Mesenchymal stem cells (MSCs) are a heterogeneous population of cells that proliferate in vitro as plastic-adherent cells, have fibroblast-like morphology and can differentiate into bone, cartilage and fat cells. Therapeutic potential of MSCs have been studied in experimental models, such as rabbit, in Laboratory of Cell Engineering of Botucatu. However, no specific markers have been reported for expanded rabbit MSCs, which hampers the isolation of pure MSC populations by immunophenotypic characterization. Thus, the objective of this study was to produce monoclonal antibodies (mAbs) to rabbit MSCs. MSCs derived from rabbit bone marrow (BM) were isolated, cultured, expanded ex vivo, and immunized into three BALB/c mices, and spleen cells subsequently harvested were used to generate hibridoma cell lines secreting antibodies against MSCs. Hybridoma cells were screened by flow cytometry and antibody-producing cells were subjected to subsequent rounds of retests. MSC1-160 obtained the best positivity for IgG expression and was cloned by limiting dilutions and micromanipulation. Ascitic fluid from ten best clones was purified by affinity chromatography in Protein A-sepharose CL-4B column and purification control was performed by electrophoresis in agarose gels. The purified IgG were tested against rabbit MSCs, obtaining high positivity by flow Cytometry. In conclusion, we developed 10 mAbs, MSC1-160 A20, A30, A41, A47, A55, A60, A63, A69, A81, and A82, that recognize rabbit MSC cell surface antigens showing potential for immunophenotypic characterization of rabbit MSC cell lines

Introduction: Tuberculosis (TB) is a granulomatous disease caused by Mycobacterium tuberculosis. The genus Mycobacteriumhas two different complexes: M. tuberculosis Complex and M. avium Complex. This is a global health epidemic and remains a major global health problem, besides, the clinical severity of TB is significantly higher in transplanted patients. The detection of these mycobacteria complexes in transplanted patients, by molecular methods, is fundamental for quick treatment of patients and can contribute for rapid and accuracy of diagnosis. Objective: To detect mycobacteria DNA of M. tuberculosis and M. avium Complexes in formalin fixed paraffin-embedded samples (FFPE) of two patients groups: non transplanted and transplanted. Materials and Methods: The study includes 40 FFPE biopsies separated in four groups: NTP – presence of epithelioid granuloma and positive ZN, non-transplanted patients – 9 samples; NTN - presence of epithelioid granuloma and negative ZN, non-transplanted patients – 10 samples; TP – positive ZN, transplanted patients – 9 samples; TN – negative ZN, transplanted patients – 7 samples. Sections were cut for DNA extraction. Samples were submitted to PCR for amplification of: a) β-actin, b) IS6110 insertion and c) IS1245 insertion. DNA evaluation was made by spectrophotometry and efficiency and PCR analysis was made by agarose gels under UV light. Results: In all samples processed, 97.1% were positive for human β-actin gene. In22.2% of NTP group were found the IS6110 insertion sequencebut the IS1245 wasn´t. In the NTN group was not found any sequence. In theTP group, 11.1% of the samples were positive for IS6110 and also 11,1% werepositive for IS1245. In the TN group, 14.3% of the samples were positive forIS6110 and for IS1245, 14.3% was also positive. Junto ao Laboratório de Fracionamento de Proteínas, desenvolveu atividades aplicando técnicas eletroforéticas (gel de agarose, eletroforese nativa em gel de poliacrilamida) em amostras de músculos de aves de interesse comercial (frangos, perus e avestruzes), tendo como co-orientador o Prof. Paulo Tadeu Figueira da Faculdade de Medicina Veterinária, PUC/PR - Toledo, onde desenvolveu atividades de análise de géis de eletroforese. Aplicou as técnicas de eletroforeses citadas em amostras de músculos e soro sanguíneo (em andamento) de tilápias (Oreochromis niloticus), bem como desenvolveu atividades de análises computacionais (VDS – PHARMACIA) e estatísticas (ASSISTAT) nos dados obtidos, resultando na participação em 7 congressos com apresentação de trabalhos. Cursou a disciplina de Genética, como aluno especial, junto à turma de Licenciatura em Ciências Biológicas, num total de 8 créditos, sendo aprovado com média 7,2

The studies of this work aimed to determine the labile fractions of manganese (Mn) in natural and drainage water samples collected around the Osamu Utsumi uranium mine, located in the municipality of Caldas, south-central region of Poços de Caldas- MG, using the technique of diffusion gradient in thin films (DGT). The DGT devices were mounted with Chelex-100 resin, polyacrylamide-agarose hydrogel (conventional porosity) and cellulose acetate membrane. The device were deployed up to 48 hours in six water samples collected from different areas around the uranium mine (075, 076, 022-E, 025-E, 014, and 041). The DGT devices immersed in each sample were gradually removed after 4, 8, 12, 24 and 48 hours. The pH of the samples ranged from 3.0 to 10.5, which influenced the lability and the sampling of the analyte by the Chelex-100 resin. The results showed a linear relationship between accumulated mass and sampling time (immersion curve) for samples 014 and 025-E (pH between 6 and 8) suggesting the ability of the DGT technique for sampling the analyte. The results obtained for samples 075 and 076 (pH<5) and samples 041 and 022-E (pH around 10) were characterized by nonlinear relationships. The values obtained by DGT were compared with Solid Phase Extraction (SPE) technique using Chelex-100. For samples 014 and 025-E, there was a good agreement between the results obtained by both techniques

The DGT technique has been used to determine and better understand the dynamics and bioavailability of metals in the sea near to marine outfalls. In this work, the DGT was assessed for the determination of Cd, Pb, Cu, Zn and Ni in marine water samples from Potiguar Basin in its conventional aspect (binding agent, Chelex-100 and diffusive agent: Diffusive polyacrylamide gel (D) and restrictive (R)) and determination of Pb and Cd through its alternative aspect (binding agent: Saccharomyces cerevisiae immobilized in agarose gels and diffusive agent: 3MM Chromatography Paper). The deployment curves for long periods of immersion in the sample, showed a good linear correlation for the conventional aspect to Zn (D: R²=0.9586, R: R²=0.9444), Ni (D: R²=0.9789, R: R²=0.9286) and Cu (D: R²=0.764, R: R²=0.8143), and alternative to Pb (R²=0.9228) and Cd (R²=0.9673). The results of the organic and inorganic composition analysis in conventional aspect, showed that for every element there is no significant fraction of organic compounds in the sample. In addition, considering the alternative aspect, the comparison of obtained and expected masses suggests that some of the Pb is not labile and that the main retention mechanism of Pb for S. Cerevisiae occurs through ion exchange

O presente trabalho analisou os parâmetros genéticos populacionais de variabilidade e diversidade genética de diferentes populações de Ocotea notata (Nees & Mart.) Mez, através de marcadores de DNA do tipo ISSR (Inter Single Sequence Repeats). Para tanto, foram amostrados 243 indivíduos de 12 populações selecionadas na Bahia e Espírito Santo, que ocorrem em campos rupestres e vegetações de restinga, a saber, morfotipos de Ocotea glaucina (Meisn.) Mez: Morro do Chapéu: populações do Tabuleiro dos Guaribas, Ferro Doido, Cria Bode e Lajes; Umburanas; Jacobina; Lençóis: população da Serra das Paridas, e os morfotipos de O. notata, Esplanada: população de Baixios; Salvador: população das Dunas do Abaeté, Alcobaça, Mucuri, e Vila Velha, ES: população de Jacarenema. O DNA total já se encontrava extraído e quantificado em gel de agarose. Foram testados 20 primers de ISSR (University of British Columbia), dos quais quatro apresentaram resultados adequados para as análises, a saber: Manny, Mao, John e UBC 844. A otimização dos protocolos de reações de PCR foi feita no Laboratório de Evolução Molecular, da UNESP de Rio Claro, com a execução das reações de PCR para cada um dos primers, para cada população, e subsequentemente foram feitas as análises dos resultados sob o arcabouço teóricometodológico da genética de populações. A Análise de Variância Molecular (AMOVA) indicou que 23% da variabilidade ocorre entre populações dentro de regiões e 76% entre indivíduos dentro de populações, com variação significativa de 1% ocorrendo entre regiões (populações de Restinga vs. Campos Rupestres)

It is known that there is a wide variety of bioavailable trace metals in aquatic systems, and its determination is fundamental to predict impacts to organisms in these environments. However, the determination of the total concentration of chemicals in aquatic systems, despite its importance, does not provide necessary information for decision making or even may lead to misinterpretation of the procedures and potential risks to the system in question. This study aimed to use the technique of diffusion in thin films by concentration gradients (DGT) to evaluate the anion exchange membrane Whatman DE 81 as binder alternative in determining Cr (VI). Experiments were carried out to determine the diffusion coefficients for the material used diffusion (agarose gel). The behaviors of the binders were analyzed before the main variables of the systems, immersion time, pH and ionic strength. Then was made the assessment of potential interferences, to finally proceed with testing using actual samples in the laboratory and in situ

Lipases have important applications in biotechnological processes, motivating us to produce, purify, immobilize and perform a biochemical characterization of the lipase from Rhizomucor pusillus. The fungus was cultivated by solid state fermentation producing lipolytic activity of about 0.5 U/mL(4U/g). A partial purification by gel filtration chromatography in Se-phacryl S-100 allowed obtaining a yield of about 85% and a purification factor of 5.7. Our results revealed that the purified enzyme is very stable with some significant differences in its properties when compared to crude extract. The crude enzyme extract has an optimum pH and temperature of 7.5 ° C and 40 ° C, respectively. After purification, a shift of the optimum pH from 7 to 8 was observed, as well as a rise in optimumtemperature to 60 ° C and an increase in stability. The enzyme was immobilized on CNBr-Agarose and Octyl-Agarose supports, having the highest immobilization yield of 94% in the second resin. The major advantage of immobilization in hydrophobic media such as Octyl is in its hyper activation, which in this case was over 200%, a very interesting finding. Another advantage of this type of immobilization is the possibility of using the derivatives in biotechnological applications, such as in oil enriched with omega-3 as the results obtained in this study display the hydrolysis of 40% EPA and 7% DHA from sardine oil, promising results compared to the literature.

Sabendo-se do potencial de toxicidade do mercúrio, mais especificamente de sua forma orgânica, o metilmercúrio (MeHg), faz-se necessária a busca por métodos analíticos eficientes e precisos para a determinação e especiação do metal. A técnica de difusão por gradiente em filmes finos (DGT) já vem sendo utilizada para tal fim de maneira eficiente tendo apresentado resultados satisfatórios em diversos estudos. Porém, a utilização de ligantes alternativos aos convencionais, que tornem a aplicação da técnica menos custosa em termos laborais e financeiros, faz-se importante para um monitoramento constante e eficiente do metilmercúrio no ambiente. No presente trabalho buscou-se avaliar o potencial da levedura Saccharomyces cerevisiae imobilizada em agarose em combinação com a poliacrilamida como agente difusivo para determinação seletiva de MeHg. Os resultados demonstraram-se positivo, com um coeficiente de difusão médio de 7,03 ± 0,77 x 10-6 cm2 s-1. Os resultados demonstraram também baixa influencia do pH, força iônica e outros íons na retenção do analito pelos dispositivos. As recuperações de Hg2+ demonstraram-se insatisfatórios, evidenciando a seletividade dos dispositivos
