In order to study peptide growth factor action in a three-dimensional cellular environment, aggregating cell cultures prepared from 15-day fetal rat telencephalon were grown in a chemically defined medium and treated during an early developmental stage with either bovine fibroblast growth factor (bFGF) or platelet-derived growth factor (PDGF homodimers AA and BB). A single dose (5-50 ng/ml) of either growth factor given to the cultures on day 3 greatly enhanced the developmental increase of the two glia-specific enzyme activities, 2',3'-cyclic nucleotide 3'-phosphohydrolase (CNP) and glutamine synthetase (GS), whereas it had relatively little effect on total protein and DNA content. Distinct patterns of dose-dependency were found for CNP and GS stimulation. At low concentrations of bFGF (0.5-5 ng/ml) and at all PDGF concentrations applied, the oligodendroglial marker enzyme CNP was the most affected. A relatively small but significant mitogenic effect was observed after treatment with PDGF, particularly at higher concentrations or after repetitive stimulation. The two PDGF homodimers AA and BB were similar in their biological effects and potency. The present results show that under histotypic conditions both growth factors, bFGF and PDGF, promote the maturation rather than the proliferation of immature oligodendrocytes and astrocytes.

Antecedentes: Existe la percepción de que los pacientes con trastorno mental severo (TMS) presentan una mayor prevalencia de factores de riesgo cardiovascular (FRCV) y un menor control de los mismos en comparación con la población general. Objetivos: Conocer la prevalencia de los FRCV y hábitos tóxicos en pacientes con TMS y evaluar el seguimiento por la Atención Primaria (AP) en los que presentan un riesgo vascular elevado. Diseño y metodología: Se realizó un estudio descriptivo de prevalencia y de seguimiento en una muestra representativa de los pacientes con diagnóstico de TMS seguidos en el Centro de salud mental de Mataró. Se registraron los parámetros antropométricos, los FRCV, hábitos tóxicos, perfil lipídico y tratamientos farmacológicos. Se calculó el RCV mediante las tablas de REGICOR. Los pacientes con un RCV & 10% fueron derivados a AP para valorar la estrategia terapéutica. Se evaluó el seguimiento por parte de los especialistas a los 4 meses y en Atención Primaria a los 6-9 meses. Resultados: Se realizaron 112 entrevistas en el centro de Salud Mental de Mataró; 62% eran hombres y 38% mujeres, con una edad media de 43 ± 11,9 años. La obesidad abdominal fue el FRCV más prevalente (62%), le siguió el tabaquismo en un 58,9%, el sedentarismo con un 48,2% y la dislipemia 42%. El 66,8% de los pacientes eran portadores de tratamiento con antipsicóticos atípicos, lo que se asocia con un mayor RCV. Un total de 102 pacientes (91% de la muestra) presentaban uno o más FRCV, 98 de los mismos tenían un RCV & 10 y 4 presentaron un RCV & 10. Se derivaron 6 pacientes a la AP para control de los FRCV, de los que 3 nunca habían sido atendidos en AP. A los 9 meses fueron visitados el 50% de los derivados a AP, y se consiguió reducir el RCV & 10 en un 50% de los que se le hizo la recomendación de seguimiento. Conclusión: Los pacientes con TMS acuden habitualmente a las consultas de AP para el control de los FRCV. La prevalencia de FRCV en pacientes con TMS es superior a la población general. El porcentaje de pacientes con RCV elevado según el REGICOR es bajo.

Background: Evidence of a role of brain-derived neurotrophic factor (BDNF) in the pathophysiology of eating disorders (ED) has been provided by association studies and by murine models. BDNF plasma levels have been found altered in ED and in psychiatric disorders that show comorbidity with ED. Aims: Since the role of BDNF levels in ED-related psychopathological symptoms has not been tested, we investigatedthe correlation of BDNF plasma levels with the Symptom Checklist 90 Revised (SCL-90R) questionnaire in a total of 78 ED patients. Methods: BDNF levels, measured bythe enzyme-linked immunoassay system, and SCL-90R questionnaire, were assessed in a total of 78 ED patients. The relationship between BDNF levels and SCL-90R scales was calculated using a general linear model. Results: BDNF plasma levels correlated with the Global Severity Index and the Positive Symptom Distress Index global scales and five of the nine subscales in the anorexia nervosa patients. BDNF plasma levels were able to explain, in the case of the Psychoticism subscale, up to 17% of the variability (p = 0.006). Conclusion: Our data suggest that BDNF levels could be involved in the severity of the disease through the modulation of psychopathological traits that are associated with the ED phenotype.

Huntington's disease (HD) is a monogenic neurodegenerative disease that affects the efferent neurons of the striatum. The protracted evolution of the pathology over 15 to 20 years, after clinical onset in adulthood, underscores the potential of therapeutic tools that would aim at protecting striatal neurons. Proteins with neuroprotective effects in the adult brain have been identified, among them ciliary neurotrophic factor (CNTF), which protected striatal neurons in animal models of HD. Accordingly, we have carried out a phase I study evaluating the safety of intracerebral administration of this protein in subjects with HD, using a device formed by a semipermeable membrane encapsulating a BHK cell line engineered to synthesize CNTF. Six subjects with stage 1 or 2 HD had one capsule implanted into the right lateral ventricle; the capsule was retrieved and exchanged for a new one every 6 months, over a total period of 2 years. No sign of CNTF-induced toxicity was observed; however, depression occurred in three subjects after removal of the last capsule, which may have correlated with the lack of any future therapeutic option. All retrieved capsules were intact but contained variable numbers of surviving cells, and CNTF release was low in 13 of 24 cases. Improvements in electrophysiological results were observed, and were correlated with capsules releasing the largest amount of CNTF. This phase I study shows the safety, feasibility, and tolerability of this gene therapy procedure. Heterogeneous cell survival, however, stresses the need for improving the technique.

The production of extracellular soluble proteins was studied in serum-free aggregating cell cultures of fetal rat telencephalon labeled on culture day 7 with a mixture of radioactive amino acid precursors. Cultures treated continuously with epidermal growth factor (EGF; 20 ng/ml) showed a generally increased protein secretion and a particularly enhanced production of a few distinct extracellular proteins. The time lag of this response after an initial dose of EGF (25 ng/ml) on day 7 was 48 h. The total macromolecular radioactivity that accumulated within 96 h of labeling in the media of EGF-treated cultures was 175% of untreated controls, whereas no difference was found in the proportions of intracellular amino acid incorporation. Cultures which received a single dose of EGF (25 ng/ml) on day 1 showed still a greatly increased protein secretion on day 7. Prevention of extracellular protein accumulation by reducing the initial cell number and increasing the rate of media changes did not affect the EGF-induced stimulation of the two glial enzymes, glutamine synthetase and 2',3'-cyclic nucleotide 3'-phosphohydrolase. The results suggest that both the increased production of extracellular proteins and the enhanced development of glial enzymatic activities reflect the stimulated phenotypic expression of EGF-sensitive brain cells.

We show that the welfare of a representative consumer can be related to observable aggregatedata. To a first order, the change in welfare is summarized by (the present value of) the Solowproductivity residual and by the growth rate of the capital stock per capita. We also show thatproductivity and the capital stock suffice to calculate differences in welfare across countries, withboth variables computed as log level deviations from a reference country. These results hold forarbitrary production technology, regardless of the degree of product market competition, and applyto open economies as well if TFP is constructed using absorption rather than GDP as the measureof output. They require that TFP be constructed using prices and quantities as perceived byconsumers. Thus, factor shares need to be calculated using after-tax wages and rental rates, andwill typically sum to less than one. We apply these results to calculate welfare gaps and growthrates in a sample of developed countries for which high-quality TFP and capital data are available.We find that under realistic scenarios the United Kingdom and Spain had the highest growth ratesof welfare over our sample period of 1985-2005, but the United States had the highest level ofwelfare.

Este trabalho tem por finalidade analisar a relação existente entre a comunicação interna e a motivação dos colaboradores dos TACV Cabo Verde Airlines. A comunicação interna é uma ferramenta fundamental para a elevação da motivação de colaboradores de qualquer organização. O sistema organizacional se viabiliza graças ao sistema de comunicação nele existente, pois permitirá sua continua realimentação e sua sobrevivência. Assim esse sistema é fundamental para o processamento das funções administrativas internas. (KUNSCH, 2003). Esta pesquisa cujo tema é Comunicação Interna como Factor de Motivação, tem como objectivo principal, descobrir a influência da comunicação interna na motivação dos colaboradores dos TACV. Propôs-se ainda descobrir o grau de motivação dos colaboradores bem como analisar a importância da comunicação para a elevação da motivação dos mesmos. De forma a esclarecer a hipótese de que a comunicação interna pode ou não influenciar a motivação, será feito o estudo com base na teoria dos Dois Factores de Frederick Herzeberg. Foi nessa óptica que se despertou o interesse por esse tema no sentido de dar algum contributo para a organização em estudo. Para a aquisição de informações, foi aplicado questionários do tipo misto, fechado, aplicados aos 42 colaboradores com a idade compreendia entre os 21 e os 55, tendo em conta a amostra estratificada de 10% sobre o total dos colaboradores de cinco departamentos escolhidos. Os departamentos escolhidos para o estudo foram: Comercial, Financeiro, Handling, Manutenção e Operações. No questionário destacou-se primeiramente com a identificação e caracterização dos colaboradores mediante sexo, idade, profissão e nível de escolaridade, seguido de, questões relacionadas com a comunicação, relacionamentos, valorização, imagem da empresa e finalmente, questões motivacionais. As informações recolhidas foram primeiramente introduzidas no Excel e depois no programa SPSS como forma de serem melhores analisadas e obter resultados satisfatórios capazes de responder as inquietações surgidas anteriormente, se de facto a comunicação veiculada nos TACV, influencia a motivação dos colaboradores. Após as análises saiu a conclusão de que realmente a comunicação veiculada nos TACV é satisfatória e influencia a motivação dos colaboradores.

Serum-free aggregating cell cultures of fetal rat telencephalon treated with low doses (0.5 nM) of epidermal growth factor (EGF) showed a small, transient increase in DNA synthesis but no significant changes in total DNA and protein content. By contrast, treatment with high doses (13 nM) of EGF caused a marked stimulation of DNA synthesis as well as a net increase in DNA and protein content. The expression of the astrocyte-specific enzyme, glutamine synthetase, was greatly enhanced both at low and at high EGF concentrations. These results suggest that at low concentration EGF stimulates exclusively the differentiation of astrocytes, whereas at high concentration, EGF has also a mitogenic effect. Nonproliferating astrocytes in cultures treated with 0.4 microM 1-beta-D-arabinofuranosyl-cytosine were refractory to EGF treatment, indicating that their responsiveness to EGF is cell cycle-dependent. Binding studies using a crude membrane fraction of 5-day cultures showed a homogeneous population of EGF binding sites (Kd approximately equal to 2.6 nM). Specific EGF binding sites were found also in non-proliferating (and nonresponsive) cultures, although they showed slightly reduced affinity and binding capacity. This finding suggests that the cell cycle-dependent control of astroglial responsiveness to EGF does not occur at the receptor level. However, it was found that the specific EGF binding sites disappear with progressive cellular differentiation.

The Stages of Change Readiness and Treatment Eagerness Scale (SOCRATES), a 19-item instrument developed to assess readiness to change alcohol use among individuals presenting for specialized alcohol treatment, has been used in various populations and settings. Its factor structure and concurrent validity has been described for specialized alcohol treatment settings and primary care. The purpose of this study was to determine the factor structure and concurrent validity of the SOCRATES among medical inpatients with unhealthy alcohol use not seeking help for specialized alcohol treatment. The subjects were 337 medical inpatients with unhealthy alcohol use, identified during their hospital stay. Most of them had alcohol dependence (76%). We performed an Alpha Factor Analysis (AFA) and Principal Component Analysis (PCA) of the 19 SOCRATES items, and forced 3 factors and 2 components, in order to replicate findings from Miller and Tonigan (Miller, W. R., & Tonigan, J. S., (1996). Assessing drinkers' motivations for change: The Stages of Change Readiness and Treatment Eagerness Scale (SOCRATES). Psychology of Addictive Behavior, 10, 81-89.) and Maisto et al. (Maisto, S. A., Conigliaro, J., McNeil, M., Kraemer, K., O'Connor, M., & Kelley, M. E., (1999). Factor structure of the SOCRATES in a sample of primary care patients. Addictive Behavior, 24(6), 879-892.). Our analysis supported the view that the 2 component solution proposed by Maisto et al. (Maisto, S.A., Conigliaro, J., McNeil, M., Kraemer, K., O'Connor, M., & Kelley, M.E., (1999). Factor structure of the SOCRATES in a sample of primary care patients. Addictive Behavior, 24(6), 879-892.) is more appropriate for our data than the 3 factor solution proposed by Miller and Tonigan (Miller, W. R., & Tonigan, J. S., (1996). Assessing drinkers' motivations for change: The Stages of Change Readiness and Treatment Eagerness Scale (SOCRATES). Psychology of Addictive Behavior, 10, 81-89.). The first component measured Perception of Problems and was more strongly correlated with severity of alcohol-related consequences, presence of alcohol dependence, and alcohol consumption levels (average number of drinks per day and total number of binge drinking days over the past 30 days) compared to the second component measuring Taking Action. Our findings support the view that the SOCRATES is comprised of two important readiness constructs in general medical patients identified by screening.

En un total de 28,265 individuos de Sardina se estudiaron el factor de condición, el coeficiente alimentario y los ítems alimentarios entre 1975 y 1986. Los cambios observados comparados a la situación “normal” en 1980. Durante El Niño 1976-77 la sardina ingirió más fitoplancton que zooplancton, aumentando el coeficiente alimentario pero decreciendo el factor condición.

O presente trabalho tem como objectivo mostrar a importância que a Gestão económica de Stock tem no processo de redução de custo, por via da implementação de um modelo. E para isso far-se-á um estudo de caso numa empresa inserida no ramo de transformação de pescado – FRESCOMAR. A metodologia adoptada para alcançar os objectivos propostos passou pela pesquisa bibliográfica, recolha de dados e de informação relevante junto da empresa e tratamento dos dados numa folha de Excel. O modelo de gestão de Stock a ser implementado depende essencialmente da componente procura, onde que para uma empresa com procura constante utiliza-se os modelos determinísticos e para uma com procura aleatória utiliza-se os modelos estocásticos (Nível de Encomenda e Revisão Cíclica). Nos modelos determinísticos temos 4 modelos diferenciados pela forma de reposição e a admissibilidade da procura. Com as informações recolhidas junto da empresa em estudo escolhemos implementar o modelo de reposição não instantânea com ruptura não permitida e obtivemos a optimização do Stock com níveis reduzidos de produção e consequentemente houve uma diminuição do custo. Com resultados obtiveram-se ainda valores da Quantidade Óptima a ser Produzida, do Stock Máximo e do Custo Total Óptimo por unidade de tempo. Devido a diversidade de produtos propomos à empresa a utilização da análise ABC para classificar os produtos reagrupando-os em classes A, B e C, conforme o peso no consumo de Stock.

Leaf-litter amount as a factor in the structure of a ponerine ants community (Hymenoptera, Formicidae, Ponerinae) in an eastern Amazonian rainforest, Brazil. Leaf-litter may be an important factor in structuring ponerine ant communities (Hymenoptera, Formicidae, Ponerinae) in tropical rainforests. We specifically examined how leaf-litter affects the structure of a ponerine ant community in primary Amazonian rainforest sites at the Ferreira Penna Scientific Station, Pará, Brazil. A total of 53 species belonging to eight genera of three ponerine tribes were collected with mini-Winkler extractors. The amount of leaf-litter positively affected the abundance and richness of the ponerine ant community, and also influenced species composition. Nearby samples often had low species similarity, especially when adjacent samples differed in the amount of leaf-litter. Leaf-litter availability in Amazonian primary forests is a key factor for distribution of ground-dwelling ponerine species, even at small scales.

Nesting substrata, colony success and productivity of the wasp Mischocyttarus cassununga. Colonies of the wasp Mischocyttarus cassununga (von Ihering, 1903) are easily found in urban areas. However, in spite of the massive presence of this species in cities, little is known about its nesting habits, colony success and productivity. The present study aimed at answering the following questions: What are the substrates used for nesting by M. cassununga? What is the main foundation strategy adopted by M. cassununga in urban areas: a solitary female or associative foundation? Is there a relationship between foundation strategies and colony success? Is the total number of cells per nest related to the number of adults produced? The study was conducted in Juiz de Fora, southeastern Brazil, from December 2006 to November 2007. Nesting in man-made substrata seems to be a common strategy in M. cassununga (90.9%), with preference for nest building with a horizontal comb facing north. The colonies were established mainly by groups of foundresses (67.6%), with a success of 84%. The number of brood cells produced per nest was 71.74 ± 45.25 (18-203), and it was positively correlated with the number of adults produced. Hence, we can say that the nests founded by M. cassununga are located mainly in man-made substrata and mostly founded by a group of females. The cell reuse behavior increases the number of adults produced, as it optimizes foraging. These characteristics together with its behavior and nesting habits promote the success of this species in thriving in urban environments.

Undocumented migrants, meaning migrants without a legal residency permit, come to Geneva from countries with high tuberculosis (TB) incidence. We estimate here whether being undocumented is a determinant of TB, independently of origin. Cross-sectional study including undocumented migrants in a TB screening program in 2002; results were compared to 12,904 age and frequency matched participants in a general TB screening program conducted at various workplaces in Geneva, Switzerland from 1992 to 2002. A total of 206 undocumented migrants (36% male, 64% female, mean age 37.8 years (SD 11.8), 82.5% from Latin America) participated in the TB screening program. Compared to legal residents, undocumented migrants had an adjusted OR for TB-related fibrotic signs of 1.7 (95% CI 0.8;3.7). The OR of TB-related fibrotic signs for Latin American (vs. other) origin was 2.7 (95% CI 1.6;4.7) among legal residents and 5.5 (95% CI 2.8;10.8) among undocumented migrants. Chest X-ray screening identified a higher proportion of TB-related fibrotic signs among Latin Americans, independently of their residency status.

Rapport de synthèse : Introduction : La perfusion isolée de membre (isolated limb perfusion, ou ILP) par TNF-alpha et melphalan, utilisés en association, est une stratégie de prise en charge chirurgicale des sarcomes non opérables des extrémités. Elle a été en partie développée au CHUV dans les années 1990, sous l'impulsion du Professeur F. Lejeune, ancien Chef du Service d'oncologie médicale (CePO). Les résultats des 31 premiers patients ont été publiés en 2000 dans l'European Journal of Surgical Oncology. Les données dans la littérature manquant sur les résultats à long terme, nous avons revu tous les patients traités au CHUV depuis 1992 pour tenter des de déterminer ces résultats à long terme, en se focalisant sur l'efficacité du traitement, symbolisée par le taux de sauvetage de membres, autrement condamnés à l'amputation ou à une chirurgie mutilante. Matériel et méthode : Etude rétrospective. De 1992 à mars 2006, 51 patients ont été traités par ILP dans notre institution, certains à deux reprises (58 ILP au total). Quatre-vingt-huit pour cent présentaient un sarcome de haut grade de malignité, et 84% une tumeur localement avancée (T2b NO Mo ou plus). Résultats : Le follow-up moyen est de 38.9 mois (4-159, médiane 22 mois), on note 21 % de complications immédiates et 23% de complications tardives ou chroniques. Une réponse complète (nécrose totale ou disparition de la tumeur) a été observée dans 25% des cas, une réponse partielle (>50% de nécrose ou de diminution de taille tumorale) dans 42%, une stabilité de la maladie dans 14% et une progression tumorale dans 14%. Un traitement adjuvant a été entrepris dans 31 % des cas, une résection des résidus tumoraux a pu être effectuée chez 65% des patients. On note un taux de récidive locale de 35% (après 20,3 mois en moyenne) et un taux de récidive à distance de 45% (après 13,4 mois en moyenne). Le disease-free survival est de 14,9 mois et la survie à 5 ans de 43,5%. Le taux d'amputation s'élève à 24%. Conclusion : La perfusion isolée de membre est un traitement grevé d'un taux élevé de complications, mais il peut étre entrepris dans les sarcomes les plus sévères avec un succès significatif. Ainsi, dans notre série, une chirurgie mutilante (en général l'amputation) a pu être épargnée à 76% des patients.
