El río Grande de la Magdalena marcó la historia del Caribe colombiano. Las grandes ciudades de la región nacieron y crecieron a sus orillas y vivieron a merced de sus caprichos. En un país enmarcado por cordilleras intransitables, la única vía de penetración y el camino más seguro para cualquier proceso de intercambio político o comercial era el cauce del río. Salir hacia las costas implicaba ciertas dificultades, pero remontar para llevar el comercio, la información o para el tránsito humano resultaba un proceso mucho más largo, costoso y complicado. Era necesario acudir a los bogas y subir el cauce con tracción humana, en contra de la corriente (Posada Carbó, 1989; Nichols, 1973).

Since the advent of the postgenomic era, efforts have focused on the development of rapid strategies for annotating plant genes of unknown function. Given its simplicity and rapidity, virus-induced gene silencing (VIGS) has become one of the preeminent approaches for functional analyses. However, several problems remain intrinsic to the use of such a strategy in the study of both metabolic and developmental processes. The most prominent of these is the commonly observed phenomenon of ""sectoring"" the tissue regions that are not effectively targeted by VIGS. To better discriminate these sectors, an effective marker system displaying minimal secondary effects is a prerequisite. Utilizing a VIGS system based on the tobacco rattle virus vector, we here studied the effect of silencing the endogenous phytoene desaturase gene (pds) and the expression and subsequent silencing of the exogenous green fluorescence protein (gfp) on the metabolism of Arabidopsis (Arabidopsis thaliana) leaves and tomato (Solanum lycopersicum) fruits. In leaves, we observed dramatic effects on primary carbon and pigment metabolism associated with the photobleached phenotype following the silencing of the endogenous pds gene. However, relatively few pleiotropic effects on carbon metabolism were observed in tomato fruits when pds expression was inhibited. VIGS coupled to gfp constitutive expression revealed no significant metabolic alterations after triggering of silencing in Arabidopsis leaves and a mild effect in mature green tomato fruits. By contrast, a wider impact on metabolism was observed in ripe fruits. Silencing experiments with an endogenous target gene of interest clearly demonstrated the feasibility of cosilencing in this system; however, carefully constructed control experiments are a prerequisite to prevent erroneous interpretation.

A presente monografia dentro de um tema abrangente como a Saúde Pública no Brasil, propõe-se, neste estudo, realizar uma análise de como a organização "Secretaria de Estado da Saúde de São Paulo" procurou equacionar os aspectos administrativos para que a programação substantiva de saúde pudesse ser implantada e executada. Retrata a conduta da administração no complexo: dirigente e subordinados, analisando em que bases racionais estão sendo desenvolvidos, no interior da Unidades de Saúde pesquisadas, as complexas atividades e Funções da Instituição. Aborda o tema "Desenvolvimento Organizacional", interpretando as mudanças ambientais, elaborando opções de inovação para as organizações evoluírem. Analisa as condições necessárias à inovação e adaptação organizacional, conduzindo a maior satisfação dos indivíduos participantes e maior eficiência das unidades estudadas. Examina, do ponto de vista de execução das atividades, sob que prisma os conflitos e a consequente relação de poder são percebidos e aceitos pela Equipe de Saúde e pela comunidade usuária.

The late eighteenth-century author Frances Burney is best known for popularizing the “comedy of manners,” a literary style later adopted by Jane Austen. Burney’s novels, journals, and plays offer an intriguing commentary on contemporary social customs and etiquette. In particular, she voices the concerns and desires of women, leading scholars to focus on the feminist overtones of her writing. Although she carefully examined female roles in the household and family structure, Burney also provided an insider’s perspective into London high life. As an acclaimed author and member of the royal court, Burney offers a rare insight into the lives of the urban elite. For these reasons, I have chosen to examine three of her works within the context of their London setting. In Evelina, Cecilia, and The Witlings, Burney examines women’s struggle for independence against the backdrop of the city. These works offer a new interpretation of the female Bildungsroman, or coming of age story. Burney shows how London life influences her heroines’ expectations, ambitions and desires. Evelina’s coming of age centers around the quest for family and social acceptance, while the two Cecilias of Cecilia and The Witlings confront the financial pressures that accompany their inheritance. Ultimately, the three protagonists learn important lessons that are specific to city life. Although Burney concludes each story with the heroine’s marriage, her focus is not on romance, as has been suggested, but on the cultural landscape of the city. Coming of age in her stories is inextricably connected to the diverse challenges and opportunities presented to urban women.
