Adaptor proteins play an important role in signaling pathways by providing a platform on which many other proteins can interact. Malfunction or mislocalization of these proteins may play a role in the development of disease. Lipoma preferred partner (LPP) is a nucleocytoplasmic shuttling adaptor protein. Previous work shows that LPP plays a role in the function of smooth muscle cells and in atherosclerosis. In this study we wanted to determine whether LPP has a role in the myocardium. LPP expression increased by 56% in hearts from pressure overload aortic-banded rats (p < 0.05 n = 4), but not after myocardial infarction, suggesting hemodynamic load regulates its expression. In vitro, LPP expression was 87% higher in cardiac fibroblasts than myocytes (p < 0.05 n = 3). LPP expression was downregulated in the absence of the actin cytoskeleton but not when microtubules were disassembled. We mechanically stretched cardiac fibroblasts using the Flexcell 4000 for 48 h (1 Hz, 5% maximum strain), which decreased total LPP total expression and membrane localization in subcellular fractions (p < 0.05, n = 5). However, L-NAME, an inhibitor of nitric oxide synthase (NOS), significantly upregulated LPP expression. These findings suggest that LPP is regulated by a complex interplay between NO and mechanical cues and may play a role in heart failure induced by increased hemodynamic load.

Bismuth germanate glasses are interesting materials due to their physical properties and their unique structural characteristics caused by the coordination changes of bismuth and germanium atoms. Glasses of the bismuth germanate system were prepared by melting/molding method and were investigated concerning their thermal and structural properties. The structural analysis of the samples was carried out by micro-Raman and Fourier transform infrared spectroscopes. It was observed that the glass structure is formed basically by GeO(4) tetrahedral units also having the formation of the GeO(6) octahedral units. BiO(2) was considered a network former by observing the presence of octahedral BiO(6) and pyramidal BiO(3) groups in the local structure of the samples. An absorption band observed at 1103 cm(-1) in the IR spectrum of the undoped glass was attributed to the Bi-O-Ge and/or Bi-O-Bi linkage vibration. The said band shifted to lower wavenumbers after the CeO(2) addition thus reflecting changes in the glass network. Cerium oxide was an efficient oxidant agent to prevent the darkening of the glasses which was probably associated to the reduction of Bi ions. However, CeO(2) was incorporated as a local network modifier in the glass structure even at concentrations of 0.2 mol%. (C) 2010 Elsevier B.V. All rights reserved.

A queda prematura dos frutos cítricos (QPFC), causada por Colletotrichum acutatum, dados os grandes prejuízos que têm causado aos produtores, constitui-se numa doença de grande importância econômica. O controle da doença é feito predominantemente mediante uso de fungicidas, que eleva o custo de produção e afeta negativamente o meio ambiente. Diante disso, este trabalho teve por objetivo buscar um método alternativo de controle da QPFC, mediante o uso de agentes de biocontrole ou de biofertilizantes. Diferentes concentrações de biofertilizantes (originários de duas fontes distintas e denominados de Bio1 e Bio 2); três isolados de Bacillus subtilis (ACB-69; 72 e 77) e três isolados de Trichoderma spp. (ACB-14; 37 e 39) foram testados, isoladamente ou em combinação, sob condições de laboratório, quanto à capacidade inibitória da germinação de conídios de C. acutatum. Estudaram-se, ainda, a produção de metabólitos termoestáveis por B. subtilis e o efeito sobre a germinação do patógeno. Quinze isolados de B. subtilis foram testados quanto à capacidade de prevenir a infecção por C. acutatum em flores destacadas de lima- ácida 'Tahiti' e, no campo, foram instalados dois experimentos, visando a testar ACBs e biofertilizantes no controle da doença. Verificou-se que o isolado ACB-72 (B. subtilis) e ACB-37 (T. pseudokoningii) foram os que mais inibiram a germinação do patógeno. Quanto à produção de metabólitos termoestáveis, ACB-69 e 77 foram os mais eficientes em produzir substâncias antifúngicas, e em quantidades suficientes para inibirem a germinação do patógeno. A mistura dos quatro isolados de Bacillus (ACBs: 69; 72; 77 e AP3) foi o que apresentou maior porcentagem de inibição (73%). Os biofertilizantes (Bio1 e Bio2), em concentrações acima de 10% e, quando em associação com isolados de Trichoderma spp., promoveram maiores inibições na germinação de C. acutatum. em testes com flores destacadas, verificou-se que, onde foram aplicados os ACBs 69; 76; 74 e 77, as porcentagens de pétalas sem sintomas de infecção por C. acutatum foram de 83; 92; 92 e 97%, respectivamente. Mediante avaliações a campo, verificou-se a potencialidade de B. subtilis e de biofertilizantes em controlar a doença.

A structural study of the thermal evolution of Ni0.69Cr0.31(OH)(2)(CO3)(0.155)(.)nH(2)O into NiO and tetragonal NiCr2O4 is reported. The characteristic structural parameters of the two coexisting crystalline phases, as well as their relative abundance, were determined by Rietveld refinement of powder x-ray diffraction (PXRD) patterns. The results of the simulations allowed us to elucidate the mechanism of the demixing process of the oxides. It is demonstrated that nucleation of a metastable nickel chromite within the common oxygen framework of the parent Cr-III-doped bunsenite is the initial step of the cationic redistribution. The role that trivalent cations play in the segregation of crystalline spinels is also discussed.

Manganese nodules and manganese carbonate concretions occur in the upper 10-15 cm of the Recent sediments of Loch Fyne, Argyllshire in water depths of 180-200 m. The nodules are spherical, a few mm to 3 cm in diameter, and consist of a black, Mn-rich core and a thin, red, Fe-rich rim. The carbonate occurs as irregular concretions, 0.5-8 cm in size, and as a cement in irregular nodule and shell fragment aggregates. It partially replaces some nodule material and clastic silicate inclusions, but does not affect aragonitic and calcitic shell fragments. The nodules are approximately 75% pure oxides and contain 30% Mn and 4% Fe. In the cores, the principal mineral phase is todorokite, with a Mn/Fe ratio of 17. The rim consists of X-ray amorphous Fe and Mn oxides with a Mn/Fe ratio of 0.66. The cores are enriched, relative to Al, in K, Ba, Co, Mo, Ni and Sr while the rims contain more P, Ti, As, Pb, Y and Zn. The manganese carbonate has the composition (Mn47.7 Ca45.1 Mg7.2) CO3. Apart from Cu, all minor elements are excluded from significant substitution in the carbonate lattice. Manganese nodules and carbonates form diagenetically within the Recent sediments of Loch Fyne. This accounts for the high Mn/Fe ratios in the oxide phases and the abundance of manganese carbonate concretions. Mn concentrations in the interstitial waters of sediment cores are high (ca. 10 ppm) as also, by inference, are the dissolved carbonate concentrations.

Bivalve shells can provide excellent archives of past environmental change but have not been used to interpret ocean acidification events. We investigated carbon, oxygen and trace element records from different shell layers in the mussels Mytilus galloprovincialis combined with detailed investigations of the shell ultrastructure. Mussels from the harbour of Ischia (Mediterranean, Italy) were transplanted and grown in water with mean pHT 7.3 and mean pHT 8.1 near CO2 vents on the east coast of the island. Most prominently, the shells recorded the shock of transplantation, both in their shell ultrastructure, textural and geochemical record. Shell calcite, precipitated subsequently under acidified seawater responded to the pH gradient by an in part disturbed ultrastructure. Geochemical data from all test sites show a strong metabolic effect that exceeds the influence of the low-pH environment. These field experiments showed that care is needed when interpreting potential ocean acidification signals because various parameters affect shell chemistry and ultrastructure. Besides metabolic processes, seawater pH, factors such as salinity, water temperature, food availability and population density all affect the biogenic carbonate shell archive.

The glacial climate system transitioned rapidly between cold (stadial) and warm (interstadial) conditions in the Northern Hemisphere. This variability, referred to as Dansgaard-Oeschger variability, is widely believed to arise from perturbations of the Atlantic Meridional Overturning Circulation. Evidence for such changes during the longer Heinrich stadials has been identified, but direct evidence for overturning circulation changes during Dansgaard-Oeschger events has proven elusive. Here we reconstruct bottom water [CO3]2- variability from B/Ca ratios of benthic foraminifera and indicators of sedimentary dissolution, and use these reconstructions to infer the flow of northern-sourced deep water to the deep central sub-Antarctic Atlantic Ocean. We find that nearly every Dansgaard-Oeschger interstadial is accompanied by a rapid incursion of North Atlantic Deep Water into the deep South Atlantic. Based on these results and transient climate model simulations, we conclude that North Atlantic stadial-interstadial climate variability was associated with significant Atlantic overturning circulation changes that were rapidly transmitted across the Atlantic. However, by demonstrating the persistent role of Atlantic overturning circulation changes in past abrupt climate variability, our reconstructions of carbonate chemistry further indicate that the carbon cycle response to abrupt climate change was not a simple function of North Atlantic overturning.

The Pliocene period is the most recent time when the Earth was globally significantly (~3°C) warmer than today. However, the existing pCO2 data for the Pliocene are sparse and there is little agreement between the various techniques used to reconstruct palaeo-pCO2. Moreover, the temporal resolution of the published records does not allow a robust assessment of the role of declining pCO2 in the intensification of the Northern Hemisphere Glaciation (INHG) and a direct comparison to other proxy records are lacking. For the first time, we use a combination of foraminiferal (delta11B) and organic biomarker (alkenone-derived carbon isotopes) proxies to determine the concentration of atmospheric CO2 over the past 5 Ma. Both proxy records show that during the warm Pliocene pCO2 was between 330 and 400 ppm, i.e. similar to today. The decrease to values similar to pre-industrial times (275-285 ppm) occurred between 3.2 Ma and 2.8 Ma - coincident with the INHG and affirming the link between global climate, the cryosphere and pCO2.

Ocean acidification (OA), resulting from increasing dissolved carbon dioxide (CO2) in surface waters, is likely to affect many marine organisms, particularly those that calcify. Recent OA studies have demonstrated negative and/or differential effects of reduced pH on growth, development, calcification and physiology, but most of these have focused on taxa other than calcareous benthic macroalgae. Here we investigate the potential effects of OA on one of the most common coral reef macroalgal genera,Halimeda. Species of Halimeda produce a large proportion of the sand in the tropics and are a major contributor to framework development on reefs because of their rapid calcium carbonate production and high turnover rates. On Palmyra Atoll in the central Pacific, we conducted a manipulative bubbling experiment to investigate the potential effects of OA on growth, calcification and photophysiology of 2 species of Halimeda. Our results suggest that Halimeda is highly susceptible to reduced pH and aragonite saturation state but the magnitude of these effects is species specific. H. opuntiasuffered net dissolution and 15% reduction in photosynthetic capacity, while H. taenicola did not calcify but did not alter photophysiology in experimental treatments. The disparate responses of these species to elevated CO2 partial -pressure (pCO2) may be due to anatomical and physiological differences and could represent a shift in their relative dominance in the face of OA. The ability for a species to exert biological control over calcification and the species specific role of the carbonate skeleton may have important implications for the potential effects of OA on ecological function in the future.

The effect of decreasing aragonite saturation state (Omega Arag) of seawater (elevated pCO2) on calcification rates of Acropora muricata was studied using nubbins prepared from parent colonies located at two sites of La Saline reef (La Réunion Island, western Indian Ocean): a back-reef site (BR) affected by nutrient-enriched groundwater discharge (mainly nitrate), and a reef flat site (RF) with low terrigenous inputs. Protein and chlorophyll a content of the nubbins, as well as zooxanthellae abundance, were lower at RF than BR. Nubbins were incubated at ~27°C over 2 h under sunlight, in filtered seawater manipulated to get differing initial pCO2 (1,440-340 µatm), Omega Arag (1.4-4.0), and dissolved inorganic carbon (DIC) concentrations (2,100-1,850 µmol/kg). Increasing DIC concentrations at constant total alkalinity (AT) resulted in a decrease in Omega Arag and an increase in pCO2. AT at the beginning of the incubations was kept at a natural level of 2,193 ± 6 µmol/kg (mean ± SD). Net photosynthesis (NP) and calcification were calculated from changes in pH and AT during the incubations. Calcification decrease in response to doubling pCO2 relative to preindustrial level was 22% for RF nubbins. When normalized to surface area of the nubbins, (1) NP and calcification were higher at BR than RF, (2) NP increased in high pCO2 treatments at BR compared to low pCO2 treatments, and (3) calcification was not related to Omega Arag at BR. When normalized to NP, calcification was linearly related to Omega Arag at both sites, and the slopes of the relationships were not significantly different. The increase in NP at BR in the high pCO2 treatments may have increased calcification and thus masked the negative effect of low Omega Arag on calcification. Removing the effect of NP variations at BR showed that calcification declined in a similar manner with decreased Omega Arag (increased pCO2) whatever the nutrient loading.

A study of chemical and isotopic composition of coastal thermal springs and waters of the Kraternaya Bay, Yankicha Island, revealed that the total mineralization and concentrations of d18O and d2H decrease from the thermal spring site I to V. These waters are of marine origin with various proportions of local meteoric water. Thermal waters of the site VI have considerably altered chemical and isotopic composition due to high temperatures of surrounding rocks. Base temperatures calculated for this area were 130-200°C. Coastal thermal springs affect isotopic composition of water throughout the bay.

Lysocline reconstructions play an important role in scenarios purporting to explain the lowered atmospheric CO2 content of glacial time. These reconstructions are based on indicators such as the CaCO3 content, the percent of coarse fraction, the ratio of fragments to whole foraminifera shells, the ratio of solution-susceptible to solution-resistant species, and the ratio of coarse to fine CaCO3. All assume that changes with time in the composition of the input material do not bias the result. However, as the composition of the input material does depend on climate, none of these indicators provides an absolute measure of the extent of dissolution. In this paper we evaluate the reliability of the ratio of >63 µm CaCO3 to total CaCO3 as a dissolution indicator. We present here results that suggest that in today's tropics this ratio appears to be determined solely by CO3= ion concentration and water depth (i.e., the saturation state of bottom waters). This finding offers the possibility that the size fraction index can be used to reconstruct CO3= ion concentrations for the late Quaternary ocean to an accuracy of ±5 µmol/kg.

The ocean plays a major role in the global carbon cycle, and attempts to reconstruct past changes in the marine carbonate system are increasing. The speciation of dissolved uranium is sensitive to variations in carbonate system parameters, and previous studies have shown that this is recorded in the uranium-to-calcium ratio (U/Ca) of the calcite shells of planktonic foraminifera. Here we test whether U/Ca ratios of deep-sea benthic foraminifera are equally suited as an indicator of the carbonate system. We compare U/Ca in two common benthic foraminifer species (Planulina wuellerstorfi and Cibicidoides mundulus) from South Atlantic core top samples with the calcite saturation state (Delta [CO3**2-] = [CO3**2-]in situ - [CO3**2-]sat) of the ambient seawater and find significant negative correlations for both species. Compared with planktonic foraminifera, the sensitivity of U/Ca in benthic foraminifera to changes in Delta [CO3**2-] is about 1 order of magnitude higher. Although Delta [CO3**2-] exerts the dominant control on the average foraminiferal U/Ca, the intertest and intratest variability indicates the presence of additional factors forcing U/Ca.

Increasing anthropogenic CO2 emissions to the atmosphere are causing a rise in pCO2 concentrations in the ocean surface and lowering pH. To predict the effects of these changes, we need to improve our understanding of the responses of marine primary producers since these drive biogeochemical cycles and profoundly affect the structure and function of benthic habitats. The effects of increasing CO2 levels on the colonisation of artificial substrata by microalgal assemblages (periphyton) were examined across a CO2 gradient off the volcanic island of Vulcano (NE Sicily). We show that periphyton communities altered significantly as CO2 concentrations increased. CO2 enrichment caused significant increases in chlorophyll a concentrations and in diatom abundance although we did not detect any changes in cyanobacteria. SEM analysis revealed major shifts in diatom assemblage composition as CO2 levels increased. The responses of benthic microalgae to rising anthropogenic CO2 emissions are likely to have significant ecological ramifications for coastal systems.
