In this work evaluated the content of vitamin C and total carotenoid in tomatoes fresh and dehydrated for conserve, with posterior immersion in sauce, verifying its sensorial acceptability. Dehydration was performed in dryer with airflow at 70 °C to obtain a product with final moisture of 45%. Determinations were carried out ascorbic acid, soluble solids, acidity, sugars and carotenoid before and after the dehydration process. The dehydrated tomatoes had been immersed in three types of sauces (pepper sauce, oregano with parrot leaves sauce and garlic sauce) per 25 days. After this period, the dry tomatoes in conserve had been submitted to the sensorial analysis in the pate form, applying themselves preference test. The results had been showed that retention of vitamin C and total carotenoid in the dehydrated product showed retention of 14.0% and 44.3%, respectively, in tomatoes after the dehydration process. Background: The liver is an important organ for its ability to transform xenobiotics, making the liver tissue a prime target for toxic substances. The carotenoid bixin present in annatto is an antioxidant that can protect cells and tissues against the deleterious effects of free radicals. In this study, we evaluated the protective effect of bixin on liver damage induced by carbon tetrachloride (CCl4) in rats.Results: The animals were divided into four groups with six rats in each group. CCl4 (0.125 mL kg(-1) body wt.) was injected intraperitoneally, and bixin (5.0 mg kg(-1) body wt.) was given by gavage 7 days before the CCl4 injection. Bixin prevented the liver damage caused by CCl4, as noted by the significant decrease in serum aminotransferases release. Bixin protected the liver against the oxidizing effects of CCl4 by preventing a decrease in glutathione reductase activity and the levels of reduced glutathione and NADPH. The peroxidation of membrane lipids and histopathological damage of the liver was significantly prevented by bixin treatment.Conclusion: Therefore, we can conclude that the protective effect of bixin against hepatotoxicity induced by CCl4 is related to the antioxidant activity of the compound.

A new species of Moenkhausia is described from Rio Xingu and Rio Tapajos basins, Brazil. The new species is distinguished from its congeners, except from Moenkhausia moisae, by having more scales in the lateral series, 43-47 (v. 23-41 in the remaining congeners). The new species is distinguished from M. moisae by its colour pattern, which consists of a dark midlateral stripe, and an asymmetrical caudal blotch (inconspicuous or faded in specimens from the Rio Arinos) continuous with the midlateral stripe (v. narrow dark midlateral line and conspicuous, regularly rounded and symmetrical blotch not continuous with the midlateral line). The new species is putatively assumed to be mimetic to Jupiaba apenima, in the Rio Xingu and Rio Teles Pires drainages, and to Jupiaba yarina in the Rio Arinos. The two species of Jupiaba are sympatric and remarkably similar in size, general external morphology and colouration to the new species. A small difference occurs in the colouration between the two species of Jupiaba and is also observed in the two respectively sympatric morphotypes of the new species of Moenkhausia. Papayas have a very short green life as a result of their rapid pulp softening as well as their susceptibility to physical injury and mold growth. The ripening-related changes take place very quickly, and there is a continued interest in the reduction of postharvest losses. Proteins have a central role in biological processes, and differential proteomics enables the discrimination of proteins affected during papaya ripening. A comparative analysis of the proteomes of climacteric and pre-climacteric papayas was performed using 2DE-DIGE. Third seven proteins corresponding to spots with significant differences in abundance during ripening were submitted to MS analysis, and 27 proteins were identified and classified into six main categories related to the metabolic changes occurring during ripening. Proteins from the cell wall (alpha-galactosidase and invertase), ethylene biosynthesis (methionine synthase), climacteric respiratory burst, stress response, synthesis of carotenoid precursors (hydroxymethylbutenyl 4-diphosphate synthase, GcpE), and chromoplast differentiation (fibrillin) were identified. There was some correspondence between the identified proteins and the data from previous transcript profiling of papaya fruit, but new, accumulated proteins were identified, which reinforces the importance of differential proteomics as a tool to investigate ripening and provides potentially useful information for maintaining fruit quality and minimizing postharvest losses. beta-Carotene (BC) is one of the natural pigments that is most commonly added to food; however, the utilization of BC is limited due to its instability. Microencapsulation techniques are commonly used because they can protect the microencapsulated material from oxidization. Nevertheless, the properties of the encapsulated compounds must be studied. We compared the antigenotoxic potential of pure and microencapsulated beta-carotene (mBC) in Wistar rats. Two doses of BC or mBC (2.5 or 5.0 mg/kg) were administered by gavage over a period of 14 days. The final gavage was followed by an injection of doxorubicin (DXR). After 24 h the animals were euthanized. The micronucleus test results showed that when both mBC and DXR were given, only the higher dose was antigenotoxic. The results of the comet assay show that when given in association with DXR, mBC had protective effects in the liver. The differences between the results obtained with BC and mBC suggest that possibly the carotenoid biodisponibility was modified by the process of microencapsulation. In conclusion, mBC does not lose its protective properties, but higher doses must be used to observe antigenotoxic effects. This is the first time that the genotoxicity and antigenotoxicity of a microencapsulated compound was evaluated in vivo. (C) 2012 Elsevier Ltd. All rights reserved.

Abscisic acid (ABA) is an important regulator of plant responses to environmental stresses and an absolute requirement for stress tolerance. Recently, a third phytoene synthase (PSY3) gene paralog was identified in monocots and demonstrated to play a specialized role in stress-induced ABA formation, thus suggesting that the first committed step in carotenogenesis is a key limiting step in ABA biosynthesis. To examine whether the ectopic expression of PSY, other than PSY3, would similarly affect ABA level and stress tolerance, we have produced transgenic tobacco containing a fruit-specific PSY (CpPSY) of grapefruit (Citrus paradisi Macf.). The transgenic plants contained a single- or double-locus insertion and expressed CpPSY at varying transcript levels. In comparison with the wild-type plants, the CpPSY expressing transgenic plants showed a significant increase on root length and shoot biomass under PEG-, NaCl- and mannitol-induced osmotic stress. The enhanced stress tolerance of transgenic plants was correlated with the increased endogenous ABA level and expression of stress-responsive genes, which in turn was correlated with the CpPSY copy number and expression level in different transgenic lines. Collectively, these results provide further evidence that PSY is a key enzyme regulating ABA biosynthesis and that the altered expression of other PSYs in transgenic plants may provide a similar function to that of the monocot's PSY3 in ABA biosynthesis and stress tolerance. The results also pave the way for further use of CpPSY, as well as other PSYs, as potential candidate genes for engineering tolerance to drought and salt stress in crop plants.

Bixin is the main carotenoid found in annatto seeds (Bixa orellana L.) and is responsible for their reddish-orange color. The antioxidant properties of this compound are associated with its ability to scavenge free radicals, which may reduce damage and protect tissues against toxicity caused by anticancer drugs such as cisplatin. In this study, the genotoxicity and antigenotoxicity of bixin on cisplatin-induced toxicity in PC12 cells was assessed. Cytotoxicity was evaluated using the mu assay, mutagenicity, genotoxicity, and protective effect of bixin were evaluated using the micronucleus test and comet assay. PC12 cells were treated with bixin (0.05, 0.08, and 0.10 mu g/mL), cisplatin (0.1 mu g/mL) or a combination of both bixin and cisplatin. Bixin was neither cytotoxic nor genotoxic compared to the controls. In the combined treatment bixin significantly reduced the percentage of DNA in tail and the frequency of micronuclei induced by cisplatin. This result suggests that bixin can function as a protective agent, reducing cisplatin-induced DNA damage in PC12 cells, and it is possible that this protection could also extend to neuronal cells. Further studies are being conducted to better understand the mechanisms involved in the activity of this protective agent prior to using it therapeutically. (C) 2011 Elsevier Ltd. Gracilaria domingensis (Kützing) Sonder ex Dickie and Gracilaria birdiae (Plastino & Oliveira) (Gracilariales, Rhodophyta) are seaweeds that occur on the Brazilian coast. Based on their economic and pharmaceutical importance, we investigated the antioxidant activity of the methanolic, ethyl acetate and hexane extracts of both species. The hexane extracts display a high antioxidant activity and comparative analyses indicated G. birdiae as the most active species. Chemical investigation of these fractions showed several carotenoids and fatty acids, as well as cholesterol and sitosterol derivatives. HPLC-DAD analysis of G. birdiae showed violaxanthin (0.04 μg.mg-1 of dry material), antheraxanthin (5.31 μg.mg-1), aloxanthin (0.09 μg.mg-1), zeaxanthin (0.45 μg.mg-1) and β-carotene (0.37 μg.mg-1) as the major carotenoids. G. domingensis showed a similar carotenoid profile, however, with much lower concentration than G. birdiae. Gas chromatography coupled to mass spectrometry was used to determine other nonpolar compounds of these seaweeds. The main compounds detected in both studied species were the fatty acids 16:0; 18:1 Δ9; 20:3 Δ6,9,12, 20:4 Δ5,8,11,14. We found no specificity of compounds in either species. However, G. birdiae, presented higher contents of carotenoids and arachidonic acid than G. domingensis.

Die Lichtsammelantenne des PSI (LHCI) ist hinsichtlich ihrer Protein- und Pigmentzusammensetzung weniger gut untersucht als die des PSII. Im Rahmen dieser Arbeit wurde deshalb zunächst die Isolation von nativen LHCI-Subkomplexen optimiert und deren Pigmentzusammensetzung untersucht. Zusätzlich wurde die Pigmentbindung analysiert sowie das Pigment/Protein-Verhältnis bestimmt. Die Analyse der Proteinzusammensetzung des LHCI erfolgte mittels einer Kombination aus ein- oder zweidimensionaler Gelelektrophorese mit Westernblotanalysen mit Lhca-Protein-spezifischen Antikörpern und massenspektrometrischen Untersuchungen. Dabei stellte sich heraus, dass der LHCI mehr Proteine bzw. Proteinisoformen enthält als bisher vermutet. So gelang durch die massenspektrometrischen Untersuchungen die Identifizierung zweier bisher noch nicht nachgewiesener Lhca-Proteine. Bei diesen handelt es sich um eine Isoform des Lhca4 und ein zusätzliches Lhca-Protein, das Tomaten-Homolog des Lhca5 von Arabidopsis thaliana. Außerdem wurden in 1D-Gelen Isoformen von Lhca-Proteinen mit unterschiedlichem elektrophoretischen Verhalten beobachtet. In 2D-Gelen trat zusätzlich eine große Anzahl an Isoformen mit unterschiedlichen isoelektrischen Punkten auf. Es ist zu vermuten, dass zumindest ein Teil dieser Isoformen physiologischen Ursprungs ist, und z.B. durch differentielle Prozessierung oder posttranslationale Modifikationen verursacht wird, wenn auch die Spotvielfalt in 2D-Gelen wohl eher auf die Probenaufbereitung zurückzuführen ist. Mittels in vitro-Rekonstitution mit anschließenden biochemischen Untersuchungen und Fluoreszenzmessungen wurde nachgewiesen, dass Lhca5 ein funktioneller LHC mit spezifischen Pigmentbindungseigenschaften ist. Außerdem zeigten in vitro-Dimerisierungsexperimente eine Interaktion zwischen Lhca1 und Lhca5, wodurch dessen Zugehörigkeit zur Antenne des PSI gestützt wird. In vitro-Dimerisierungsexperimente mit Lhca2 und Lhca3 führten dagegen nicht zur Bildung von Dimeren. Dies zeigt, dass die Interaktion in potentiellen Homo- oder Heterodimeren aus Lhca2 und/oder Lhca3 schwächer ist als die zwischen Lhca1 und Lhca4 oder Lhca5. Die beobachtete Proteinheterogenität deutet daraufhin, dass die Antenne des PSI eine komplexere Zusammensetzung hat als bisher angenommen. Für die Integration „neuer“ LHC in den PSI-LHCI-Holokomplex werden zwei Modelle vorgeschlagen: geht man von einer festen Anzahl von LHCI-Monomeren aus, so kann sie durch den Austausch einzelner LHC-Monomere erreicht werden. Als zweites Szenario ist die Bindung zusätzlicher LHC vorstellbar, die entweder indirekt über bereits vorhandene LHC oder direkt über PSI-Kernuntereinheiten mit dem PSI interagieren. In Hinblick auf die Pigmentbindung der nativen LHCI-Subfraktionen konnte gezeigt werden, dass sie Pigmente in einer spezifischen Stöchiometrie und Anzahl binden, und sich vom LHCIIb vor allem durch eine verstärkte Bindung von Chlorophyll a, eine geringere Anzahl von Carotinoiden und die Bindung von ß-Carotin an Stelle von Neoxanthin unterscheiden. Der Vergleich von nativem LHCI mit rekonstituierten Lhca-Proteinen ergab, dass Lhca-Proteine Pigmente in einer spezifischen Stöchiometrie binden, und dass sie Carotinoidbindungsstellen mit flexiblen Bindungseigenschaften besitzen. Auch über die Umwandlung des an die einzelnen Lhca-Proteine gebundenen Violaxanthins (Vio) im Xanthophyllzyklus war nur wenig bekannt. Deshalb wurden mit Hilfe eines in vitro-Deepoxidationssystems sowohl native als auch rekonstituierte LHCI hinsichtlich ihrer Deepoxidationseigenschaften untersucht und der Deepoxidationsgrad von in vivo deepoxidierten Pigment-Protein-Komplexen bestimmt. Aus den Deepoxidationsexperimenten konnte abgeleitet werden, dass in den verschiedenen Lhca-Proteinen unterschiedliche Carotinoidbindungsstellen besetzt sind. Außerdem bestätigten diese Experimente, dass der Xanthophyllzyklus auch im LHCI auftritt, wobei jedoch ein niedrigerer Deepoxidationsgrad erreicht wird als bei LHCII. Dies konnte durch in vitro-Deepoxidationsversuchen auf eine geringere Deepoxidierbarkeit des von Lhca1 und Lhca2 gebundenen Vio zurückgeführt werden. Damit scheint Vio in diesen Lhca-Proteinen eher eine strukturelle Rolle zu übernehmen. Eine photoprotektive Funktion von Zeaxanthin im PSI wäre folglich auf Lhca3 und Lhca4 beschränkt. Damit enthält jede LHCI-Subfraktion ein LHC-Monomer mit langwelliger Fluoreszenz, das möglicherweise am Lichtschutz beteiligt ist. Insgesamt zeigten die Untersuchungen der Pigmentbindung, der Deepoxidierung und der Fluoreszenzeigenschaften, dass sich die verschiedenen Lhca-Proteine in einem oder mehreren dieser Parameter unterscheiden. Dies lässt vermuten, dass schon durch leichte Veränderungen in der Proteinzusammensetzung des LHCI eine Anpassung an unterschiedliche Licht-verhältnisse erreicht werden kann.
