Surface pollen assemblages and their relationhips with the modern vegetation and climate provide a foundation for investigating palaeo-environment conditions by fossil pollen analysis. A promising trend of palynology is to link pollen data more closely with ecology. In this study, I summarized the characteristics of surface pollen assemblages and their quantitative relation with the vegetation and climate of the typical ecological regions in northern China, based on surface pollen analysis of 205 sites and investigating of modern vegetation and climate. The primary conclusions are as follows:The differences in surface pollen assemblages for different vegetation regions are obvious. In the forest communities, the arboreal pollen percentages are more than 30%, herbs less than 50% and shrubs less than 10%; total pollen concentrations are more than 106 grains/g. In the steppe communities, arboreal pollen percentages are generally less than 5%; herb pollen percentages are more than 90%, and Artemisia and Chenopodiaceae are dominant in the pollen assemblages; total pollen concentrations range from 103 to 106 grains/g. In the desert communities, arboreal pollen percentages are less than 5%. Although Chenopodiaceae and Artemisia still dominate the pollen assemblages, Ephedra, Tamaricaceae and Nitraria are also significant important in the pollen assemblages; total pollen concentrations are mostly less than 104grains/g. In the sub-alpine or high and cold meadow communities, arboreal pollen percentages are less than 30%. and Cyperaceae is one of the most significant-taxa in the pollen assemblages. In the shrub communities, the pollen assemblages are consistent with the zonal vegetation; shrub pollen percentages are mostly less than 20%, except for Artemisia and Hippophae rhamnoides communities.There are obvious trends for the pollen percentage ratios of Artemisia to Chenopodiaceae (A/C), Pinus to Artemisia (P/A) and arbor to non-arbor (AP/NAP) in the different ecological regions. In the temperate deciduous broad-leaved forest region, the P/A ratios are generally higher than 0.1, the A/C ratios higher than 2 and the AP/NAP ratios higher than 0.3. In the temperate steppe regions, the P/A ratios are generally less than 0.1, the A/C ratios higher than 1 and the AP/NAP ratios less than 0.1. In the temperate desert regions, the P/A ratios are generally less than 0.1, the A/C ratios less than 1, and the AP/NAP ratios less than 0.1.The study on the representation and indication of pollen to vegetation shows that Pinus, Artemisia, Betula, Chenopodiaceae, Ephedra, Selaginella sinensis etc. are over-representative in the pollen assemblages and can only indicate the regional vegetation. Some pollen types, such as Quercus, Carpinus, Picea, Abies, Elaeagus, Larix, Salix, Pterocelis, Juglans, Ulmus, Gleditsia, Cotinus, Oleaceae, Spiraea, Corylus, Ostryopsis, Vites, Tetraena, Caragana, Tamaricaceae, Zygophyllum, Nitraria, Cyperaceae, Sanguisorba etc. are under-representative in the pollen assemblages, and can indicate the plant communities well. Populus, Rosaceae, Saxifranaceae, Gramineae, Leguminosae, Compositae, Caprifoliaceae etc. can not be used as significant indicators to the plants.The study on the relation of pollen percentages with plant covers shows that Pinus pollen percentages are more than 30% where pine trees exist in the surrounding region. The Picea+Abies pollen percentages are higher than 20% where the Picea+Abies trees are dominant in the communities, but less than 5% where the parent plants are sparse or absent. Larix pollen percentages vary from 5% to 20% where the Larix trees are dominant in the communities, but less than 5% where the parent plants are sparse or absent. Betula pollen percentages are higher than 40% where the Betula trees are dominant in the communities" but less than 5% where the parent plants are sparse or absent. Quercus pollen percentages are higher than 10% where the Quercus trees are dominant in the communities, but less than 1% where the parent plants sparse or absent. Carpinus pollen percentages vary from 5% to 15% where the Carpinus trees are dominant in the communities, but less than 1% where the parent plants are sparse or absent. Populus pollen percentages are about 0-5% at pure Populus communities, but cannot be recorded easily where the Populus plants mixed with other trees in the communities. Juglans pollen accounts for 25% to 35% in the forest of Juglans mandshurica, but less than 1% where the parent plants are sparse or absent. Pterocelis pollen percentages are less than 15% where the Pterocelis trees are dominant in the communities, but cannot be recorded easily where the parent plants are sparse or absent. Ulmus pollen percentages are more than 8% at Ulmus communities, but less than 1% where the Ulmus plants mixed with other trees in the communities. Vitex pollen percentages increase along with increasing of parent plant covers, but the maximum values are less than 10 %. Caragana pollen percentages are less than 20 % where the Caragana plant are dominant in the communities, and cannot be recorded easily where the parent plants are sparse or absent. Spiraea pollen percentages are less than 16 % where the Spiraea plant are dominant in the communities, and cannot be recorded easily where the parent plants are sparse or absent.The study on the relation of surface pollen assemblages with the modern climate shows that, in the axis 1 of DCA, surface samples scores have significant correlation with the average annual precipitations, and the highest determination coefficient (R2) is 0.8 for the fitting result of the third degree polynomial functions. In the axis 2 of DCA, the samples scores have significant correlation with the average annual temperatures, average July temperatures and average January temperatures, and the determination coefficient falls in 0.13-0.29 for the fitting result of the third degree polynomial functions with the highest determination coefficient for the average July temperature.The sensitivity of the different pollen taxa to climate change shows that some pollen taxa such as Pinus, Quercus, Carpinus, Juglans, Spiraea, Oleaceae, Gramineae, Tamariaceae and Ephedra are only sensitive to the change in precipitation.

Performance of comprehensive two-dimensional liquid chromatography system is greatly improved than we reported previously by using a silica monolithic column as for the second dimensional separation. Due to the increase of the elution speed on the second dimensional monolithic column, the first dimensional column efficiency and analysis rate can be greatly improved as comparing with conventionally second dimensional column. The developed system was applied to analysis of methanol extraction of two umbelliferae herbs Ligusticum chuanxiong Hort. and Angelica sinensis (Oliv.) Diels by using CN column as for the first dimensional separation and a silica monolithic ODS column for the second dimensional separation, and the obtained three-dimensional chromatograms were treated by normalization of peak heights with the value of the highest peak or setting a certain value using a software written in-house. It was observed that much more peaks for low-abundant components in TCM extract can clearly be detected here than we reported before, due to the large difference for the amount of components in TCMs' extract. With the above improvements in separation performance and data treatment, totally about 120 components in methanol extraction of Rhizoma chuanxiong and 100 in A. sinensis were separated with UV detection within 130 min. This result meant that both the number of peaks detected increase twice but the analysis time decease twice if comparing with the previously reported result. (c) 2005 Published by Elsevier B.V.

Immobilized liposome chromatography (ILC), the stationary phase of which has been regarded as a mimic biomembranes system was used to separate and analyze compounds interacting with liposome membrane in Danggui Buxue decoction, a combined prescription of traditional Chinese medicines (CPTCMs), and its compositions Radix Astragli and Radix Angelica Sinensis. More than 10 main peaks in the extract of Danggui Buxue decoction were resolved on the ILC column, suggesting that more than 10 components in the prescription have significant retention on ILC column. Ligustilide, astragaloside, TV and formononetin, three main bioactive ingredients in Danggui Buxue decoction, were found to have relatively significant, while ferulic acid, another bioactive ingredient in the prescription, relatively weak retention on ILC column. Effects of the eluent pH and amount of immobilized phosphatidylcholine (PC) on separation of interactional compounds in the extract of Danggui Buxue decoction were also investigated. It was found that these two factors strongly affected the retention of some interactional compounds. In addition, the fractions partitioned with different solvents from water extract of this combined prescription were evaluated with this ILC column system. (c) 2005 Elsevier B.V. 1. The changes in the composition and distribution of the plankton of the southern North Sea have been investigated month by month, from June 1932 to December 1937; the present report deals with the phytoplankton. The survey was carried out by the Continuous Plankton Recorder, towed at a standard depth of 10 metres, by ships on regular steamship lines across the North Sea from Hull towards the Skagerrak, to Bremen and to Rotterdam, and later between London and Esbjerg. 2. The material and methods are described, together with a discussion on the validity of this type of survey and some comparison of its results with those obtained by other methods (pp. 76-86). 3. Particular attention has been paid to Rhizosolenia styliformis (pp. 92- 107), Biddulphia sinensis (pp. 108-115), Phaeocystis (pp. 149-153), and the Dinoflagellates (pp. 134-149); of these the first three are known to be of particular importance in relation to the herring fisheries. More generalised data are available for the principal diatoms other than R. styliformis and B. sinensis (pp. 116-134). 4. The main part of the work is an ecological study of the phytoplankton changes in time and space over the 5½ years. Each year is marked by some distinct variations in the abundance and the times of increase, maximum numbers and decline as recorded in the different forms. These variations in the annual cycles are compared on the different lines by a series of graphs arranged against a time scale of months, a set for each year being placed side by side (Plates I-XXI). More detailed studies by more frequent records were made in the autumns of 1934, 1935, 1936 and 1937 (cf. Figs. 3 and 4). The changes in spatial distribution are shown by a series of monthly maps arranged in a similar manner for each year (Plates XXII-LXIV). These intensive studies of the changes in time and space are also intended to form the basis for correlations with other features in the general ecology of the area (e. g. the zooplankton, hydrology, meteorology and fisheries) to be made in later publications. 5. Whilst each form has shown its own peculiar features, a trend towards a general increase in the phytoplankton as a whole has been observed during the period, although the years 1934 and 1936 have in some respects shown deviations and regressive features, and not all organisms have revealed the same trend. The possible relation of this gradual trend to other events observed in recent years in these and neighbouring waters is discussed (pp. 162-167). 6. The application of these results to the study of patchiness (pp. 154-158), inter-relationships in the plankton (pp. 159-160) and to water movements (pp. 160-162) is briefly discussed.

An 18.2 kDa protein from the liver fluke, Fasciola hepatica has been identified and characterised. The protein shows strongest sequence similarity to egg antigen proteins from Schistosoma mansoni, Schistosoma japonicum and Clonorchis sinensis. The protein is predicted to adopt a calmodulin-like fold; it thus represents the third calmodulin-like protein to be characterised in F. hepatica and has been named FhCaM3. Compared to the classical calmodulin structure there are some variations. Most noticeably, the central, linker helix is disrupted by a cysteine residue. Alkaline native gel electrophoresis showed that FhCaM3 binds calcium ions. This binding event increases the ability of the protein to bind the hydrophobic fluorescent probe 8-anilinonaphthalene-1-sulphonate, consistent with an increase in surface hydrophobicity as seen in other calmodulins. FhCaM3 binds to the calmodulin antagonists trifluoperazine and W7, but not to the myosin regulatory light chain binding compound praziquantel. Immunolocalisation demonstrated that the protein is found in eggs and vitelline cells. Given the critical role of calcium ions in egg formation and hatching this suggests that FhCaM3 may play a role in calcium signalling in these processes. Consequently the antagonism of FhCaM3 may, potentially, offer a method for inhibiting egg production and thus reducing the spread of infection.

Functional genomics have not been reported for Opisthorchis viverrini or the related fish-borne fluke, Clonorchis sinensis. Here we describe the introduction by square wave electroporation of Cy3-labeled small RNA into adult O. viverrini worms. Adult flukes were subjected to square wave electroporation employing a single pulse for 20 ms of 125V in the presence of 50 µg/ml of Cy3-siRNA. The parasites tolerated this manipulation and, at 24 and 48 h after electroporation, fluorescence from the Cy3-siRNA was evident throughout the parenchyma of the worms, with strong fluorescence evident in the guts and reproductive organs of the adult worms. Second, other worms were treated using the same electroporation settings with double stranded RNA targeting an endogenous papain-like cysteine protease, cathepsin B. This manipulation resulted in a significant reduction in specific mRNA levels encoding cathepsin B, and a significant reduction in cathepsin B activity against the diagnostic peptide, Z-Arg-Arg-AMC. This appears to be the first report of introduction of reporter genes into O. viverrini and the first report of experimental RNA interference (RNAi) in this fluke. The findings indicated the presence of an intact RNAi pathway in these parasites which, in turn, provides an opportunity to probe gene functions in this neglected tropical disease pathogen.

Bradykinin-related peptides (BRPs) are significant components of the defensive skin secretions of many anuran amphibians, and these secretions represent the source of the most diverse spectrum of such peptides so far encountered in nature. Of the many families of bioactive peptides that have been identified from this source, the BRPs uniquely appear to represent homologues of counterparts that have specific distributions and receptor targets within discrete vertebrate taxa, ranging from fishes through mammals. Their broad spectra of actions, including pain and inflammation induction and smooth muscle effects, make these peptides ideal weapons in predator deterrence. Here, we describe a novel 12-mer BRP (RVALPPGFTPLR-RVAL-(L1, T6, L8)-bradykinin) from the skin secretion of the Fujian large-headed frog (Limnonectes fujianensis). The C-terminal 9 residues of this BRP (-LPPGFTPLR) exhibit three amino acid substitutions (L/R at Position 1, T/S at Position 6 and L/F at Position 8) when compared to canonical mammalian bradykinin (BK), but are identical to the kinin sequence present within the cloned kininogen-2 from the Chinese soft-shelled turtle (Pelodiscus sinensis) and differ from that encoded by kininogen-2 of the Tibetan ground tit (Pseudopodoces humilis) at just a single site (F/L at Position 8). These data would imply that the novel BRP is an amphibian defensive agent against predation by sympatric turtles and also that the primary structure of the avian BK, ornithokinin (RPPGFTPLR), is not invariant within this taxon. Synthetic RVAL-(L1, T6, L8)-bradykinin was found to be an antagonist of BK-induced rat tail artery smooth muscle relaxation acting via the B2-receptor.

Invasive alien aquatic species, including marine and freshwater macroinvertebrates, have become increasingly important in terms of both environmental and socio-economic impacts. In order to assess their environmental and economic costs, we applied the Generic Impact Scoring System (GISS) and performed a comparison with other taxa of invaders in Europe. Impacts were scored into six environmental and six socio-economic categories, with each category containing five impact levels. Among 49 aquatic macroinvertebrates, the most impacting species were the Chinese mitten crab, Eriocheir sinensis (Milne-Edwards, 1853) and the zebra mussel, Dreissena polymorpha (Pallas, 1771). The highest impacts found per GISS impact category were, separately; on ecosystems, through predation, as competitors, and on animal production. Eleven species have an impact score > 10 (high impact) and seven reach impact level 5 in at least one impact category (EU blacklist candidates), the maximum score that can be given is 60 impact points. Comparisons were drawn between aquatic macroinvertebrates and vertebrate invaders such as fish, mammals and birds, as well as terrestrial arthropods, revealing invasive freshwater macroinvertebrates to be voracious predators of native prey and damaging to native ecosystems compared with other taxa. GISS can be used to compare these taxa and will aid policy making and targeting of invasive species for management by relevant agencies, or to assist in producing species blacklist candidates.
