In der vorliegenden Arbeit wurden Derivate des Ionentransporters Valinomycin synthetisiert und als Sensorelemente in Modellmembransysteme inkorporiert. Als Modellmembranen wurden festkörpergestützte Lipiddoppelschichten (tBLMs, tethered bilayer lipid membranes) verwendet. rnValinomycin transportiert selektiv Kalium-Ionen durch Membranen, was durch einen Rückgang des Widerstandes über elektrochemische Messmethoden nachgewiesen werden kann. Es ist ein zyklisches Dodecadepsipeptid, das aus zwei verschiedenen Aminosäuren (L- und D-Valin) und Hydroxysäuren (L Milchsäure und D Hydroxyisovaleriansäure) besteht. In dieser Arbeit wurde ein L Valin durch ein L-Lysin ausgetauscht, um eine freie Aminogruppe zum Anbinden verschiedenster Liganden zu erhalten. rnDie Synthese erfolgte in Lösung über einen linearen Decadepsipeptid-Precursor, hierbei wurde hauptsächlich mit Benzyl- und Boc-Schutzgruppen gearbeitet. An den Precursor wurden dann unterschiedlich modifizierte Lysin-Didepside gebunden und das daraus erhaltene lineare Dodecadepsipeptid zyklisiert.rnInsgesamt wurden sechs verschiedene Derivate synthetisiert, deren Ionentransportfähigkeit mit Hilfe eines angebundenen Liganden blockiert wurde. Diese Blockade kann entweder mechanisch durch Festhalten des Ionencarriers an der Oberfläche der Membran oder chemisch durch Einbringen einer Ladung erfolgen, da geladene Moleküle eine Membran nicht überwinden können. rnAcetyl-Lysin-Valinomycin wurde als Testsystem hergestellt, um zu zeigen, dass die Synthese einen funktionsfähigen Ionencarrier ergeben hatte. Im nächsten Schritt wurde Lysin-Valinomycin mit freier Aminogruppe synthetisiert, um es als pH-Sensor zu nutzen und damit zu überprüfen, ob das chemische Einbringen einer Ladung möglich ist. Es konnte ein pH abhängiger Kalium-Transport nachgewiesen werden, die Blockade der Ionentransportfähigkeit über eine eingebrachte Ladung ist somit möglich. rnAuf dem gleichen Konzept beruht Ferrocen-Valinomycin. Wird der Ferrocen-Ligand oxidiert, liegt eine positive Ladung vor und der Ionencarrier kann die Membran nicht mehr überwinden. Eine Reduktion macht diesen Prozess reversibel. Ferrocen-Valinomycin konnte innerhalb einer tBLM chemisch oxidiert und reduziert werden, dieses System kann somit als chemischer Redox-Sensor eingesetzt werden.rnEine mechanische Blockade liegt dem Biotin- und dem Sulfonamid-Valinomycin zugrunde. Dabei soll die Zugabe von Streptavidin bzw. BCA II (bovine Carboanhydrase) den Ionentransport durch die Membran stoppen. Beide Valinomycin-Derivate zeigten aber keine Ionentransportfähigkeit, eine Inkorporation in tBLMs konnte jedoch über SPR gezeigt werden. rnDie Synthese eines fluoreszenz-markierten (FITC) Valinomycins ergab zwar auch keinen transportfähigen Ionencarrier, aber mit diesem Derivat konnte der Diffusionskoeffizient von Valinomycin in sBLMs mit Hilfe von Fluorescence Recovery after photobleaching (FRAP) bestimmt werden.rn

Membranproteine spielen eine wichtige Rolle bei physiologischen Prozessen wie Signalweiterleitung oder Immunreaktion. Deshalb stehen sie im Fokus der pharmakologischen Wirkstoffentwicklung und es besteht großes Interesse, Membranproteinbasierte Biosensoren zu entwickeln, die sich z.B. als Screening-Plattformen eignen. Allerdings stellt die Handhabung von Membranproteinen wegen ihrer amphiphilen Struktur eine große Herausforderung dar. Membranproteine werden meist in Zellkultur oder in bakteriellen Expressionssystemen synthetisiert. Diese Verfahren liefern aber oft nur eine geringe Ausbeute und erlauben wenig Kontrolle über die Expressionsbedingungen. Als alternativer Ansatz bietet sich stattdessen die in vitro Synthese von Proteinen an, die in einer zellfreien Umgebung stattfindet. Ziel der vorliegenden Arbeit war die Etablierung eines miniaturisierten Analysesystems, das Aktivitätsmessungen an in vitro synthetisierten Ionenkanälen erlaubt. Dafür wurde ein Labon- Chip entwickelt, der elektrochemische und optische Nachweismethoden in parallelen Anätzen ermöglicht. Als amphiphile Umgebung für die Inkorporation von Membranproteinen wurden vier verschieden biomimetische Membranaufbauten hinsichtlich ihrer Dichtigkeit und ihrer Reproduzierbarkeit untersucht. Als Methode fanden insbesondere die Impedanzspektroskpie und die Oberflächenplasmonen-Resonanzspektroskopie Anwendung. Die peptide cushioned Bilyer Lipid Membranes (pcBLM) eignete sich dabei am besten für Untersuchungen an Membranproteinen. Zur Detektion der Ionenkanalaktivität wurde eine neue Messmethode etabliert, die auf der Messung der Impedanz bei fester Frequenz basiert und u.a. eine Aussage über die Änderung des Membranwiderstandes bei Aktivierung erlaubt. Am Beispiel des nicotinischen Acetylcholinrezeptors (nAchR) konnte gezeigt werden, dass sich die Aktivität von Ionenkanälen mit dem entwickelten Chip-System nachweisen ließ. Die Spezifität der Methode konnte durch verschiedene Kontrollen wie die Zugabe eines nicht-aktivierenden Liganden oder Inhibition des Rezeptors nachgewiesen werden. Weiterhin konnte die in vitro Synthese des Ionenkanals a7 nAchR durch Radioaktivmarkierung nachgewiesen werden. Die Inkorporation des Rezeptors in die biomimetischen Membranen wurde mit Immunodetektion und elektrochemischen Methoden untersucht. Es zeigte sich, dass die funktionelle Inkorporation des a7 nAchR davon abhing, welcher biomimetische Membranaufbau verwendet wurde.

OBJECTIVES: Many flow-cytometric cell characterization methods require costly markers and colour reagents. We present here a novel device for cell discrimination based on impedance measurement of electrical cell properties in a microfluidic chip, without the need of extensive sample preparation steps and the requirement of labelling dyes. MATERIALS AND METHODS, RESULTS: We demonstrate that in-flow single cell measurements in our microchip allow for discrimination of various cell line types, such as undifferentiated mouse fibroblasts 3T3-L1 and adipocytes on the one hand, or human monocytes and in vitro differentiated dendritic cells and macrophages on the other hand. In addition, viability and apoptosis analyses were carried out successfully for Jurkat cell models. Studies on several species, including bacteria or fungi, demonstrate not only the capability to enumerate these cells, but also show that even other microbiological life cycle phases can be visualized. CONCLUSIONS: These results underline the potential of impedance spectroscopy flow cytometry as a valuable complement to other known cytometers and cell detection systems.

The generation of collimated electron beams from metal double-gate nanotip arrays excited by near infrared laser pulses is studied. Using electromagnetic and particle tracking simulations, we showed that electron pulses with small rms transverse velocities are efficiently produced from nanotip arrays by laser-induced field emission with the laser wavelength tuned to surface plasmon polariton resonance of the stacked double-gate structure. The result indicates the possibility of realizing a metal nanotip array cathode that outperforms state-of-the-art photocathodes.

Recently, sub-wavelength-pitch stacked double-gate metal nanotip arrays have been proposed to realize high current, high brightness electron bunches for ultrabright cathodes for x-ray free-electron laser applications. With the proposed device structure, ultrafast field emission of photoexcited electrons is efficiently driven by vertical incident near infrared laser pulses, via near field coupling of the surface plasmon polariton resonance of the gate electrodes with the nanotip apex. In this work, in order to gain insight in the underlying physical processes, the authors report detailed numerical studies of the proposed device. The results indicate the importance of the interaction of the double-layer surface plasmon polariton, the position of the nanotip, as well as the incident angle of the near infrared laser pulses.

Conductive nanoparticles, especially elongated ones such as carbon nanotubes, dramatically modify the electrical behavior of liquid crystal cells. These nanoparticles are known to reorient with liquid crystals in electric fields, causing significant variations of conductivity at minute concentrations of tens or hundreds ppm. The above notwithstanding, impedance spectroscopy of doped cells in the frequency range customarily employed by liquid crystal devices, 100 Hz?10 kHz, shows a relatively simple resistor/capacitor response where the components of the cell can be univocally assigned to single components of the electrical equivalent circuit. However, widening the frequency range up to 1 MHz or beyond reveals a complex behavior that cannot be explained with the same simple EEC. Moreover, the system impedance varies with the application of electric fields, their effect remaining after removing the field. Carbon nanotubes are reoriented together with liquid crystal reorientation when applying voltage, but barely reoriented back upon liquid crystal relaxation once the voltage is removed. Results demonstrate a remarkable variation in the impedance of the dielectric blend formed by liquid crystal and carbon nanotubes, the irreversible orientation of the carbon nanotubes and possible permanent contacts between electrodes.

As a second-order nonlinear optical process, sum-frequency generation is highly surface-specific and accordingly has been developed into a very powerful and versatile surface spectroscopic tool. It has found many unique applications in different disciplines and thus provided many exciting new research opportunities in surface and surface-related science. Selected examples are discussed here to illustrate the power of the technique.

A rapid direct assay for polymerase-induced elongation along a given template is an obligate requirement for understanding the processivity of polymerization and the mode of action of drugs and inhibitors on this process. Surface plasmon resonance can be used to follow the association and the dissociation rates of a given reverse transcriptase on DNA.RNA and DNA.DNA hybrids immobilized on a biotin-streptavidin surface. The addition of nucleotides complementary to the template strand produces an increase in the local mass, as deduced from an increase in the measured signal, due to elongation of the primer strand that allows an estimation of both the extent and rate of the polymerization process. The terminator drug 3'-deoxy-3'-azidothymidine triphosphate completely abolishes the increase in signal as would be expected from an inhibition of elongation. This technique provides a sensitive assay for the affinities of different polymerases for specific templates and for the effects of terminators of the elongation process.

The moisture content and its spatial distribution has a great influence on the durability properties of concrete structures. Several non-destructive techniques have been used for the determination of the total water content, but moisture distribution is difficult to determine. In this paper impedance spectroscopy is used to study the water distribution in concrete samples with controlled and homogeneously distributed moisture contents. The technique is suitable for the determination of water distribution inside the sample, using the appropriate equivalent circuits. It is shown that using the selected drying procedures there is no change in the solid phase of the samples, although the technique can only be used for the qualitative study of variations in the solid phase when samples are too thick. The results of this work show that for a wide range of concrete percentages of saturation, from full to 18 % saturation, practically all the pores keep at least a thin layer of electrolyte covering their walls, since the capacitance measurement results are practically independent of the saturation degree.

In this work, the microstructure of mortars made with an ordinary Portland cement and slag cement has been studied. These mortars were exposed to four different constant temperature and relative humidity environments during a 180-day period. The microstructure has been studied using impedance spectroscopy, and mercury intrusion porosimetry as a contrast technique. The impedance spectroscopy parameters make it possible to analyze the evolution of the solid fraction formation for the studied mortars and their results are confirmed with those obtained using mercury intrusion porosimetry. The development of the pore network of mortars is affected by the environment. However, slag cement mortars are more influenced by temperature while the relative humidity has a greater influence on the OPC mortars. The results show that slag cement mortars hardened under non-optimal environments have a more refined microstructure than OPC mortars for the studied environmental conditions.

In this study we have demonstrated the interactions of kalata B1 and its naturally occurring analogue kalata B6 with five model lipid membranes and have analyzed the binding kinetics using surface plasmon resonance. Two kalata peptides showed a higher affinity for the phosphatidylethanolamine-containing membranes, indicating that the peptides would bind selectively to bacterial membranes. Also we have optimized the procedure for the immobilization of five liposome mixtures and have shown that the procedure provides reproducible levels of immobilized liposomes and could be used to screen the selective binding of putative antimicrobial peptides to model mammalian or microbial phospholipid membranes. A series of surface plasmonic fibre devices were fabricated using multiple coatings deposited on a lapped section of a single mode fibre. Coupling from the guided mode to surface plasmons was promoted following UV laser irradiation of the coated region through a phase mask, which generated a surface relief grating structure. The devices showed high spectral sensitivities and strong coupling for low refractive indices as compared to other grating-type fibre devices. The plasmonic devices were used to detect the variation in the refractive indices of alkane gases with measured wavelength and coupling sensitivity to index of 3400 nm RIU-1 and 8300 dB RIU-1, respectively. As a demonstration of the performance of these gas sensors, a minimum concentration of 2% by volume of butane in ethane was achieved.

We demonstrate surface plasmon resonance (SPR) fiber devices based upon ultraviolet inscription of a grating-type structure into both single-layered and multilayered thin films deposited on the flat side of a lapped D-shaped fiber. The single-layered devices were fabricated from germanium, while the multilayered ones comprised layers of germanium, silica, and silver. Some of the devices operated in air with high coupling efficiency in excess of 40 dB and an estimated index sensitivity of Delta lambda/Delta n = 90 mn from 1 to 1.15 index range, while others provided an index sensitivity of Delta lambda/Delta n = 6790 mn for refractive indices from 1.33 to 1.37. A series of surface plasmonic fibre devices were fabricated by depositing multiple thin coatings on a lapped section of a standard single mode telecoms fibre forming a D-shaped section and then inscribing a grating-type structure using UV light. The coatings consisted of base coatings of semi-conductor (germanium) and dielectric (silicon dioxide) materials, followed by different metals. These fibre devices showed high spectral refractive index sensitivity with high coupling efficiency in excess of 40 dB for indices in the aqueous regime and below, with estimated index sensitivities of Lambda lambda/Lambda n = 90-800 nm from 1 to 1.15 index range and Lambda lambda/Lambda n = 1200-4000 nm for refractive indices from 1.33 to 1.39. We demonstrate the use of tilted fiber gratings to assist with the generation of infrared surface plasmons on a metal film coating the flat of a D-shaped fiber. The wavelength of the strong (>25 dB) resonance is tunable over similar to 1000 nm by adjusting the polarization state of the light and is highly sensitive to the refractive index of any aqueous medium surrounding the fiber (sensitivity= 3365 nm).
