Objectives. This study characterized the feldspathic ceramic surfaces after various silanization protocols.Methods. Ceramic bars (2 mm x 4 mm x 10 mm) (N = 18) of feldpathic ceramic (VM7, VITA Zahnfabrik) were manufactured and finished. Before silane application, the specimens were ultrasonically cleaned in distilled water for 10 min. The ceramic specimens were randomly divided into nine groups (N = 2 per group) and were treated with different silane protocols. MPS silane (ESPE-Sil, 3M ESPE) was applied to all specimens and left to react at 20 degrees C for 2 min (G20). After drying, the specimens were subjected to heat treatment in an oven at 38 degrees C (G38), 79 degrees C (G79) or 100 degrees C (G100) for 1 min. Half of the specimens of each group were rinsed with water at 80 degrees C for 15 s (G20B, G38B, G79B, G100B). The control group (GC) received no silane. Attenuated total reflection infrared Fourier transform analysis (ATR FT-IR) was performed using a spectrometer. Thickness of silane layer was measured using a spectroscopic ellip-someter working in the lambda = 632.8 nm (He-Ne laser) at 70 degrees incidence angle. Surface roughness was evaluated using an optical profilometer. Specimens were further analyzed under the Scanning Electron Microscopy (SEM) to observe the topographic patterns.Results. ATR FT-IR analysis showed changes in Si-O peaks with enlarged bands around 940 cm(-1). Ellipsometry measurements showed that all post-heat treatment actions reduced the silane film thickness (30.8-33.5 nm) compared to G20 (40 nm). The groups submitted to rinsing in hot water (B groups) showed thinner silane films (9.8-14.4 nm) than those of their corresponding groups (without washing) (30.8-40 nm). Profilometer analysis showed that heat treatments (Ra approximate to 0.10-0.19 mu m; Rq approximate to 0.15-0.26 mu m) provided a smoother surface than the control group (Ra approximate to 0.48 mu m; Rq approximate to 0.65 mu m). Similar patterns were also observed in SEM images.Significance. Heat treatment after MPS silane application improved the silane layer network. Rinsing with boiling water eliminated the outmost unreacted regions of the silane yielding to thinner film thicknesses. (C) 2011 Academy of Dental Materials. Published by Elsevier Ltd. Herein, it was investigated for the first time the electro-oxidation of ethanol on Pt and PtRu electrodeposits in acidic media by using in situ surface enhanced infrared absorption spectroscopy with attenuated total reflection (ATR-SEIRAS). The experimental setup circumvents the weak absorbance signals related to adsorbed species, usually observed for rough, electrodeposited surfaces, and allows a full description of the CO coverage with the potential for both catalysts. The dynamics of adsorption-oxidation of CO was accessed by ATR-SEIRAS experiments (involving four ethanol concentrations) and correlated with expressions derived from a simple kinetic model. Kinetic analysis suggests that the growing of the CO adsorbed layer is nor influenced by the presence of Ru neither by the concentration of ethanol. The results suggest that the C-C scission is not related to the presence of Ru and probably happens at Pt sites.

This work describes the study of the ethanol oxidation reaction of a Pt/C Etek electrocatalyst that was supported on different substrates, such as gold, glassy carbon and carbon cloth treated with PTFE. In the ethanol oxidation reaction, the activity varies with the substrate, as well as the pathways for ethanol oxidation, as studied by an ATR-FTIR in situ setup using the carbon cloth as the electrocatalyst support. The electrocatalyst Pt/C supported on gold starts acetaldehyde production from ethanol oxidation at an onset potential of 0.1 V less than that observed for the same process on Teflon-treated carbon cloth. The Pt/C supported on the carbon cloth starts its CO2 production for the same oxidation process at 0.2 V less than on the Pt/C supported on gold substrate. The differences in catalytic activity for the ethanol oxidation reaction depend not only on the electrocatalyst but also on various electrode factors, such as the substrate, the roughness of the electrode and the charge transfer resistance.

FIR spectroscopy is an alternative way of collecting spectra of many inorganic pigments and corrosion products found on art objects, which is not normally observed in the MIR region. Most FIR spectra are traditionally collected in transmission mode but as a real novelty it is now also possible to record FIR spectra in ATR (Attenuated Total Reflectance) mode. In FIR transmission we employ polyethylene (PE) for preparation of pellets by embedding the sample in PE. Unfortunately, the preparation requires heating of the PE in order to produces at transparent pellet. This will affect compounds with low melting points, especially those with structurally incorporated water. Another option in FIR transmission is the use of thin films. We test the use of polyethylene thin film (PETF), both commercial and laboratory-made PETF. ATR collection of samples is possible in both the MIR and FIR region on solid, powdery or liquid samples. Changing from the MIR to the FIR region is easy as it simply requires the change of detector and beamsplitter (which can be performed within a few minutes). No preparation of the sample is necessary, which is a huge advantage over the PE transmission method. The most obvious difference, when comparing transmission with ATR, is the distortion of band shape (which appears asymmetrical in the lower wavenumber region) and intensity differences. However, the biggest difference can be the shift of strong absorbing bands moving to lower wavenumbers in ATR mode. The sometimes huge band shift necessitates the collection of standard library spectra in both FIR transmission and ATR modes, provided these two methods of collecting are to be employed for analyses of unknown samples. Standard samples of 150 pigment and corrosion compounds are thus collected in both FIR transmission and ATR mode in order to build up a digital library of spectra for comparison with unknown samples. XRD, XRF and Raman spectroscopy assists us in confirming the purity or impurity of our standard samples. 24 didactic test tables, with known pigment and binder painted on the surface of a limestone tablet, are used for testing the established library and different ways of collecting in ATR and transmission mode. In ATR, micro samples are scratched from the surface and examined in both the MIR and FIR region. Additionally, direct surface contact of the didactic tablets with the ATR crystal are tested together with water enhanced surface contact. In FIR transmission we compare the powder from our test tablet on the laboratory PETF and embedded in PE. We also compare the PE pellets collected using a 4x beam condenser, focusing the IR beam area from 8 mm to 2 mm. A few samples collected from a mural painting in a Nepalese temple, corrosion products collected from archaeological Chinese bronze objects and samples from a mural paintings in an Italian abbey, are examined by ATR or transmission spectroscopy.

Oggetto di questa tesi di laurea è la riqualificazione funzionale ed energetica di un'autorimessa per corriere costruita a Forlì nel 1935 dal geom. Alberto Flamigni e di proprietà dell' ATR, Agenzia per la Mobilità della provincia di Forlì-Cesena. Al deposito per corriere sono annessi dei piccoli capanni adibiti a magazzini ed una palazzina per uffici costruita negli anni '50, non facente parte del progetto originale. Oggi l'intero complesso risulta in disuso e la richiesta espressa dall'Amministrazione Comunale di Forlì è quella di adattare il comparto per ospitare funzioni musicali, d'intrattenimento e cultura, pensando anche ad un collegamento col manufatto storico dell'Arena Forlivese; quest'ultima, costruita negli anni '20, è situata ai margini del lotto in esame, risulta di proprietà privata ed oggi versa in condizioni di grave degrado. Uno dei fini del progetto, sul filo conduttore delle richieste dell'Amministrazione, è quello di mantenere l'involucro originale dell'edificio, su cui grava anche un vincolo storico, essendo stato progettato durante il periodo fascista ed avendo forti richiami alle soluzioni architettoniche adottate da Marcello Piacentini. Si è quindi deciso di lavorare al suo interno, al fine di creare dei nuclei indipendenti che ospitano le nuove funzioni di auditorium, mediateca, spazio espositivo, sale prova, camerini, mantenendo invece intatto il perimetro in mattoni facciavista con basamento in travertino. Il fronte esposto a sud, essendo stato originariamente pensato come mero elemento di chiusura, senza basamento e sistema di rivestimento ma semplicemente intonacato, si distacca dal resto dell'involucro ed è stato perciò oggetto di maggiori modifiche, in relazione anche al nuovo orientamento d'ingresso pensato per il comparto: l'Amministrazione Comunale ha infatti espresso il desiderio di modificare il percorso di accesso all'edificio, dal fronte nord su piazza Savonarola al fronte ovest su via Ugo Bassi. Il progetto ha adottato un approccio integrato dal punto di vista formale e costruttivo, ponendo particolare attenzione al rispetto e alla valorizzazione della struttura esistente: uno dei punti forti dell'ex deposito è infatti la sua copertura in travi reticolari in c.a. con shed vetrati orientati a nord. Tale sistema di copertura è stato mantenuto per favorire l'illuminazione degli spazi interni, isolato termicamente ed integrato con dei pannelli diffusori che garantiscono una luce uniforme e ben distribuita. Dal punto di vista funzionale e distributivo il progetto ha risposto a criteri di massima flessibilità e fruibilità degli ambienti interni, assecondando le esigenze dell'utenza. Mantenendo la finalità del minimo intervento sull'involucro esistente, nel piano terra si è adottata una tipologia di ambienti open space che delimitano il doppio volume dello spazio espositivo, pensato come un semplice e neutro contenitore, allestibile in base al tipo di mostra ed alla volontà degli organizzatori. Particolare attenzione è stata rivolta alla scelta della tipologia costruttiva per l'auditorium ed i volumi adibiti a sale prova, camerini e depositi, adottando elementi prefabbricati in legno assemblati a secco. Si sono studiati anche i sistemi impiantistici al fine di garantire un elevato livello di comfort interno e nel contempo un considerevole risparmio dal punto di vista energetico. Durante le varie fasi di avanzamento e messa a punto del progetto è stata posta grande attenzione all'aspetto acustico, dalla scelta della forma della sala al trattamento superficiale per garantire un'ottima resa prestazionale, parametro imprescindibile nella progettazione di un adeguato spazio musicale. Altro elemento preso in considerazione a scala locale e urbana è stato quello della sistemazione del cortile a sud, oggi asfaltato ed utilizzato come semplice parcheggio di autobus, al fine di trasformarlo in parco pubblico fruibile dagli utenti del complesso culturale e nel contempo elemento di connessione con l'Arena, tramite un nuovo sistema di orientamenti e percorsi.

In der vorliegenden Arbeit wurde der Einfluss der DNA-Reparaturenzyme NBN, ATM und ATR, die wichtige Funktionen während der Reparatur von DNA-Doppelstrangbrüchen (DSBs) besitzen, auf die Alkylanzien-induzierte Toxizität untersucht. Dabei konnte gezeigt werden, dass verschiedene menschliche Zelllinien, welche eine Beeinträchtigung in einem dieser drei Gene aufweisen, eine erhöhte Sensitivität gegenüber N-Methyl-N'-Nitro-N-Nitrosoguanidin (MNNG) und dem Chemotherapeutikum Temozolomid (TMZ) zeigen. Da das DNA-Reparaturenzym MGMT die Zellen vor der Induktion des Zelltods schützt, kann geschlussfolgert werden, dass die Hypersensitivität der mutierten Zelllinien auf die O6-MeG-Läsion zurückzuführen ist. Es konnte gezeigt werden, dass Mutationen von NBN oder ATM nicht zu einer verminderten Kapazität der Basen-Exzisions-Reparatur (BER) führen. Somit ist die erhöhte Sensitivität der mutierten Zellen sehr wahrscheinlich auf eine verminderte Reparatur der DSBs zurückzuführen, welche durch die O6-MeG-Läsion induziert werden. Damit konnte NBN, ATM und ATR als neue Faktoren in der Abwehr gegen Alkylanzien-induzierte Toxizität identifiziert werden. Dies ist von großer klinischer Bedeutung, da einerseits die drei Proteine als therapeutisches Angriffsziel Bedeutung gewinnen und andererseits verschiedene Tumore, die in der Klinik mit alkylierenden Agenzien behandelt werden, Mutationen in diesen Genen tragen.rnrnWeiterhin wurde beobachtet, dass NBN- und ATM-defiziente Zellen nach Behandlung mit methylierenden Agenzien eine ungewöhnlich hohe Nekrose-Rate aufweisen. Es konnte gezeigt werden, dass diese unabhängig von einer PARP1-Aktivierung induziert wird. Dennoch wurde in den NBN- und ATM-mutierten Zelllinien im Gegensatz zum Wildtyp eine sehr starke Verminderung der ATP-Menge nach MNNG-Behandlung beobachtet. Diese wird durch das Fehlen einer effektiven Aktivierung der AMP-Kinase in diesen Zellen verursacht. Somit kann angenommen werden, dass die hohe Nekrose-Rate auf eine ATP-Depletion zurückzuführen ist, welche durch die nicht ausreichende AMP-Kinase-Aktivierung in diesen Zellen bedingt wird. Daher konnte NBN und ATM als Faktoren des zellulären Schutzes gerichtet gegen die Induktion der „programmierten Nekrose“ identifiziert werden. Dies ist ebenfalls von klinischer Bedeutung. Tragen Tumorzellen von Tumoren, welche mit methylierenden Agenzien behandelt werden, Mutationen in einem dieser Gene, so muss mit einer vermehrten Induktion von Nekrose und daher mit einer Stimulierung des Immunsystems während der Chemotherapie gerechnet werden. Dies wäre einerseits mit erhöhten Nebenwirkungen, die sich insbesondere durch Entzündungsreaktionen äußern, verbunden. Andererseits zeigen verschiedene Arbeiten, dass die Stimulation des Immunsystems durch sterbende Tumorzellen während der Chemotherapie die Tumorregression positiv beeinflussen kann.

Die Gesundheitseffekte von Aerosolpartikeln werden stark von ihren chemischen und physikalischen Eigenschaften und somit den jeweiligen Bildungsprozessen und Quellencharakteristika beeinflusst. Während die Hauptquellen der anthropogenen Partikelemissionen gut untersucht sind, stellen die spezifischen Emissionsmuster zahlreicher kleiner Aerosolquellen, welche lokal und temporär zu einer signifikanten Verschlechterung der Luftqualität beitragen können, ein Forschungsdesiderat dar.rnIn der vorliegenden Arbeit werden in kombinierten Labor- und Feldmessungen durch ein integratives Analysekonzept mittels online (HR-ToF-AMS ) und filterbasierter offline (ATR-FTIR-Spektroskopie ) Messverfahren die weitgehend unbekannten physikalischen und chemischen Eigenschaften der Emissionen besonderer anthropogener Aerosolquellen untersucht. Neben einem Fußballstadion als komplexe Mischung verschiedener Aerosolquellen wie Frittieren und Grillen, Zigarettenrauchen und Pyrotechnik werden die Emissionen durch Feuerwerkskörper, landwirtschaftliche Intensivtierhaltung (Legehennen), Tief- und Straßenbauarbeiten sowie abwasserbürtige Aerosolpartikel in die Studie mit eingebunden. Die primären Partikelemissionen der untersuchten Quellen sind vorrangig durch kleine Partikelgrößen (dp < 1 µm) und somit eine hohe Lungengängigkeit gekennzeichnet. Dagegen zeigen die Aerosolpartikel im Stall der landwirtschaftlichen Intensivtierhaltung sowie die Emissionen durch die Tiefbauarbeiten einen hohen Masseanteil von Partikeln dp > 1 µm. Der Fokus der Untersuchung liegt auf der chemischen Charakterisierung der organischen Partikelbestandteile, welche für viele Quellen die NR-PM1-Emissionen dominieren. Dabei zeigen sich wichtige quellenspezifische Unterschiede in der Zusammensetzung der organischen Aerosolfraktion. Die beim Abbrand von pyrotechnischen Gegenständen freigesetzten sowie die abwasserbürtigen Aerosolpartikel enthalten dagegen hohe relative Gehalte anorganischer Substanzen. Auch können in einigen spezifischen Emissionen Metallverbindungen in den AMS-Massenspektren nachgewiesen werden. Über die Charakterisierung der Emissionsmuster und -dynamiken hinaus werden für einige verschiedenfarbige Rauchpatronen sowie die Emissionen im Stall der Intensivtierhaltung Emissionsfaktoren bestimmt, die zur quantitativen Bilanzierung herangezogen werden können. In einem weiteren Schritt werden anhand der empirischen Daten die analytischen Limitierungen der Aerosolmassenspektrometrie wie die Interferenz organischer Fragmentionen durch (Hydrogen-)Carbonate und mögliche Auswertestrategien zur Überwindung dieser Grenzen vorgestellt und diskutiert.rnEine umfangreiche Methodenentwicklung zur Verbesserung der analytischen Aussagekraft von organischen AMS-Massenspektren zeigt, dass für bestimmte Partikeltypen einzelne Fragmentionen in den AMS-Massenspektren signifikant mit ausgewählten funktionellen Molekülgruppen der FTIR-Absorptionsspektren korrelieren. Bedingt durch ihre fehlende Spezifität ist eine allgemeingültige Interpretation von AMS-Fragmentionen als Marker für verschiedene funktionelle Gruppen nicht zulässig und häufig nur durch die Ergebnisse der komplementären FTIR-Spektroskopie möglich. Des Weiteren wurde die Verdampfung und Ionisation ausgewählter Metallverbindungen im AMS analysiert. Die Arbeit verdeutlicht, dass eine qualitative und quantitative Auswertung dieser Substanzen nicht ohne Weiteres möglich ist. Die Gründe hierfür liegen in einer fehlenden Reproduzierbarkeit des Verdampfungs- und Ionisationsprozesses aufgrund von Matrixeffekten sowie der in Abhängigkeit vorangegangener Analysen (Verdampferhistorie) in der Ionisationskammer und auf dem Verdampfer statt-findenden chemischen Reaktionen.rnDie Erkenntnisse der Arbeit erlauben eine Priorisierung der untersuchten anthropogenen Quellen nach bestimmten Messparametern und stellen für deren Partikelemissionen den Ausgangpunkt einer Risikobewertung von atmosphärischen Folgeprozessen sowie potentiell negativen Auswirkungen auf die menschliche Gesundheit dar. rn

Fourier transform infrared (FTIR) spectroscopy was applied to determine the type of surface treatment and dose used on cork stoppers and to predict the friction between stopper and bottleneck. Agglomerated cork stoppers were finished with two different doses and using two surface treatments: P (paraffin and silicone), 15 and 25 mg/stopper, and S (only silicone), 10 and 15 mg/stopper. FTIR spectra were recorded at five points for each stopper by attenuated total reflectance (ATR). Absorbances at 1,010, 2,916, and 2,963 cm -1 were obtained in each spectrum. Discriminant analysis techniques allowed the treatment, and dose applied to each stopper to be identified from the absorbance values. 91.2% success rates were obtained from individual values and 96.0% from the mean values of each stopper. Spectrometric data also allowed treatment homogeneity to be determined on the stopper surface, and a multiple regression model was used to predict the friction index (If = Fe/Fc) (R 2 = 0.93)

El presente trabajo busca el desarrollo de una planificación en fútbol comparando los diferentes modelos de planificación. Se basa en la contraposición entre los modelos tradicionales y los contemporáneos, siendo el modelo ATR el más adecuado para la planificación de una temporada de fútbol. El trabajo se desarrolla desde un marco teórico englobando las diferentes características y fases en el desarrollo de una planificación así como los pasos a seguir para su correcto desarrollo incluyendo la técnica de marketing actualmente más utilizada, la técnica DAFO, y un diseño de plan de ejecución adecuado.

In fission yeast, the rad3 gene product plays a critical role in sensing DNA structure defects and activating damage response pathways. A structural homologue of rad3 in humans (ATR) has been identified based on sequence similarity in the protein kinase domain. General information regarding ATR expression, protein kinase activity, and cellular localization is known, but its function in human cells remains undetermined. In the current study, the ATR protein was examined by gel filtration of protein extracts and was found to exist predominantly as part of a large protein complex. A kinase-inactivated form of the ATR gene was prepared by site-directed mutagenesis and was used in transfection experiments to probe the function of this complex. Introduction of this kinase-dead ATR into a normal fibroblast cell line, an ATM-deficient fibroblast line derived from a patient with ataxia–telangiectasia, or a p53 mutant cell line all resulted in significant losses in cell viability. Clones expressing the kinase-dead ATR displayed increased sensitivity to x-rays and UV and a loss of checkpoint control. We conclude that ATR functions as a critical part of a protein complex that mediates responses to ionizing and UV radiation in human cells. These responses include effects on cell viability and cell cycle checkpoint control.

In some cases external morphology is not sufficient to discern between populations of a species, as occurs in the dung beetle Canthon humectus hidalgoensis Bates; and much less to determine phenotypic distances between them. FTIR-ATR spectroscopy show several advantages over other identification techniques (e.g. morphological, genetic, and cuticular hydrocarbons analysis) due to the non-invasive manner of the sample preparation, the relative speed of sample analysis and the low-cost of this technology. The infrared spectrum obtained is recognized to give a unique ‘fingerprint’ because vibrational spectra are specific and unique to the molecular nature of the sample. In our study, results showed that proteins, amino acids and aromatic ethers of insect exocuticle have promising discriminative power to discern between different populations of C. h. hidalgoensis. Furthermore, the correlation between geographic distances between populations and the chemical distances obtained by proteins + amino acids + aromatic ethers was statistically significant, showing that the spectral and spatial information available of the taxa together with appropriated chemometric methods may help to a better understanding of the identity, structure, dynamics and diversity of insect populations.

In the current study, the relationship between current and biomass and bio-adhesion mechanism of electrogenic biofilm on electrode were investigated using EQCM and ATR-SEIRAS linking electrochemistry. The results indicated that cellular biomass of biofilm on QCM-crystal surface showed maximum value of 6.0 μg/cm2 in initial batch and 11.5 μg/cm2 in the second batch on mature biofilm, producing a similar maximum current density of 110 μA/μg. Especially, the optimum cell biomass linking high electricity production ratio (110 μA/μg) occurred before maximum biomass coming, implying that over-growth mature biofilm is not an optimum state for enhancing power output of MFCs. On the other hand, the spectra using ATR-SEIRAS technique linking electrochemistry obviously exhibited water structure adsorption change at early biofilm formation and meanwhile the water adsorption accompanied the adsorbed bacteria and the bound cells population on the electrode increased with time. Meanwhile, the direct contact of bacteria and electrode via outer-membrane protein can be confirmed via a series spectra shift at amide I and amide II modes and water movement from negative bands displacing by adsorbed bacteria. Our study provided supplementary information about the interaction between the microbes and electrode beyond traditional electrochemistry.
