Telomerase activity is developmentally regulated in mammals. Here we examine telomerase activity in plants, whose development differs in fundamental ways from that of animals. Using a modified version of the telomere repeat amplification protocol (TRAP) assay, we detected an activity in extracts from carrots, cauliflower, soybean, Arabidopsis, and rice with all the characteristics expected for a telomerase synthesizing the plant telomere repeat sequence TTTAGGG. The activity was dependent on RNA and protein components, required dGTP, dATP, and dTTP, but not dCTP, and generated products with a seven nucleotide periodicity. Telomerase activity was abundant in cauliflower meristematic tissue and undifferentiated cells from Arabidopsis, soybean, and carrot suspension cultures, but was low or not detectable in a sampling of differentiated tissues from mature plants. Telomerase from cauliflower meristematic tissues exhibited relaxed DNA sequence requirements, which might reflect the capacity to form telomeres on broken chromosomes in vivo. The dramatic differences in telomerase expression and their correlation with cellular proliferation capacity mirror changes in human telomerase levels during differentiation and immortalization. Hence, telomerase activation appears to be a conserved mechanism involved in conferring long-term proliferation capacity.

Using an event-related functional MRI design, we explored the relative roles of dorsal and ventral prefrontal cortex (PFC) regions during specific components (Encoding, Delay, Response) of a working memory task under different memory-load conditions. In a group analysis, effects of increased memory load were observed only in dorsal PFC in the encoding period. Activity was lateralized to the right hemisphere in the high but not the low memory-load condition. Individual analyses revealed variability in activation patterns across subjects. Regression analyses indicated that one source of variability was subjects’ memory retrieval rate. It was observed that dorsal PFC plays a differentially greater role in information retrieval for slower subjects, possibly because of inefficient retrieval processes or a reduced quality of mnemonic representations. This study supports the idea that dorsal and ventral PFC play different roles in component processes of working memory.

“Catch,” a state where some invertebrate muscles sustain high tension over long periods of time with little energy expenditure (low ATP hydrolysis rate) is similar to the “latch” state of vertebrate smooth muscles. Its induction and release involve Ca2+-dependent phosphatase and cAMP-dependent protein kinase, respectively. Molecular mechanisms for catch remain obscure. Here, we describe a quantitative microscopic in vitro assay reconstituting the catch state with proteins isolated from catch muscles. Thick filaments attached to glass coverslips and pretreated with ≈10−4 M free Ca2+ and soluble muscle proteins bound fluorescently labeled native thin filaments tightly in catch at ≈10−8 M free Ca2+ in the presence of MgATP. At ≈10−4 M free Ca2+, the thin filaments moved at ≈4 μm/s. Addition of cAMP and cAMP-dependent protein kinase at ≈10−8 M free Ca2+ caused their release. Rabbit skeletal muscle F-actin filaments completely reproduced the results obtained with native thin filaments. Binding forces >500 pN/μm between thick and F-actin filaments were measured by glass microneedles, and were sufficient to explain catch tension in vivo. Synthetic filaments of purified myosin and twitchin bound F-actin in catch, showing that other components of native thick filaments such as paramyosin and catchin are not essential. The binding between synthetic thick filaments and F-actin filaments depended on phosphorylation of twitchin but not of myosin. Cosedimentation experiments showed that twitchin did not bind directly to F-actin in catch. These results show that catch is a direct actomyosin interaction regulated by twitchin phosphorylation.

Despite considerable evidence that ethanol can enhance chloride flux through the gamma-aminobutyric acid type A (GABA/A/) receptor-channel complex in several central neuron types, the effect of ethanol on hippocampal GABAergic systems is still controversial. Therefore, we have reevaluated this interaction in hippocampal pyramidal neurons subjected to local monosynaptic activation combined with pharmacological isolation of the various components of excitatory and inhibitory synaptic potentials, using intracellular current- and voltage-clamp recording methods in the hippocampal slice. In accord with our previous findings, we found that ethanol had little effect on compound inhibitory postsynaptic potentials/currents (IPSP/Cs) containing both GABA/A/ and GABA/B/ components. However, after selective pharmacological blockade of the GABA/B/ component of the IPSP (GABA/B/-IPSP/C) by CGP-35348, low concentrations of ethanol (22-66 mM) markedly enhanced the peak amplitude, and especially the area, of the GABA/A/ component (GABA/A/-IPSP/C) in most CA1 pyramidal neurons. Ethanol had no significant effect on the peak amplitude or area of the pharmacologically isolated GABA/B/-inhibitory postsynaptic current (IPSC). These results provide new data showing that activation of GABAB receptors can obscure ethanol enhancement of GABA/A/ receptor function in hippocampus and suggest that similar methods of pharmacological isolation might be applied to other brain regions showing negative or mixed ethanol-GABA interactions.

We have previously shown beneficial effects of dietary protein restriction on transforming growth factor beta (TGF-beta) expression and glomerular matrix accumulation in experimental glomerulonephritis. We hypothesized that these effects result from restriction of dietary L-arginine intake. Arginine is a precursor for three pathways, the products of which are involved in tissue injury and repair: nitric oxide, an effector molecule in inflammatory and immunological tissue injury; polyamines, which are required for DNA synthesis and cell growth; and proline, which is required for collagen production. Rats were fed six isocaloric diets differing in L-arginine and/or total protein content, starting immediately after induction of glomerulonephritis by injection of an antibody reactive to glomerular mesangial cells. Mesangial cell lysis and monocyte/macrophage infiltration did not differ with diet. However, restriction of dietary L-arginine intake, even when total protein intake was normal, resulted in decreased proteinuria, decreased expression of TGF-beta 1 mRNA and TGF-beta 1 protein, and decreased production and deposition of matrix components. L-Arginine, but not D-arginine, supplementation to low protein diets reversed these effects. These results implicate arginine as a key component in the beneficial effects of low protein diet.

Aims. We investigated in detail the system WDS 19312+3607, whose primary is an active M4.5Ve star previously inferred to be young (τ ~ 300–500 Ma) based on its high X-ray luminosity. Methods. We collected intermediate- and low-resolution optical spectra taken with 2 m-class telescopes, photometric data from the B to 8 μm bands, and data for eleven astrometric epochs with a time baseline of over 56 years for the two components in the system, G 125–15 and G 125–14. Results. We derived the M4.5V spectral types of both stars, confirmed their common proper motion, estimated their heliocentric distance and projected physical separation, determined their Galactocentric space velocities, and deduced a most-probable age of older than 600 Ma. We discovered that the primary, G 125–15, is an inflated, double-lined, spectroscopic binary with a short period of photometric variability of 1.6 d, which we associated with orbital synchronisation. The observed X-ray and Hα emissions, photometric variability, and abnormal radius and effective temperature of G 125–15 AB are indicative of strong magnetic activity, possibly because of the rapid rotation. In addition, the estimated projected physical separation between G 125–15 AB and G 125–14 of about 1200 AU ensures that WDS 19312+3607 is one of the widest systems with intermediate M-type primaries. Conclusions. G 125–15 AB is a nearby (d ≈ 26 pc), bright (J ≈ 9.6 mag), active spectroscopic binary with a single proper-motion companion of the same spectral type at a wide separation. They are thus ideal targets for specific follow-ups to investigate wide and close multiplicity or stellar expansion and surface cooling because of the lower convective efficiency.

Gasoline coming from refinery fluid catalytic cracking (FCC) unit is a major contributor to the total commercial grade gasoline pool. The contents of the FCC gasoline are primarily paraffins, naphthenes, olefins, aromatics, and undesirables such as sulfur and sulfur containing compounds in low quantities. The proportions of these components in the FCC gasoline invariable determine its quality as well as the performance of the associated downstream units. The increasing demand for cleaner and lighter fuels significantly influences the need not only for novel processing technologies but also for alternative refinery and petrochemical feedstocks. Current and future clean gasoline requirements include increased isoparaffins contents, reduced olefin contents, reduced aromatics, reduced benzene, and reduced sulfur contents. The present study is aimed at investigating the effect of processing an unconventional refinery feedstock, composed of blend of vacuum gas oil (VGO) and low density polyethylene (LDPE) on FCC full range gasoline yields and compositional spectrum including its paraffins, isoparaffins, olefins, napthenes, and aromatics contents distribution within a range of operating variables of temperature (500–700 °C) and catalyst-feed oil ratio (CFR 5–10) using spent equilibrium FCC Y-zeolite based catalyst in a FCC pilot plant operated at the University of Alicante’s Research Institute of Chemical Process Engineering (RICPE). The coprocessing of the oil-polymer blend led to the production of gasoline with very similar yields and compositions as those obtained from the base oil, albeit, in some cases, the contribution of the feed polymer content as well as the processing variables on the gasoline compositional spectrum were appreciated. Carbon content analysis showed a higher fraction of the C9–C12 compounds at all catalyst rates employed and for both feedstocks. The gasoline’s paraffinicity, olefinicity, and degrees of branching of the paraffins and olefins were also affected in various degrees by the scale of operating severity. In the majority of the cases, the gasoline aromatics tended toward the decrease as the reactor temperature was increased. While the paraffins and iso-paraffins gasoline contents were relatively stable at around 5 % wt, the olefin contents on the other hand generally increased with increase in the FCC reactor temperature.

Commercial off-the-shelf microprocessors are the core of low-cost embedded systems due to their programmability and cost-effectiveness. Recent advances in electronic technologies have allowed remarkable improvements in their performance. However, they have also made microprocessors more susceptible to transient faults induced by radiation. These non-destructive events (soft errors), may cause a microprocessor to produce a wrong computation result or lose control of a system with catastrophic consequences. Therefore, soft error mitigation has become a compulsory requirement for an increasing number of applications, which operate from the space to the ground level. In this context, this paper uses the concept of selective hardening, which is aimed to design reduced-overhead and flexible mitigation techniques. Following this concept, a novel flexible version of the software-based fault recovery technique known as SWIFT-R is proposed. Our approach makes possible to select different registers subsets from the microprocessor register file to be protected on software. Thus, design space is enriched with a wide spectrum of new partially protected versions, which offer more flexibility to designers. This permits to find the best trade-offs between performance, code size, and fault coverage. Three case studies have been developed to show the applicability and flexibility of the proposal.

Human tremor can be defined as a somewhat rhythmic and quick movement of one or more body parts. In some people, it is a symptom of a neurological disorder. From the mathematical point of view, human tremor can be defined as a weighted contribution of different sinusoidal signals which causes oscillations of some parts of the body. This sinusoidal is repeated over time, but its amplitude and frequency change slowly. This is why amplitude and frequency are considered important factors in the tremor characterization, and thus for its diagnosis. In this paper, a tool for the prediagnosis of the human tremor is presented. This tool uses a low cost device (<$40) and allows to compute the main factors of the human tremor accurately. Real cases have been tested using the algorithms developed in this investigation. The patients suffered from different tremor severities, and the components of amplitude and frequency were computed using a series of tests. These additional measures will help the experts to make better diagnoses allowing them to focus on specific stages of the test or get an overview of these tests. From the experimental, we stated that not all tests are valid for every patient to give a diagnosis. Guided by years of experience, the expert will decide which test or set of tests are the most appropriate for a patient.

The studied Cenozoic sedimentary successions consist of deposits from continental/shallow-water to deep-marine environments of the Malaguide Complex (Betic Cordillera) outcropping in the Sierra Espuña area (SE Spain). The aim of this study is to characterize the composition, source area(s) provenance and weathering processes of these sedimentary successions from the pre-orogenic (Paleocene-Early Oligocene) to the syn-orogenic (Late Oligocene-Early Miocene) stage using petrological and geochemical methodologies. The studied sandstones are mainly quartzolithic with abundant metamorphic and sedimentary lithic fragments. In particular, the composition of samples from the pre-orogenic cycle is mainly carbonate with important siliciclastic components that occur within the medium to fine grained arenites. The composition of samples from the syn-orogenic cycle is characterized by a sharp change from carbonate to siliciclastic terms. Thus, the composition of the overall sandstone samples is very heterogeneous and suggests a source area mainly characterized by the Malaguide basement and lower units of the Internal Betic Zone, that partially compose the Mesomediterranean Microplate. The geochemical proxies suggest a provenance mainly from felsic source area with a minor supply from mafic rocks in some samples of the syn-orogenic stage. Furthermore, palaeoweathering indices indicate low to moderate weathering conditions for the sources. The Cenozoic sedimentary successions of the Malaguide Complex played an important role in the geodynamic evolution of the Betic Cordillera that represents the key tectonic element of the western domains of the Mesomediterranean Microplate.

This Policy Brief describes the interaction between three approaches that are effective in driving innovation in low-carbon technologies. Based on that, the author provides four recommendations for making low-carbon technology support smarter. THE ISSUE Combating climate change on the global level will be much easier when abundant low-carbon technologies that are competitive in their cost and capabilities are available. But private companies underinvest in low-carbon innovation because they cannot capture the climate benefits. There are three policies to address this issue: pricing carbon, supporting deployment of as-yet uncompetitive technologies and supporting research and development.

Investigation of the Middle Miocene-Pleistocene succession in cores at ODP Site 817A (Leg 133), drilled on the slope south of the Queensland Plateau, identified the various material fluxes contributing to sedimentation and has determined thereby the paleogeographic events which occurred close to the studied area and influenced these fluxes. To determine proportions of platform origin and of plankton origin of carbonate mud, two reference sediments were collected: (1) back-reef carbonate mud from the Young Reef area (Great Barrier Reef); and (2) Late Miocene chalk from the Loyalty Basin, off New Caledonia. Through their biofacies and mineralogical and geochemical characters, these reference sediments were used to distinguish the proportions of platform and basin components in carbonate muds of 25 core samples from Hole 817A. Two "origin indexes" (i1 and i2) relate the proportion in platform and basin materials. The relative sedimentation rate is inferred from the high-frequency cycles determined by redox intervals in the cores. Bulk carbonate deposited in each core has been calculated in two ways with close results: (1) from calcimetric data available in the Leg 133 preliminary reports (Davies et al., 1991); and (2) from average magnetic susceptibility of cores, a value negatively correlated to the average carbonate content. Vertical changes in sedimentation rates, in carbonate content, in origin indexes and in "linear fluxes" document the evolution of sediment origins from platform carbonates, planktonic carbonates and insoluble material through time. These data are augmented with the variations in organic-matter content through the 817A succession. The observed changes and their interpretation are not modified by compaction, and are compatible with major paleogeographic events including drowning of the Queensland Plateau (Middle Miocene-Early Pliocene) and the renewal of shallow carbonate production, (1) during the Late Pliocene, and (2) from the Early Pleistocene. The birth and growth of the Great Barrier Reef is also recorded from 0.5 Ma by a strengthening of detrital carbonate deposition and possibly by a lack of clay minerals in the 4 upper cores, a response to trapping of terrigenous material behind this barrier. In addition, a maximum of biological silica production is displayed in the Middle Miocene. These changes constrain the time of events and the sequence-stratigraphy framework some components of which are transgression surface, maximum flooding surface and low-stand turbidites. Sedimentation rates and material fluxes show cycles lasting 1.75 Myr. Whatever their origin (climatic and/or eustatic) these cycles affected the planktonic production primarily. The changes also show that major carbonate variations in the deposits are due to a dilution effect by insoluble material (clay, biogenic silica and volcanic glasses) and that plankton productivity, controlling the major fraction of carbonate sedimentation, depends principally on terrigenous supplies, but also on deep-water upwelling. Accuracy of the method is reduced by redeposition, reworking, and probable occurrence of hiatuses.

Volcanic ash layers (1-3 cm thick) are abundant in the North Aoba Basin drill sites but less common at forearc sites. Ash deposited on the forearc slopes is liable to be redistributed as turbidites. In addition, the westerly upper winds also minimize ash-fall on the western (forearc) side of the New Hebrides Island Arc. Crystalline components in the ashes are primarily plagioclase (An90-An44), clinopyroxene (Ca46Mg49Fe5-Ca43Mg33Fe24), olivine (Fo87-Fo62), and titanomagnetite. There are also small amounts of orthopyroxene, magnetite, apatite, and quartz. Glass shards occur in most of the ashes and range in composition from basalt to rhyolite. There is often a variety of glass compositions within a single ash layer. One explanation for this is that the rate of accumulation of ash from several different eruptions or eruptive phases exceeded the background sedimentation rate: there may also have been a certain amount of reworking. The high-K and low-K trends previously recognized in volcanic rocks from the New Hebrides Island Arc are clearly represented in the Leg 134 glasses. All of the ashes investigated here are thought to have originated from the Central Chain volcanoes. The source of the high-K group was probably the Central Basin volcanoes of Santa Maria, Aoba, and Ambrym. The lower-K series includes a distinctive group of dacites and is likely to have originated from the Epi-Tongoa-Tongariki sector of the arc where major pyroclastic eruptions, associated with caldera collapse, have occurred during the Holocene, perhaps as recently as 400 yr ago.

Distribution of diatoms, radiolarians, planktonic and benthic foraminifers, and sediment components in fraction >0.125 mm was analyzed in a core obtained from the central Sea of Okhotsk within frameworks of the Russian-German KOMEX Project. The core section characterizes the period 190-350 ka, which corresponds to marine-isotopic stages (MIS) 7 to 10. During glacial MIS 10 and MIS 8, the basin accumulated terrigenous material lacking microfossils or containing them in low abundance, which reflects, along with their composition, heavy sea-ice conditions, suppressed bioproductivity, and bottom environment aggressive toward calcium carbonate. Interglacial MIS 9 was characterized by elevated bioproductivity with accumulation of diatomaceous ooze during the climatic optimum (328 to 320 ka). Water exchange with the Pacific was maximal from 328 to 324 ka ago. Environment became moderate and close to the present-day one at the end of the optimum exhibiting possible existence of a dichothermal layer with substantial amounts of surface Pacific water still flowing into the basin. Similar to interglacial MIS 5e and MIS 1, ''old'' Pacific water determined near-bottom environment in the central Sea of Okhotsk during that period, although influx of terrigenous material was higher, probably reflecting more humid climate of the region. Slight warming marked the terminal MIS 8 (approximately 260 ka ago). Paleoceanographic situation during the interglacial MIS 7 was highly variable: from warm-water to almost glacial. The main climatic optimum of MIS 7 occurred within 220-210 ka, when subsurface stratification increased and the dichothermal layer developed. Bottom environment during the studied time interval, except for the optimum of interglacial MIS 9, resembled those characteristic of glacial periods: actively formed ''young'' Okhotsk water displaced ''old'' Pacific deep water.
