Conventional sample holder cells used to the electric characterization of ceramics at high temperature consists of an alumina tube and platinum wires and plates using a complex design. The high cost materials used in the conventional sampler holder cell were replaced by stainless steel and conventional ceramics. The sample holder was validated by characterizing yttria-stabilized-zirconia in a temperature range of 25 to 700 ºC. The results do not present variations, discontinuity or unusual noise in the electric signals. Several samples were characterized without maintenance, which demonstrates that the sample holder is electric and mechanic adequate to be used to electrical characterization of ceramics up to 700 ºC.

In this review is presented an innovative technology for use of animal and vegetable waste with high pollution levels in microbial fuel cell (MFC) as an alternative to waste remediation and simultaneously producing electricity and fertilizer for agriculture. A brief history of MFC, the studies about the electron transfer mechanisms, discussion of the biological nanowires in bacteria and the use of chemical mediators or carriers of electrons are explained. The factors influencing the performance of MFCs, the application in waste and sewage treatment and power generation are also discussed.

Proton exchange membrane fuel cell (PEMFC) requires membrane electrode assemblies (MEA) to generate electrical energy from hydrogen and oxygen. In this study a MEA production process by sieve printing and an ink composition were developed to produce catalyst layers of MEAs. The deposition of the exact catalyst content was possible on cathodes and anodes with only one print step. The optimal ink developed shown viscosity of 2.75 Pa s, density 1.27 g cm-3, total solid content of 33.76 % and tack of 92 U.T. The electrodes prepared in only one printing step showed higher performance than those prepared in several steps.

Significant functions in the Proton Exchange Membrane Fuel Cells (PEMFCs) rely on Gas Diffusion Layers (GDLs), such as control the water balance in the membrane electrode assembly (MEA), allow suitable gas permeability and porosity, etc. Aware of the GDL importance in the cell performance and its great demand in scale-up projects, the fuel cell research group at Instituto de Pesquisas Energéticas e Nucleares (IPEN) has developed a Sieve Printing method (innovative in Brazil) as a strategic solution for producing GDL and electrodes used in high power PEMFC stacks. The method has shown to be adequate to fabricate low cost electrodes, GDLs of different dimensions and to produce any amount of MEAs for power stacks.

Polttokennojen, erityisesti SOFC-kennojen, tutkimuksessa on viime vuosina saavutettu merkittäviä edistysaskelia ja mahdollisuudet kennojen laajamittaiseen hyödyntämiseen paranevat koko ajan. Polttokennojen yleistyessä tarvitaan tehoelektroniikkaa muokkaamaan kennojen tasajännite verkkoon sopivaksi vaihtojännitteeksi. Verkkovaihtosuuntaaja vaatii korkeamman jännitetason, kuin polttokennosta on saatavissa, joten tasajännitetasoa on ensin nostettava. Tässä diplomityössä esitellään kolme eri hakkuritopologiaa ja perehdytään kokosiltahakkurin optimointiin. Hakkurin pääasialliset häviölähteet olivat toision diodisilta ja transistorit. Diodien vaihtaminen piidiodeista piikarbididiodeihin ei parantanut hyötysuhdetta, koska toision jännite tarkastellussa sovelluksessa oli matala. Muuntajan käämiminen litz-johtimella paransi hyötysuhdetta merkittävästi.

The maximum realizable power throughput of power electronic converters may be limited or constrained by technical or economical considerations. One solution to this problemis to connect several power converter units in parallel. The parallel connection can be used to increase the current carrying capacity of the overall system beyond the ratings of individual power converter units. Thus, it is possible to use several lower-power converter units, produced in large quantities, as building blocks to construct high-power converters in a modular manner. High-power converters realized by using parallel connection are needed for example in multimegawatt wind power generation systems. Parallel connection of power converter units is also required in emerging applications such as photovoltaic and fuel cell power conversion. The parallel operation of power converter units is not, however, problem free. This is because parallel-operating units are subject to overcurrent stresses, which are caused by unequal load current sharing or currents that flow between the units. Commonly, the term ’circulatingcurrent’ is used to describe both the unequal load current sharing and the currents flowing between the units. Circulating currents, again, are caused by component tolerances and asynchronous operation of the parallel units. Parallel-operating units are also subject to stresses caused by unequal thermal stress distribution. Both of these problemscan, nevertheless, be handled with a proper circulating current control. To design an effective circulating current control system, we need information about circulating current dynamics. The dynamics of the circulating currents can be investigated by developing appropriate mathematical models. In this dissertation, circulating current models aredeveloped for two different types of parallel two-level three-phase inverter configurations. Themodels, which are developed for an arbitrary number of parallel units, provide a framework for analyzing circulating current generation mechanisms and developing circulating current control systems. In addition to developing circulating current models, modulation of parallel inverters is considered. It is illustrated that depending on the parallel inverter configuration and the modulation method applied, common-mode circulating currents may be excited as a consequence of the differential-mode circulating current control. To prevent the common-mode circulating currents that are caused by the modulation, a dual modulator method is introduced. The dual modulator basically consists of two independently operating modulators, the outputs of which eventually constitute the switching commands of the inverter. The two independently operating modulators are referred to as primary and secondary modulators. In its intended usage, the same voltage vector is fed to the primary modulators of each parallel unit, and the inputs of the secondary modulators are obtained from the circulating current controllers. To ensure that voltage commands obtained from the circulating current controllers are realizable, it must be guaranteed that the inverter is not driven into saturation by the primary modulator. The inverter saturation can be prevented by limiting the inputs of the primary and secondary modulators. Because of this, also a limitation algorithm is proposed. The operation of both the proposed dual modulator and the limitation algorithm is verified experimentally.

Concerns about condition of the environment and rising fossil fuel prices have accelerated the research of finding new cheap and environmental friendly energy source. Fuel cells are one of the most promising green technologies, especially in the field of distributed energy generation, backup power systems, transportation and mobile power generation. In this bachelor’s thesis use of fuel cells is studied, especially from the DC-DC converter point of view. This bachelor’s thesis concentrates on study of two different DC-DC converters. The aim of this thesis is to study design and steering methods for proposed converters. The ultimate aim of this thesis is to determine which one of proposed converters is most suitable for fuel cell applications.

Huoli ympäristön tilasta ja fossiilisten polttoaineiden hinnan nousu ovat vauhdittaneet tutkimusta uusien energialähteiden löytämiseksi. Polttokennot ovat yksi lupaavimmista tekniikoista etenkin hajautetun energiantuotannon, varavoimalaitosten sekä liikennevälineiden alueella. Polttokenno on tehonlähteenä kuitenkin hyvin epäideaalinen, ja se asettaa tehoelektroniikalle lukuisia erityisvaatimuksia. Polttokennon kytkeminen sähköverkkoon on tavallisesti toteutettu käyttämällä galvaanisesti erottavaa DC/DC hakkuria sekä vaihtosuuntaajaa sarjassa. Polttokennon kulumisen estämiseksi tehoelektroniikalta vaaditaan tarkkaa polttokennon lähtövirran hallintaa. Perinteisesti virran hallinta on toteutettu säätämällä hakkurin tulovirtaa PI (Proportional and Integral) tai PID (Proportional, Integral and Derivative) -säätimellä. Hakkurin epälineaarisuudesta johtuen tällainen ratkaisu ei välttämättä toimi kaukana linearisointipisteestä. Lisäksi perinteiset säätimet ovat herkkiä mallinnusvirheille. Tässä diplomityössä on esitetty polttokennon jännitettä nostavan hakkurin tilayhtälökeskiarvoistusmenetelmään perustuva malli, sekä malliin perustuva diskreettiaikainen integroiva liukuvan moodin säätö. Esitetty säätö on luonteeltaan epälineaarinen ja se soveltuu epälineaaristen ja heikosti tunnettujen järjestelmien säätämiseen.

This dissertation considers the impact of technology foresight in innovation within the context of a technology driven development. The main hypothesis made was that by using different methods of foresight in the industry level significant value could be created. The question was approached through a case study in portable fuel cell technology. The theoretical background of the study draws from Innovation, Product Development, Management of Technology, and Technology Foresight. The connection within the topics is made by analyzing foresight, not in a policy view as often done in Europe, but in a micro-level. Focusing mostly on how a technology driven development scenario could be analyzed. The study is based on a bibliometric, extrapolation and patent analysis within the context of a case study. In addition, a large two-year Delphi study was conducted. The study was finalized with a scenario work on the future possibilities of the case study technology. Original publications also consider several methodological issues. In the context of the case study, the study questions the practicality of establishing a portable fuel cell technology in Finland showing several impractical assumptions has been made. In a more conceptual level, the study makes notions on two underlying factors: policy-push technologies and growth of data. Policy-push questions in which level a policy effort towards a single technology is practical. The European foresight effort is more directed towards policy decisions in contrast to US foresight, which is to some extent corporate driven. Although the policy-based foresight has produced significant results in the European context, policy led efforts towards a single technology are challenging. Growth of data argues on the challenges produced by the large-scale application of quantitative measures of foresight. Bibliometric studies and trend extrapolations have been taken advantage of the increasing number of databases made available, and used these as the basis for forecasts. However, the relationship with actual development and quantitative evidence is still unproven.

Fuel cells are a promising alternative for clean and efficient energy production. A fuel cell is probably the most demanding of all distributed generation power sources. It resembles a solar cell in many ways, but sets strict limits to current ripple, common mode voltages and load variations. The typically low output voltage from the fuel cell stack needs to be boosted to a higher voltage level for grid interfacing. Due to the high electrical efficiency of the fuel cell, there is a need for high efficiency power converters, and in the case of low voltage, high current and galvanic isolation, the implementation of such converters is not a trivial task. This thesis presents galvanically isolated DC-DC converter topologies that have favorable characteristics for fuel cell usage and reviews the topologies from the viewpoint of electrical efficiency and cost efficiency. The focus is on evaluating the design issues when considering a single converter module having large current stresses. The dominating loss mechanism in low voltage, high current applications is conduction losses. In the case of MOSFETs, the conduction losses can be efficiently reduced by paralleling, but in the case of diodes, the effectiveness of paralleling depends strongly on the semiconductor material, diode parameters and output configuration. The transformer winding losses can be a major source of losses if the windings are not optimized according to the topology and the operating conditions. Transformer prototyping can be expensive and time consuming, and thus it is preferable to utilize various calculation methods during the design process in order to evaluate the performance of the transformer. This thesis reviews calculation methods for solid wire, litz wire and copper foil winding losses, and in order to evaluate the applicability of the methods, the calculations are compared against measurements and FEM simulations. By selecting a proper calculation method for each winding type, the winding losses can be predicted quite accurately before actually constructing the transformer. The transformer leakage inductance, the amount of which can also be calculated with reasonable accuracy, has a significant impact on the semiconductor switching losses. Therefore, the leakage inductance effects should also be taken into account when considering the overall efficiency of the converter. It is demonstrated in this thesis that although there are some distinctive differences in the loss distributions between the converter topologies, the differences in the overall efficiency can remain within a range of a few percentage points. However, the optimization effort required in order to achieve the high efficiencies is quite different in each topology. In the presence of practical constraints such as manufacturing complexity or cost, the question of topology selection can become crucial.

Ilmastonmuutos ja fossiilisten polttoaineiden ehtyminen ovat edesauttaneet uusiutuvien energialähteiden tutkimusta huomattavasti. Lisäksi alati kasvava sähköenergian tarve lisää hajautetun sähköntuotannon ja vaihtoehtoisten energialähteiden kiinnostavuutta. Yleisimpiä hajautetun sähköntuotannon energialähteitä ovat tuulivoima, aurinkovoima ja uutena tulokkaana polttokennot. Polttokennon kytkeminen sähköverkkoon vaatii tehoelektroniikkaa, ja yleensä yksinkertaisessa polttokennosovelluksessa polttokenno kytketään galvaanisesti erottavan yksisuuntaisen DC/DC-hakkurin ja vaihtosuuntaajan kanssa sarjaan. Polttokennon rinnalla voidaan käyttää akkua tasaamaan polttokennon syöttämää jännitettä, jolloin akun ja polttokennon väliin tarvitaan kaksisuuntainen DC/DC-hakkuri, joka pystyy siirtämään energiaa molempiin suuntiin. Tässä diplomityössä on esitetty kaksisuuntaisen DC/DC-hakkurin tilayhtälökeskiarvoistusmenetelmään perustuva malli sekä mallin perusteella toteutettu virtasäätö. Tutkittava hakkuritopologia on kokosilta-tyyppinen boost-hakkuri, ja säätömenetelmä keskiarvovirtasäätö. Työn tuloksena syntyi tilayhtälömalli kaksisuuntaiselle FB boost -hakkurille sekä sen tulokelan virran säätämiseen soveltuva säädin. Säädin toimii normaalitilanteissa hyvin, mutta erikoistilanteissa, kuten hakkurin tulojännitteen äkillisessä muutostilanteessa, vaadittaisiin tehokkaampi säädin, jolla saavutettaisiin nopeampi nousuaika ilman ylitystä ja oskillointia.

Electric energy demand has been growing constantly as the global population increases. To avoid electric energy shortage, renewable energy sources and energy conservation are emphasized all over the world. The role of power electronics in energy saving and development of renewable energy systems is significant. Power electronics is applied in wind, solar, fuel cell, and micro turbine energy systems for the energy conversion and control. The use of power electronics introduces an energy saving potential in such applications as motors, lighting, home appliances, and consumer electronics. Despite the advantages of power converters, their penetration into the market requires that they have a set of characteristics such as high reliability and power density, cost effectiveness, and low weight, which are dictated by the emerging applications. In association with the increasing requirements, the design of the power converter is becoming more complicated, and thus, a multidisciplinary approach to the modelling of the converter is required. In this doctoral dissertation, methods and models are developed for the design of a multilevel power converter and the analysis of the related electromagnetic, thermal, and reliability issues. The focus is on the design of the main circuit. The electromagnetic model of the laminated busbar system and the IGBT modules is established with the aim of minimizing the stray inductance of the commutation loops that degrade the converter power capability. The circular busbar system is proposed to achieve equal current sharing among parallel-connected devices and implemented in the non-destructive test set-up. In addition to the electromagnetic model, a thermal model of the laminated busbar system is developed based on a lumped parameter thermal model. The temperature and temperature-dependent power losses of the busbars are estimated by the proposed algorithm. The Joule losses produced by non-sinusoidal currents flowing through the busbars in the converter are estimated taking into account the skin and proximity effects, which have a strong influence on the AC resistance of the busbars. The lifetime estimation algorithm was implemented to investigate the influence of the cooling solution on the reliability of the IGBT modules. As efficient cooling solutions have a low thermal inertia, they cause excessive temperature cycling of the IGBTs. Thus, a reliability analysis is required when selecting the cooling solutions for a particular application. The control of the cooling solution based on the use of a heat flux sensor is proposed to reduce the amplitude of the temperature cycles. The developed methods and models are verified experimentally by a laboratory prototype.

Streptococcus du Groupe B (GBS) et Streptococcus suis sont deux pathogènes encapsulés qui induisent des pathologies similaires dont la méningite et la septicémie chez les animaux et/ou les humains. Les sérotypes III et V du GBS et les sérotypes 2 et 14 du S. suis (utilisés dans cette étude) sont parmi les plus prévalents et/ou les plus virulents. La capsule polysaccharidique (CPS) définit le sérotype et est considérée comme un facteur de virulence essentiel pour les deux espèces bactériennes. Malgré que plusieurs études aient été réalisées au niveau des interactions entre ces streptocoques et les cellules de l’immunité innée, aucune information n’est disponible sur la régulation de la réponse immunitaire contre ces pathogènes par les cellules dendritiques (DCs) et leur interactions avec d’autres cellules, notamment les cellules ‘natural killer’ (NK). Dans cette étude, différentes approches (in vitro, ex vivo et in vivo) chez la souris ont été développées pour caractériser les interactions entre les DCs, les cellules NK et GBS ou S. suis. L’utilisation de mutants non encapsulés a permis d’évaluer l’importance de la CPS dans ces interactions. Les résultats in vitro avec les DCs infectées par GBS ou S. suis ont démontré que ces deux pathogènes interagissent différemment avec ces cellules. GBS est grandement internalisé par les DCs, et ce, via de multiples mécanismes impliquant notamment les radeaux lipidiques et la clathrine. Le mécanisme d’endocytose utilisé aurait un effet sur la capacité du GBS à survivre intracellulairement. Quant au S. suis, ce dernier est très faiblement internalisé et, si le cas, rapidement éliminé à l’intérieur des DCs. GBS et S. suis activent les DCs via différents récepteurs et favorisent la production de cytokines et chimiokines ainsi que l’augmentation de l’expression de molécules de co-stimulation. Cette activation permet la production d’interferon-gamma (IFN-y) par les cellules NK. Cependant, GBS semble plus efficient à activer les DCs, et par conséquent, les cellules NK que S. suis. La production d’IFN-y, en réponse à la stimulation bactérienne, est principalement assurée par un contact direct entre les DCs et les cellules NK et ne dépend qu’en partie de facteurs solubles. De plus, nos résultats in vivo ont démontré que ces deux streptocoques induisent rapidement la libération d'IFN-y par les cellules NK lors de la phase aiguë de l'infection. Ceci suggère que les interactions entre les DCs et les cellules NK pourraient jouer un rôle dans le développement d’une réponse immune T auxiliaire de type 1 (T ‘helper’ 1 en anglais; Th1). Cependant, la capacité de S. suis à activer la réponse immunitaire in vivo est également plus faible que celle observée pour GBS. En effet, les CPSs de GBS et de S. suis jouent des rôles différents dans cette réponse. La CPS de S. suis empêche une activation optimale des DCs et des cellules NK alors que c’est l’opposé pour la CPS de GBS, indépendamment du sérotype évalué. En résumé, cette étude adresse pour la première fois la contribution des DCs et des cellules NK dans la réponse immunitaire innée lors d’une infection à GBS ou à S. suis et, par extension, dans le développement d’une réponse Th1. Nos résultats renforcent davantage le rôle central des DCs dans le contrôle efficace des infections causées par des bactéries encapsulées.

L’hypertrophie cardiaque représente la réponse primaire du cœur dans le but d’améliorer la fonction cardiaque qui est compromise suite à un accident ischémique ou une surcharge hémodynamique. Cependant, l’hypertrophie cardiaque a pour conséquence pathologique la fibrose réactive, qui est caractérisée par la synthèse incontrôlée et le dépôt du collagène par les myofibroblastes. Ainsi, l’accumulation accrue du collagène dans le cœur hypertrophié mène à l’augmentation de la rigidité cardiaque et la détérioration progressive de la fonction contractile du cœur. Plusieurs études ont démontré que la protéine nestine, appartenant à la famille des filaments intermédiaires, est ré-exprimée dans les myofibroblastes durant la fibrose réparative et est impliquée dans la prolifération cellulaire. Basée sur ces observations, cette étude teste l’hypothèse selon laquelle nestine est induite dans les myofibroblastes suivant le développement de la fibrose réactive dans le cœur des rats ayant subi une constriction aortique supra-rénale. Deux semaines suivant une constriction aortique supra-rénale chez le rat, un patron d’hypertrophie concentrique cardiaque a été observé et associé avec une réponse de fibrose réactive caractérisée par le dépôt accru de collagène dans le tissu interstitiel et la région péri-vasculaire de nombreux vaisseaux sanguins cardiaques. De plus, les niveaux de la protéine nestine sont augmentés significativement dans les cœurs des rats hypertrophiés, et ce, de façon corrélative avec la pression artérielle moyenne et la pression systolique du ventricule gauche. Les techniques d’immunofluorescences ont révélé une apparition accrue des cellules immunoréactives à nestine, qui présentent un phénotype mésenchymateux caractérisé par la co-expression de collagène dans le tissu interstitiel et la région péri-vasculaire des cœurs hypertrophiés. Ces données suggèrent que les fibroblastes résidents peuvent exprimer la protéine nestine ou que l’expression de nestine est induite en réponse aux facteurs pro-fibrotiques impliqués dans la fibrose réactive. En effet, l’exposition des myofibroblastes normaux et des myofibroblastes isolés des cœurs hypertrophiés à l’AII, TGF-B1 et EGF augmente significativement l’expression de la protéine nestine, tandis que l’expression de l’α-SMA demeure inchangée. De plus, de manière prédominante dans le cœur hypertrophié, des cellules non-vasculaires CD31(+) ont été détectées dans le tissu interstitiel et la région péri-vasculaire. Ces cellules co-expriment nestine et collagène suggérant une transition des cellules endothéliales vers un phénotype mésenchymateux. Finalement, la protéine nestine, sous sa forme filamenteuse, a été détectée dans les cellules endothéliales de l’artère coronaire humaine et leur exposition au TGF-B1, induit l’expression de collagène. En revanche, l’expression de collagène a été détectée dans les cellules microvasculaires de rats CD31(+), alors que l’expression de nestine est absente. En réponse aux traitements de TGF-B1 et EGF, l’expression de nestine, sous sa forme non-filamenteuse, est détectée dans les cellules microvasculaires de rats. Collectivement, ces données supportent la prémisse selon laquelle la réponse de fibrose réactive dans les cœurs hypertrophiés, suite à une constriction aortique supra-rénale, est attribuée en partie à l’augmentation de l’apparition des cellules mésenchymateuses positives à l’expression de nestine qui proviennent des fibroblastes résidents du ventricule. De plus, les données in vivo et in vitro suggèrent que les cellules endothéliales déplacées représentent une source additionnelle des cellules mésenchymateuses nestine(+) dans le cœur hypertrophié et contribuent au développement de la fibrose réactive. Cibler la protéine nestine peut représenter une approche thérapeutique afin d’atténuer la réponse de fibrose réactive indépendamment de l’origine des cellules mésenchymateuses.

The use of catalysts in chemical and refining processes has increased rapidly since 1945, when oil began to replace coal as the most important industrial raw material. Catalysis has a major impact on the quality of human life as well as economic development. The demand for catalysts is still increasing since catalysis is looked up as a solution to eliminate or replace polluting processes. Metal oxides represent one of the most important and widely employed classes of solid catalysts. Much effort has been spent in the preparation, characterization and application of metal oxides. Recently, great interest has been devoted to the cerium dioxide (CeO2) containing materials due to their broad range of applications in various fields, ranging from catalysis to ceramics, fuel cell technologies, gas sensors, solid state electrolytes, ceramic biomaterials, etc., in addition to the classical application of CeO2 as an additive in the so-called three way catalysts (TWC) for automotive exhaust treatment. Moreover, it can promote water gas shift and steam reforming reactions, favours catalytic activity at the interfacial metal-support sites. The solid solutions of ceria with Group IV transitional-metals deserve particular attention for their applicability in various technologically important catalytic processes. Mesoporous CeO2−ZrO2 solid solutions have been reported to be employed in various reactions which include CO oxidation, soot oxidation, water-gas shift reaction, and so on. Inspired by the unique and promising characteristics of ceria based mixed oxides and solid solutions for various applications, we have selected ceria-zirconia oxides for our studies. The focus of the work is the synthesis and investigation of the structural and catalytic properties of modified and pure ceria-zirconia mixed oxide.
