Activation of pyruvate dehydrogenase (PDH), which converts pyruvate into acetyl-CoA, is accomplished by a pair of specific phosphatases (PDP 1 & 2). A cross-sectional study investigating the effect of aerobic capacity on PDP activity and expression found that: 1) PDP activity and PDP! protein expression were positively correlated with most aerobic capacity measures in males (n=lS), but not females (n=12); 2) only males showed a positive correlation between PDP activity and PDPl protein expression (r=0.47; p=O.05), indicating that the increase in PDP activity in males is largely explained by increased PDPl protein expression, but that females rely on another level for PDP activity regulation; and 3) PDP} and Ela protein expression increase in unison when expressed relative to the E2 core. These data suggest that with increased aerobic capacity there is an increased capacity for carbohydrate oxidation through PDH, via El a, and an increased ability to activate PDH, via PDP, when exercising maximally.

Evidence suggests that children with developmental coordination disorder (DCD) have lower levels of cardiorespiratory fitness (CRF) compared to children without the condition. However, these studies were restricted to field-based methods in order to predict V02 peak in the determination of CRF. Such field tests have been criticised for their ability to provide a valid prediction of V02 peak and vulnerability to psychological aspects in children with DCD, such as low perceived adequacy toward physical activity. Moreover, the contribution of physical activity to the variance in V02 peak between the two groups is unknown. The purpose of our study was to determine the mediating role of physical activity and perceived adequacy towards physical activity on V02 peak in children with significant motor impairments. This prospective case-control design involved 122 (age 12-13 years) children with significant motor impairments (n=61) and healthy matched controls (n=61) based on age, gender and school location. Participants had been previously assessed for motor proficiency and classified as a probable DCD (p-DCD) or healthy control using the movement ABC test. V02 peak was measured by a progressive exercise test on a cycle ergometer. Perceived adequacy was measured using a 7 -item subscale from Children's Selfperception of Adequacy and Predilection for Physical Activity scale. Physical activity was monitored for seven days with the Actical® accelerometer. Children with p-DCD had significantly lower V02 peak (48.76±7.2 ml/ffm/min; p:50.05) compared to controls (53.12±8.2 ml/ffm/min), even after correcting for fat free mass. Regression analysis demonstrated that perceived adequacy and physical activity were significant mediators in the relationship between p-DCD and V02 peak. In conclusion, using a stringent laboratory assessment, the results of the current study verify the findings of earlier studies, adding low CRF to the list of health consequences associated with DCD. It seems that when testing for CRF in this population, there is a need to consider the psychological barriers associated with their condition. Moreover, strategies to increase physical activity in children with DCD may result in improvement in their CRF.

The primary objectives of the present study were 1) to examine the relationship between health-enhancing physical activity (HEPA) and well-being across the previous day and 2) to examine the role of basic psychological need satisfaction as a potential mediator of the HEPA – well-being relationship. Participants (N = 203) were a convenience sample of undergraduate students with data collected cross sectionally. HEPA was generally associated with well-being (r‟s ranged from .18 to .62). Multiple mediation analyses supported psychological need satisfaction as mechanisms underpinning the HEPA – well- being relationship. Subsequent analyses demonstrated that effort put forth in HEPA activities, as opposed to frequency or duration, uniquely predicted well-being. The role of effort was further highlighted in the multiple mediation analyses. As such future research may wish to investigate the utility of a HEPA program that facilitates effortful engagement and fulfillment of basic psychological needs.

Introduction: Developmental coordination disorder (DCD) is a prevalent condition characterized by poor motor proficiency that interferes with a child‟s activities of daily living. Children with DCD often experience compromised health-related fitness components such as cardiorespiratory fitness (CRF). Purpose: To better understand the physical activity and fitness characteristics of children with probable DCD (pDCD), with a particular focus on CRF. Specifically: (1) to present a synopsis of current literature; (2) to determine the longitudinal trajectories of CRF; (3) to compare the submaximal CRF of children with and without pDCD. Methods: A comprehensive, systematic literature review was conducted of the recent available data on fitness and physical activity and pDCD (Chapter 2). This review provided the background for the other two studies included in this thesis. In Chapter 3, a prospective cohort design was used to assess how CRF in children with pDCD changes over time (56 months) relative to a group of typically developing controls. Using a nested-case control design, 63 subjects with pDCD and 63 matched controls from the larger sample were recruited to participate in the lab-based component of the study (Chapter 4). In this investigation CRF was examined using the oxygen cost of work (VO2) during an incremental test on a cycle ergometer. Results: The literature review showed that fitness parameters, including CRF and physical activity levels, were consistently reduced in children with pDCD. Chapter 3 demonstrated that the difference in CRF between children with pDCD and typically developing children is substantial, and that it tends to increase over time. Results from VO2 assessments showed that children with pDCD utilized more oxygen to sustain the same submaximal workloads compared to typically developing children. Conclusions: Findings from this thesis have made several important contributions to our understanding of children with pDCD. Since differences in CRF between children with and without pDCD tend to worsen over time, this adds to the argument that interventions intended to improve CRF may be appropriate for children with motor difficulties. This thesis also presented the first evidence suggesting that DCD involves higher energy expenditure, and could help explain why children with pDCD perform poorly on tasks requiring CRF.

Objective: To identify the association of low physical activity (PA) participation in children with various motor performances (MP) and to establish the impact of social competence (SC). Methods: Sixth grade children from PHAST study at Brock University (n=1958; 50.53% males) had MP test results from Bruininks-Oseretsky Test of Motor Proficiency, Participation Questionnaire (PQ) used for PA and Harter Social Competence Scale for self-perceived SC. Comparative tests, multiple and logistic regressions were performed. Results: Significant differences in PQ measures in MP quartiles and SCs. MP and SC are independent predictors of PA (p<.05) except with SES on free play activity, making MP not significant. Lower MP increased the odds of low total PA and organized sport participation but not for free play activities (OR~1). Higher SC reduced the risk of low participation in all PA measures. Conclusions: SC improves PA participation, including free play and organized sports, despite the child’s MP.

The objective of this study was to examine the effectiveness of a 12 week weight loss intervention in a commercial fitness centre on body mass index (BMI), moderate to vigorous physical activity (MVPA) and behavioural regulations consistent with Organismic Integration Theory (OIT, Deci & Ryan, 2002). The intervention group received weekly coaching sessions and bi-weekly seminars designed to increase MVPA and improve dietary intake. The results of the mixed model analyses of variance showed a significant within-subjects main effect for BMI (F = 3.57, p = .04). Changes in MVPA were not observed over time or between conditions. Changes in behavioural regulations congruent with OIT (Deci & Ryan, 2002) favoured the intervention condition. Study results indicate that 12 week weight loss challenges in commercial fitness centres may be effective to support the internalization process of exercise behavioural regulations but ineffective at producing sustainable weight loss or behavioural changes.

This research takes a phenomenological approach through narratives to investigate the meanings females attribute to their experiences with physical activity. Nine females across the age ranges of 10-80 were asked to share three meaningful physical activity stories. These experiences could be positive or negative in nature as long as they were reflective of a meaningful experience for the participant. The aim of the research was to explore the types of physical activity experiences that females find meaningful. The results indicate that participants perceived the following to be meaningful within their experience: social support, benefits to personal health and well-being, recognition and acknowledgment, empowerment, body control, early positive experience and identity formation. Participants’ stories offered an enlightened understanding of the meanings active females associate with physical activity across their lifespans that perhaps can assist in formulating and developing strategies to encourage and promote involvement in physical activity.

The purpose of this study was to look at individuals living on a gluten-free diet (GFD), their dietary adherence, PA levels and the reasons why they engage in these lifestyle behaviours consistent with Organismic Integration Theory (Deci & Ryan, 2002). Participants (N = 202; Mage = 42.35 years, SDage = 12.43 years) completed a series of online questionnaires. GFD adherence (74.7%) across the previous week was consistent with existing literature (Dowd et al., 2013), but participant physical activity scores were higher than reported normative values (p = .00; Wilson et al., 2010). Specific motives predicted gluten-free dietary adherence (i.e., integrated and identified regulations) and PA (i.e., intrinsic and identified regulations; p < .05). Findings may be used by health professionals to inform behavioural interventions consistent with OIT (Deci & Ryan, 2002).

Objectif: Examiner le lien entre la participation des parents aux activités physiques (AP) de leur enfant et les habitudes de vie et le statut pondéral de ces derniers. Méthode: Les données proviennent de l’Enquête Sociale et Santé des Enfants et des Adolescent Québécois (1999), comprenant des échantillons représentatifs de jeunes de 9, 13 et 16 ans (n=2511). L’implication des parents est définie par aucun, 1 seul, ou 2 parents faisant de l’AP avec leur enfant ≥1/semaine. Un rappel 7 jours a servi à classer les jeunes selon leur niveau d’AP, soit faible, modéré ou élevé. Le temps d’écran a été défini par: ≤14 vs. >14 heures/semaine. Le statut pondéral a été défini selon les critères de Cole. Résultats: Lorsque les deux parents participent aux AP du jeune, le niveau d’AP des adolescents de 13 (OR 3.89, IC 95%: 1.85-8.18) et 16 ans (OR 3.45, IC 95%: 1.32-9.01) est davantage élevé, et le temps d’écran moindre (OR 2.36, IC 95%: 1.30-4.25) chez ceux de 13 ans. Des analyses secondaires montrent que le lien entre l’implication des parents et le niveau d’AP des jeunes est présent chez les familles biparentales seulement; le lien avec le temps d’écran est présent dans les quartiers sécuritaires seulement. Aucune association n’est observée pour le statut pondéral. Conclusion: Les stratégies de promotion de la santé ciblant la participation des parents aux AP de leurs enfants pourraient réduire le fardeau des maladies chroniques, étant donné l’association favorable entre leur implication et les habitudes de vie des jeunes.

To investigate the role of muscles in the development of adolescent idiopathic scoliosis (AIS), our group was initially interested in Duchenne muscular dystrophy (DMD) diseases where a muscular degeneration often leads to scoliosis. Few years ago the studies with those patients provided interesting results but were obtained only from few patients. To increase that number, the present project was initiated but recruitment of new DMD patients from Marie-Enfant hospital was found impossible. As an alternative, patients with Friedreich’s ataxia (FA) were recruited since they also suffer from a muscular deficiency which often induces a scoliosis. So, 4 FA patients and 4 healthy controls have been chosen to closely match the age, weight and body mass indexes (BMI) of the patients were enrolled in our experiments. As in the previous study, electromyography (EMG) activity of paraspinal muscles were recorded on each side of the spine during three types of contraction at 2 different maximum voluntary contractions (MVC). Moreover, the volume and skinfold thickness of these muscles were determined from ultrasound images (US) in order to facilitate the interpretation of EMG signals recorded on the skin surface. For the 3 FA right scoliotic patients, EMG activity was most of the time larger on the concave side of the deviation. The opposite was found for the 4th one (P4, left scoliosis, 32°) for whom EMG activity was larger on the convex side; it should however be noted that all his signals were of small amplitude. This was associated to a muscle weakness and a large skinfold thickness (12 mm) vs 7 mm for the 3 others. As for the paraspinal muscle volume, it was present on the convex side of P1, P3 and P4 and on the concave side for P2. As for skinfold thickness over this muscle, it was larger on the concave side for P1 and P2 and the opposite for P3 and P4. At the apex of each curve, the volume and skinfold thickness differences were the largest. Although the study covers only a small number of FA patients, the presence of larger EMG signals on the concave side of a spinal deformation is similar to pre-scoliotic DMD patients for whom the deformation is in its initial stage. It thus seems that our FA patients with more EMG activity on their concave side could see progression of their spinal deformation in the coming months in spite of their already important Cobb angle.

A series of novel naphthyridine derivatives 3 and 4 was prepared from substituted pyridine 2 and ketones using ZnCl2 as catalyst under microwave irradiation conditions. All the compounds were evaluated for AChE inhibitory activity and promising compounds 3d, 3e, 4b, and 4g was identified. Representative compounds 3d and 3e were found to show insignificant THLE-2 liver cell viability/toxicity. The binding mode between X-ray crystal structure of human AChE and compounds was studied using molecular docking method and fitness scores were found to be in good correlation with the activity data.

Pollution by toxic compounds is one of the most relevant environmental damages to ecosystems produced by human activity and, therefore, it must be considered in environmental protection and restoration of contaminated sites. According to this purposes, the main goal of this doctoral thesis has been to analyse the impact of several chlorophenols and heavy metals on the microbial communities of two typical Mediterranean soils. The ecological risk concentrations of each pollutant, which have been determined according to respirometric activity and changes in the microbial community composition, and the factors that influence on their effective toxic concentrations (bioavailable pollutants) have been analysed in order to predict their potential impact on different soil ecosystems and provide scientific data for the regulation of the soil protection policies. Moreover, resistant microorganisms with pollutant removal capacities have been isolated from artificially contaminated soil microcosms and tested in axenic cultures, to infer their potential usefulness for bioremediation.

This special report analyses legislative activity in the European Union and coalition formation in the European Parliament (EP) during the first half of the 7th legislative term, 2009-14. Co-decision is now the ordinary legislative procedure, not by name only: it was deployed on 90% of new proposals in 2010 and 86% in 2011, which suggests that the EP is now more influential than ever. There are differences in the degree of empowerment across committees, however. This report looks at the legislative workload of selected committees as an indicator of change in their influence, identifying which of them won and which lost out in terms of the quantity and type of legislation they tackle.
