Gene targeting by microRNAs is important in health and disease. We developed a functional assay for identifying microRNA targets and applied it to the K+ channel Kir2.1 (KCNJ2) which is dysregulated in cardiac and vascular disorders. The 3'UTR was inserted downstream of the mCherry red fluorescent protein coding sequence in a mammalian expression plasmid. MicroRNA sequences were inserted into the pSM30 expression vector which provides enhanced green fluorescent protein as an indicator of microRNA expression. HEK293 cells were co-transfected with the mCherry-3'UTR plasmid and a pSM30-based plasmid with a microRNA insert. The principle of the assay is that functional targeting of the 3'UTR by the microRNA results in a decrease in the red/green fluorescence intensity ratio as determined by automated image analysis. The method was validated with miR-1, a known downregulator of Kir2.1 expression, and was used to investigate targeting of the Kir2.1 3'UTR by miR-212. Red/green ratio was lower in miR-212-expressing cells compared to non-targeting controls, an effect that was attenuated by mutating the predicted target site. MiR-212 also reduced inward rectifier current and Kir2.1 protein in HeLa cells. This novel assay has several advantages over traditional luciferase-based assays including larger sample size, amenability to time course studies and adaptability to high-throughput screening.

Flexile single layer electromagnetic wave absorbers were designed by incorporating appropriate amounts of carbon black in a nitrile butadiene rubber matrix along with an optimized amount of magnetic counterpart, namely, barium hexaferrite for applications in S, C, and X-bands. Effective dielectric permittivity and magnetic permeability were measured using cavity perturbation method in the frequency range of 2–12 GHz. The microwave absorbing characteristics of the composites were studied in the S, C, and X-bands employing a model in which an electromagnetic wave is incident normally on a metal terminated single layer. Reflection loss exceeding 20 dB is obtained for all the samples in a wide frequency range of 2–12 GHz when an appropriate absorber thickness between 5 and 9mm is chosen. A Constituição Federal do Brasil determina que os Municípios e Estados apliquem não menos que 25% do somatório das suas receitas tributárias e transferências constitucionais nas ações relacionadas ao ensino público. Para demonstrar o atendimento desta regra constitucional cada Ente deve publicar um demonstrativo contábil constante no Relatório Resumido da Execução Orçamentária, instituído pela Secretaria do Tesouro Nacional – STN desde 2000, após a vigência da Lei de Responsabilidade Fiscal – LC 101/2000. A Portaria STN nº 560, de 14 de dezembro de 2001, instituiu o Manual de Elaboração do Relatório Resumido da Execução Orçamentária, composto de vários quadros contábeis, onde tem no seu Anexo X o Demonstrativo das Receitas e Despesas com Manutenção e Desenvolvimento do Ensino – MDE, para evidenciar os valores destinados à educação. Não obstante a comprovação da aplicação se dar por meio de um demonstrativo contábil, isto não garante que os recursos destinados às ações do ensino obedeçam a critérios de qualidade. Não se apresenta neste demonstrativo indicadores sociais que permitam verificar a evolução no nível de ensino da rede pública, o que levou à instituição de novos indicadores para avaliar os resultados destes investimentos públicos. O Índice de Desenvolvimento da Educação Brasileira, Ideb, veio estabelecer metas de critérios sociais por meio de uma pontuação que classifica os entes federativos de acordo com resultados sociais apresentados, como a taxa de aprovação e abandono escolar, por exemplo. O ranking divulgado em 2009 demonstra que os municípios que obtiveram os últimos lugares comprovaram o atendimento do art. 212 da Constituição Federal, ou seja, não há como ter uma relação entre a regra constitucional e a eficácia da administração da educação pelos municípios brasileiros, este é justamente o propósito deste trabalho.

This work has as main objective to study the application of microstrip antennas with patch and use of superconducting arrays of planar and linear phase. Was presented a study of the main theories that explain clearly the superconductivity. The BCS theory, Equations of London and the Two Fluid Model are theories that supported the implementation of the superconducting microstrip antennas. Arrangements phase was analyzed in linear and planar configuration of its antennas are reported factors such arrays to settings and criteria of phase and the spacing between the elements that make the arrayst was reviewed in order to minimize losses due to secondary lobes. The antenna used has a rectangular patch Sn5InCa2Ba4Cu10Oy the superconducting material was analyzed by the method of Transverse Transmission Line (TTL) applied in the field of Fourier transform (FTD). The TTL is a full-wave method, which has committed to obtaining the electromagnetic fields in terms of cross-cutting components of the structure. The inclusion of superconducting patch is made using the boundary condition, complex resistive. Are obtained when the resonant frequency depending on the parameters of the antenna, radiation pattern of E-Plan and H-Plan for the M-phase arrangements of antennas in the linear and planar configurations for different values of phase and spacing between the elements.
