The aim of this study was to improve cage systems for maintaining adult honey bee (Apis mellifera L.) workers under in vitro laboratory conditions. To achieve this goal, we experimentally evaluated the impact of different cages, developed by scientists of the international research network COLOSS (Prevention of honey bee COlony LOSSes), on the physiology and survival of honey bees. We identified three cages that promoted good survival of honey bees. The bees from cages that exhibited greater survival had relatively lower titers of deformed wing virus, suggesting that deformed wing virus is a significant marker reflecting stress level and health status of the host. We also determined that a leak- and drip-proof feeder was an integral part of a cage system and a feeder modified from a 20-ml plastic syringe displayed the best result in providing steady food supply to bees. Finally, we also demonstrated that the addition of protein to the bees' diet could significantly increase the level ofvitellogenin gene expression and improve bees' survival. This international collaborative study represents a critical step toward improvement of cage designs and feeding regimes for honey bee laboratory experiments.

A diapausa é um fenômeno amplamente presente nos artrópodes e é considerada como primordial para o sucesso evolutivo da Classe Insecta, pois possibilita a sobrevivência em condições adversas, como estações frias e secas. Sabe-se que durante a diapausa ocorre o silenciamento de muitos genes e que outros são unicamente expressos nesta fase. Embora existam evidências de que o processo da diapausa tenha se mantido conservado durante a evolução das espécies, ainda há lacunas no conhecimento sobre o nível de conservação dos padrões metabólicos. Um bom modelo para se estudar a diapausa é Tetrapedia diversipes, uma espécie bivoltina de abelha solitária. Os indivíduos que nascem na primeira geração seguem o desenvolvimento desde ovo até adulto em tempo bem menor do que aqueles que nascem na segunda geração; estes retardam o desenvolvimento na fase larval. Além disso, essa espécie é de fácil obtenção no seu ambiente natural, pois apresenta alta taxa de nidificação em ninhos-armadilha. O objetivo deste trabalho foi comparar o perfil de expressão de genes entre as larvas da 1ª geração (que não entram em diapausa), larvas da 2ª geração (que entrariam em diapausa) e das larvas em diapausa. Foram identificados 196 genes diferencialmente expressos, destes 87 foram anotados. Muitos destes genes já foram descritos na literatura como relacionados à diapausa em outras espécies, no entanto, o padrão de expressão não é conservado. Os genes aqui identificados foram divididos em cinco grupos: relacionados à desintoxicação celular, cutícula e citoesqueleto, metabolismo de lipídeos e esteróis, ciclo celular e outros genes relacionados à diapausa

The agriculture with the use of products with less environmental impact is expanding. In it, the producers offer their products without the use of synthetic chemical inputs, solving the phytosanitary problems with the use of biological or alternative control agents such as parasites, predators, entomopathogenic, alternative products, plant extracts and essential oils. These products can be considered safe to non-target organisms, but studies are needed to find these features on natural enemies and on the beneficial insects such as bees, common frequenter of cultures. In this sense, this study aims to evaluate the effects of control over reproductive quality queens of Apis mellifera L. (Hymenoptera: Apidae) Africanized. For this, it tested the action of control products on the production of A. mellifera queens, using the commercial entomopathogenic fungus Boveril® 1,0x108 (Beauveria bassiana) and aqueous extract of pomegranate (Punica granatum) at a concentration of 5% sterile distilled water with Tween (0.01%) and sterile distilled water (controls). The treatments were incorporated into a tissue type gauze, wrapped in an acrylic plate and packed inside minirrecrias type colonies for the production of queens on the day before the transfer of larvae. The next day were introduced battens with 30 domes with larvae to produce queens, so the workers have contacted the agent tested. From the emergence of all the queens, they were monitored to determine the measures of body weight (mg), length and width of wing and abdomen, length, width and height of the chest (mm) as well as the time of emergence of queens. The next step was evaluated the influence of the control agents in production creates, performing measurements of creating areas in cm2 for six straight weeks. It was found that the area creates Queens did not differ among the treatments. Histological analysis of hipofaringeanas of workers glands that came into contact with the control agents and the midgut of virgin queens were also held. Histological analysis differences were observed in the tissues when the treatments were compared with the respective controls.

Caste determination in Trigona spinipes Fabricius (Hymenoptera, Apidae, Meliponini) is trophogenic. Larvae that eat about 360 mu l of food become queens, while those who consume 36 mu l develop into workers. We studied the effect of larval nutrition on the number and length of ovarioles and on ovarian development in fifth instar larvae, white eyed, pink eyed and black-eyed pupae as well as newly emerged adults. All larvae have four ovarioles per ovary, while in queen pupae this number ranged from 8 to 15. Cyst formation, the cell death and other characteristics of ovary morphogenesis were the same regardless of the quantity of food consumed. These results are discussed in relation to caste differentiation in other bees.

Lia Goncalves, Claudia Ines da Silva, and Maria Luisa Tunes Buschini (2012) Collection of pollen grains by Centris (Hemisiella) tarsata Smith (Apidae: Centridini): Is C. tarsata an oligolectic or polylectic species? Zoological Studies 51(2): 195-203. Among pollinator species, bees play a prominent role in maintaining biodiversity because they are responsible, on average, for 80% of angiosperm pollination in tropical regions. The species richness of the bee genus Centris is high in South America. In Brazil, these bees occur in many types of ecosystems. Centris tarsata is an endemic species occurring only in Brazil. No previous studies considered interactions between plants and this bee species in southern Brazil, where it is the most abundant trap-nesting bee. Accordingly, the goals of this study were to investigate plants used by this species for its larval food supply and determine if this bee is polylectic or oligolectic in this region. This work was conducted in the Parque Municipal das Araucarias, Guarapuava (PR), southern Brazil, from Mar. 2002 to Dec. 2003. Samples of pollen were collected from nests of these bees and from flowering plants in grassland and swamp areas where the nests were built. All of the samples were treated with acetolysis to obtain permanent slides. The family Solanaceae was visited most often (71%). Solanum americanum Mill. (28.6%) and Sol. variabile Mart. (42.4%) were the primary pollen sources for C. tarsata in the study area. We found that although C. tarsata visited 20 species of plants, it preferred Solanum species with poricidal anthers and pollen grains with high protein levels. This selective behavior by females of C. tarsata indicates that these bees are oligolectic in their larval provisioning in this region of southern Brazil. http://zoolstud.sinica.edu.tw/Journals/51.2/195.pdf

This study describes the sperm morphology of the mayfly Hexagenia (Pseudeatonica) albivitta (Ephemeroptera). Its spermatozoon measures approximately 30 μm of which 9 μm corresponds to the head. The head is composed of an approximately round acrosomal vesicle and a cylindrical nucleus. The nucleus has two concavities, one in the anterior tip, where the acrosomal vesicle is inserted and a deeper one at its base, where the flagellum components are inserted. The flagellum is composed of an axoneme, a mitochondrion and a dense rod adjacent to the mitochondrion. A centriolar adjunct is also observed surrounding the axoneme in the initial portion of the flagellum and extends along the flagellum for at least 2 μm, surrounding the axoneme in a half-moon shape. The axoneme is the longest component of the flagellum, and it follows the 9+9+0 pattern, with no central pair of microtubules. At the posterior region of the flagellum, the mitochondrion has a dumb-bell shape in cross sections that, together with the rectangular mitochondrial-associated rod, is responsible for the flattened shape of the flagellum. An internal membrane is observed surrounding both mitochondrion and its associated structure.

Bee males (drones) of stingless bees tend to congregate near entrances of conspecific nests, where they wait for virgin queens that initiate their nuptial flight. We observed that the Neotropical solitary wasp Trachypus boharti (Hymenoptera, Cabronidae) specifically preys on males of the stingless bee Scaptotrigona postica (Hymenoptera, Apidae); these wasps captured up to 50 males per day near the entrance of a single hive. Over 90% of the wasp attacks were unsuccessful; such erroneous attacks often involved conspecific wasps and worker bees. After the capture of non-male prey, wasps almost immediately released these individuals unharmed and continued hunting. A simple behavioral experiment showed that at short distances wasps were not specifically attracted to S. postica males nor were they repelled by workers of the same species. Likely, short-range prey detection near the bees' nest is achieved mainly by vision whereas close-range prey recognition is based principally on chemical and/or mechanical cues. We argue that the dependence on the wasp's visual perception during attack and the crowded and dynamic hunting conditions caused wasps to make many preying attempts that failed. Two wasp-density-related factors, wasp-prey distance and wasp-wasp encounters, may account for the fact that the highest male capture and unsuccessful wasp bee encounter rates occurred at intermediate wasp numbers.

Members of social insect colonies employ a large variety of chemical signals during their life. Of these, cuticular hydrocarbons are of primary importance for social insects since they allow for the recognition of conspecifics, nestmates and even members of different castes. The objectives of this study were (1) to characterize the variation of the chemical profiles among workers of the stingless bee Melipona marginata, and (2) to investigate the dependence of the chemical profiles on the age and on the behavior of the studied individuals. The results showed that cuticular hydrocarbon profiles of workers were composed of alkanes, alkenes and alkadienes that varied quantitatively and qualitatively according to function of workers in the colony. Chemical compounds on the cuticle are a rich source of information used during interactions among social insects. Despite the multitude of studies on these substances and their function in ants, wasps, and honeybees, little is known about this subject in stingless bees (Hymenoptera: Apidae, Meliponini). We studied the chemical composition of the cuticle of the stingless bee, Frieseomelitta varia, by gas chromatography-mass spectrometry (GC-MS), to investigate potential chemical variation among castes, gender, age, and reproductive status. We found differences in the cuticular hydrocarbon composition among workers, males, and queens, recording both qualitative and quantitative differences among individuals of different ages and gender. The cuticle of physogastric queens presented a chemical profile that was distinct from all other groups in the analysis, with high relative abundances of alkenes and alkadienes with 27, 29, and 31 carbon atoms. We discuss the possibility that these compounds signal a queen`s presence to the colony, thereby initiating all vital worker-queen interactions.

We undertook a field study to determine whether comb cell size affects the reproductive behavior of Varroa destructor under natural conditions. We examined the effect of brood cell width on the reproductive behavior of V. destructor in honey bee colonies, under natural conditions. Drone and worker brood combs were sampled from 11 colonies of Apis mellifera. A Pearson correlation test and a Tukey test were used to determine whether mite reproduction rate varied with brood cell width. Generalized additive model analysis showed that infestation rate increased positively and linearly with the width of worker and drone cells. The reproduction rate for viable mother mites was 0.96 viable female descendants per original invading female. No significant correlation was observed between brood cell width and number of offspring of V. destructor. Infertile mother mites were more frequent in narrower brood cells.

Muitas substâncias de origem vegetal podem ser tóxicas ou apresentar potencial inseticida. Com o objetivo de diminuir a problemática da poluição ambiental alguns estudos vêm tentando substituir os inseticidas artificiais pelos inseticidas botânicos. Ricinus communis (Euphorbiaceae) apresenta uma grande variedade de substâncias sendo a ricinina o principal componente tóxico. Considerando que as abelhas são insetos benéficos por atuarem como agentes polinizadores das plantas, este estudo teve por objetivo avaliar o efeito tóxico da ricinina para as operárias e larvas de Apis mellifera (Linnaeus, 1758) (Hymenoptera, Apidae) e Scaptotrigona postica (Latreille, 1907) (Hymenoptera, Meliponini). Para isso, foram realizados testes de ingestão em operárias confinadas recebendo ricinina incorporada à dieta e testes de aplicação tópica com a substância solubilizada em metanol e aplicada no pronoto das abelhas com auxílio de uma microseringa. Para as larvas foram realizados testes de ingestão e calculada sua taxa de mortalidade. Os resultados mostram atividade tóxica significativa (p < 0.0001) da ricinina nas abelhas adultas das duas espécies para a concentração 0,1% nos testes de ingestão e 0,2% nos testes de aplicação tópica. Outrossim, uma elevada porcentagem de larvas foi afetada negativamente pela ricinina logo nos primeiros dias de vida para todas as concentrações testadas (0,25%, 0,5% e 1%).
