Protein crystallization is of strategic and commercial relevance in the post-genomic era because of its pivotal role in structural proteomics projects. Although protein structures are crucial for understanding the function of proteins and to the success of rational drug design and other biotechnology applications, obtaining high quality crystals is a major bottleneck to progress. The major means of obtaining crystals is by massive-scale screening of a target protein solution with numerous crystallizing agents. However, when crystals appear in these screens, one cannot easily know if they are crystals of protein, salt, or any other molecule that happens to be present in the trials. We present here a method based on Attenuated Total Reflection (ATR)-FT-IR imaging that reliably identifies protein crystals through a combination of chemical specificity and the visualizing capability of this approach, thus solving a major hurdle in protein crystallization. ATR-FT-IR imaging was successfully applied to study the crystallization of thaumatin and lysozyme in a high-throughput manner, simultaneously from six different solutions. This approach is fast as it studies protein crystallization in situ and provides an opportunity to examine many different samples under a range of conditions.

Protein crystallization has gained a new strategic and commercial relevance in the postgenomic era due to its pivotal role in structural genomics. Producing high quality crystals has always been a bottleneck to efficient structure determination, and this problem is becoming increasingly acute. This is especially true for challenging, therapeutically important proteins that typically do not form suitable crystals. The OptiCryst consortium has focused on relieving this bottleneck by making a concerted effort to improve the crystallization techniques usually employed, designing new crystallization tools, and applying such developments to the optimization of target protein crystals. In particular, the focus has been on the novel application of dual polarization interferometry (DPI) to detect suitable nucleation; the application of in situ dynamic light scattering (DLS) to monitor and analyze the process of crystallization; the use of UV-fluorescence to differentiate protein crystals from salt; the design of novel nucleants and seeding technologies; and the development of kits for capillary counterdiffusion and crystal growth in gels. The consortium collectively handled 60 new target proteins that had not been crystallized previously. From these, we generated 39 crystals with improved diffraction properties. Fourteen of these 39 were only obtainable using OptiCryst methods. For the remaining 25, OptiCryst methods were used in combination with standard crystallization techniques. Eighteen structures have already been solved (30% success rate), with several more in the pipeline.

The present study focuses on the development of pedagogical activities in Music Teaching, aiming to enhance the accessibility of musical knowledge for both deaf and hearing students, using a bilingual approach in regular schools. Few studies address Music and Deafness, and those that do focus exclusively on the context of special education, and specifically the deaf student, which signals the urgent need for conducting research on this issue in the context of inclusion – empirically and carried out on school grounds. Therefore, we developed our study at a Natal City Public Elementary school, in a class of 6th graders, comprised of 37 students, 3 of whom were deaf. The objective of the study was to develop a proposal for a pedagogical intervention in Music Teaching, using a bilingual approach, with deaf and hearing students, in the context of regular school classes. The research is based on the theoretical framework presented in Penna (2010), Brito (2001) and Fonterrada (2008), with reference to music education, and Haguiara-Cervellini (2003), Finck (2009) and Louro (2006), with reference to inclusion in teaching music. To achieve this objective, we developed a proposal for intervention based on the methodological dictates of intervention research, presented in studies by Jobim and Souza (2011) in light of the theoretical concepts posited by Mikhail Bakhtin, which assert that knowledge is produced through interaction between subjects, dialogically and through alterity. This methodology was carried out in pedagogical workshops, conceived as spaces for the construction of knowledge, mobilizing participants to engage in ludic activities of musical experimentation. Content covered in these workshops focused on Pulse and Rhythm – basic elements in music education – demonstrating that awareness about and sensitivity to these elements is not limited to the auditory sensory perception of the student, once the entire body is used as an agent of acquisition and expression. Thus, we began the trajectory of our research from the starting point of the identification and perception of „Pulse‟, using one‟s own body and the body of classmates, representing it through physical expressions and movement. Subsequently, this Pulse was extended from the body to a percussion instrument, and was then represented graphically as lines of rhythm, constituting a process of reading and writing; ultimately the intervention culminated in the class presentation with the musical group De Pau e Lata (Stick and Can). In our analysis, faced with the challenges and possibilities presented in our study, findings showed satisfactory results with regard to the participation of all of the students: completing the activities proposed in the class, asking questions when they did not understand, positioning themselves when they thought it necessary, expressing opinions about the work completed, evaluating the workshops given, interacting, helping in the activities, constructing knowledge collaterally, experimenting and experiencing musical elements through the body in activities that applied to both groups (deaf and hearing) in the one class. These indications elucidate the viability of teaching music to deaf and hearing students, using a bilingual approach, and based on experiences with the body and communicative and cultural specificities involved, confirming, as well, the role of Sign Language as a mediator in the teaching/learning process.

The study of diatoms in core HC11 collected from the southwestern part of Chukchi Sea, allowed to distinguish 3 diatoms ecological zones, reflecting paleoenvironmental changes during the last 2300 years. The sediment age was based on the sedimentation rates, determined by 210Pb and radiocarbon dating of mollusk shells. The environmental changes of Chukchi Sea revealed by examination of diatoms correlates with global climate changes - the warming of the early and middle Subatlantic and cooling of the late Subatlantic (Little Ice Age). Warming early and middle Subatlantic in the Chukchi Sea was probably stronger than the warming of the late 20th century and was not accompanied by significant changes in sea level.

La atención a la diversidad se ha convertido en un elemento básico de los sistemas educativos de muchos países. Las respuestas educativas que se han desarrollado en las escuelas, aquellas que entienden la diversidad como un valor enriquecer dentro del proceso educativo, han dado lugar a modelos muy diferentes. A nivel internacional, hay un movimiento que defiende la necesidad de promover los modelos inclusivos, que entienden la diferencia como una oportunidad para aprender juntos, y sitúa la metodología como uno de los pilares básicos del proceso de enseñanza-aprendizaje. En este sentido, el aprendizaje basado en la cooperación se presenta como una propuesta alternativa para trabajar en la escuela. Diferentes investigaciones realizadas a lo lardo de los últimos años ponen en evidencia las importantes contribuciones del trabajo cooperativo para la adquisición de valores y actitudes que mejoran la convivencia al mismo tiempo que facilitan un mejor desarrollo personal para todo el alumnado. Nuestro trabajo en la escuela primaria y secundaria nos ha permitido reflexionar y recoger información sobre nuestra experiencia alrededor de la atención a la diversidad en el marco de una escola que pretende caminar a la inclusión. El artículo presenta una reflexión sobre las contribuciones del aprendizaje cooperativo dentro de una escuela que busca la inclusión de todos los chicos y las chicas, basándonos para ello en nuestras experiencias profesionales propias así como en el análisis y la interpretación de las contribuciones principales recogidas en la bibliografía nacional e internacional.

Lata 2003–2004 to okres zamykający etap przygotowawczy do przystąpienia Polski do Unii Europejskiej, w szczególności w zakresie dostosowywania polskich rozwiązań prawnych i instytucjonalnych do standardów unijnych. Dwa wydarzenia z tego czasu: referendum akcesyjne i pierwsze w Polsce wybory do Parlamentu Europejskiego stanowiły przy tym swoiste podsumowanie wieloletniej debaty publicznej nad kierunkami polskiej polityki zagranicznej oraz miejscem i rolą naszego kraju w ukształtowanych na początku lat 90. nowych warunkach geopolitycznych.

Tämän tutkielman aiheena ja tutkimuskohteena on osituksen sovittelu voimassa olevan oikeuden kannalta. Avioliitto nykyisessä muodossaan on varallisuusoikeudellinen sopimus, joka perustaa avio-oikeuden puolisoiden välille. Avio-oikeus konkretisoituu avioliiton purkautuessa avioeroon tai puolison kuolemaan. Normaalisääntöjen mukainen ositus johtaa omaisuuden puolittamiseen. Avioliiton purkautuessa puolittamisperiaate on vahva pääsääntö. Sen säännönmukainen noudattaminen voi johtaa epäoikeudenmukaiseen lopputulokseen. Osituksen epäoikeudenmukaista lopputulosta varten on avioliittolain 103b §:ssä säädetty mahdollisuudesta sovitella ositusta. Osituksen sovittelun osapuolia voivat olla puolisot, puoliso ja toisen puolison perilliset, tai molempien perillistahot. Tutkielmassa kuvataan, miten avioliittolain 103b §:ssä ilmaistu osituksen sovittelusääntö sijoittuu aviovarallisuusjärjestelmän kokonaisuuteen. Tutkielma pyrkii vastaamaan, millaisten edellytysten vallitessa sovittelu on mahdollista de lege lata. Sovitteluedellytysten lisäksi tutkielmassa tarkastellaan, millaisia sovittelukeinoja voimassa oleva laki mahdollistaa, ja millaisia rajoituksia niiden käyttöön liittyy. Tutkielmassa selvitetään sovittelussa käytettyjen harkintakriteerien painoarvoa ja keskinäistä suhdetta. Mielenkiinto kohdistuu myös siihen, millaisia ongelmia sovittelusäännön tulkinnassa ja soveltamisessa on ilmennyt korkeimman oikeuden ennakkoratkaisujen perusteella säännön liki kolmekymmenvuotisen voimassaolon aikana. Osituksen sovittelusääntö on oikeudenmukaisuustavoitteen kannalta välttämätön osa voimassaolevan aviovarallisuusjärjestelmän kokonaisuutta. Sillä voidaan korjata ositusjärjestelmän heikkouksia poikkeuksellisissa tilanteissa. Tutkimusmateriaalina on lainsäädäntö, lain valmistelutyöt, oikeuskirjallisuus sekä osituksen sovittelua koskeva korkeimman oikeuden ratkaisukäytäntö.

Tekst powstał jako część wprowadzająca do książki o Lidze Narodów obejmującej lata 1926-1933. Oczekiwania wydawcy na książkę skromniejszą objętościowo wymusiło istotne zmiany koncepcyjne oraz redakcyjne fragmentów już napisanych. Ta część napisanej już pracy do zmienionej ze względów "organizacyjnych" koncepcji nie przystaje. Walka zaś i opór w istniejących na rynku wydawniczym warunkach, byłyby jałowym biegiem. Zaprezentowany materiał jest zatem wstępną wersją, której cyzelowanie, uzupełnianie i poprawianie nie wyzwalało emocji.

Lignocellulosic biomass is the most abundant renewable source of energy that has been widely explored as second-generation biofuel feedstock. Despite more than four decades of research, the process of ethanol production from lignocellulosic (LC) biomass remains economically unfeasible. This is due to the high cost of enzymes, end-product inhibition of enzymes, and the need for cost-intensive inputs associated with a separate hydrolysis and fermentation (SHF) process. Thermotolerant yeast strains that can undergo fermentation at temperatures above 40°C are suitable alternatives for developing the simultaneous saccharification and fermentation (SSF) process to overcome the limitations of SHF. This review describes the various approaches to screen and develop thermotolerant yeasts via genetic and metabolic engineering. The advantages and limitations of SSF at high temperatures are also discussed. A critical insight into the effect of high temperatures on yeast morphology and physiology is also included. This can improve our understanding of the development of thermotolerant yeast amenable to the SSF process to make LC ethanol production commercially viable.
