Le facteur de transcription Growth factor independent 1b (GFI1b) est impliqué à différents stades dans la régulation de l'hématopoïèse. Il est notamment fortement exprimé dans les cellules souches hématopoïétiques et au cours de la différenciation des cellules des lignées érythroïdes et mégacaryocytaires. Grâce à un modèle de délétion conditionnelle chez la souris par le système CreLox, nous avons montré que l'absence de GFI1b entraîne une prolifération de cellules mégacaryocytaires incapables de produire des plaquettes. Notre étude ne permet pas de confirmer formellement la prolifération des cellules souches dans la moelle osseuse précédemment observée par notre équipe dans un autre modèle murin. En revanche, les souris GFI1b "knockout" présentent une augmentation des cellules souches circulantes dans le sang périphérique. Une analyse moléculaire préliminaire montre que GFI1b pourrait influer sur la régulation par le cycle circadien de la mobilisation de ces cellules dans le sang. Finalement, notre étude des effets biologiques de la délétion de GFI1b dans le compartiment hématopoïétique des souris adultes nous a permis de définir de façon plus précise le rôle de GFI1b dans l'hématopoïèse et de confirmer son rôle majeur dans la régulation de la mégacaryopoïèse et la production de plaquettes matures.

In zebrafish, germ cells are responsible for transmitting the genetic information from one generation to the next. During the first cleavages of zebrafish embryonic development, a specialized part of the cytoplasm known as germ plasm, is responsible of committing four blastomeres to become the progenitors of all germ cells in the forming embryo. Much is known about how the germ plasm is spatially distributed in early stages of primordial germ cell development, a process described to be dependant on microtubules and actin. However, little is known about how the material is inherited after it reorganizes into a perinuclear location, or how is the symmetrical distribution regulated in order to ensure proper inheritance of the material by both daughter cells. It is also not clear whether there is a controlled mechanism that regulates the number of granules inherited by the daughter cells, or whether it is a random process. We describe the distribution of germ plasm material from 4hpf to 24hpf in zebrafish primordial germ cells using Vasa protein as marker. Vasa positive material appears to be conglomerate into 3 to 4 big spherical structures at 4hpf. While development progresses, these big structures become smaller perinuclear granules that reach a total number of approximately 30 at 24hpf. We investigated how this transformation occurs and how the minus-end microtubule dependent motor protein Dynein plays a role in this process. Additionally, we describe specific colocalization of microtubules and perinuclear granules during interphase and more interestingly, during all different stages of cell division. We show that distribution of granules follow what seems to be a regulated distribution: during cells division, daughter cells inherit an equal number of granules. We propose that due to the permanent colocalization of microtubular structures with germinal granules during interphase and cell division, a coordinated mechanism between these structures may ensure proper distribution of the material among daughter cells. Furthermore, we show that exposure to the microtubule-depolymerizing drug nocodazole leads to disassembly of the germ cell nuclear lamin matrix, chromatin condensation, and fusion of granules to a big conglomerate, revealing dependence of granular distribution on microtubules and proper nuclear structure.

Fundamentos. La eficacia de paclitaxel junto con rhG-CSF en la movilización de progenitores hematopoyéticos, se ha probada en pacientes hematológicos. Farmacogenéticamente el paclitaxel presenta una alta variabilidad inter-individual. Los genes CYP2C8 y ABCB1 involucrados en su metabolismo y transporte podrían afectar dicha variabilidad inter-individual. Objetivo. Evaluar en una cohorte retrospectiva de pacientes sometidos a TASPE, el efecto de algunos polimorfismos de nucleótido simple (del gen CYP2C8 y del gen ABCB1) sobre la eficacia en la movilización y toxicidad hematológica inducida por del paclitaxel. Materiales y Métodos. Un grupo de 107 pacientes recibieron paclitaxel y rhG-CSF como esquema movilizador. Los polimorfismos genotipados fueron para los genes ABCB1 rs1045642 A>G, ABCB1 rs2032582 C>A, ABCB1 rs2032582 C>T, CYP2C8 rs10509681 C>T, y CYP2C8 rs11572080 A>G. Resultados. El uso de paclitaxel logró éxito movilizador en más del 80% de los pacientes con linfomas o mieloma (p=0,0021), pero no lo fue en la leucemia aguda. En pacientes con mieloma la variable G>rs1045642 del gen ABCB1 se asoció con mala movilización (p= 0,018) y mayor toxicidad hematológica (p= 0,034). El alelo C>rs10509681 del gen CYP2C8 se relacionó con mayor toxicidad en pacientes con linfoma (p= 0,045) y mieloma múltiple (p=0,042), y portadores del alelo TT en homocigosis presentaron una mayor toxicidad hematológica comparada con los portadores CC o CT (p= 0,027). Conclusión. Este estudio sugiere que los SNPs de las variables alélicas analizadas en los genes CYP2C8 y ABCB1 en algunos grupos de pacientes inciden en la capacidad movilizadora y afectan el grado de toxicidad hematológica.

In this research we explore several aspects about the quality of life of youth from factors such as self-esteem, locus of control, the social perceived support, values, etc. In this article we study the correlations among factors the influence the values and vital satisfaction of adolescents aged 12-16 years old. We analyze data from boys and girls and from their parents, we explore the relationship between the factors and we explore the concordances and discrepancies of their answers

The results presented in this paper are from a research using a questionnaire about activities and evaluations of boys and girls in relation to different audio-visual media (television, computer, videoconsole, educative CD-Roms, Internet and computer and console games). Results show us that children information about any audio-visual media is systematically overestimated by parents. Generally, the media with more negative concordances is video-games, the one with more positive concordances is the computer and the one with more discrepancies between generations is TV

Aquest estudi insisteix en la importància de l’educació sanitària dirigida als pares per tal d’assegurar l’eficàcia del tractament a administrar i consolidar els coneixements i habilitats del progenitors per tal de que aquests puguin dur a terme les tècniques requerides de forma adequada. Destacar la necessitat de conscienciar a tots els professionals de la salut, sobretot els que passen més temps amb els familiars, de la importància que té la seva competència com educador en l’àmbit de la salut. Evidenciar la importància de fer una avaluació continuada dels coneixements i les habilitats adquirides pels cuidadors principals, sobre com dur a terme les tècniques del rentat de nas i administració de la medicació inhalada amb cambra i mascareta, per detectar-ne els possibles errors i poder posar-hi mesures correctores per pal·liar-los. Constatar la utilitat de preguntar i esbrinar “in situ” què saben els progenitors sobre les tècniques a avaluar (rentat de nas i administració de la medicació inhalada amb cambra i mascareta), abans de donar qualsevol explicació o iniciar l’educació sanitària per detectar les necessitats específiques de cada pare o mare i poder individualitzar el procés educatiu

Des de la perspectiva psicosocial s'analitzen alguns aspectes de les interaccions entre progenitors i fills/es adolescents entre 12 i 16 anys en relació a l'ús del telèfon mòbil en el marc de la societat acceleradament canviant. S'ha emprat un mètode plural o multimètode que s'ha articulat en dues fases d'investigació, una quantitativa i una qualitativa, per l'estudi dels fenòmens socials complexes com ho són les relacions intergeneracionals. Els resultats posen de manifest que existeixen interaccions entre les actituds dels adolescents i dels progenitors en l'ús del telèfon mòbil. També apunten que les tecnologies audiovisuals com el mòbil estan mediatitzant les relacions intergeneracionals. És a dir, no només els adolescents utilitzen molt més les tecnologies que els adults, sinó que a més sobre el context social que té impacte aquest ús és fonamentalment amb els amics. Cal un major apropament intergeneracional pel que fa a mantenir converses entre adults i adolescents al voltant dels temes que els motiven i interessen, com és el cas de les TICs.

Limb girdle muscular dystrophy type 2H (LGMD2H) is an inherited autosomal recessive disease of skeletal muscle caused by a mutation in the TRIM32 gene. Currently its pathogenesis is entirely unclear. Typically the regeneration process of adult skeletal muscle during growth or following injury is controlled by a tissue specific stem cell population termed satellite cells. Given that TRIM32 regulates the fate of mammalian neural progenitor cells through controlling their differentiation, we asked whether TRIM32 could also be essential for the regulation of myogenic stem cells. Here we demonstrate for the first time that TRIM32 is expressed in the skeletal muscle stem cell lineage of adult mice, and that in the absence of TRIM32, myogenic differentiation is disrupted. Moreover, we show that the ubiquitin ligase TRIM32 controls this process through the regulation of c-Myc, a similar mechanism to that previously observed in neural progenitors. Importantly we show that loss of TRIM32 function induces a LGMD2H-like phenotype and strongly affects muscle regeneration in vivo. Our studies implicate that the loss of TRIM32 results in dysfunctional muscle stem cells which could contribute to the development of LGMD2H.

Losses of cultivated cocoa (Theobroma cacao L.) due to diseases and continued depletion of forests that harbour the wild progenitors of the crop make ex situ conservation of cocoa germplasm of paramount importance. In order to enhance security of in situ germplasm collections, 2-3 mm floral-derived secondary somatic embryos were cryopreserved by vitrification. This work demonstrates the most uncomplicated clonal cocoa cryopreservation. Optimal post-cryostorage survival (74.5%) was achieved by 5 d preculture of SSEs on 0.5 M sucrose medium followed by 60 min dehydration in cold PVS2. To minimise free radical related cryo-injury, cation sources were removed from the embryo development solution and/or the recovery medium, the former treatment resulting in a significant benefit. After optimisation with cocoa genotype AMAZ 15, the same protocol was effective across all five additional cocoa genotypes tested. For the multiplication of clones, embryos regenerated following cryopreservation were used as explant sources, and vitrification was found to maintain their embryogenic potential.

Neural differentiation of embryonic stem cells (ESCs) requires coordinated repression of the pluripotency regulatory program and reciprocal activation of the neurogenic regulatory program. Upon neural induction, ESCs rapidly repress expression of pluripotency genes followed by staged activation of neural progenitor and differentiated neuronal and glial genes. The transcriptional factors that underlie maintenance of pluripotency are partially characterized whereas those underlying neural induction are much less explored, and the factors that coordinate these two developmental programs are completely unknown. One transcription factor, REST (repressor element 1 silencing transcription factor), has been linked with terminal differentiation of neural progenitors and more recently, and controversially, with control of pluripotency. Here, we show that in the absence of REST, coordination of pluripotency and neural induction is lost and there is a resultant delay in repression of pluripotency genes and a precocious activation of both neural progenitor and differentiated neuronal and glial genes. Furthermore, we show that REST is not required for production of radial glia-like progenitors but is required for their subsequent maintenance and differentiation into neurons, oligodendrocytes, and astrocytes. We propose that REST acts as a regulatory hub that coordinates timely repression of pluripotency with neural induction and neural differentiation.

Proneural genes such as Ascl1 are known to promote cell cycle exit and neuronal differentiation when expressed in neural progenitor cells. The mechanisms by which proneural genes activate neurogenesis--and, in particular, the genes that they regulate--however, are mostly unknown. We performed a genome-wide characterization of the transcriptional targets of Ascl1 in the embryonic brain and in neural stem cell cultures by location analysis and expression profiling of embryos overexpressing or mutant for Ascl1. The wide range of molecular and cellular functions represented among these targets suggests that Ascl1 directly controls the specification of neural progenitors as well as the later steps of neuronal differentiation and neurite outgrowth. Surprisingly, Ascl1 also regulates the expression of a large number of genes involved in cell cycle progression, including canonical cell cycle regulators and oncogenic transcription factors. Mutational analysis in the embryonic brain and manipulation of Ascl1 activity in neural stem cell cultures revealed that Ascl1 is indeed required for normal proliferation of neural progenitors. This study identified a novel and unexpected activity of the proneural gene Ascl1, and revealed a direct molecular link between the phase of expansion of neural progenitors and the subsequent phases of cell cycle exit and neuronal differentiation.

Acute intermittent porphyria is an inborn error of haem synthesis which is transmitted as a dominant character with variable phenotypic expression. The disorder is caused by a partial deficiency of porphobilinogen deaminase in all tissues so far studied. The nature of the enzymatic deficiency of porphobilinogen deaminase in haemolysates from patients with acute intermittent porphyria was examined by the use of monospecific antibody probes. In affected heterozygotes from three British pedigrees of diverse ancestry, the catalytic deficiency of porphobilinogen deaminase was accompanied by diminished enzyme protein, as determined by radial immunodiffusion. No evidence of functionally attenuated enzyme was demonstrable by kinetic studies. The molecular forms of the residual enzyme were investigated in red cell extracts and in lysed preparations of reticulocytes by a sensitive Western blotting procedure. This revealed the presence of reduced amounts of porphobilinogen deaminase polypeptide co-migrating with wild type enzyme (Mr approximately 40,000), and no evidence of variant forms in situ. The studies show that porphobilinogen deaminase deficiency in acute intermittent porphyria is commonly associated with a CRM-phenotype. The residual activity under these circumstances is thus related to expression of a single normal allele, since sensitive techniques detected neither aberrant nor degraded forms of the enzyme in erythroid tissues.

Schwann cells (SCs) are the supporting cells of the peripheral nervous system and originate from the neural crest. They play a unique role in the regeneration of injured peripheral nerves and have themselves a highly unstable phenotype as demonstrated by their unexpectedly broad differentiation potential. Thus, SCs can be considered as dormant, multipotent neural crest-derived progenitors or stem cells. Upon injury they de-differentiate via cellular reprogramming, re-enter the cell cycle and participate in the regeneration of the nerve. Here we describe a protocol for efficient generation of neurospheres from intact adult rat and murine sciatic nerve without the need of experimental in vivo pre-degeneration of the nerve prior to Schwann cell isolation. After isolation and removal of the connective tissue, the nerves are initially plated on poly-D-lysine coated cell culture plates followed by migration of the cells up to 80% confluence and a subsequent switch to serum-free medium leading to formation of multipotent neurospheres. In this context, migration of SCs from the isolated nerve, followed by serum-free cultivation of isolated SCs as neurospheres mimics the injury and reprograms fully differentiated SCs into a multipotent, neural crest-derived stem cell phenotype. This protocol allows reproducible generation of multipotent Schwann cell-derived neurospheres from sciatic nerve through cellular reprogramming by culture, potentially marking a starting point for future detailed investigations of the de-differentiation process.

Neural crest-derived stem cells (NCSCs) from the embryonic peripheral nervous system (PNS) can be reprogrammed in neurosphere (NS) culture to rNCSCs that produce central nervous system (CNS) progeny, including myelinating oligodendrocytes. Using global gene expression analysis we now demonstrate that rNCSCs completely lose their previous PNS characteristics and acquire the identity of neural stem cells derived from embryonic spinal cord. Reprogramming proceeds rapidly and results in a homogenous population of Olig2-, Sox3-, and Lex-positive CNS stem cells. Low-level expression of pluripotency inducing genes Oct4, Nanog, and Klf4 argues against a transient pluripotent state during reprogramming. The acquisition of CNS properties is prevented in the presence of BMP4 (BMP NCSCs) as shown by marker gene expression and the potential to produce PNS neurons and glia. In addition, genes characteristic for mesenchymal and perivascular progenitors are expressed, which suggests that BMP NCSCs are directed toward a pericyte progenitor/mesenchymal stem cell (MSC) fate. Adult NCSCs from mouse palate, an easily accessible source of adult NCSCs, display strikingly similar properties. They do not generate cells with CNS characteristics but lose the neural crest markers Sox10 and p75 and produce MSC-like cells. These findings show that embryonic NCSCs acquire a full CNS identity in NS culture. In contrast, MSC-like cells are generated from BMP NCSCs and pNCSCs, which reveals that postmigratory NCSCs are a source for MSC-like cells up to the adult stage.
