O presente artigo trata de avaliar em perspectiva histórica, algumas falhas na condução da política de Defesa no Brasil, fulcrais para se compreender a continuidade de um quadro de indevida participação militar e de ausência de controle civil na elaboração e implementação da mencionada pasta, incompatíveis com um contexto de governabilidade democrática. A despeito de algumas iniciativas no sentido de regulamentar os limites de atuação das Forças Armadas para que as mesmas não extrapolem suas funções e missões essenciais, e para que a política de Defesa seja revestida de um verniz público e civil - a exemplo da criação do Ministério da Defesa e da publicação da Estratégia Nacional de Defesa -, prevalece uma situação de fragilidade institucional e de vazio de poder público propícios para que a vontade militar permaneça conferindo o tom da Defesa nacional.

As condições ideais dos frangos de corte no momento do abate devem ser conhecidas a fim de possibilitar a produção de carne de excelente qualidade, uma vez que diversos fatores pré e pós-abate estão envolvidos na qualidade final. em condições normais de abate e processamento, a retirada de ração é feita de 6 a 8 horas antes da apanha das aves, resultando em um período total de jejum de 8 a 12 horas antes do abate, para esvaziar o intestino e com isso minimizar a contaminação no abatedouro. A escalda, depena e evisceração são pontos importantes de contaminação cruzada no abatedouro devido à grande quantidade de microorganismos aderidos às penas, pele e patas das aves e ao rompimento das vísceras durante a evisceração. Entretanto, a desidratação da carcaça começa imediatamente após o início do jejum. Períodos prolongados de jejum podem afetar o pH das diversas partes do intestino, aumentando a presença de Salmonella e outros microorganismos patogênicos. Além disso, determinam uma maior contaminação pela bile, e são, subjetivamente, associados à fragilidade dos intestinos durante a evisceração mecânica. Portanto, os esquemas de processamento devem ser estabelecidos levando-se em conta a integridade e o esvaziamento do intestino e da vesícula biliar, bem como a desidratação e os seus efeitos sobre o bem estar das aves, contaminação da carcaça e qualidade da carne. Como alguns efeitos do jejum ainda não são bem conhecidos, sugerem-se pesquisas nas seguintes áreas: definir o tempo ótimo de jejum para atender o bem estar das aves, minimizar a contaminação e otimizar os parâmetros de qualidade de carcaça; estudar os efeitos de períodos prolongados de jejum sobre o pH e a colonização do papo, pró-ventrículo, moela, intestino delgado, intestino grosso e cecos por enterobactérias, como Salmonella, por exemplo; efeito do jejum sobre o tamanho e cor do fígado. O resultado esperado é um aumento na qualidade final dos produtos aliado a uma redução nas perdas e no custo de produção.

We studied cytogenetically 48 male patients with Duchenne or Becker muscular dystrophy. All of them showed normal X chromosomes. Fragility of Xp21 was investigated in 1400 G-banded chromosomes of 28 patients and only one break was observed at this band (0.07%). This low frequency of breakage excludes Xp21 as a fragile site in these patients.

The cytogenetic study of 182 river buffalo (Bubalus bubalis L., 2n=50) of Murrah, Mediterranean and Jaffarabadi breeds, from the State of São Paulo, was carried out to characterize their chromosomes and to detect possible chromosomal abnormalities. The karyotypes were indistinguishable with conventional staining as well as with C and replication R banding techniques. In about 44% of the sample (8 males and 72 females), an X marker chromosome due to a fragile site was shown. The frequency of metaphases expressing the fragility site on the X was highly variable, from 2.86 to 41.03%. In females, the fragile site, rarely appeared on both X chromosomes. Most of the metaphases showed only 1 marker chromosome. In R-banded metaphases using 5-bromodeoxyuridine (BrdU) treatment, it corresponded in general to the late replicating X chromosome. No correlation between the X fragile site and altered phenotype was found. Structural and numerical chromosome rearrangements were ruled out in the present sample of buffalo. Although insects lack the adaptive immune response of the mammalians, they manifest effective innate immune responses that include both cellular and humoral components. Cellular responses are mediated by hemocytes and Immoral responses include the activation of proteolytic cascades that initiate many events, including NO production. In this work, we determined NO production in Chrysomya megaccphala hemolymph and hemocytes after yeast inoculation. Assays were performed with non-infected controls (NIL), saline-injected larvae (SIL) or larvae injected with Saccharomyces cerevisiae (YIL). The hemolymph of injected groups was collected 0.5, 1, 2, 4, 12, 24 or 48 h post-injection. NO levels in SIL were comparable to those measured in NIL until 12 h, which might be considered the basal production, increasing at 24 and 48 h post-injection, probably in response to the increased larval fragility after cuticle rupture. YIL exhibited significantly higher levels of NO than were found in other groups, peaking at 24 h. L-NAME and EDTA caused a significant reduction of NO production in YIL at this time, suggesting the activity of a Ca2+ -dependent NOS. Plasmatocytes and granular cells phagocytosed the yeasts. Plasmatocytes initiated the nodule formation and granular cells were the only hemocyte type to produce NO. These results permit us to conclude that yeasts induced augmented NO production in C. megacephala hemolymph and granular cells are the hemocyte type involved with the generation of this molecule. The efficacy of sodium heparin and tripotassium EDTA as anticoagulant and their effect on the hematological parameters of tambaqui (Colossoma macropomum) were evaluated in this study. Ten fish weighing 384.9 +/- 85.71 g and measuring 27.90 +/- 2.10 cm were used for heparin 5.000 IU, heparin 100 IU and K3EDTA 10% evaluation. Clotting inhibition after 10 h, erythrogram and osmotic fragility of erythrocytes were observed. The results were submitted to variance analysis and means compared by Tukey test (P < 0.05). Heparin 5.000 IU, heparin 100 IU and K3EDTA 10% were effective in preventing coagulation for more than 10 h. However, tripotassium EDTA caused hemolysis since first moments. In erythrogram there was no difference (P > 0.05) in erythrocyte count, hematocrit, hemoglobin and MCHC. on the other hand, an increase in MCV (P < 0.05) in samples kept with K3EDTA10% was observed. This anticoagulant provoked a significant increase (P < 0.01) in the osmotic fragility of erythrocytes when compared to pure heparin, diluted heparin and the control group. Heparin as an anticoagulant is more appropriate for tambaqui since it was effective in preventing coagulation for more than 10 h, without causing hemolysis, changes on hematological parameters or osmotic fragility of erythrocytes.

Dapsone (DDS) is useful in the treatment of a number of inflammatory conditions which are characterized by neutrophil infiltration. It is the drug of choice for the treatment of leprosy and prophylaxis of malaria. Haematological side effects of methaemoglobinaemia and haemolysis have been long recognized. However, the frequency and severity of these side effects in patients already treated with DDS as a single drug or as part of a multidrug therapy (MDT) have not been well documented. We report herein an investigation of the effect of dapsone long-term treatment on the haematological and biochemical alterations in leprosy patients undergoing dapsone as a single drug (DDS group) or as part of a multidrug therapy in combination with rifampin and clofazimine (MDT group). Methaemoglobinaemia and haemolytic anaemia were the principal side effects observed. Reticulocytes were found to be elevated (> 1.5%) in 90% of the patients. Heinz bodies were also detected (6.6% of the patients). The osmotic fragility test showed a reduction in cell resistance and in the evaluation of white cells a severe eosinophilia was found. Hepatic, pancreatic and renal evaluation by the determination of biochemical parameters showed rare and occasional changes of no apparent clinical significance. We conclude that haematological side effects of dapsone are significant even at doses currently used to treat leprosy (100 mg/day) and that rifampin and clofazimine do not increase the incidence of these effects during long-term treatment.

A comparative study was made regarding the clinical and hematological alterations caused by isolates of Babesia bigemina from southeastern, northeastern and northern Brazil in experimentally infected Nelore calves. Eighteen calves between 7 and 9 months of age, without antibodies against Babesia sp and raised free from ticks, were used. Three animals were previously inoculated with 2.0×109 parasitic erythrocytes (PE) for each stabilate. The other 15 calves were subdivided into three groups, with five animals each, that were subinoculated with 1.0×1010 PE of the respective isolates. The clinical and hematological alterations were evaluated by the determination of parasitaemia, haemogram, plasmatic fibrinogen, reticulocyte count, descriptive analysis of the bone marrow and erythrocytic osmotic fragility, for 30 days, totalizing seven moments of observation. The follow-up of the immunological response by the indirect fluorescent antibody test was carried out daily until the 10th day after inoculation (DAI) and after that, on the 15th, 20th, 25th and 30th DAI. A mild clinical manifestation of the disease was observed. The laboratory findings revealed low levels of parasitaemia; decrease of the erythrogram values; absence of reticulocytes, initial decrease in the total count of leukocytes, neutrophils and lymphocytes with a posterior elevation of the number of these cells; hypercellularity of the erythrocytic series and decrease of the myeloid: erythroid relation which was more accentuated between the 8th and 12th DAI, and an increase of the erythrocytic osmotic fragility in the groups inoculated with the Southeast and Northeast isolates. None of the three isolates of B. bigemina gave rise to the clinical characteristic form of the disease, although they induced an humoral immune response.

Rearrangements involving chromosomes 2 and 22 were described not only as acquired abnormalities in a variety of human neoplasias but also in the constitutional karyotype suggesting the existence of a greater fragility in some specific regions in these chromosomes. Patients with DiGeorge and Velocardiofacial syndromes have a deletion on 22q11 leading to haploinsufficiency for one or more gene(s). We report a patient with velocardiofacial syndrome in which cytogenetic and fluorescence in situ hybridization analysis showed a rare t(2;22) and deletion in the 22q11 region. © 2007 Lippincott Williams & Wilkins, Inc.

Aging is associated with decline in muscle mass and strength and reduced bone density. Age-related bone loss is a primary factor in osteoporosis and all individuals are potential candidates for osteoporosis because bone loss with aging occurs in men and women, but less studied in men. To examine the appropriateness of hindlimb elevation, by tail suspension as a model for diminished mechanical loading, and to determine the influence of age on bone responsiveness to skeletal unloading, we use dual X ray absorptiometry (DXA) and digital radiographic images to analyze the response of the femur from mature rats to biomechanical loads. Femurs from male Wistar rats (9-mo-old) were scanned using DXA and DIGORA and measures obtained in ephipyseal and diaphyseal regions of interest. The mechanical testing was divided into compression load to fracture the head and a three-point bending load to fracture the femur middiaphysis. In femoral epiphysis from hindlimb unload (HU), animals presented significant differences between mineral bone content and density assessed by DXA. Detailed regions of femoral epiphysis (head, throcanteric fossa, throcanter and metaphysis) presented significant lower values from radiographic density. Only compressive load necessary to fracture the femoral head neck was also significantly diminished in HU animals. Disuse induced, as in elderly patients, deterioration of the trabecular bone architecture with critical effect on bone fragility. Rats with 21 days of hindlimb unloading can simulate disuse, suggesting that certain sub-regions of their aging bones are more susceptible to fracture, while other, i.e. diaphyses, are not.

Soil use for the development of activities as agriculture and livestock has been causing great alterations in the environment, mainly when these are practiced intensively, disrespecting the fragility and aptitude of the natural resources. Therefore, it is essential that the planning of the agricultural activities is done, taking into consideration the several environmental criteria involved in the decision-making process. Thus, this study aimed to analyze the agricultural aptitude classes of lands from a watershed through geographical information system (GIS). The Arroio Ajuricaba watershed is located in the Municipality of Marechal Cândido Rondon - PR among the coordinates UTM 787309m E and 793892m E; 7275026m N and 7281310m N, in the Spindle 21, presenting an area of 1681ha. Soil maps, in semi detail scale, was the digital cartographic base used. The results allowed to conclude that 42.41% of the basin presented a good aptitude for farming in handling level of high technology (handling level C), regular aptitude for B, and restricted aptitude for A [class 1(a)bC] and that 12% of the area had regular aptitude for farming in the three handling levels (class 1abc). The watershed lands (14.24%) presented regular aptitude for farming in handling level C, restricted aptitude for B, and inapt for A [class of aptitude 2(b)c]; 15.85% presented good aptitude for planted pasture (class 4P) and 12.21% were considered without aptitude for agricultural use (class 6). We can say that 71.94% of the lands has aptitude for farming, although they present different degrees of limitations that request different treatments for its conservation.
