Lipid rafts are highly ordered membrane domains rich in cholesterol and sphingolipids that provide a scaffold for signal transduction proteins; altered raft structure has also been implicated in cancer progression. We have shown that 25 mu M 10-(octyloxy) decyl-2-(trimethylammonium) ethyl phosphate (ODPC), an alkylphospholipid, targets high cholesterol domains in model membranes and induces apoptosis in leukemia cells but spares normal hematopoietic and epithelial cells under the same conditions. We performed a quantitative (SILAC) proteomic screening of ODPC targets in a lipid-raft-enriched fraction of leukemic cells to identify early events prior to the initiation of apoptosis. Six proteins, three with demonstrated palmitoylation sites, were reduced in abundance. One, the linker for activation of T-cell family member 2 (LAT2), is an adaptor protein associated with lipid rafts in its palmitoylated form and is specifically expressed in B lymphocytes and myeloid cells. Interestingly, LAT2 is not expressed in K562, a cell line more resistant to ODPC-induced apoptosis. There was an early loss of LAT2 in the lipid-raft-enriched fraction of NB4 cells within 3 h following treatment with 25 mu M ODPC. Subsequent degradation of LAT2 by proteasomes was observed. Twenty-five mu M ODPC inhibited AKT activation via myeloid growth factors, and LAT2 knockdown in NB4 cells by shRNA reproduced this effect. LAT2 knockdown in NB4 cells also decreased cell proliferation and increased cell sensitivity to ODPC (7.5X), perifosine (3X), and arsenic trioxide (8.5X). Taken together, these data indicate that LAT2 is an early mediator of the anti-leukemic activity of alkylphospholipids and arsenic trioxide. We recently demonstrated that Angiotensin-(3-4) [Ang-(3-4)], an Ang II-derived dipeptide, overcomes inhibition of plasma membrane Ca2+-ATPase promoted by nanomolar concentrations of Ang II in basolateral membranes of renal proximal tubule cells, with involvement of a so far unknown AT(2)R-dependent and NO-independent mechanism. The present study investigates the signaling pathway triggered by Ang-(3-4) that is responsible for counteracting the inhibitory effect of Ang II, and attempts to elucidate the functional interaction of the dipeptide with Ang II at the level of AT(2)R. Stimulation by cholera toxin of G(s)alpha protein structurally linked to AT(2)R as revealed by their co-immunoprecipitation mimicked the effect of Ang-(3-4) on Ca2+-ATPase activity. Furthermore, addition of dibutyril-cAMP (db-cAMP) mimicked Ang-(3-4), whereas the specific PKA inhibitor, PKAi((5-24)) peptide, suppressed the counter-regulatory effect of Ang-(3-4) and the AT(2)R agonist, CGP42112A. Membrane-associated PKA activity was stimulated by Ang-(3-4) or CGP42112A to comparable levels as db-cAMP, and the Ang-(3-4) effect was abrogated by the AT(2)R antagonist PD123319, whereas the AT(1)R antagonist Losartan had no effect. Ang-(3-4) stimulated PKA-mediated phosphorylation of Ca2+-ATPase and activated PKA to comparable levels. Binding assays demonstrated that Ang-(3-4) could not displace H-3-Ang II from HEK 293T cells expressing AT(2)R, but 10(-10) mol/L Ang-(3-4) resulted in the appearance of a probable higher-affinity site (picomolar range) for Ang II. The results presented herein demonstrate that Ang-(3-4), acting as an allosteric enhancer, suppresses Ang II-mediated inhibition of Ca2+-ATPase through an AT(2)R/cAMP/PKA pathway, after inducing conformational changes in AT(2)R that results in generation of higher-affinity sites for Ang II. We examined the interaction of the cationic antimicrobial peptide (AMP) tritrpticin (VRRFPWWWPFLRR, TRP3) with Langmuir monolayers of zwitterionic (dipalmitoyl phosphatidylcholine, DPPC, and dipalmitoyl phosphatidylethanolamine, DPPE) and negatively charged phospholipids (dipalmitoyl phosphatidic acid, DPPA, and dipalmitoyl phosphatidylglycerol, DPPG). Both surface pressure and surface potential isotherms became more expanded upon addition of TRP3 (DPPE similar to DPPC << DPPA < DPPG). The stronger interaction with negatively charged phospholipids agrees with data for vesicles and planar lipid bilayers, and with AMPs greater activity against bacterial membranes versus mammalian cell membranes. Considerable expansion of negatively charged monolayers occurred at 10 and 30 mol% TRP3, especially at low surface pressure. Moreover, a difference was observed between PA and PG, demonstrating that the interaction, besides being modulated by electrostatic interactions, displays specificity with regard to headgroup, being more pronounced in the case of PG, present in large quantities in bacterial membranes. In previous studies, it was proposed that the peptide acts by a toroidal pore-like mechanism [1,2]. Considering the evidence from the literature that PG shows a propensity to form a positive curvature as do toroidal pores, the observation of TRP3's preference for the PG headgroup and the dramatic increase in area promoted by this interaction represent further support for the toroidal pore mechanism of action proposed for TRP3. The recent biomedical applications of natural rubber (NR) latex, mostly in dry membranes, have motivated research into novel, more noble uses of this low-cost biomaterial. In this article, we provide the first report on the fabrication of layer-by-layer (LbL) films of NR alternated with the polyelectrolytes polyethylenimine (PEI) and polyallylamine hydrochloride (PAH). Stable (PAH/NR)n and (PEI/NR)n LbL films displayed similar physicochemical properties, but differed in terms of film morphology according to atomic force microscopy (AFM) and scanning electron microscopy (SEM) data. Most significantly, (PEI/NR)5 LbL films were made of smaller and flattened particles, which were not efficient for the growth and proliferation of normal human fibroblasts (NHF). In contrast, efficient NHF proliferation could be obtained with (PAH/NR)n LbL films, with the fibroblasts exhibiting the expected elongated morphology. Furthermore, cell growth did not occur for cast films of NR, thus demonstrating the suitability of the LbL method for this biologically related application. The differences between the two polyelectrolytes illustrate the importance of the film architecture and morphology, which open the way for exploiting the molecular control inherent in the LbL technique for further applications of NR-containing films. O estresse oxidativo, decorrente de uma atividade física, leva a peroxidação lipídica de membranas celulares, além de danos protéicos e em ácidos nucléicos, e um dos produtos finais desta reação é o malondialdeído (MDA). A glutationa reduzida (GSH), considerada um antioxidante multifuncional, está presente no plasma e principalmente nas hemácias e tem importância pelo fato de ser um dos índices da capacidade total antioxidante do corpo após um estresse oxidativo. Com o objetivo de avaliar o estresse oxidativo em diferentes condições de treinamento físico, determinaram-se a concentração de MDA sérico e GSH eritrocitária em 45 cavalos da raça American Trotter e mestiços divididos em três grupos: G1 (sem treinamento), G2 (até 6 meses de treinamento) e G3 (treinamento há mais de 12 meses). Observou-se que o MDA teve um valor significativamente menor no grupo de animais sem treinamento físico. Não houve diferença estatística significante para GSH corrigida pela Hb e para GSH corrigida pelo VG entre os grupos analisados, mas houve uma aparente tendência a maiores valores no G2, no qual o sistema antioxidante está em fase de adaptação ao treinamento físico constante e suas consequentes injúrias. Conclui-se que a atividade física acarreta danos celulares frente ao estresse oxidativo, mas o sistema antioxidante tem papel fundamental nesta homeostasia observando uma adaptação às injúrias causadas pelos radicais livres.

In biological world, life of cells is guaranteed by their ability to sense and to respond to a large variety of internal and external stimuli. In particular, excitable cells, like muscle or nerve cells, produce quick depolarizations in response to electrical, mechanical or chemical stimuli: this means that they can change their internal potential through a quick exchange of ions between cytoplasm and the external environment. This can be done thanks to the presence of ion channels, proteins that span the lipid bilayer and act like switches, allowing ionic current to flow opening and shutting in a stochastic way. For a particular class of ion channels, ligand-gated ion channels, the gating processes is strongly influenced by binding between receptive sites located on the channel surface and specific target molecules. These channels, inserted in biomimetic membranes and in presence of a proper electronic system for acquiring and elaborating the electrical signal, could give us the possibility of detecting and quantifying concentrations of specific molecules in complex mixtures from ionic currents across the membrane; in this thesis work, this possibility is investigated. In particular, it reports a description of experiments focused on the creation and the characterization of artificial lipid membranes, the reconstitution of ion channels and the analysis of their electrical and statistical properties. Moreover, after a chapter about the basis of the modelling of the kinetic behaviour of ligand gated ion channels, a possible approach for the estimation of the target molecule concentration, based on a statistical analysis of the ion channel open probability, is proposed. The fifth chapter contains a description of the kinetic characterisation of a ligand gated ion channel: the homomeric α2 isoform of the glycine receptor. It involved both experimental acquisitions and signal analysis. The last chapter represents the conclusions of this thesis, with some remark on the effective performance that may be achieved using ligand gated ion channels as sensing elements.

The work of this thesis has been focused on the characterization of metallic membranes for the hydrogen purification from steam reforming process and also of perfluorosulphonic acid ionomeric (PFSI) membranes suitable as electrolytes in fuel cell applications. The experimental study of metallic membranes was divided in three sections: synthesis of palladium and silver palladium coatings on porous ceramic support via electroless deposition (ELD), solubility and diffusivity analysis of hydrogen in palladium based alloys (temperature range between 200 and 400 °C up to 12 bar of pressure) and permeation experiments of pure hydrogen and mixtures containing, besides hydrogen, also nitrogen and methane at high temperatures (up to 600 °C) and pressures (up to 10 bar). Sequential deposition of palladium and silver on to porous alumina tubes by ELD technique was carried out using two different procedures: a stirred batch and a continuous flux method. Pure palladium as well as Pd-Ag membranes were produced: the Pd-Ag membranes’ composition is calculated to be close to 77% Pd and 23% Ag by weight which was the target value that correspond to the best performance of the palladium-based alloys. One of the membranes produced showed an infinite selectivity through hydrogen and relatively high permeability value and is suitable for the potential use as a hydrogen separator. The hydrogen sorption in silver palladium alloys was carried out in a gravimetric system on films produced by ELD technique. In the temperature range inspected, up to 400°C, there is still a lack in literature. The experimental data were analyzed with rigorous equations allowing to calculate the enthalpy and entropy values of the Sieverts’ constant; the results were in very good agreement with the extrapolation made with literature data obtained a lower temperature (up to 150 °C). The information obtained in this study would be directly usable in the modeling of hydrogen permeation in Pd-based systems. Pure and mixed gas permeation tests were performed on Pd-based hydrogen selective membranes at operative conditions close to steam-reforming ones. Two membranes (one produced in this work and another produced by NGK Insulators Japan) showed a virtually infinite selectivity and good permeability. Mixture data revealed the existence of non negligible resistances to hydrogen transport in the gas phase. Even if the decrease of the driving force due to polarization concentration phenomena occurs, in principle, in all membrane-based separation systems endowed with high perm-selectivity, an extensive experimental analysis lack, at the moment, in the palladium-based membrane process in literature. Moreover a new procedure has been introduced for the proper comparison of the mass transport resistance in the gas phase and in the membrane. Another object of study was the water vapor sorption and permeation in PFSI membranes with short and long side chains was also studied; moreover the permeation of gases (i.e. He, N2 and O2) in dry and humid conditions was considered. The water vapor sorption showed strong interactions between the hydrophilic groups and the water as revealed from the hysteresis in the sorption-desorption isotherms and thermo gravimetric analysis. The data obtained were used in the modeling of water vapor permeation, that was described as diffusion-reaction of water molecules, and in the humid gases permeation experiments. In the dry gas experiments the permeability and diffusivity was found to increase with temperature and with the equivalent weight (EW) of the membrane. A linear correlation was drawn between the dry gas permeability and the opposite of the equivalent weight of PFSI membranes, based on which the permeability of pure PTFE is retrieved in the limit of high EW. In the other hand O2 ,N2 and He permeability values was found to increase significantly, and in a similar fashion, with water activity. A model that considers the PFSI membrane as a composite matrix with a hydrophilic and a hydrophobic phase was considered allowing to estimate the variation of gas permeability with relative humidity on the basis of the permeability in the dry PFSI membrane and in pure liquid water.

Ionic Liquids (ILs) constituted by organic cations and inorganic anions are particular salts with a melting point below 100°C. Their physical properties such as melting point and solubility can be tuned by altering the combination of their anions and cations. In the last years the interest in ILs has been centered mostly on their possible use as “green” alternatives to the traditional volatile organic solvents (VOCs) thanks to their low vapour pressure and the efficient ability in catalyst immobilization. In this regard, the subject of the present thesis is the study of the oxodiperoxomolybdenum catalyzed epoxidation of olefins in ILs media with hydrogen peroxide as the oxidant. In particular N-functionalized imidazolium salts, such as 1-(2-t-Butoxycarbonylamino-ethyl)-3-methylimidazolium (1), were synthesized with different counterions [I]-, [PF6]-, [NO3]-, [NTf2]- and [ClO4]– and tested as reaction solvents. The counterion exchange with [Cl]-, [NTf2]- and [NO3]- was also performed in unfuctionalized imidazolium salts such as 3-butyl-1-methylimidazol-3-ium (3). All the prepared ILs were tested in catalytic epoxidation of olefins exploiting oxodiperoxomolybdenum complexes [MoO(O2)2(C4H6N2)2] (4) and [MoO(O2)2(C5H8N2)2] (5) as catalysts. The IL 3[NTf2] and the catalysts 5 give rise to the best results leading to the selective formation of the epoxide of cis-cyclooctene avoiding hydrolysis side reaction. A preliminary study on the synthesis of novel NHC oxodiperoxomolybdenum complexes starting from imidazolium salts was also developed.

Escherichia coli α-Hämolysin (HlyA) ist ein Prototyp der RTX-Toxine, die zu den α-porenbildenden Toxinen gehören. HlyA bildet Poren in einer Vielzahl eukaryontischer Zielzellen. Das 107 kDa große Protein besteht aus 1024 Aminosäuren, die gemeinsam mit den Proteinen für posttranslationale Modifikation und Sekretion in einem Operon codiert werden. Die N-terminale Hälfte von HlyA besteht aus mehreren amphipathischen α –Helices, die mit der Porenbildung assoziiert werden, gefolgt von der Calcium-bindenden RTX-Domäne in der C-terminalen Hälfte des Moleküls. Über den porenbildenen Mechanismus ist wenig bekannt. Die vorliegende Arbeit fokussierte sich auf die Frage, ob dieser Prozess eine Oligomerisierung mehrerer HlyA-Moleküle beinhaltet, oder ob die membranschädigende Struktur von einem Monomer gebildet wird. Drei unabhängige biochemische Methoden wurden in dem Versuch eingesetzt, HlyA-Oligomere in permeabilisierten Membranen zu detektieren. In allen drei Ansätzen wurden negative Ergebnisse erreicht, was das Konzept bestätigt, dass die Pore von HlyA von einem Monomer gebildet wird. PCR-basierte Cysteinsubstitutionen wurden durchgeführt, um den N-terminus von HlyA zu charakterisieren. Einzelne Cysteinreste wurden an 21 Positionen innerhalb der Aminosäuresequenz 13-55 eingeführt, und mit dem umgebungssensitiven Fluorophor Badan markiert. Spektrofluorimetrische Messungen zeigten, dass alle untersuchten Aminosäuren innerhalb dieser Domäne unabhängig von der porenbildenden Aktivität in die Membran inserieren. Deletionen der Aminosäuren 1-50 hatten keinen Einfluß auf die lytische Aktivität, während die Deletion der Aminosäuren 1-100 in einer fast vollständig inaktiven Toxinmutante resultierte. Die Einführung von Prolinen durch PCR-basierte Mutagenese wurde durchgeführt, um die Beteiligung vorhergesagter α-Helices innerhalb der N-terminalen Hälfte von HlyA an der hämolytischen Aktivität zu untersuchen. Die Ergebnisse deuten darauf hin, dass die Struktur von mindestens vier vorhergesagten Helices bedeutend für die hämolytische Aktivität ist.

Membranen spielen eine essentielle Rolle bei vielen wichtigen zellulären Prozessen. Sie ermöglichen die Erzeugung von chemischen Gradienten zwischen dem Zellinneren und der Umgebung. Die Zellmembran übernimmt wesentliche Aufgaben bei der intra- und extrazellulären Signalweiterleitung und der Adhäsion an Oberflächen. Durch Prozesse wie Endozytose und Exozytose werden Stoffe in oder aus der Zelle transportiert, eingehüllt in Vesikel, welche aus der Zellmembran geformt werden. Zusätzlich bietet sie auch Schutz für das Zellinnere. Der Hauptbestandteil einer Zellmembran ist die Lipiddoppelschicht, eine zweidimensionale fluide Matrix mit einer heterogenen Zusammensetzung aus unterschiedlichen Lipiden. In dieser Matrix befinden sich weitere Bausteine, wie z.B. Proteine. An der Innenseite der Zelle ist die Membran über Ankerproteine an das Zytoskelett gekoppelt. Dieses Polymernetzwerk erhöht unter anderem die Stabilität, beeinflusst die Form der Zelle und übernimmt Funktionenrnbei der Zellbewegung. Zellmembranen sind keine homogenen Strukturen, je nach Funktion sind unterschiedliche Lipide und Proteine in mikrsokopischen Domänen angereichert.Um die grundlegenden mechanischen Eigenschaften der Zellmembran zu verstehen wurde im Rahmen dieser Arbeit das Modellsystem der porenüberspannenden Membranen verwendet.Die Entwicklung der porenüberspannenden Membranen ermöglicht die Untersuchung von mechanischen Eigenschaften von Membranen im mikro- bis nanoskopischen Bereich mit rasterkraftmikroskopischen Methoden. Hierbei bestimmen Porosität und Porengröße des Substrates die räumliche Auflösung, mit welcher die mechanischen Parameter untersucht werdenrnkönnen. Porenüberspannende Lipiddoppelschichten und Zellmembranen auf neuartigen porösen Siliziumsubstraten mit Porenradien von 225 nm bis 600 nm und Porositäten bis zu 30% wurden untersucht. Es wird ein Weg zu einer umfassenden theoretischen Modellierung der lokalen Indentationsexperimente und der Bestimmung der dominierenden energetischen Beiträge in der Mechanik von porenüberspannenden Membranen aufgezeigt. Porenüberspannende Membranen zeigen eine linear ansteigende Kraft mit zunehmender Indentationstiefe. Durch Untersuchung verschiedener Oberflächen, Porengrößen und Membranen unterschiedlicher Zusammensetzung war es für freistehende Lipiddoppelschichten möglich, den Einfluss der Oberflächeneigenschaften und Geometrie des Substrates, sowie der Membranphase und des Lösungsmittels auf die mechanischen Eigenschaften zu bestimmen. Es ist möglich, die experimentellen Daten mit einem theoretischen Modell zu beschreiben. Hierbei werden Parameter wie die laterale Spannung und das Biegemodul der Membran bestimmt. In Abhängigkeit der Substrateigenschaften wurden für freitragende Lipiddoppelschichten laterale Spannungen von 150 μN/m bis zu 31 mN/m gefunden für Biegemodulde zwischen 10^(−19) J bis 10^(−18) J. Durch Kraft-Indentations-Experimente an porenüberspannenden Zellmembranen wurde ein Vergleich zwischen dem Modell der freistehenden Lipiddoppelschichten und nativen Membranen herbeigeführt. Die lateralen Spannungen für native freitragende Membranen wurden zu 50 μN/m bestimmt. Weiterhin konnte der Einfluss des Zytoskeletts und der extrazellulä-rnren Matrix auf die mechanischen Eigenschaften bestimmt und innerhalb eines basolateralen Zellmembranfragments kartiert werden, wobei die Periodizität und der Porendurchmesser des Substrates das räumliche Auflösungsvermögen bestimmen. Durch Fixierung der freistehenden Zellmembran wurde das Biegemodul der Membran um bis zu einem Faktor 10 erhöht. Diese Arbeit zeigt wie lokal aufgelöste, mechanische Eigenschaften mittels des Modellsystems der porenüberspannenden Membranen gemessen und quantifiziert werden können. Weiterhin werden die dominierenden energetischen Einflüsse diskutiert, und eine Vergleichbarkeit zurnnatürlichen Membranen hergestellt.rn

This dissertation deals with two specific aspects of a potential hydrogen-based energy economy, namely the problems of energy storage and energy conversion. In order to contribute to the solution of these problems, the structural and dynamical properties of two promising materials for hydrogen storage (lithium imide/amide) and proton conduction (poly[vinyl phosphonic acid]) are modeled on an atomistic scale by means of first principles molecular dynamics simulation methods.rnrnrnIn the case of the hydrogen storage system lithium amide/imide (LiNH_2/Li_2NH), the focus was on the interplay of structural features and nuclear quantum effects. For these calculations, Path-Integral Molecular Dynamics (PIMD) simulations were used. The structures of these materials at room temperature were elucidated; in collaboration with an experimental group, a very good agreement between calculated and experimental solid-state 1H-NMR chemical shifts was observed. Specifically, the structure of Li_2NH features a disordered arrangement of the Li lattice, which was not reported in previous studies. In addition, a persistent precession of the NH bonds was observed in our simulations. We provide evidence that this precession is the consequence of a toroid-shaped effective potential, in which the protons in the material are immersed. This potential is essentially flat along the torus azimuthal angle, which might lead to important quantum delocalization effects of the protons over the torus.rnrnOn the energy conversion side, the dynamics of protons in a proton conducting polymer (poly[vinyl phosphonic acid], PVPA) was studied by means of a steered ab-initio Molecular Dynamics approach applied on a simplified polymer model. The focus was put on understanding the microscopic proton transport mechanism in polymer membranes, and on characterizing the relevance of the local environment. This covers particularly the effect of water molecules, which participate in the hydrogen bonding network in the material. The results indicate that these water molecules are essential for the effectiveness of proton conduction. A water-mediated Grotthuss mechanism is identified as the main contributor to proton conduction, which agrees with the experimentally observed decay on conductivity for the same material in the absence of water molecules.rnrnThe gain in understanding the microscopic processes and structures present in this materials can help the development of new materials with improved properties, thus contributing to the solution of problems in the implementation of fuel cells.
