Im Rahmen dieser Arbeit konnte in dem marinen Schwamm Suberites domuncula ein Gen identifiziert werden, dessen C-terminale konservierte Domäne eine hohe Sequenzähnlichkeit zu den Zinkfingerdomänen der Nanos-Proteine aufweist. Weiter konnte ein N-terminales Sequenzmotiv identifiziert werden, das eine hohe Sequenzidentität zu den konservierten NIM Motiven (CNOT1-interagierendes-Motiv) von Nanos zeigt. Nach der Klonierung der cDNA erfolgte die Expression des als Sd_nrp bezeichneten Proteins in E. coli Bakterien, für dessen 231 Aminosäuren umfassende Polypeptidkette eine theoretische Molekülmasse von 25.8 kDa berechnet wurde. Anschließend gelang ein Nachweis des Proteins mithilfe eines polyklonalen, gegen Sd_nrp gerichteten Antikörpers in drei Gewebetypen, dem Pinacoderm, den Primmorphen (3D-Zellaggregate) und den Gemmulae (Dauerstadien der Schwämme). Dabei konnte die höchste Expression von Sd_nrp in den als totipotent geltenden Stammzellen der Schwämme, den Archaeocyten innerhalb der Gemmulae beobachtet werden. Die Identifizierung der Zellstrukturen, erfolgte dabei aufgrund morphologischer Vergleiche. Speziell die Merkmale der Zellen in den Gemmulae, der große Nukleus, die amöboide Form sowie die granulären Reservesubstanzen, entsprechen den typischen morphologischen Eigenschaften der Archaeocyten, und bestätigen die Interpretation der Ergebnisse. Weiter konnte mit Hilfe des Anti-Sd_nrp Antikörpers das native Protein in Proteinextrakten aus Gewebe adulter Tiere nachgewiesen werden. Die vergleichende Sequenzanalyse von Sd_nrp mit dem Nanos-verwandten Protein der Schwammspezies Ephydatia fluviatilis und die phylogenetische Stammbaum-Analyse mit Nanos-Homologen unterschiedlicher Invertebraten und Vertebraten lässt die Schlussfolgerung zu, dass es sich bei dem hier identifizierten Protein Sd_nrp um ein Nanos-verwandtes Protein handelt. Darüber hinaus konnte anhand eines Homologiemodells bestätigt werden, dass es sich bei der konservierten C-terminalen Domäne des Proteins Sd_nrp um die für Nanos-Proteine spezifische Zinkfingerstruktur mit dem konservierten Sequenzmotiv CCHC handelt. Dieses Ergebnis konnte auch bei einem Vergleich der Zinkfingerdomäne von Sd_nrp mit den Zinkfingerdomänen der Nanos-Homologen unterschiedlicher Invertebraten- und Vertebratenspezies bestätigt werden.

Philosophers and laymen alike have often used morality to invite misconceptions of human life into ethics, and also of ethics into human life. The Kant/Williams discourse provides a rich backdrop on which to consider these misconceptions. But the misconceptionsof morality involved are just as numerous and just as serious. One thing that the Kant/Williams discourse shows is this: that ethics can be neither contained by nor cultivated without morality. Though much of Williams’ critique of Kantian morality is quite astute, thephilosophical and ethical wisdoms of morality abound in spite of these. Morality understands the fundamental condition of moral loss, and the sometimes irreducible quandaries that this condition places human beings in. It understands the nature of the moral law, and theintricacies that the levels of letter and spirit invite into human life. Perhaps more importantly, it understands the uncompromising relationship between moral loss and moral law, and how the human navigation of this relationship leads into the ethical realm via giving rise to ethical conviction. Finally, for all of its pressures, morality abounds in valuable wisdoms for the one discovering that the human soul occupies a place of ethical significance in the world. It is responsible for pointing out, grounding and providing a framework for some of the most fundamental truths about the world and human beings; and these are essential to any viable ethical theory and sensible conception of human life.

Zunic investigated the relationship between nationalism and Serbian literature. He first analysed and evaluated the justifications for a number of critical accusations against Serbian literature. These included mythological and epic foundations (traditionalism), an obsession with national history and with the motif of the Serbian people as an eternal victim, the domination of the collective over the individual (populism), an anachronistic romantic conception of the social function of literature. In order to gain an unbiased judgement of the nationalistic role of contemporary Serbian literature, Zunic prepared a list of those books with the greatest number of copies issued in the decade 1985-1995, and constructed an appropriate hermeneutic procedure of understanding the meaning of the content and form of these works. He concluded that contemporary Serbian literature is in fact occupied with national history, and also with the unmasking of communist totalitarianism. The most influential books express and document either nationally-oriented or civil-oriented world views. The former, although mostly not militant works (with their realistic "closed" form), might have had an ideological influence on the Serbian national consciousness, while the civil-oriented works (with their "open" modern or post-modern form) could not neutralise all these extra-literary effects of the nationally-oriented works, since the predominant way of reading is a communication with the literary content (realised as a testimony of historical facts), and not with the literary form (as a carrier of the artistic value and a "moderator" of any content).

CopY of Enterococcus hirae is a well characterized copper-responsive repressor involved in copper homeostasis. In the absence of copper, it binds to the promoter. In high copper, the CopZ copper chaperone donates copper to CopY, thereby releasing it from the promoter and allowing transcription of the downstream copper homeostatic genes of the cop operon. We here show that the CopY-like repressors from E. hirae, Lactococcus lactis, and Streptococcus mutans have similar affinities not only for their native promoters, but also for heterologous cop promoters. CopZ of L. lactis accelerated the release of CopY from the promoter, suggesting that CopZ of L. lactis acts as copper chaperone, similar to CopZ in E. hirae. The consensus binding motif of the CopY-like repressors was shown to be TACAxxTGTA. The same binding motif is present in promoters controlled by BlaI of Bacillus licheniformis, MecI of Staphylococcus aureus and related repressors. BlaI and MecI have known structures and belong to the family of 'winged helix' proteins. In the N- terminal domain, they share significant sequence similarity with CopY of E. hirae. Moreover, they bind to the same TACAxxTGTA motif. NMR analysis of the N-terminal DNA binding domain of CopY of L. lactis showed that it contained the same alpha-helical content like the same regions of BlaI and MecI. These findings suggest that the DNA binding domains of CopY-like repressors are also of the 'winged helix' type.

A partir de los aportes críticos de Enrique Anderson Imbert y de Umberto Eco acerca de las posibles relaciones existentes entre los escritos de Borges y Chesterton es que, en el presente trabajo, me propongo estudiar el motivo del cuarto cerrado y su relación con el detective en Seis problemas para don Isidro Parodi de Honorio Bustos Domecq (seudónimo de Adolfo Bioy Casares y Jorge Luis Borges). En tal sentido, intento fundamentar la relación paródica que a nivel de procedimientos se establece con la saga del padre Brown de Gilbert R. Chesterton. Con tal finalidad, reseño el itinerario diacrónico que los tres elementos fundamentales de este tipo de relatos -el cuarto cerrado, el detective y la víctima-, han tenido a lo largo de la historia de la literatura de índole policial con el objetivo preciso de señalar la peculiar textualización de los mismos en la obra analizada. La originalidad del tratamiento de estos motivos típicos, tanto por parte de Biorges como por parte de Chesterton, ha evitado su posible petrificación en el tiempo como tópico literario.

En este trabajo mostramos que, si hubo un culto solar en las Etiópicas de Heliodoro, no es el de Émesa del siglo III d.C. de los Severos, sino el de la antigua Heliópolis egipcia, que provee la base de la astronomía antigua. El gran sacerdote del templo del Sol (Ra) de Heliópolis se llamaba, en hebreo, "Putifar"; en griego, "Heliodoros": extraordinaria coincidencia, ya que el motivo de la mujer de Putifar de Génesis 39.7-20 ocupa un lugar destacado en la trama de la novela, pero ésta atiende estrictamente a sus modelos griegos clásicos. Proponemos, además, que la composición de la obra coincide con o surge del Carmen Sæculare de Horacio.

Oxidation of molecular hydrogen catalyzed by [NiFe] hydrogenases is a widespread mechanism of energy generation among prokaryotes. Biosynthesis of the H2-oxidizing enzymes is a complex process subject to positive control by H2 and negative control by organic energy sources. In this report we describe a novel signal transduction system regulating hydrogenase gene (hox) expression in the proteobacterium Alcaligenes eutrophus. This multicomponent system consists of the proteins HoxB, HoxC, HoxJ*, and HoxA. HoxB and HoxC share characteristic features of dimeric [NiFe] hydrogenases and form the putative H2 receptor that interacts directly or indirectly with the histidine protein kinase HoxJ*. A single amino acid substitution (HoxJ*G422S) in a conserved C-terminal glycine-rich motif of HoxJ* resulted in a loss of H2-dependent signal transduction and a concomitant block in autophosphorylating activity, suggesting that autokinase activity is essential for the response to H2. Whereas deletions in hoxB or hoxC abolished hydrogenase synthesis almost completely, the autokinase-deficient strain maintained high-level hox gene expression, indicating that the active sensor kinase exerts a negative effect on hox gene expression in the absence of H2. Substitutions of the conserved phosphoryl acceptor residue Asp55 in the response regulator HoxA (HoxAD55E and HoxAD55N) disrupted the H2 signal-transduction chain. Unlike other NtrC-like regulators, the altered HoxA proteins still allowed high-level transcriptional activation. The data presented here suggest a model in which the nonphosphorylated form of HoxA stimulates transcription in concert with a yet unknown global energy-responsive factor.

The specificity of protein–protein interactions in cellular signaling cascades is dependent on the sequence and intramolecular location of distinct amino acid motifs. We used the two-hybrid interaction trap to identify proteins that can associate with the PDZ motif-rich segment in the protein tyrosine phosphatase PTP-BL. A specific interaction was found with the Lin-11, Isl-1, Mec-3 (LIM) domain containing protein RIL. More detailed analysis demonstrated that the binding specificity resides in the second and fourth PDZ motif of PTP-BL and the LIM domain in RIL. Immunohistochemistry on various mouse tissues revealed a submembranous colocalization of PTP-BL and RIL in epithelial cells. Remarkably, there is also an N-terminal PDZ motif in RIL itself that can bind to the RIL-LIM domain. We demonstrate here that the RIL-LIM domain can be phosphorylated on tyrosine in vitro and in vivo and can be dephosphorylated in vitro by the PTPase domain of PTP-BL. Our data point to the presence of a double PDZ-binding interface on the RIL-LIM domain and suggest tyrosine phosphorylation as a regulatory mechanism for LIM-PDZ associations in the assembly of multiprotein complexes. These findings are in line with an important role of PDZ-mediated interactions in the shaping and organization of submembranous microenvironments of polarized cells.

A 16-amino acid residue peptide derived from a consensus motif of natural ferredoxins incorporates a tetranuclear iron sulfur cluster under physiological conditions. Successful assembly of the [4Fe–4S]2+/1+ cluster within a monomeric peptide was demonstrated using size exclusion chromatography, UV-visible, visible CD, and cryogenic EPR spectroscopies. The robustness of [4Fe–4S]2+/1+ formation was tested using peptides with either the ligating cysteine exchanged for alanine or with the intervening amino acids replaced by glycine. The small size of the peptide allows for modular incorporation into more complex protein structures. In one larger structure, we describe a tetra-α-helix bundle that self-assembles both iron–sulfur clusters and hemes, thereby demonstrating feasibility for the general synthesis of maquettes containing multiple, juxtaposed redox cofactors. This is a motif common to the catalytic sites of native oxidoreductases.

The RegA proteins from the bacteriophage T4 and RB69 are translational repressors that control the expression of multiple phage mRNAs. RegA proteins from the two phages share 78% sequence identity; however, in vivo expression studies have suggested that the RB69 RegA protein binds target RNAs with a higher affinity than T4 RegA protein. To study the RNA binding properties of T4 and RB69 RegA proteins more directly, the binding sites of RB69 RegA protein on synthetic RNAs corresponding to the translation initiation region of two RB69 target genes were mapped by RNase protection assays. These assays revealed that RB69 RegA protein protects nucleotides –9 to –3 (relative to the start codon) on RB69 gene 44, which contains the sequence GAAAAUU. On RB69 gene 45, the protected site (nucleotides –8 to –3) contains a similar purine-rich sequence: GAAAUA. Interestingly, T4 RegA protein protected the same nucleotides on these RNAs. To examine the specificity of RNA binding, quantitative RNA gel shift assays were performed with synthetic RNAs corresponding to recognition elements (REs) in three T4 and three RB69 mRNAs. Comparative gel shift assays demonstrated that RB69 RegA protein has an ∼7-fold higher affinity for T4 gene 44 RE RNA than T4 RegA protein. RB69 RegA protein also binds RB69 gene 44 RE RNA with a 4-fold higher affinity than T4 RegA protein. On the other hand, T4 RegA exhibited a higher affinity than RB69 RegA protein for RB69 gene 45 RE RNA. With respect to their affinities for cognate RNAs, both RegA proteins exhibited the following hierarchy of affinities: gene 44 > gene 45 > regA. Interestingly, T4 RegA exhibited the highest affinity towards RB69 gene 45 RE RNA, whereas RB69 RegA protein had the highest affinity for T4 gene 44 RE RNA. The helix–loop groove RNA binding motif of T4 RegA protein is fully conserved in RB69 RegA protein. However, homology modeling of the structure of RB69 RegA protein reveals that the divergent residues are clustered in two areas of the surface, and that there are two large areas of high conservation near the helix–loop groove, which may also play a role in RNA binding.

Aminoacyl-tRNA synthetases (tRNA synthetases) of higher eukaryotes form a multiprotein complex. Sequence elements that are responsible for the protein assembly were searched by using a yeast two-hybrid system. Human cytoplasmic isoleucyl-tRNA synthetase is a component of the multi-tRNA synthetase complex and it contains a unique C-terminal appendix. This part of the protein was used as bait to identify an interacting protein from a HeLa cDNA library. The selected sequence represented the internal 317 amino acids of human bifunctional (glutamyl- and prolyl-) tRNA synthetase, which is also known to be a component of the complex. Both the C-terminal appendix of the isoleucyl-tRNA synthetase and the internal region of bifunctional tRNA synthetase comprise repeating sequence units, two repeats of about 90 amino acids, and three repeats of 57 amino acids, respectively. Each repeated motif of the two proteins was responsible for the interaction, but the stronger interaction was shown by the native structures containing multiple motifs. Interestingly, the N-terminal extension of human glycyl-tRNA synthetase containing a single motif homologous to those in the bifunctional tRNA synthetase also interacted with the C-terminal motif of the isoleucyl-tRNA synthetase although the enzyme is not a component of the complex. The data indicate that the multiplicity of the binding motif in the tRNA synthetases is necessary for enhancing the interaction strength and may be one of the determining factors for the tRNA synthetases to be involved in the formation of the multi-tRNA synthetase complex.

In a search for regulatory proteins that interact with the leucine zipper motif of c-Fos in the yeast two-hybrid screen, we have identified a protein (FZA-B) that has extensive sequence similarity to SUG1 of Saccharomyces cerevisiae. Here we show that FZA-B can functionally substitute for SUG1 in yeast and that FZA-B interacts with Fos proteins in vitro through their leucine zippers. In rat liver and in HeLa cells, FZA-B is present in the 26S proteasome complex, as is c-Fos. Immobilized antibody raised against an FZA-B-specific peptide depleted peptidase activity, proteasomal proteins, FZA-B, and c-Fos from a 26S proteasome preparation. FZA-B is found predominantly in the nuclear fraction of COS cells expressing an FZA-B transgene and in the nuclear 26S proteasome of HeLa cells. We conclude that FZA-B is the mammalian homolog of SUG1 (mSug1) and that it is present in the nuclear 26S proteasome of cells. Our results suggest that mSug1 may be involved in the degradation of c-Fos and other transcription factors.

Gonadotropin-releasing hormone (GnRH) is known and named for its essential role in vertebrate reproduction. Release of this decapeptide from neurons in the hypothalamus controls pituitary gonadotropin levels which, in turn, regulate gonadal state. The importance of GnRH is underscored by its widespread expression and conservation across vertebrate taxa: five amino acids are invariant in all nine known forms, whereas two others show only conservative changes. In most eutherian mammals, only one form, expressed in the hypothalamus, is thought to exist, although in a recent report, antibody staining in developing primates suggests an additional form. In contrast, multiple GnRH forms and expression loci have been reported in many non-mammalian vertebrates. However, evidence based on immunological discrimination does not always agree with analysis of gene expression, since GnRH forms encoded by different genes may not be reliably distinguished by antibodies. Here we report the expression of three distinct GnRH genes in a teleost fish brain, including the sequence encoding a novel GnRH preprohormone. Using in situ hybridization, we show that this form is found only in neurons that project to the pituitary and exhibit changes in soma size depending on social and reproductive state. The other two GnRH genes are expressed in other, distinct cell populations. All three genes share the motif of encoding a polypeptide consisting of GnRH and a GnRH-associated peptide. Whereas the GnRH moiety is highly conserved, the GnRH-associated peptides are not, reflecting differential selective pressure on different parts of the gene. GnRH forms expressed in nonhypothalamic regions may serve to coordinate reproductive activities of the animal.
