The developing seeds of Actinodaphne hookeri were investigated to delineate their ability to synthesize large amounts of trilaurin. Until 88 days after flowering the embryos contained 71% neutral lipids (NL) and 29% phospholipids (PL) and both these components contained C-16:0, C-18:0, C-18:2, and C-18:3 as the major fatty acids (FA). At 102 days after flowering the seeds began to accumulate triacylglycerols (TAG) and to synthesize lauric acid (C-12:0). By 165 days after flowering, when the seeds were mature, they contained about 99% NL and 1% FL. At this stage the TAG contained exclusively C-12:0, while the PL consisted of long-chain fatty acids (LCFA) only. Leaf lipids in contrast did not contain any C-12:0. Experiments on [1-C-14]acetate incorporation into developing seed slices showed that at 88 days after flowering only 4% of the label was in TAG, 1% in diacylglycerols (DAG), and 87% in FL. One hundred two days after flowering seeds incorporated only 2% of the label into TAG, 30% into DAG, and 64% into FL. In contrast at 114 days after flowering 71% of the label was incorporated into TAG, 25% into DAG, and only 2% into FL. Analysis of labeled FA revealed that up to 102 days after flowering it was incorporated only into LCFA, whereas at 114 days after flowering it was incorporated exclusively into C-12:0. Furthermore, 67% of the label in PL at 114 days after flowering was found to be dilaurylglycerophosphate. Analysis of the label in DAG at this stage showed that it was essentially in dilaurin species. These observations indicate the induction of enzymes of Kennedy pathway for the specific synthesis of trilaurin at about 114 days after flowering, Homogenates of seeds (114 days after flowering) incubated with labeled FA in the presence of glycerol-3-phosphate and coenzymes A and ATP incorporated 84% of C-12:0 and 61% of C-14:0, but not C-16:0, C-18:2, and C-18:3, into TAG. In contrast the LCFA were incorporated preferentially into FL. It is concluded that, between 102 and 114 days after flowering, a switch occurs in A. hookeri for the synthesis of C-12:0 and trilaurin which is tissue specific. Since the seed synthesizes exclusively C-12:0 at 114 days after flowering onwards and incorporates specifically into TAG, this system appears to be ideal for identifying the enzymes responsible for medium-chain fatty acid as well as trilaurin synthesis and for exploiting them for genetic engineering. Acyl carrier proteins (ACP) were purified to homogeneity in the active form from developing seeds of pisa (Actinodaphne hookeri) which synthesizes exclusively trilaurin and from ground nut (Arachis hypogaea) which synthesizes triacylglycerols containing long chain fatty acids. Two major isoforms of ACPs were purified from developing pisa seeds using DEAE-cellulose, Superose-6 FPLC and C-4 reversed phase HPLC chromatographic methods. In contrast, only a single form of ACP was present in ground nut seeds which was purified by anion-exchange and activated thiol-Sepharose 4B affinity chromatography. The two isoforms of ACPs from pisa showed nearly the same specific activity of 6,706 and 7,175 pmol per min per mg protein while ground nut ACP showed a specific activity of 3,893 pmol per min per mg protein when assayed using E. coli acyl-ACP synthetase and [1-C-14]palmitic acid. When compared with E. coli ACP, the purified ACPs from both the seeds showed considerable difference in their mobility in native PAGE, but showed similar mobility in SDS-PAGE under reducing conditions. In the absence of reducing agents formation of dimers was quite prominent. The ACPs from both the seed sources were acid- and heat-stable. The major isoform of pisa seed ACP and the ground nut ACP contain 91 amino acids with M(r) 11,616 and 1,228 respectively. However, there is significant variation in their amino acid composition. A comparision of the amino acid sequence in the N-terminal region of pisa and ground nut seed ACPs showed considerable homology between themselves and with other plant ACPs but not with E. coli ACP.

XVII foi, para Portugal, um tempo de lutas, tanto na Europa como no Brasil e na África. Foi, ainda, um período de reorganização jurídica e econômica da sociedade.

El presente estudio trata en primera instancia de acercar el informe de educación internacional “Pisa” al lector, detallando de forma clara en qué consiste el mismo. Una vez que se ha introducido la temática y se han expuesto las nociones básicas, el objetivo principal consistirá en el análisis del informe Pisa del 2012 a nivel nacional a través del programa estadístico R (R Core Team 2014), de dónde se obtendrán los datos necesarios para el mismo análisis. Por un lado, se contrastarán los resultados del informe atendiendo a una serie de características, para posteriormente localizar y reflejar las diferencias en los resultados en el territorio español. Por otro lado, se examinará la posible relación entre estos resultados y una serie de variables externas al informe que se estimen relevantes. De modo que lo que finalmente se logra con este estudio es, un recorrido completo dentro del área de evaluación educativa.

Este trabalho investigou a formação científica brasileira a partir dos dados do PISA 2006. O PISA - Programa Internacional de Avaliação dos Estudantes - é um programa internacional de avaliação comparada, aplicado a uma amostra de estudantes de 15 anos de idade. O ensino adequado de ciências estimula o raciocínio lógico e a curiosidade, ajuda a formar cidadãos mais aptos a enfrentar os desafios da sociedade contemporânea e fortalece a democracia, dando à população, em geral, melhores condições para participar dos debates cada vez mais comuns sobre temas científicos que afetam nosso cotidiano. Partindo da importância e pertinência de se discutir a formação científica brasileira e fazendo uso dos dados do PISA 2006, buscou-se responder a três questões: (a) qual a percepção dos alunos brasileiros que participaram do PISA 2006 sobre Ciências? (b) como o Brasil se situa no contexto internacional no que se refere às competências científicas priorizadas por esta avaliação? (c) que características dos estudantes estão associadas aos seus resultados no teste de Ciências aplicado pelo PISA 2006? Para a realização do estudo, conduzimos nossas análises baseando-nos em duas lógicas principais: descrição (estatística univariada e bivariada) e explicação (teste de modelos de regressão linear). Os resultados indicam que os alunos brasileiros tem relativo interesse pela Ciência, no entanto apresentam uma visão bastante estereotipada e demonstram não associá-la a si. Aparentemente, a ciência tem relação com fenômenos que estão muito além da compreensão e configura-se em algo abstrato que não está diretamente ligado aos alunos brasileiros. Estes apontam a escola como principal fonte de aprendizagem de tópicos relacionados a Ciência, porém, a escola parece não converter o potencial interesse dos alunos por Ciências em letramento científico. O Brasil ocupa a 52 posição entre os 57 países participantes e está alocado no nível mais baixo de desempenho do PISA 2006. Entre os principais aspectos que impactam significativamente os resultados dos alunos brasileiros no programa estão a Rede de Ensino, o Atraso Escolar, o Nível Socioeconômico e o Nível de Escolaridade dos Pais.

El presente trabajo tiene como objeto acercar en primera instancia el Informe PISA 2012 al lector, de modo que se pueda hacer una idea clara del mismo. A su vez se presentan los resultados globales y en función del sexo para los 65 países participantes en PISA. A continuación se estudian diferentes indicadores de desarrollo y se analiza su relación con el rendimiento académico observado en PISA 2012. Las operaciones econométricas pertinentes se realizan a través del programa Gretel.

The putative 5-HT6 receptor agonist ST1936 has been shown to increase extracellular dopamine (DA) in the n.accumbens (NAc) Shell and in the medial prefrontal cortex (PFCX). These observations suggest that 5-HT6 receptors modulate DA transmission in mesolimbic and mesocortical terminal DA areas. To investigate the behavioral counterpart of this interaction I studied in rats the effect of 5-HT6 receptor blockade on cocaine stimulated overflow of DA in dialysates from the PFCX and from the NAc Shell and on cocaine i.v. selfadministration. Pretreatment with the 5-HT6 antagonist SB271046 reduced cocaine-induced increase of dialysate DA in the NAc Shell but not in the PFCX and impaired i.v. cocaine selfadministration. These suggest that 5-HT6 receptors play a role in cocaine reinforcement via their facilitatore interaction with DA projections to the NAc Shell. This 5-HT/DA interaction might provide the basis for a new pharmacotherapeutic strategy of cocaine addiction. Caffeine is one of the psychoactive substances most widely used as adulterant in illicit drugs, such as cocaine. Animal studies have demonstrated that caffeine is able to potentiate cocaine actions, although the enhancement of the cocaine reinforcing property by caffeine is less reported, and the results depend on the paradigms and experimental protocols used. In the present study I examined the ability of caffeine to enhance the motivational and rewarding properties of cocaine using the intravenous self-administration paradigm in rats. Additionally, the role of caffeine as a primer cue during extinction was evaluated. To this end, we assessed in naïve rats: 1) the ability of the combination of cocaine (0,125 mg/kg/infusion) and caffeine (0,0625 mg/kg/infusion) to maintain self-administration in fixed ratio (FR) and progressive ratio (PR) schedules of reinforcement compared with cocaine and caffeine alone; 2) the effect of caffeine in the maintenance of responding in the animals exposed to the combination of the drugs during cocaine extinction. Cocaine and the combination of cocaine and caffeine were self-administered on a FR and PR schedules of reinforcement, and the responding for the combination of the drugs was higher than cocaine alone. Caffeine was not reliably self-administered, but was able to maintain a drug-seeking behavior in rats previously exposed to cocaine plus caffeine. These findings suggest that the presence of caffeine enhances the reinforcing effects of cocaine and the motivational value of the drug. Our results highlight the role of active adulterants commonly used in illicit street drugs.
