temperature of 3 degrees C was observed, and fruit with a superior appearance and higher soluble solids content was obtained.
