This work focuses on the design of high-efficient DC-DC converters based on WBG power devices. The first objective is the development of an isolated bidirectional converter for the distribution network of future electrical aircrafts. A SiC-based Dual Active Bridge converter is designed and fabricated. Control strategies for individual and parallel operations are investigated and implemented into a FPGA platform. Experimental results on 1.2kW 270V/28V prototype are presented to confirm the proper behavior of the proposed solution. The second project belongs to the field of photovoltaic systems and aims to develop a three-port converter with multiple power elements interfacing capability. A GaN-based Triple Active Bridge has been designed, regarding both the controller and the hardware realization.

Az írás célja, hogy rámutasson: egy táj természetitőke-ellátottsága meghatározható annak vízháztartási alapfolyamatai alapján. Az ökoszisztéma-szolgáltatások értékelésének a tájhasználati különbségekből érdemes kiindulnia, ehhez nyújt első lépésben közelítést a természetitőke-ellátottságról nyerhető információ. A cikk gondolatmenete a Kárpát-medence területére alkalmazza a Millennium Ecosystem Assessment (MEA) és a The Economics of Ecosystems and Biodiversity (TEEB) kutatások során kialakított elemzési keretet, a ökoszisztéma-szolgáltatások és a természeti tőke szemléletmódját. Az alkalmazott példa segítségével továbblép a természeti tőke értelmezésében, rámutat annak szoros kapcsolatára a kis-vízkör táji folyamatával. Ez a kapcsolat lehetőséget nyújt arra, hogy egy táj vízháztartási folyamatairól rendelkezésre álló információk alapján elhelyezzük a táj jelenlegi, megvalósított állapotát a természeti adottságaiból fakadó, potenciális természetitőke-szinthez képest. A Tisza magyarországi vízgyűjtőjére alkalmazva a vízháztartás alapú összevetést kiderül, hogy a jelenlegi természetitőke-szint a megújuló természeti erőforrások biztosította potenciál felén áll. A szélsőséges vízháztartási helyzetek gyakoribbá válása jelentős részben ezen alacsony természetitőke-szintből fakadó hiányjelenség, és emellett nagyok a helyzetből fakadó haszonlehetőség-költségek (elszalasztott lehetőségek) is. E kedvezőtlen helyzetből a területhasználat differenciálását lehetővé tevő és azt egyben igénylő tájfenntartási-szolgáltatások (vízkárelhárítás, területi vízgazdálkodás) alkalmazkodásán keresztül lehet továbblépni. Ezt az elkerülhetetlen folyamatot akkor lehet a legolcsóbban véghezvinni, ha mind a területhasználat, mind az azt kiszolgáló tájfenntartási rendszerek alkalmazkodása során figyelembe veszik a természetitőke-szint emelésének követelményét.

Miniature direct methanol fuel cells (DMFCs) are promising micro power sources for portable appliction. Low temperature cofired ceramic (LTCC), a competitive technology for current MEMS based fabrication, provides cost-effective mass manufacturing route for miniature DMFCs. Porous silver tape is adapted as electrodes to replace the traditional porous carbon electrodes due to its compatibility to LTCC processing and other electrochemical advantages. Electrochemical evaluation of silver under DMFCs operating conditions demonstrated that silver is a good electrode for DMFCs because of its reasonable corrosion resistance, low passivating current, and enhanced catalytic effect. Two catalyst loading methods (cofiring and postfiring) of the platinum and ruthenium catalysts are evaluated for LTCC based processing. The electrochemical analysis exhibits that the cofired path out-performs the postfiring path both at the anode and cathode. The reason is the formation of high surface area precipitated whiskers. Self-constraint sintering is utilized to overcome the difficulties of the large difference of coefficient of thermal expansion (CTE) between silver and LTCC (Dupont 951) tape during cofiring. The graphite sheet employed as a cavity fugitive insert guarantees cavity dimension conservation. Finally, performance of the membrane electrode assembly (MEA) with the porous silver electrode in the regular graphite electrode based cell and the integrated cofired cell is measured under passive fuel feeding condition. The MEA of the regular cell performs better as the electrode porosity and temperature increased. The power density of 10 mWcm-2 was obtained at ambient conditions with 1M methanol and it increased to 16 mWcm -2 at 50°C from an open circuit voltage of 0.58V. For the integrated prototype cell, the best performance, which depends on the balance methanol crossover and mass transfer at different temperatures and methanol concentrations, reaches 1.13 mWcm-2 at 2M methanol solution at ambient pressure. The porous media pore structure increases the methanol crossover resistance. As temperature increased to 60°C, the device increases to 2.14 mWcm-2.

Deep-water coral ecosystems are hot spots of biodiversity and provide habitats and refuges for several deep-sea species. However, their role in shaping the biodiversity of the surrounding open slopes is still poorly known. We investigated how meiofaunal biodiversity varies with and is related to the occurrence of deep-water living scleractinian corals and coral rubble in two deep-sea areas (the Rockall Bank, northeastern Atlantic) and the Santa Maria di Leuca (central Mediterranean). In both areas, replicated sampling on alive and dead coral areas and from the adjacent slope sediments without corals (at the same and increasing depths) allowed us to demonstrate that sediments surrounding the living corals and coral rubble were characterised by higher meiofaunal biodiversity (as number of higher taxa, and nematode species richness) than the slope sediments. Despite the soft sediments surrounding the living coral having a higher nutritional value than those not associated with corals, with the opposite seen for coral rubble, the presence of both alive and dead corals had a significant effect on nematode assemblages. Our data suggest that, due particularly to the effects on habitat heterogeneity/complexity, both living coral and coral rubble promoted higher biodiversity levels than in surrounding slope sediments. We conclude that the protection of deep-water corals can be crucial to preserve the biodiversity of surrounding open slopes, and that the protection of dead corals, a so-far almost neglected habitat in terms of biological conservation, can further contribute to the maintenance of a high deep-sea biodiversity along continental margins.

Wind- induced exposure is one of the major forces shaping the geomorphology and biota in coastal areas. The effect of wave exposure on littoral biota is well known in marine environments (Ekebon et al., 2003; Burrows et al., 2008). In the Cabrera Archipelago National Park wave exposure has demostrated to have an effect on the spatial distribution of different stages of E.marginatus (Alvarez et al., 2010). Standarized average wave exposures during 2008 along the Cabrera Archipelago National park coast line were calculated to be applied in studies of littoral species distribution within the archipelago. Average wave exposure (or apparent wave power) was calculated for points located 50 m equidistant on the coastline following the EXA methodology (EXposure estimates for fragmented Archipelagos) (Ekebon et al., 2003). The average wave exposures were standardized from 1 to 100 (minimum and maximum in the area), showing coastal areas with different levels of mea wave exposure during the year. Input wind data (direction and intensity) from 2008 was registered at the Cabrera mooring located north of Cabrera Archipelago. Data were provided by IMEDEA (CSIC-UIB, TMMOS http://www.imedea.uib-csic.es/tmoos/boyas/). This cartography has been developed under the framework of the project EPIMHAR, funded by the National Park's Network (Spanish Ministry of Environment, Maritime and Rural Affairs, reference: 012/2007 ). Human genetics has been experiencing a wave of genetic discoveries thanks to the development of several technologies, such as genome-wide association studies (GWAS), whole-exome sequencing, and whole genome sequencing. Despite the massive genetic discoveries of new variants associated with human diseases, several key challenges emerge following the genetic discovery. GWAS is known to be good at identifying the locus associated with the patient phenotype. However, the actually causal variants responsible for the phenotype are often elusive. Another challenge in human genetics is that even the causal mutations are already known, the underlying biological effect might remain largely ambiguous. Functional evaluation plays a key role to solve these key challenges in human genetics both to identify causal variants responsible for the phenotype, and to further develop the biological insights from the disease-causing mutations.

We adopted various methods to characterize the effects of variants identified in human genetic studies, including patient genetic and phenotypic data, RNA chemistry, molecular biology, virology, and multi-electrode array and primary neuronal culture systems. Chapter 1 is a broader introduction for the motivation and challenges for functional evaluation in human genetic studies, and the background of several genetics discoveries, such as hepatitis C treatment response, in which we performed functional characterization.

Chapter 2 focuses on the characterization of causal variants following the GWAS study for hepatitis C treatment response. We characterized a non-coding SNP (rs4803217) of IL28B (IFNL3) in high linkage disequilibrium (LD) with the discovery SNP identified in the GWAS. In this chapter, we used inter-disciplinary approaches to characterize rs4803217 on RNA structure, disease association, and protein translation.

Chapter 3 describes another avenue of functional characterization following GWAS focusing on the novel transcripts and proteins identified near the IL28B (IFNL3) locus. It has been recently speculated that this novel protein, which was named IFNL4, may affect the HCV treatment response and clearance. In this chapter, we used molecular biology, virology, and patient genetic and phenotypic data to further characterize and understand the biology of IFNL4. The efforts in chapter 2 and 3 provided new insights to the candidate causal variant(s) responsible for the GWAS for HCV treatment response, however, more evidence is still required to make claims for the exact causal roles of these variants for the GWAS association.

Chapter 4 aims to characterize a mutation already known to cause a disease (seizure) in a mouse model. We demonstrate the potential use of multi-electrode array (MEA) system for the functional characterization and drug testing on mutations found in neurological diseases, such as seizure. Functional characterization in neurological diseases is relatively challenging and available systematic tools are relatively limited. This chapter shows an exploratory research and example to establish a system for the broader use for functional characterization and translational opportunities for mutations found in neurological diseases.

Overall, this dissertation spans a range of challenges of functional evaluations in human genetics. It is expected that the functional characterization to understand human mutations will become more central in human genetics, because there are still many biological questions remaining to be answered after the explosion of human genetic discoveries. The recent advance in several technologies, including genome editing and pluripotent stem cells, is also expected to make new tools available for functional studies in human diseases.

Feed can easily be contaminated and colonized by fungi that use up the nutrients for their own metabolism and growth, producing secondary metabolites such as mycotoxins that are not eliminated throughout the feed processing. The major problems associated with mycotoxin contaminated animal feed are metabolic disturbances resulting in poor animal productivity. In addition, handling contaminated animal feed can also raise health issues regarding workers exposure to fungi and mycotoxins. The scope of this work was to characterize fungal distribution in 11 poultry feed samples. Twenty grams of feed were suspended in 180 mL of distilled water and homogenized during 20 minutes at 200 rpm. The washed supernatant was plated in malt extract agar (MEA) and dichloran glycerol agar base (DG18) media for morphological identification of the mycobiota present. Using macro- and microscopic analysis of the colonies, fungal contamination was evident in 72.7% of the analyzed poultry feed samples. Fungal load ranged from 0 to 13140 CFU/g, and the most prevalent species/genera were F. graminearum complex (71.1%), Penicillium sp. (11.6%), Cladosporium sp. (8.8%), and Fusarium poae (3.6%). In addition to these species, we also isolated Aspergillus sections Circumdati, Nigri and Aspergilli, and Mucor and Rhizopus genus albeit at a lower abundance. The data obtained showed that, besides high fungal contamination, mycotoxins contamination is probably a reality, particularly in the final product since mycotoxins resist to all the processing operations including thermal treatment. Additionally, data claimed attention for the probable co-exposure to fungi and mycotoxins of the workers in feed industries.

The presence of microorganisms in ophthalmic instruments and surfaces can lead to the exposure of patients to several infections. However, there is no information regarding fungal and bacteria contamination in optical shops. This study aims to characterize fungi and bacteria contamination in air and surfaces from 10 optical shops covering also ophthalmic instruments. Air samples were collected through an impaction method onto malt extract agar (MEA) supplemented with chloramphenicol (0.05%) used for fungi and Tryptic Soy Agar (TSA) supplemented with nystatin (0.2%) used for bacteria. Outdoor samples were also performed to be used as reference. Surface and equipment’s swab samples were also collected side-by-side. All the collected samples were incubated at 27ºC for 5 to 7 days (fungi) or at 30º for 7 days (bacteria). Regarding fungal distribution, thirteen different species/genera were found in the air, being the most common Alternaria sp. (62.0%). Eight different species/genera were identified in the surfaces, ranging from 2 to 5x104 CFU/m2, being the most common A. versicolor complex and Penicillium sp. (40.0%). The trial frames were the most contaminated equipment, since 50.0% of the collected samples were with countless colonies. The airborne bacterial population indicated higher concentrations in the contactology office (average: 133 CFU/m3) than in the client’s waiting rooms (average: 126 CFU/m3). The surface samples indicated bacterial concentrations ranging from 2x104 to 1x106 CFU/m2, pointing out the automatic refractometer as the surface with higher bacterial load.

El presente trabajo de tesis tuvo como finalidad evaluar la flora micológica del maíz seco y su harina producidos en la parroquia San Juan - Cantón Gualaceo con el fin de aportar con información sobre la calidad de estos alimentos. Además se evaluaron los siguientes factores para el desarrollo de la flora micológica: temperatura, actividad acuosa, almacenamiento; así como también el crecimiento en distintos medios de cultivos y el uso de la desinfección del grano como parte de la técnica de siembra. La detección, recuento y aislamiento de hongos se realizó de acuerdo a la técnica de recuento en placa por siembra en profundidad para la harina y en el caso del grano por la técnica de siembra directa, con y sin desinfección de la superficie del grano utilizándose el agar MEA (Agar Extracto de Malta), PDA (Agar Papa Dextrosa) y DRBC (Agar Rosa de Bengala Diclorán) y sometiendo a dos tratamientos térmicos (18°C vs 25°C). Se encontró que la micoflora presente en las muestras analizadas corresponde a los géneros Penicillium, Fusarium, Rhizopus, Aspergillus y levaduras, siendo el género predominante Penicillium tanto en el grano como en la harina. La temperatura óptima para el crecimiento fue 25°C; y a su vez el crecimiento fue mejor sin desinfectar las superficies del grano. El crecimiento micológico varió dependiendo del medio, siendo PDA el más óptimo para Penicillium y Rhizopus, mientras que para Fusarium y Aspergillus no hubo diferencia. Igualmente se encontró que la actividad acuosa influyó en forma proporcional al crecimiento micológico y que el almacenamiento controlado influyó positivamente en su disminución.

O presente estudo pretende avaliar a exposição ocupacional a contaminação fúngica e bacteriana em quartos de hotel, mais precisamente em dois quartos com características diferentes, nomeadamente, com pavimento em alcatifa e outro sem alcatifa. Doze amostras de ar de 250L foram colhidas pelo método de impacto, em meio agar de extracto de malte (MEA) suplementado com cloranfenicol (0,05%) para fungos e em meio de TSA (agar de soja tríptica) com nistatina (0,2%) para bactérias. Foram também realizadas amostras de superfície nos mesmos locais. Em ambos os quartos apenas uma amostra de ar, no quarto sem alcatifa, apresentou contagens de fungos mais elevadas do que no exterior. No entanto, as concentrações de bactérias no ar interior foram superiores às do ar exterior. Em relação às superfícies, o quarto sem alcatifa apresentou diferenças estatisticamente significativas em relação ao quarto com alcatifa, sendo que o primeiro apresentou concentrações mais elevadas de fungos. Todas as superfícies analisadas apresentaram contaminação bacteriana, mas não houve diferenças estatisticamente significativas entre os quartos. Os géneros de fungos mais prevalentes no ar foram idênticos em ambos os quartos (Penicillium sp. 40,7% - 12,3% e Cladosporium sp. 43,5% - 55,4%). Nas superfícies analisadas, os isolados pertencentes ao complexo Aspergillus fumigatus foram os únicos encontrados no quarto com alcatifa, enquanto no outro quarto os géneros mais prevalentes foram Penicillium sp. (63,6%) e Aspergillus sp. (13,6%).

Study developed in order to know the carpet influence when used in the floor of a hotel room. Twelve air samples of 250L (six in a room with carpet and six more in a room with wood floor) were collected through an impaction method with a flow rate of 140 L/min onto malt extract agar (MEA) supplemented with chloramphenicol (0.05%), using the Millipore air Tester (Millipore), during cleaning activities. Outdoor sample was also performed to be used as a reference. Surface samples from floor and desks, taken at the same time, were collected by the swabbing method. to 7 days. Besides fungal contamination, we also assessed particulate matter contamination in both rooms during the same cleaning tasks. In the analyzed sur- faces, isolates belonging to Aspergillus fumigatus complex were the only fungi found in the carpeted room, whereas in the other room we found Penicllium sp. (63.6%) and Aspergillus sp. (13.6%) as the most frequent genera. In the case of particles the room with carpet obtained significant higher values for both metrics (PMC and PNC), showing that carpet may has influence on particles’ contamination of the room.

Miniaturization of power generators to the MEMS scale, based on the hydrogen-air fuel cell, is the object of this research. The micro fuel cell approach has been adopted for advantages of both high power and energy densities. On-board hydrogen production/storage and an efficient control scheme that facilitates integration with a fuel cell membrane electrode assembly (MEA) are key elements for micro energy conversion. Millimeter-scale reactors (ca. 10 µL) have been developed, for hydrogen production through hydrolysis of CaH2 and LiAlH4, to yield volumetric energy densities of the order of 200 Whr/L. Passive microfluidic control schemes have been implemented in order to facilitate delivery, self-regulation, and at the same time eliminate bulky auxiliaries that run on parasitic power. One technique uses surface tension to pump water in a microchannel for hydrolysis and is self-regulated, based on load, by back pressure from accumulated hydrogen acting on a gas-liquid microvalve. This control scheme improves uniformity of power delivery during long periods of lower power demand, with fast switching to mass transport regime on the order of seconds, thus providing peak power density of up to 391.85 W/L. Another method takes advantage of water recovery by backward transport through the MEA, of water vapor that is generated at the cathode half-cell reaction. This regulation-free scheme increases available reactor volume to yield energy density of 313 Whr/L, and provides peak power density of 104 W/L. Prototype devices have been tested for a range of duty periods from 2-24 hours, with multiple switching of power demand in order to establish operation across multiple regimes. Issues identified as critical to the realization of the integrated power MEMS include effects of water transport and byproduct hydrate swelling on hydrogen production in the micro reactor, and ambient relative humidity on fuel cell performance.

La Geometría del Espacio es una rama de la Matemática que estudia las propieda-des y medidas de figurasque se relacionan con la mayoría de objetos tridimensionales que tenemos a nuestro alrededor; por esto nuestro trabajo titulado “Elaboración de una guía y material didáctico de la Geometría del Espacio para el Laboratorio de Matemática de la carrera de Matemáticas y Física de la Universidad de Cuenca” da lugar a una nueva estrategia que puede utilizar el docente en el proceso de enseñanza-aprendizaje de esta asignatura. En el capítulo uno de nuestro trabajo de graduación se analizan los aspectos generales de la educación así como las corrientes pedagógicas que están presentes dentro del proceso educativo, para luego hablar de la didáctica y la importancia de trabajar con material concreto en el área de Matemática, específicamente en la Geometría del Espacio así como los recursos que son óptimos para trabajar esta asignatura. En el capítulo dos, se demuestra mediante un muestreo no probabilístico que existe dificultad en la comprensión de los contenidos de la Geometría del Espacio y que una buena opción para desarrollar el proceso de enseñanza-aprendizaje sobre esta asignatura es la utilización de material concreto y de una guía didáctica que facilite la comprensión de los contenidos. Por último en el capítulo tres se presenta un conjunto de diez prácticas sobre pla-nos y sólidos, las cuales, siguen los pasos que exige una práctica de laboratorio innovadora, de una manera ordenada y secuencial para reforzar el proceso de enseñanza-aprendizaje.
