A pesquisa teve como objetivo estudar o processo de maturação das sementes de calêndula. A partir de 50% dos botões florais em antese, foi feita a marcação das flores, sendo a coleta das sementes realizada em seis épocas: 20, 24, 28, 32, 36 e 40 dias após a antese (DAA). Em cada coleta, as sementes foram separadas pelo tamanho (maior e menor), e classificadas conforme a Tabela de Munsell (Munsell,1977) conforme as colorações observadas: verde, verde claro, creme, marrom claro e marrom escuro. Em seguida, foram realizadas as determinações de teor de água, peso da matéria seca das sementes, germinação e avaliações de qualidade fisiológica: primeira contagem da germinação e emissão de raiz primária. O maior acúmulo de matéria seca ocorreu entre 28 e 32 DAA, quando ainda havia sementes com coloração verde claro, entretanto, aos 36 DAA as sementes mostraram maior viabilidade e vigor (primeira contagem da germinação e emissão de raiz primária) não havendo diferença entre sementes de maior e menor tamanho, que apresentavam colorações creme, marrom claro e marrom escuro. Embora não tenha ocorrido coincidência temporal entre acúmulo de matéria seca, germinação e vigor, a maturidade fisiológica ocorre entre 28 e 32 DAA, estando as sementes com teor de água médio de 36%, sendo indicada a colheita aos 36 DAA com teor de água médio de 20%, antes de ocorrer a deiscência dos frutos aos 40 DAA.

As plantas sofrem agressões por agentes bióticos e abióticos e apesar de não apresentarem defesas através de movimentos ágeis, podem ocorrer adaptações e profundas alterações no metabolismo celular, entre elas a síntese de proteínas de defesas, ativada através de mecanismos complexos. A aplicação exógena ou o estímulo à síntese endógena de ácidos orgânicos como o ácido salicílico, pode agir como indutor de proteínas de tolerância aos diferentes estresses, bem como para elevar a atividade de enzimas de desintoxicação celular, especialmente às envolvidas na degradação de radicais ativos oxigenados. O objetivo deste trabalho foi estudar o efeito do ácido salicílico sobre a germinação e o vigor de sementes de calêndula (Calendula officinalis L.) em condições ideais e sob estresse térmico e hídrico. As sementes foram colocadas para germinar em papel embebido em soluções crescentes de ácido salicílico (zero, 0,0125, 0,025, 0,05, 0,1 e 0,2mM); medindo-se as variáveis: percentagem de germinação; índice de velocidade de germinação e primeira contagem da germinação. Ficou constatado através do teste de Tukey que apenas a germinação foi significativa, sendo que as melhores dosagens de sementes germinadas ficou entre 0,025 e 0,05mM de ácido salicílico. Três outros experimentos foram feitos, um com água acidulada aos pH respectivos às concentrações de ácido salicílico (6,0; 4,8; 4,2; 3,6 e 3,2), um com diferentes potenciais hídricos induzidos por manitol (0; -0,3; -0,6; -0,9 e -1,2MPa), e outro com temperaturas (20, 25, 30 e 35ºC). O ácido salicílico na dose de 0,025mM interferiu positivamente na percentagem de germinação e no índice de velocidade de germinação de sementes da calêndula em condições ideais e sob efeito de estresse hídrico e térmico a 35º.

Alloxan induced animal model was used to evaluate the potential antidiabetic effect of A . marmelose leaf extract. The diabetic animals were given insulin injection and another group A . marmelose leaf extract orally. It maintained the weight of the animals near to the control rats but a significant decrease in weight was noted it diabetic animals without any treatment . The blood elucose level in 'treated animals were near to that of control ones. Also a significantly increased glucose tolerance was observed in animals orally given the leaf extract prior to the experiment . A significant decrease in liver glycogen ( 1.24 -.07 s!/ 100 g of wet tissue) was observed in diabetic rats which was brought to almost the normal level (I.84- . 14 g; 100 g) with Icaf extract treatment. Blood urea and serum cholesterol increased ( 62.66 - 3.50 and 192.67± 13.64 mg' dl) significantly in alloxxan diabetic rats . The leaf extract treatment decreased the blood urea and serum . cholesterol (37.83 - 3.97 and 99?0±8.43 mg/dl ) to that of control ones . A similar effect was seen with insulin treatment . The results indicate that the active principle in .4. marmelose leaf extract has similar hypoglycaemic activity to insulin treatment.

Alloxan induced diabetic animal model was sed to evaluate the potential antidiabetic effect of ie aqueous leaf extract of Aegle marmelose. Insuinjection and oral treatment of Aegle marmelose of extract to the alloxan induced rats maintained ;e animal weight near to the control rats whereas ,,ere was a significant decrease in weight in alloxan •duced rats. Insulin injection and oral treatment the leaf extract reversed the decrease of protein )ntent in all the organs studied except the pancas. Glucose level was maintained near to the co- :rol levels in the insulin injected and leaf extract Bated alloxan diabetic rats. A significant increased ucose tolerance was observed in animals orally ven the leaf extract prior to the experiment. The suits indicate that there is an increased utilization glucose in the Aegle marmelose leaf extract treed rats.

The functional basis of diabetes-mellitus to a certain extent, can be elucidated by studying diabetes-induced changes in metabolic enzymes. Malate dehydrogenase (MDH), is an enzyme directly involved in glucose metabolism. The kinetic parameters of MDH and its purified cytosolic isozyme, S-MDH, have been studied in the liver of streptozotocin- diabetic rats; also the potential of the leaf extract of A. marmelose as an was investigated. The Km of the liver enzyme increased significantly, in both crude and purified preparations in the diabetic state when compared to Lhe respective controls. Insulin as well as leaf- •extract treatment of the diabetic rats brought about a reversal of K. values to near normal. Vmax of purified S-MDH was significantly higher in the diabetic state when compared to the control. Insulin and leaf extract treatment did not reverse this change. Since MDH is an important enzyme in glucose metabolism, the variation in its quantitative and qualitative nature may contribute to the pathological status of diabetes. The fact that leaf extract of A. marmelose was found to be as effective as insulin in restoration of blood glucose and body weight to normal levels, the use of A. marmelose as potential hypoglycemic agent is suggested.

Epilepsy is a syndrome of episodic brain dysfunction characterized by recurrent unpredictable, spontaneous seizures. Cerebellar dysfunction is a recognized complication of temporal lobe epilepsy and it is associated with seizure generation, motor deficits and memory impairment. Serotonin is known to exert a modulatory action on cerebellar function through 5HT2C receptors. 5-HT2C receptors are novel targets for developing anticonvulsant drugs. In the present study, we investigated the changes in the 5-HT2C receptors binding and gene expression in the cerebellum of control, epileptic and Bacopa monnieri treated epileptic rats. There was a significant down regulation of the 5-HT content (pb0.001), 5-HT2C gene expression (pb0.001) and 5-HT2C receptor binding (pb0.001) with an increased affinity (pb0.001). Carbamazepine and B. monnieri treatments to epileptic rats reversed the down regulated 5-HT content (pb0.01), 5-HT2C receptor binding (pb0.001) and gene expression (pb0.01) to near control level. Also, the Rotarod test confirms the motor dysfunction and recovery by B. monnieri treatment. These data suggest the neuroprotective role of B. monnieri through the upregulation of 5-HT2C receptor in epileptic rats. This has clinical significance in the management of epilepsy

White spot syndrome virus (WSSV), the most contagious pathogen of cultured shrimp, causes mass mortality, leading to huge economic loss to the shrimp industry. The lack of effective therapeutic or prophylactic measures has aggravated the situation, necessitating the development of antiviral agents. With this objective, the antiviral activity in the aqueous extract of a mangrove plant Ceriops tagal in Penaeus monodon was evaluated. The Ceriops tagal aqueous extract (CTAE) was non-toxic to shrimps at 50 mg/ml when injected intramuscularly at a dosage of 10 lL/animal (0.5 mg/animal) and showed a protective effect against WSSV at 30 mg/ml when mixed with WSSV suspension at a 1:1 ratio. When the extract was administered along with the diet and the animals were challenged orally, there was a dose-dependent increase in survival, culminating in 100 % survival at a concentration of 500 mg/kg body weight/day. Neither hypertrophied nuclei nor the viral envelope protein VP28 could be demonstrated in surviving shrimps using histology and indirect immunofluorescence histochemistry (IIFH), respectively. To elucidate the mode of action, the temporal expression of WSSV genes and shrimp immune genes, including antimicrobial peptides, was attempted. None of the viral genes were found to be expressed in shrimps that were fed with the extract and challenged or in those that were administered CTAE-exposed WSSV. The overall results suggest that the aqueous extract from C. tagal can protect P. monodon from white spot syndrome virus infection.

Heilkräuter sind während des Trocknungsprozesses zahlreichen Einflüssen ausgesetzt, welche die Qualität des Endproduktes entscheidend beeinflussen. Diese Forschungsarbeit beschäftigt sich mit der Trocknung von Zitronenmelisse (Melissa officinalis .L) zu einem qualitativ hochwertigen Endprodukt. Es werden Strategien zur Trocknung vorgeschlagen, die experimentelle und mathematische Aspekte mit einbeziehen, um bei einer adäquaten Produktivität die erforderlichen Qualitätsmerkmale im Hinblick auf Farbeänderung und Gehalt an ätherischen Ölen zu erzielen. Getrocknete Zitronenmelisse kann zurzeit, auf Grund verschiedener Probleme beim Trocknungsvorgang, den hohen Qualitätsanforderungen des Marktes nicht immer genügen. Es gibt keine standardisierten Informationen zu den einzelnen und komplexen Trocknungsparametern. In der Praxis beruht die Trocknung auf Erfahrungswerten, bzw. werden Vorgehensweisen bei der Trocknung anderer Pflanzen kopiert, und oftmals ist die Trocknung nicht reproduzierbar, oder beruht auf subjektiven Annäherungen. Als Folge dieser nicht angepassten Wahl der Trocknungsparameter entstehen oftmals Probleme wie eine Übertrocknung, was zu erhöhten Bruchverlusten der Blattmasse führt, oder eine zu geringe Trocknung, was wiederum einen zu hohen Endfeuchtegehalt im Produkt zur Folge hat. Dies wiederum mündet zwangsläufig in einer nicht vertretbaren Farbänderung und einen übermäßigen Verlust an ätherischen Ölen. Auf Grund der unterschiedlichen thermischen und mechanischen Eigenschaften von Blättern und Stängel, ist eine ungleichmäßige Trocknung die Regel. Es wird außerdem eine unnötig lange Trocknungsdauer beobachtet, die zu einem erhöhten Energieverbrauch führt. Das Trocknen in solaren Tunneln Trocknern bringt folgendes Problem mit sich: wegen des ungeregelten Strahlungseinfalles ist es schwierig die Trocknungstemperatur zu regulieren. Ebenso beeinflusst die Strahlung die Farbe des Produktes auf Grund von photochemischen Reaktionen. Zusätzlich erzeugen die hohen Schwankungen der Strahlung, der Temperatur und der Luftfeuchtigkeit instabile Bedingungen für eine gleichmäßige und kontrollierbare Trocknung. In Anbetracht der erwähnten Probleme werden folgende Forschungsschwerpunkte in dieser Arbeit gesetzt: neue Strategien zur Verbesserung der Qualität werden entwickelt, mit dem Ziel die Trocknungszeit und den Energieverbrauch zu verringern. Um eine Methodik vorzuschlagen, die auf optimalen Trocknungsparameter beruht, wurden Temperatur und Luftfeuchtigkeit als Variable in Abhängigkeit der Trocknungszeit, des ätherischer Ölgehaltes, der Farbänderung und der erforderliche Energie betrachtet. Außerdem wurden die genannten Parametern und deren Auswirkungen auf die Qualitätsmerkmale in solaren Tunnel Trocknern analysiert. Um diese Ziele zu erreichen, wurden unterschiedliche Ansätze verfolgt. Die Sorption-Isothermen und die Trocknungskinetik von Zitronenmelisse und deren entsprechende Anpassung an verschiedene mathematische Modelle wurden erarbeitet. Ebenso wurde eine alternative gestaffelte Trocknung in gestufte Schritte vorgenommen, um die Qualität des Endproduktes zu erhöhen und gleichzeitig den Gesamtenergieverbrauch zu senken. Zusätzlich wurde ein statistischer Versuchsplan nach der CCD-Methode (Central Composite Design) und der RSM-Methode (Response Surface Methodology) vorgeschlagen, um die gewünschten Qualitätsmerkmalen und den notwendigen Energieeinsatz in Abhängigkeit von Lufttemperatur und Luftfeuchtigkeit zu erzielen. Anhand der gewonnenen Daten wurden Regressionsmodelle erzeugt, und das Verhalten des Trocknungsverfahrens wurde beschrieben. Schließlich wurde eine statistische DOE-Versuchsplanung (design of experiments) angewandt, um den Einfluss der Parameter auf die zu erzielende Produktqualität in einem solaren Tunnel Trockner zu bewerten. Die Wirkungen der Beschattung, der Lage im Tunnel, des Befüllungsgrades und der Luftgeschwindigkeit auf Trocknungszeit, Farbänderung und dem Gehalt an ätherischem Öl, wurde analysiert. Ebenso wurden entsprechende Regressionsmodelle bei der Anwendung in solaren Tunneltrocknern erarbeitet. Die wesentlichen Ergebnisse werden in Bezug auf optimale Trocknungsparameter in Bezug auf Qualität und Energieverbrauch analysiert.
