We present here the characterization of a new gene family, awr, found in all sequenced Ralstonia solanacearum strains and in other bacterial pathogens. We demonstrate that the five paralogues in strain GMI1000 encode type III-secreted effectors and that deletion of all awr genes severely impairs its capacity to multiply in natural host plants. Complementation studies show that the AWR (alanine-tryptophanarginine tryad) effectors display some functional redundancy, although AWR2 is the major contributor to virulence. In contrast, the strain devoid of all awr genes (¿awr1-5) exhibits enhanced pathogenicity on Arabidopsis plants. A gain-of-function approach expressing AWR in Pseudomonas syringae pv. tomato DC3000 proves that this is likely due to effector recognition, because AWR5 and AWR4 restrict growth of this bacterium in Arabidopsis. Transient overexpression of AWR in nonhost tobacco species caused macroscopic cell death to varying extents, which, in the case of AWR5, shows characteristics of a typical hypersensitive response. Our work demonstrates that AWR, which show no similarity to any protein with known function, can specify either virulence or avirulence in the interaction of R. solanacearum with its plant hosts.

A novel method to synthesize cyclic oligonucleotides (5- to 26-mer) using the thiol-maleimide reaction is described. The target molecules were obtained after subsequent removal of thiol and maleimide protecting groups from 5′-maleimido-3′-thiol-derivatized linear precursors. Retro-Diels-Alder conditions deprotecting the maleimide simultaneously promoted cyclization cleanly and in high yield.

Pollen allergy has a remarkable clinical impact all over Europe, and there are evidences that the prevalence of allergic respiratory induced reactions by pollen in Europe has been increasing in recent decades. The presence of potentially allergenic ornamental species in the town of Tiana, representing 41% of total species recorded. Approximately 28% of these species are Platanus acerifolia. This study attempts to relate the effect it has on the people’s health, the presence of certain urban ornamental plants by identifying and locating the ornamental trees in the town.

Many Gram-negative, cold-adapted bacteria from the Antarctic environment produce large amounts of extracellular matter with potential biotechnological applications. Transmission electron microscopy (TEM) analysis after high-pressure freezing and freeze substitution (HPF-FS) showed that this extracellular matter is structurally complex, appearing around cells as a netlike mesh, and composed of an exopolymeric substance (EPS) containing large numbers of outer membrane vesicles (OMVs). Isolation, purification and protein profiling via 1D SDS-PAGE confirmed the outer membrane origin of these Antarctic bacteria OMVs. In an initial attempt to elucidate the role of OMVs in cold-adapted strains of Gram-negative bacteria, a proteomic analysis demonstrated that they were highly enriched in outer membrane proteins and periplasmic proteins associated with nutrient processing and transport, suggesting that the OMVs may be involved in nutrient sensing and bacterial survival. OMVs from Gram-negative bacteria are known to play a role in lateral DNA transfer, but the presence of DNA in these vesicles has remained difficult to explain. A structural study of Shewanella vesiculosa M7T using TEM and Cryo-TEM revealed that this Antarctic Gram-negative bacterium naturally releases conventional one-bilayer OMVs, together with a more complex type of OMV, previously undescribed, which on formation drags along inner membrane and cytoplasmic content and can therefore also entrap DNA.

Marine microorganisms, including Aeromonas, are a source of compds. for drug development that have generated great expectations in the last decades. Aeromonas infections produce septicemia, and ulcerative and haemorrhagic diseases in fish. Among the pathogenic factors assocd. with Aeromonas, the lipopolysaccharides (LPS), a surface glyconconjugate unique to Gram-neg. bacteria consisting of lipid A (lipid anchor of the mol.), core oligosaccharide and O-specific polysaccharide (O antigen), are key elicitors of innate immune responses. The chem. structure of these three parts has been characterized in Aeromonas. Based on the high variability of repeated units of O-polysaccharides, a total of 97 O-serogroups have been described in Aeromonas species, of which four of them (O:11; O:16; O:18 and O:34) account for more than 60% of the septicemia cases. The core of LPS is subdivided into two regions, the inner (highly conserved) and the outer core. The inner core of Aeromonas LPS is characterized by the presence of 3-deoxy-d-manno-oct-2-ulosonic (ketodeoxyoctonic) acid (Kdo) and l-glycero-d-manno-Heptoses (l,d-Hep), which are linked to the outer core, characterized by the presence of Glc, GlcN, Gal, and GalNAc (in Aeromonas salmonicida), d,d-Hep (in Aeromonas salmonicida), and l,d-Hep (in Aeromonas hydrophila). The biol. relevance of these differences in the distal part of the outer core among these species has not been fully assessed to date. The inner core is attached to the lipid A, a highly conserved structure that confers endotoxic properties to the LPS when the mol. is released in blood from lysed bacteria, thus inducing a major systemic inflammatory response known as septic or endotoxic shock. In Aeromonas salmonicida subsp. salmonicida the Lipid A components contain three major lipid A mols., differing in acylation patterns corresponding to tetra-, penta- and hexa-acylated lipid A species and comprising of 4'-monophosphorylated β-2-amino-2-deoxy-d-glucopyranose-(1→6)-2-amino-2-deoxy-d-glucopyranose disaccharide. In the present review, we discuss the structure-activity relationships of Aeromonas LPS, focusing on its role in bacterial pathogenesis and its possible applications.

Background: Colorectal cancer is a major health problem worldwide and many efforts have been done to delineate risk factors and develop screening strategies to reduce its incidence and mortality. Colorectal adenomas have been clearly considered preneoplastic lesions due to their potential malignant transformation via the adenoma-carcinoma sequence. Com a resultat de les politiques i estratègies de col·laboració entre la universitat de Vic i de l’hospital i de la voluntat de realitzar activitats formatives conjuntes , s’estableix un línia de treball orientada a l’estudi i anàlisi de la situació logística interna actual del laboratori d’anàlisis clíniques de l’Hospital General de Vic. El treball es centra en el procés intern del laboratori i l’abast de l’estudi es troba limitat a les àrees especifiques d’hematologia i coagulació i bioquí… [+]mica. D’aquestes dues àrees el treball realitza un estudi exhaustiu del seu procés intern, identifica les seves activitats i la seva metodologia de treball amb l’objectiu d’elaborar el Value Stream Map de cadascuna de les àrees. Les àrees de Microbiologia, Banc de Sang i Urgències resten fora d’aquest estudi exhaustiu tot i que són presents en el treball per la inevitable interacció que tenen en la globalitat del procés. El treball es centra bàsicament en els processos automatitzats tot i que els processos que es duen a terme en el laboratori són tant automatitzats com manuals. També es limita al sistema productiu intern del laboratori tot i la interacció que té aquest sistema intern amb altres centres productius del sistema com ara són els centres d’atenció primària, els diversos hospitals i centres d’atenció sociosanitària. El laboratori es troba immers en el moment de l’elaboració d’aquest treball en un situació de canvi i millora del seus processos interns que consisteixen principalment en la substitució de part la maquinària actual que obliguen a la definició d’un nou layout i d’una nova distribució de la producció a cada màquina. A nivell extern també s’estan produint millores en el sistema informàtic de gestió que afecten a part del seu procés. L’objectiu del treball és donar visibilitat total al procés de logística interna actual del laboratori, identificant clarament com són i quina seqüència tenen els processos logístics interns i els mètodes de treball actuals, tant de recursos màquina com recursos persona, per poder identificar sota una perspectiva de generació de valor, aquells punts concrets de la logística interna que poden ser millorats en quant a eficiència i productivitat amb l’objectiu que un cop identificats es puguin emprendre accions i/o projectes de millora. El treball finalitza amb un anàlisis final del procés logística interna des d’una òptica Lean. Per fer-ho, identifica aquelles activitats que no aporten valor al procés o MUDA i les classifica en set categories i es realitzen diverses propostes de millora com són la implantació d’un flux continu , anivellat i basat en un concepte pull , identifica activitats que poden ser estandarditzades i/o simplificades i proposa modificacions en les infraestructures físiques per donar major visibilitat al procés. L’aspecte humà del procés es planteja des d’un punt de vist de metodologia, formació, comunicació i aplicació de les 5S.

Objetivo: Se comparó in vitro la acción antimicrobiana de diversas medicaciones intraconducto frente a Enterococcus faecalis y Actinomyces israelii. Material y métodos: para evaluar las zonas de inhibición microbiana se utilizó el test de difusión en agar frente a diversas pastas, incluyendo una pasta con base de metronidazol (Grinazole ®), una pasta con base de dexametasona, tiretrocina, polimixana y neomicina (Septomixine forte ®) y otra de hidróxido de calciio (Calcipulpe ®) y paroclorofenol alcanforado (KRI-3 ®), frente a E. faecalis y A. israelii. Resultados: La acción inhibitoria frente a E. faecalis fue significativamente mayor (p< 0,05) con Septomixine forte ® (30,7 mm) seguido de KRI- 3 ® (22,5 mm), Calcipulpe ® (16,6 mm) y Grinazole ® (11,4 mm). La inhibición microbiana frente a E. israelii fue significativamente mejor con Septomixine forte ® (p< 0,05) (40,1 mm) seguido de KRI-3 ® (36,6 mm), Grinazole ® (7,6 mm) y Calcipulpe ® (6 mm). Conclusión: Septomixine forte ® fue la pasta antibiótica más efectiva frente a E. faecalis y A. israelii comunmente hallados en las infecciones endodóncicas.

A geração de radicais livres, resultado dos processos metabólicos necessários para a produção de energia, causam um grande número de doenças, o que aumenta o interesse pelos compostos antioxidantes presentes em frutos e vegetais e que são responsáveis pela captura destes radicais. Além disso, a aplicação de óleos e extratos de frutos no controle microbiológico justifica o crescimento dos estudos científicos dessas matérias-primas. Nesse contexto o presente trabalho analisou o óleo essencial de diferentes partes do fruto de uvaia (Eugenia pyriformis) em relação à atividade microbiológica, fenóis totais e antioxidantes. O óleo essencial apresentou rendimento maior na casca (1,23 %), seguido pela fração casca com polpa, a fração com menor rendimento foi à semente, sendo quantificados a partir do fruto refrigerado. Os resultados obtidos na microbiologia frente as cepas de Scherichia coli ATCC25922, Staphylococcus aureus ATCC 6538, Pseudomonas aeruginosas ATCC 27 853 e Enterococcus faecalis ATCC99212 foram de ação bacteriostática em todas as cepas testadas, exceto a bactéria P. aeruginosas. Intervalo de inibição superior a 24 h foi observado para o óleo essencial da casca frente a bactéria P. aeruginosas. A quantificação de fenóis totais não foi possível pois os valores médios encontrados foram inferiores ao LQ (5 μg/mL), por espectrofotometria a 760 nm e as atividade antioxidante mostraram-se não significativas nas concentrações analisadas, em todas as frações.

Tumores - sintomas hiperplásicos em plantas - incitados por espécies de Agrobacterium sp. sempre exerceram fascínio sobre fitopatologistas desde o início do Século XX, quando Erwin Smith e colaboradores demonstraram serem eles de etiologia bacteriana. No início, imaginava-se que os tumores eram decorrentes de alterações hormonais na planta provocadas pela bactéria. Contudo, até recentemente, a microbiologia e a biologia molecular não eram suficientemente avançadas para que os cientistas pudessem compreender e deduzir a forma através da qual o patógeno incitava os tumores. Demorou quase um século para que se deslindassem os complexos mecanismos bioquímicos, genéticos e fisiológicos através dos quais o patógeno transforma a planta, inserindo no genoma desta uma região de seu megaplasmídeo de modo a criar para si mesmo um nicho ecológico específico. Neste trabalho é apresentada uma súmula histórica da evolução do conhecimento a respeito, das características genômicas do plasmídeo Ti, dos eventos e requerimentos atinentes ao processo infectivo bem como é discutida a dinâmica da transformação da planta pelo patógeno.

Objetivou-se, com este trabalho, avaliar a eficiência de isolados de rizobactérias sobre o enraizamento e crescimento de clones de eucalipto sob diferentes condições de propagação clonal. Os resultados evidenciaram que os incrementos em enraizamento e em biomassa radicular variaram de acordo com o isolado de rizobactéria e o clone de eucalipto, não sendo observado efeito deletério em nenhuma das combinações testadas. O ganho médio no enraizamento e biomassa radicular foi de 20,4 e 73,0%, respectivamente. Para enraizamento, o isolado mais promissor foi o S2 e na biomassa radicular, os isolados S1 e S2.

RESUMO A inoculação de micro-organismos solubilizadores de fosfato, em conjunto ou não com fungos micorrízicos, pode ser alternativa para a redução dos custos de produção de eucalipto por meio da diminuição dos gastos com fertilizantes fosfatados. Dessa forma, objetivou-se com este trabalho isolar e caracterizar bactérias solubilizadoras de fosfato da rizosfera de Eucalyptussp., visando à sua coinoculação com fungos ectomicorrízicos. Entre os 24 isolados de bactérias originários do solo rizosférico de eucalipto, 12 são do filo γ-Proteobacteria e pertencentes à família Enterobacteriaceae (Enterobacter, Kluyvera e Klebsiella); 9 do filo β-Proteobacteria (Burkholderia); 1 do filo Actinobacteria (Curtobacterium); e 1 do filo Firmicutes (Enterococcus). O índice de solubilização de fosfato, calculado dividindo-se o diâmetro do halo de clareamento pelo diâmetro da colônia, variou de 0 a 11, sendo Enterococcus avium a espécie com o maior potencial de solubilização de CaHPO4in vitro. A produção de acidez em meio glicose-extrato de levedura pelos isolados bacterianos rizosféricos foi significativa, no entanto a capacidade de solubilização de CaHPO4 não se correlacionou com a acidificação do meio. Alguns isolados bacterianos promoveram forte inibição do crescimento de Pisolithus sp., isolado H4111, enquanto outros não causaram esse fenômeno. Os isolados RE 56 (Enterococcus avium), RE 41 (Burkholderia cepacea), RE 52 e RE 30 (ambos Burkholderia pyrrocinia) foram aqueles que apresentaram maior potencial para utilização em experimentos de coinoculação com fungos ectomicorrízicos.
