To confer abscisic acid (ABA) and/or stress-inducible gene expression, an ABA-response complex (ABRC1) from the barley (Hordeum vulgare L.) HVA22 gene was fused to four different lengths of the 5′ region from the rice (Oryza sativa L.) Act1 gene. Transient assay of β-glucuronidase (GUS) activity in barley aleurone cells shows that, coupled with ABRC1, the shortest minimal promoter (Act1–100P) gives both the greatest induction and the highest level of absolute activity following ABA treatment. Two plasmids with one or four copies of ABRC1 combined with the same Act1–100P and HVA22(I) of barley HVA22 were constructed and used for stable expression of uidA in transgenic rice plants. Three Southern blot-positive lines with the correct hybridization pattern for each construct were obtained. Northern analysis indicated that uidA expression is induced by ABA, water-deficit, and NaCl treatments. GUS activity assays in the transgenic plants confirmed that the induction of GUS activity varies from 3- to 8-fold with different treatments or in different rice tissues, and that transgenic rice plants harboring four copies of ABRC1 show 50% to 200% higher absolute GUS activity both before and after treatments than those with one copy of ABRC1.

Maize (Zea mays L.) plants were grown to the nine-leaf stage. Despite a saturating N supply, the youngest mature leaves (seventh position on the stem) contained little NO3− reserve. Droughted plants (deprived of nutrient solution) showed changes in foliar enzyme activities, mRNA accumulation, photosynthesis, and carbohydrate and amino acid contents. Total leaf water potential and CO2 assimilation rates, measured 3 h into the photoperiod, decreased 3 d after the onset of drought. Starch, glucose, fructose, and amino acids, but not sucrose (Suc), accumulated in the leaves of droughted plants. Maximal extractable phosphoenolpyruvate carboxylase activities increased slightly during water deficit, whereas the sensitivity of this enzyme to the inhibitor malate decreased. Maximal extractable Suc phosphate synthase activities decreased as a result of water stress, and there was an increase in the sensitivity to the inhibitor orthophosphate. A correlation between maximal extractable foliar nitrate reductase (NR) activity and the rate of CO2 assimilation was observed. The NR activation state and maximal extractable NR activity declined rapidly in response to drought. Photosynthesis and NR activity recovered rapidly when nutrient solution was restored at this point. The decrease in maximal extractable NR activity was accompanied by a decrease in NR transcripts, whereas Suc phosphate synthase and phosphoenolpyruvate carboxylase mRNAs were much less affected. The coordination of N and C metabolism is retained during drought conditions via modulation of the activities of Suc phosphate synthase and NR commensurate with the prevailing rate of photosynthesis.

Transformed (cauliflower mosaic virus 35S promoter [35S]) tobacco (Nicotiana plumbaginifolia L.) plants constitutively expressing nitrate reductase (NR) and untransformed controls were subjected to drought for 5 d. Drought-induced changes in biomass accumulation and photosynthesis were comparable in both lines of plants. After 4 d of water deprivation, a large increase in the ratio of shoot dry weight to fresh weight was observed, together with a decrease in the rate of photosynthetic CO2 assimilation. Foliar sucrose increased in both lines during water stress, but hexoses increased only in leaves from untransformed controls. Foliar NO3− decreased rapidly in both lines and was halved within 2 d of the onset of water deprivation. Total foliar amino acids decreased in leaves of both lines following water deprivation. After 4 d of water deprivation no NR activity could be detected in leaves of untransformed plants, whereas about 50% of the original activity remained in the leaves of the 35S-NR transformants. NR mRNA was much more stable than NR activity. NR mRNA abundance increased in the leaves of the 35S-NR plants and remained constant in controls for the first 3 d of drought. On the 4th d, however, NR mRNA suddenly decreased in both lines. Rehydration at d 3 caused rapid recovery (within 24 h) of 35S-NR transcripts, but no recovery was observed in the controls. The phosphorylation state of the protein was unchanged by long-term drought. There was a strong correlation between maximal extractable NR activity and ambient photosynthesis in both lines. We conclude that drought first causes increased NR protein turnover and then accelerates NR mRNA turnover. Constitutive NR expression temporarily delayed drought-induced losses in NR activity. 35S-NR expression may therefore allow more rapid recovery of N assimilation following short-term water deficit.

A irrigação quando bem manejada, pode minimizar os riscos econômicos da atividade sucroalcooleira, particularmente em safras com presença de instabilidade climática onde a restrição hídrica, promovida pela diminuição no volume de chuvas, pode reduzir a produtividade dos canaviais. Dentre as ferramentas disponíveis para a gestão eficiente da água na agricultura irrigada, a técnica de irrigação sob déficit pode se tornar uma escolha acertada para a cana-de-açúcar, desde que sejam identificadas as fases fenológicas e épocas de cultivo onde a limitação da oferta de água não implique em reduções antieconômicas no rendimento da cultura. Diante disso, a hipótese que norteia essa pesquisa, é a de que existe uma estratégia de irrigação sob déficit, que associada a uma variedade com características específicas, possibilite a expressão de indicadores de produtividade tão satisfatórios quanto os obtidos em condições de irrigação plena. Nessa linha, os objetivos da pesquisa envolveram o estudo da dinâmica foliar, acúmulo e particionamento de biomassa e ainda, índices de produtividade da água para biomassa, açúcar e etanol de 1ª e 2ª geração de oito variedade de cana-de-açúcar, submetidas a diferentes condições de disponibilidade hídrica no solo em dois ciclos de cultivo (cana-planta e cana-soca). A pesquisa foi realizada na Escola Superior de Agricultura \"Luiz de Queiroz\", em Piracicaba/SP, onde foram estudados os dois primeiros ciclos de cultivo da cana-de-açúcar, sendo estes abordados nesta tese como Experimento 1 (cana-planta) e Experimento 2 (cana-soca). O delineamento experimental adotado para ambos os ciclos foi o de blocos casualizados, com três blocos completos. Os tratamentos foram compostos por três fatores em esquema de parcelas sub-subdivididas. Estas parcelas foram formadas por duas plantas (touceiras) alocadas em um vaso com aproximadamente 330 litros de solo. No Experimento 1, foram estudados três fatores, sendo o primeiro e segundo com quatro níveis e o terceiro com oito (4x4x8), totalizando assim 128 tratamentos, sendo eles: quatro níveis de irrigação ao longo do ciclo (125, 100, 75 e 50% da ETc); oito variedades comerciais de cana-de-açúcar e quatro procedimentos de maturação, impostos por meio de variações na intensidade do déficit hídrico aplicado. Para o Experimento 2, substitui-se o fator Maturação por Épocas de Corte, o qual consistiu em colheitas de um quarto do experimento a cada 90 dias. Os resultados encontrados apontaram que a área foliar responde positivamente a maior disponibilidade hídrica no solo, tendo sido verificado uma relação proporcional entre estes. Quanto ao acúmulo de biomassa, verificou-se que para as oito variedades estudadas houve incremento de biomassa a medida em que se aumentou o volume de água disponibilizado às variedades. No tocante ao particionamento, as folhas foram os drenos principais de fotoassimilados da planta até os 100 dias de cultivo, sendo que após este período, os colmos ocuparam o lugar de dreno preferencial. Os indicadores de produtividade da água apresentaram diferenças significativas para o fator lâmina, o que indica a existência de cultivares de cana-de-açúcar mais eficientes no uso da água. Por fim, observou-se que a produtividade da água para etanol total apresentou valores expressivos, com média para essa variável igual a 1,81 L m-3, o que denota o potencial de rendimento de etanol (1G + 2G) a partir da cana-de-açúcar quando é adotado o aproveitamento integral das plantas.

A cana-de-açúcar é uma espécie amplamente cultivada em regiões tropicais e subtropicais. Sua propagação é realizada através do plantio de porções caulinares contendo uma média de três gemas. Tal prática requer grande quantidade de material vegetal, o que reduz o ganho dos produtores. Adicionalmente, a utilização de grande quantidade de material vegetal para o plantio dificulta algumas práticas em relação ao manejo da cultura, como transporte e armazenamento. A utilização de mini-toletes, contendo uma única gema, representa uma alternativa ao plantio convencional. Existem limitações impostas à utilização de mini-toletes, relacionadas à baixa disponibilidade de reservas de nutrientes e de água, devido ao reduzido tamanho dos toletes. O presente trabalho teve o objetivo de avaliar o vigor e o desenvolvimento de plantas de cana-de-açúcar provenientes de mini-toletes. No primeiro experimento, comparou-se plantas provenientes de diferentes tipos de propágulos e gemas. Foram avaliados o número de brotações, a porcentagem de brotações, a altura das plantas e as massas de folhas e raízes. No segundo experimento, avaliou-se o efeito da aplicação de biorreguladores em mini-toletes provenientes de gemas apicais e basais. Foram realizadas as determinações do número de brotações, da altura das plantas, da área foliar e das massas secas de folhas e colmos. No terceiro experimento, avaliou-se a aplicação de ureia como fonte de adubação nitrogenada e tiametoxam, um inseticida sistêmico com ação bioativadora, no desenvolvimento de plantas originárias de mini-toletes. Realizaram-se as seguintes determinações: número de brotações, altura das plantas, área foliar e massas secas de colmos, folhas e raízes. No quarto experimento, plantas de cana-de-açúcar originárias de mini-toletes tratados com agroquímicos foram submetidas ao déficit hídrico. Foram avaliadas a altura das plantas, a área foliar e as massas de raiz, folha e caule. Foi possível concluir que plantas provenientes de gemas superiores e de toletes contendo três e duas gemas apresentaram um melhor desenvolvimento. De maneira geral, os resultados indicaram que a cana-de-açúcar não responde de maneira evidente ao uso de reguladores vegetais em mini-toletes. A utilização de ureia aumenta o desenvolvimento de plantas originárias de mini-toletes. Em conjunto com diferentes doses de ureia, a utilização de tiametoxam incrementa aspectos do desenvolvimento da cana-de-açúcar. Adicionalmente, foi possível concluir que a aplicação de agroquímicos em mini-toletes alivia os efeitos negativos do déficit hídrico no desenvolvimento radicular. A partir dos resultados obtidos no presente trabalho, foi possível concluir que a utilização de ureia, tiametoxam e agroquímicos melhora o desenvolvimento de plantas de cana-de-açúcar originárias de mini-toletes.

Este estudio pretende profundizar en el conocimiento sobre el funcionamiento de pequeños humedales mediterráneos, en estado de degradación, que, por razones históricas y socioeconómicas, han quedado confinados en un espacio urbano, olvidando la importancia ambiental de estas anomalías hídricas positivas en espacios sometidos a déficit hídrico. Este tipo de humedales, como el de Calpe (Alicante), suelen localizarse en áreas costeras de alta densidad urbana, lo que supone un importante valor añadido desde la ordenación y gestión territorial, ya que contribuyen a mantener la conectividad ambiental a una escala mucho más amplia, funcionando como auténticos reservorios de biodiversidad. Así, se ha llevado a cabo una aproximación holística, analizando el estado actual de la lámina de agua y del espacio circundante, donde se han incluido parámetros del medio físico, del medio biótico y los usos del suelo. Para ello se han combinado técnicas de muestreo de campo con el uso de GPS y sistemas de información geográfica (SIG). Una vez detectadas las amenazas, y estudiado el potencial ecológico del humedal, se ha desarrollado un plan de restauración y conservación de la zona con la pretensión de implicar a la población local para devolver a este espacio el protagonismo que merece. En definitiva, se pretende proporcionar las herramientas teóricas y técnicas necesarias para garantizar la conservación y gestión sostenible de los humedales costeros urbanos y sus recursos naturales.

La Huerta de Alicante, prototipo de los denominados regadíos deficitarios o secanos mejorados, ofrece uno de los mejores ejemplos de cómo la escasez hídrica activó la búsqueda de soluciones técnicas, propició la inversión en infraestructuras hidráulicas y desarrolló unas peculiares relaciones entre los labradores regantes y los propietarios del agua. La gestión de los recursos hídricos y su distribución por estricto cómputo horario por parte del municipio alicantino hasta 1739 permitió, pese a los condicionantes físicos y climáticos, el desarrollo de una actividad agrícola significativa durante los siglos modernos. La temprana separación del agua de la tierra a la que estaba adscrita en los primeros repartos medievales provocó numerosos conflictos a lo largo de la historia e intentos de unificar ambas propiedades, algo que nunca se consiguió.

. Separating continuously measured stem radius (SR) fluctuations into growth-induced irreversible stem expansion (GRO) and tree water deficit-induced reversible stem shrinkage (TWD) requires a concept to decide on potential growth processes during periods of shrinking and expanding SR below a precedent maximum. Here we investigated two physiological concepts: the linear growth (LG) concept assuming linear growth vs. the zero growth (ZG) concept assuming no growth during periods of shrunken stems. . We evaluated the physiological mechanisms underlying these two concepts and assessed the respective plausibility with SR data obtained from 15 deciduous and evergreen trees. . The LG concept showed steady growth rates, whereas the ZG concept showed strongly varying growth rates over time, more in accordance with mechanistic expectations. Further, growth increased for maximally 120 min after periods of shrunken stems, indicating limited growth activity during that period. However, the fraction of this extra growth was found to be small. Furthermore, TWD of the ZG concept was better explained by a hydraulic plant model than TWD of the LG concept. . We conclude that periods of shrunken stems allow for very little growth in the four tree species investigated. However, further studies should focus on obtaining independent growth data to ultimately validate these findings.

Abscisic acid (ABA) supplied in the vase solution can induce stomatal closure in the leaves of cut flowers, including roses (Rosa hybrida L.). This effect may be beneficial in reducing water deficit stress. Extracellular pH can affect active ABA concentrations in the apoplast of guard cells, with sap alkalisation enhancing the physiological activity of ABA. Accordingly, it was hypothesized that vase solution pH may affect ABA-mediated stomatal closure of cut roses. Two experiments were conducted to study the interaction of vase solution pH and ABA. In the first, cut 'Baccara' roses were held in vase solutions with +/- 10(-5) M ABA at pH 6, pH 7 and pH 8. In the second experiment, roses were held with +/- 10(-5) M ABA at pH 6 and pH 8 in the presence and absence of 1 mg l(-1) AgNO3 as a bactericide. Supply of ABA increased vase life and reduced vase solution usage of flowers held in low pH 6 solutions, indicating induction of stomatal closure. Conversely, ABA supplied at pH 8 was associated with reduced vase life. This negative result was associated with enhanced development of vase solution microbes at high pH, which overrode any potential pH-mediated ABA efficacy effects.

The recent summary report of a Department of Energy Workshop on Plant Systems Biology (P.V. Minorsky [2003] Plant Physiol 132: 404-409) offered a welcomed advocacy for systems analysis as essential in understanding plant development, growth, and production. The goal of the Workshop was to consider methods for relating the results of molecular research to real-world challenges in plant production for increased food supplies, alternative energy sources, and environmental improvement. The rather surprising feature of this report, however, was that the Workshop largely overlooked the rich history of plant systems analysis extending over nearly 40 years (Sinclair and Seligman, 1996) that has considered exactly those challenges targeted by the Workshop. Past systems research has explored and incorporated biochemical and physiological knowledge into plant simulation models from a number of perspectives. The research has resulted in considerable understanding and insight about how to simulate plant systems and the relative contribution of various factors in influencing plant production. These past activities have contributed directly to research focused on solving the problems of increasing biomass production and crop yields. These modeling approaches are also now providing an avenue to enhance integration of molecular genetic technologies in plant improvement (Hammer et al., 2002).

Screening for drought resistance of rainfed lowland rice using drought score (leaf death) as a selection index has a long history of use in breeding programs. Genotypic variation for drought score during the vegetative stage in two dry season screens was examined among 128 recombinant inbred lines from four biparental crosses. The genotypic variation detected for drought score in the dry season was used to examine the reliability of the dry season screening method to estimate relative grain yield of genotypes under different types of drought stress in the wet season. Large genotypic variation for drought score existed in two experiments (A and B). However, there was no relationship between the drought scores of genotypes determined in these two experiments. Different patterns of development and severity of drought stress in these two experiments, i.e. slow development and mild plant water deficit in experiment A and fast development and severe plant water deficit in experiment B, were identified as the major factors contributing to the genotypes responding differently. Larger drought score in the dry season experiments was associated with lower grain yield under specific drought stress conditions in the wet season, but the association was weak to moderate and significant only in particular drought conditions. In most cases, a significant phenotypic and moderate genetic correlation between drought score in the dry season and grain yield in the wet season existed only when both drought score and grain yield of genotypes were affected by similar patterns and severity of drought stress in their respective experimental environments. The dry season environments used to measure genotypic variation for drought score should be managed to correspond to relevant types of drought environment that are frequent in the wet season. The efficiency of using the drought score as an indirect selection criterion for improving grain yield for drought conditions was lower than the direct selection for grain yield, and hence wet season screening with grain yield as a selection criterion would be more efficient. However, using drought score as a selection index, a larger number of genotypes can be evaluated than for wet season grain yield. Therefore, it is possible to apply higher selection intensities using the drought score system, and the selected lines can be further tested for grain yield in the wet season. (C) 2004 Elsevier B.V. Drought is a major constraint for rice production in the rainfed lowlands in Southeast Asia and Eastern India. The breeding programs for tainted lowland rice in these regions focus on adaptation to a range of drought conditions. However, a method of selection of drought tolerant genotypes has not been established and is considered to be one of the constraints faced by rice breeders. Drought response index (DRI) is based on grain yield adjusted for variation in potential yield and flowering date, and has been used recently, but its consistency among drought environments and hence its usefulness is not certain. In order to establish a selection method and subsequently to identify donor parents for drought resistance breeding, a series of experiments with 15 contrasting genotypes was conducted under well-watered and managed drought conditions at two sites for 5 years in Cambodia. Water level in the field was recorded and used to estimate the relative water level (WLREL) around flowering as an index of the severity of water deficit at the time of flowering for each entry. This was used to determine if DRI or yield reduction was due to drought tolerance or related to the amount of available water at flowering, i.e. drought escape. Grain yield reduction due to drought ranged from 12 to 46%. The drought occurred mainly during the reproductive phase, while four experiments had water stress from the early vegetative stage. There was significant variation for water availability around flowering among the nine experiments and this was associated with variation in mean yield reduction. Genotypic variation in DRI was consistent among most experiments, and genotypic mean DRI ranged from -0.54 to 0.47 (LSD 5% = 0.47). Genotypic variation in DRI was not related to WLREL around flowering in the nine environments. It is concluded that selection for DRI under drought conditions would allow breeders to identify donor lines with high drought tolerance as an important component of breeding better adapted varieties for the rainfed lowlands; two genotypes were identified with high DRI and low yield reduction and were subsequently used in the breeding program in Cambodia. The mid-Holocene (6000 calibrated years before present) is a key period in palaeoclimatology because incoming summer insolation was lower than during the late Holocene in the Southern Hemisphere, whereas the opposite happened in the Northern Hemisphere. However, the effects of the decreased austral summer insolation over South American climate have been poorly discussed by palaeodata syntheses. In addition, only a few of the regional studies have characterised the mid-Holocene climate in South America through a multiproxy approach. Here, we present a multiproxy compilation of mid-Holocene palaeoclimate data for eastern South America. We compiled 120 palaeoclimatological datasets, which were published in 84 different papers. The palaeodata analysed here suggest a water deficit scenario in the majority of eastern South America during the mid-Holocene if compared to the late Holocene, with the exception of northeastern Brazil. Low mid-Holocene austral summer insolation caused a reduced land-sea temperature contrast and hence a weakened South American monsoon system circulation. This scenario is represented by a decrease in precipitation over the South Atlantic Convergence Zone area, saltier conditions along the South American continental margin, and lower lake levels.
