Brazil is one of the largest mango producers and the third largest mango exporter worldwide. Irradiation treatment and its commercial feasibility have been studied in our country to make it possible to develop new markets and, consequently, to compete with the major exporters of mangoes, Mexico and India. This work was designed to compare irradiation treatment with the hot water dip treatment in mangoes cv. Tommy Atkins for export and to verify that the main attributes for acceptance, color and texture, as well as carbohydrate and organic acid contents, were maintained. In this study, the fruit was divided into groups: control, hot water dip-treated (46 degrees C for 90 min), and irradiation-treated at doses of 0.4 kGy and 1.0 kGy. The fruit was stored at low temperature (11 degrees C +/- 2) for 14 days and then at room temperature (23 degrees C +/- 2) until the end of the study. The results indicated that the fruit given a dose of 1.0 kGy remained in a less advanced stage of ripening (stage 3) throughout the storage period, but experienced a greater loss of texture in the beginning of the experiment. It was noted that only the control group had higher levels of citric acid and succinic acid on the last day of the experiment. There were no significant differences in the total sugar content between any treatment groups. Gamma radiation can be used as a quarantine treatment and does not interfere negatively with the quality attributes of mangoes. (C) 2012 Elsevier Ltd. All rights reserved.

Trigeneration systems have been used with advantage in the last years in distributed electricity generation systems as a function of a growth of natural gas pipeline network distribution system, tax incentives, and energy regulation policies. Typically, a trigeneration system is used to produce electrical power simultaneously with supplying heating and cooling load by recovering the combustion products thermal power content that otherwise would be driven to atmosphere. Concerning that, two small scale trigeneration plants have been tested for overall efficiency evaluation and operational comparison. The first system is based on a 30 kW (ISO) natural gas powered microturbine, and the second one uses a 26 kW natural gas powered internal combustion engine coupled to an electrical generator as a prime mover. The stack gases from both machines were directed to a 17.6 kW ammonia-water absorption refrigeration chiller for producing chilled water first and next to a water heat recovery boiler in order to produce hot water. Experimental results are presented along with relevant system operational parameters for appropriate operation including natural gas consumption, net electrical and thermal power production, i.e., hot and cold water production rates, primary energy saving index, and the energy utilization factor over total and partial electrical load operational conditions.

The research activity described in this thesis is focused mainly on the study of finite-element techniques applied to thermo-fluid dynamic problems of plant components and on the study of dynamic simulation techniques applied to integrated building design in order to enhance the energy performance of the building. The first part of this doctorate thesis is a broad dissertation on second law analysis of thermodynamic processes with the purpose of including the issue of the energy efficiency of buildings within a wider cultural context which is usually not considered by professionals in the energy sector. In particular, the first chapter includes, a rigorous scheme for the deduction of the expressions for molar exergy and molar flow exergy of pure chemical fuels. The study shows that molar exergy and molar flow exergy coincide when the temperature and pressure of the fuel are equal to those of the environment in which the combustion reaction takes place. A simple method to determine the Gibbs free energy for non-standard values of the temperature and pressure of the environment is then clarified. For hydrogen, carbon dioxide, and several hydrocarbons, the dependence of the molar exergy on the temperature and relative humidity of the environment is reported, together with an evaluation of molar exergy and molar flow exergy when the temperature and pressure of the fuel are different from those of the environment. As an application of second law analysis, a comparison of the thermodynamic efficiency of a condensing boiler and of a heat pump is also reported. The second chapter presents a study of borehole heat exchangers, that is, a polyethylene piping network buried in the soil which allows a ground-coupled heat pump to exchange heat with the ground. After a brief overview of low-enthalpy geothermal plants, an apparatus designed and assembled by the author to carry out thermal response tests is presented. Data obtained by means of in situ thermal response tests are reported and evaluated by means of a finite-element simulation method, implemented through the software package COMSOL Multyphysics. The simulation method allows the determination of the precise value of the effective thermal properties of the ground and of the grout, which are essential for the design of borehole heat exchangers. In addition to the study of a single plant component, namely the borehole heat exchanger, in the third chapter is presented a thorough process for the plant design of a zero carbon building complex. The plant is composed of: 1) a ground-coupled heat pump system for space heating and cooling, with electricity supplied by photovoltaic solar collectors; 2) air dehumidifiers; 3) thermal solar collectors to match 70% of domestic hot water energy use, and a wood pellet boiler for the remaining domestic hot water energy use and for exceptional winter peaks. This chapter includes the design methodology adopted: 1) dynamic simulation of the building complex with the software package TRNSYS for evaluating the energy requirements of the building complex; 2) ground-coupled heat pumps modelled by means of TRNSYS; and 3) evaluation of the total length of the borehole heat exchanger by an iterative method developed by the author. An economic feasibility and an exergy analysis of the proposed plant, compared with two other plants, are reported. The exergy analysis was performed by considering the embodied energy of the components of each plant and the exergy loss during the functioning of the plants.

In recent years the hot water treatment (HW) represents an effective and safe approach for managing postharvest decay. This study reported the effect of an HW (60°C for 60 s and 45°C for 10 min) on brown rot and blue mould respectively. Peaches was found more thermotolerant compared to apple fruit, otherwise Penicillium expansum was more resistant to heat with respect to Monilinia spp. In semi-commercial and commercial trials, the inhibition of brown rot in naturally infected peaches was higher than 78% after 6 days at 0°C and 3 days at 20°C. Moreover, in laboratory trials a 100% disease incidence reduction was obtained by treating artificially infected peaches at 6-12 h after inoculation revealing a curative effect of HW. The expression levels of some genes were evaluated by qRT-PCR. Specifically, the cell wall genes (β-GAL, PL, PG, PME) showed a general decrease of expression level whereas PAL, CHI, HSP70 and ROS-scavenging genes were induced in treated peaches compared to the control ones. Contrarily, HW applied on artificially infected fruit before the inoculum was found to increase brown rot susceptibility. This aspect might be due to an increase of fruit VOCs emission as revealed by PTR-ToF-MS analysis. In addition a microarray experiment was conducted to analyze molecular mechanisms underneath the apple response to heat. Our results showed a largest amount of induced Heat shock proteins (HSPs), Heat shock cognate proteins (HSCs), Heat shock transcription factors (HSTFs) genes found at 1 and 4 hours from the treatment. Those genes required for the thermotolerance process could be involved in induced resistance response. The hypothesis was confirmed by 30% of blue mold disease reduction in artificially inoculated apple after 1 and 4 hours from the treatment. In order to improve peaches quality and disease management during storage, an innovative tool was also used: Da-meter.

A comprehensive knowledge of cell wallstructure and function throughout the plant kingdom is essential to understanding cell wall evolution. The fundamental understanding of the charophycean green algal cell wall is broadening. The similarities and differences that exist between land plant and algal cell walls provide opportunities to understand plant evolution. A variety of polymers previously associated with higher plants were discovered in the charophycean green algae (CGA), including homogalacturonans, cross-linking glycans, arabinogalactan protein, β-glucans, and cellulose. The cellulose content of CGA cell walls ranged from 6% to 43%, with the higher valuescomparable to that found in the primary cell wall of land plants (20-30%). (1,3)β-glucans were found in the unicellular Chlorokybus atmophyticus, Penium margaritaceum, and Cosmarium turpini, the unbranched filamentous Klebsormidium flaccidum, and the multicellular Chara corallina. The discovery of homogalacturonan in Penium margaritaceum representsthe first confirmation of land plant-type pectinsin desmids and the second rigorous characterization of a pectin polymer from the charophycean algae. Homogalacturonan was also indicated from the basal species Chlorokybus atmophyticus and Klebsormidium flaccidum. There is evidence of branched pectins in Cosmarium turpini and linkage analysis suggests the presence of type I rhamnogalacturonan (RGI). Cross-linking β-glucans are associated with cellulose microfibrils during land plant cell growth, and were found in the cell wall of CGA. The evidence of mixed-linkage glucan (MLG) in the 11 charophytesis both suprising and significant given that MLG was once thought to be specific to some grasses. The organization and structure of Cosmarium turpini and Chara corallina MLG was found to be similar to that of Equisetumspp., whereas the basal species of the CGA, Chlorokybus atmophyticus and Klebsormidium flaccidum, have unique organization of alternating of 3- and 4-linkages. The significance of this result on the evolution of the MLG synthetic pathway has yet to be determined. The extracellular matrix (ECM) of Chlorokybus atmophyticus, Klebsormidium flaccidum, and Spirogyra spp. exhibits significant biochemical diversity, ranging from distinct “land plant” polymers to polysaccharides unique to these algae. The neutral sugar composition of Chlorokybus atmophyticus hot water extract and Spirogyra extracellular polymeric substance (EPS), combined with antibody labeling results, revealed the distinct possibility of an arabinogalactan protein in these organisms. Polysaccharide analysis of Zygnematales (desmid) EPS, indicated a probable range of different EPS backbones and substitution patterns upon the core portions of the molecules. Desmid EPS is predominately composed of a complex matrix of branched, uronic acid containing polysaccharides with ester sulfate substitutions and, as such, has an almost infinite capacity for various hydrogen bonding, hydrophobic interaction and ionic cross-bridging motifs, which characterize their unique function in biofilms. My observations support the hypothesis that members of the CGA represent the phylogenetic line that gave rise to vascular plants and that the primary cell wall of vascular plants many have evolved directly from structures typical of the cell wall of filamentous green algae found in the charophycean green algae.

The effect of three different aging methods (immersion in hot water, freeze–thaw cycles and wet–dry cycles) on the mechanical properties of GRC were studied and compared. Test results showed that immersion in hot water may be an unreliable method for modified GRC formulations, with it being in probability a very harmful procedure. A new aging method, mixing freeze–thaw cycles and wet–dry cycles, seems to be the most accurate simulation of weather conditions that produce a noticeable change in GRC mechanical properties. Future work should be carried out to find a correlation between real weather and the proposed aging method.

ABSTRACT The aim of this thesis is to contribute to the development and use of solar energy as an alternative for producing hot water in the agribusiness sector. Hot water supply is a key issue for a great many agribusinesses and agricultural holdings. Both hot water demand times and required tank water temperature (rTWT) are highly variable, where the demand profile tends to differ from domestic use. Further research is needed on how differences in rTWT influence the performance and feasibility of these systems. The main objective of this thesis is to characterize the performance and test the feasibility of an evacuated tube collector (ETC) solar water heating (SWH) system providing water at a higher temperature than is usual for such systems. The aim is to determine what influence the rTWT has on the system’s energy efficiency, quantify the volume of hot water that the system is capable of supplying at the respective rTWT and establish whether SWH is feasible as a booster supply system for the different analysed rTWTs. To do this, a prototype water heating system has been designed, installed and commissioned and its performance monitored at different rTWTs under real operating conditions. The quantitative results show that a higher rTWT results in a lower energy supply to the tank, where the differences may be greater than 1000 Wh m-2 d-1 from 40 ºC to 80 ºC for insolation values of around 8000 Wh m-2 d-1 (system efficiency ranges from 73% to 56%). The drop in supply is due to lower collector efficiency and greater heat losses from the pipe system. As regards water supplied at the rTWT, the insolation required for the first withdrawal of water to take place is greater at higher rTWTs, where the time between withdrawals increases and the number of withdrawals decreases throughout the day. As rTWT increases, the volume of water supplied at the rTWT decreases due to factors such as lower collector efficiency, pipe system heat losses, energy stored in the water at below the rTWT and more energy being extracted from the system by water heating. For a rTWT of 80 ºC, much of the energy is stored in the tank at below the rTWT at the end of the day. A booster supply system would be required to take advantage of this energy, as overnight tank heat losses may significantly reduce the usable energy available on the following day. It is often not feasible to use the solar system as a single supply system, especially at high rTWTs, as, unlike the supply from the solar heating system which does not produce hot water on many days of the year because insolation is below the required minimum, hot water demand is not seasonal. On the other hand, investment in a solar system as a booster system to meet part of a plant’s heat energy demand is highly recommended. The solar system’s estimated annual usable energy ranges from 1322 kWh m-2 to 1084 kWh m-2. Cost efficiency would be greatest if there were an existing electric boiler, where the payback period would be just a few years —from 5.7 years at 40 ºC to 7.2 years at 80 ºC—. Cost efficiency is also high if there is an existing diesel boiler with payback periods of under 10 years. In a fictitious industry with a demand of 100 kWh day-1 and an existing diesel boiler, the investment in the solar plant would be highly recommended at any rTWT, with a net present value similar to investment costs —12000 € at 80 ºC and 15000 € at 40 ºC— and a payback period of 10 years. The results of this study are potentially very useful for determining the feasibility of using similar systems for meeting the hot water demand of agribusinesses and arable and livestock farms or for other applications demanding water at temperatures not typical of domestic demand (60ºC). Performance and cost efficiency will be determined by the regional insolation, the required water temperature and the demand curve of the specific processes in each case.

Energy consumption accounts for a significant part of the emissions of CO2, which is one of the main causes of the greenhouse effect on our planet. The use of solar energy for hot water production. can save energy and reduce the impact of energy consumption on the environment and therefore a reduced environmental impact. The objective of this research is to study the solar utilization for heating the sludge in anaerobic digesters by hot water circulating inside a coil surrounding the surface of digester, to support conventional heating methods sludge as the electrical resistance or heat exchanger by energy generated by the methane gas produced in the anaerobic digestion. To study 3 digesters used two models which are heated with hot water within a coil (one insulated by a layer of fiberglass and polyurethane and other uninsulated) .The third digester had no external heating in order to observe their behavior and compare their evolution with the rest of the .The comparison digesters digesters 1 and 2 allowed us to study the advisability of providing insulation to the digester. Heat transfer through copper coil gave values between 83 and 92%. The installation application to a treatment for maintaining full scale within the digester at T = 32ºC in different climates: temperate, warm and cold climates, consisted of calculating the surface area of solar collectors and coil required to cover the annual energy needs of the digester, and the study of profitability of the installation, giving the best results for hot climates with return periods of 12 years and an internal rate of return (IRR) of 16% achieving an annual coverage of 79 % of energy needs with solar energy.

Achnanthes longipes is a marine, biofouling diatom that adheres to surfaces via adhesive polymers extruded during motility or organized into structures called stalks that contain three distinct regions: the pad, shaft, and collar. Four monoclonal antibodies (AL.C1–AL.C4) and antibodies from two uncloned hybridomas (AL.E1 and AL.E2) were raised against the extracellular adhesives of A. longipes. Antibodies were screened against a hot-water-insoluble/hot-bicarbonate-soluble-fraction. The hot-water-insoluble/hot-bicarbonate-soluble fraction was fractionated to yield polymers in three size ranges: F1, ≥ 20,000,000 Mr; F2, ≅100,000 Mr; and F3, <10,000 Mr relative to dextran standards. The ≅100,000-Mr fraction consisted of highly sulfated (approximately 11%) fucoglucuronogalactans (FGGs) and low-sulfate (approximately 2%) FGGs, whereas F1 was composed of O-linked FGG (F2)-polypeptide (F3) complexes. AL.C1, AL.C2, AL.C4, AL.E1, and AL.E2 recognized carbohydrate complementary regions on FGGs, with antigenicity dependent on fucosyl-containing side chains. AL.C3 was unique in that it had a lower affinity for FGGs and did not label any portion of the shaft. Enzyme-linked immunosorbent assay and immunocytochemistry indicated that low-sulfate FGGs are expelled from pores surrounding the raphe terminus, creating the cylindrical outer layers of the shaft, and that highly sulfated FGGs are extruded from the raphe, forming the central core. Antibody-labeling patterns and other evidence indicated that the shaft central-core region is related to material exuded from the raphe during cell motility.
