In recent years, a new paradigm for communication called cooperative communications has been proposed for which initial information theoretic studies have shown the potential for improvements in capacity over traditional multi-hop wireless networks. Extensive research has been done to mitigate the impact of fading in wireless networks, being mostly focused on Multiple-Input Multiple-Output (MIMO) systems. Recently, cooperative relaying techniques have been investigated to increase the performance of wireless systems by using diversity created by different single antenna devices, aiming to reach the same level of performance of MIMO systems with low cost devices. Cooperative communication is a promising method to achieve high spectrum efficiency and improve transmission capacity for wireless networks. Cooperative communications is the general idea of pooling the resources of distributed nodes to improve the overall performance of a wireless network. In cooperative networks the nodes cooperate to help each other. A cooperative node offering help is acting like a middle man or proxy and can convey messages from source to destination. Cooperative communication involves exploiting the broadcast nature of the wireless medium to form virtual antenna arrays out of independent singleantenna network nodes for transmission. This research aims at contributing to the field of cooperative wireless networks. The focus of this research is on the relay-based Medium Access Control (MAC) protocol. Specifically, I provide a framework for cooperative relaying called RelaySpot which comprises on opportunistic relay selection, cooperative relay scheduling and relay switching. RelaySpot-based solutions are expected to minimize signaling exchange, remove estimation of channel conditions, and improve the utilization of spatial diversity, minimizing outage and increasing reliability.

A fidelidade da síntese proteica é fundamental para a estabilidade do proteoma e para a homeostasia celular. Em condições fisiológicas normais as células têm uma taxa de erro basal associada e esta muitas vezes aumenta com o envelhecimento e doença. Problemas na síntese das proteínas estão associados a várias doenças humanas e aos processos de envelhecimento. De facto, a incorporação de erros nas proteínas devido a tRNAs carregados pelas aminoacil-tRNA sintetases com o amino ácido errado causa doenças neurodegenerativas em humanos e ratos. Ainda não é claro como é que estas doenças se desenvolvem e se são uma consequência directa da disrupção do proteoma ou se são o resultado da toxicidade produzida pela acúmulação de proteínas mal traduzidas ao nível do ribossoma. Para elucidar como é que as células eucarióticas lidam com proteínas aberrantes e agregados proteicos (stress proteotóxico) desenvolvemos uma estratégia para destabilizar o proteoma. Para isso estabelecemos um sistema de erros de tradução em embriões de peixe zebra que assenta em tRNAs mutantes capazes de incorporar erradamente serina nas proteínas. As proteínas produzidas neste sistema despoletam as vias de resposta ao stress, nomeadamente a via da ubiquitina-proteassoma (UPP – “ubiquitin protesome pathway”) e a via do retículo endoplasmático (UPR – “unfolded protein response”). O stress proteotóxico gerado pelos erros de tradução altera a expressão génica e perfis de expressão de miRNAs, o desenvolvimento embrionário e viabilidade, aumenta a produção de espécies reactivas de oxigénio (ROS), leva ainda à acumulação de agregados proteicos e à disfunção mitocondrial. As malformações embrionárias e fenótipos de viabilidade que observámos foram revertidos por antioxidantes, o que sugere que os ROS desempenham papéis importantes nos fenótipos degenerativos celulares induzidos pela produção de proteínas aberrantes e agregação proteica. Estabelecemos ainda uma linha de peixe zebra transgénica para o estudo do stress proteotóxico. Este trabalho mostra que a destabilização do proteoma em embriões de peixe zebra com tRNAs mutantes é uma boa metodologia para estudar a biologia do stress proteotóxico visto que permite a agregação controlada do proteoma, mimetizando os processos de agregação de proteínas que ocorrem naturalmente durante o envelhecimento e em doenças conformacionais humanas.

This work investigates low cost localization systems (LS) based on received signal strength (RSS) and integrated with different types of antennas with main emphasis on sectorial antennas. The last few years have witnessed an outstanding growth in wireless sensor networks (WSN). Among its various possible applications, the localization field became a major area of research. The localization techniques based on RSS are characterized by simplicity and low cost of integration. The integration of LS based on RSS and sectorial antennas (SA) was proven to provide an effective solution for reducing the number of required nodes of the networks and allows the combination of several techniques, such as RSS and angle of arrival (AoA). This PhD thesis focuses on studying techniques, antennas and protocols that best meet the needs of each LS with main focus on low cost systems based on RSS and AoA. Firstly there are studied localization techniques and system that best suit the requirements of the user and the antennas that are most appropriate according to the nature of the signal. In this step it is intended to provide a fundamental understanding of the undertaken work. Then the developed antennas are presented according to the following categories: sectorial and microstrip antennas. Two sectorial antennas are presented: a narrowband antenna operating at 2.4 to 2.5 GHz and a broadband antenna operating at 800MHz-2.4GHz. The low cost printed antennas were designed to operate at 5 GHz, which may be used for vehicular communication. After presenting the various antennas, several prototypes of indoor/outdoor LS are implemented and analyzed. Localization protocols are also proposed, one based on simplicity and low power, and the other on interoperability with different types of antennas and system requirements.

In this work physical and behavioral models for a bulk Reflective Semiconductor Optical Amplifier (RSOA) modulator in Radio over Fiber (RoF) links are proposed. The transmission performance of the RSOA modulator is predicted under broadband signal drive. At first, the simplified physical model for the RSOA modulator in RoF links is proposed, which is based on the rate equation and traveling-wave equations with several assumptions. The model is implemented with the Symbolically Defined Devices (SDD) in Advanced Design System (ADS) and validated with experimental results. Detailed analysis regarding optical gain, harmonic and intermodulation distortions, and transmission performance is performed. The distribution of the carrier and Amplified Spontaneous Emission (ASE) is also demonstrated. Behavioral modeling of the RSOA modulator is to enable us to investigate the nonlinear distortion of the RSOA modulator from another perspective in system level. The Amplitude-to-Amplitude Conversion (AM-AM) and Amplitude-to-Phase Conversion (AM-PM) distortions of the RSOA modulator are demonstrated based on an Artificial Neural Network (ANN) and a generalized polynomial model. Another behavioral model based on Xparameters was obtained from the physical model. Compensation of the nonlinearity of the RSOA modulator is carried out based on a memory polynomial model. The nonlinear distortion of the RSOA modulator is reduced successfully. The improvement of the 3rd order intermodulation distortion is up to 17 dB. The Error Vector Magnitude (EVM) is improved from 6.1% to 2.0%. In the last part of this work, the performance of Fibre Optic Networks for Distributed and Extendible Heterogeneous Radio Architectures and Service Provisioning (FUTON) systems, which is the four-channel virtual Multiple Input Multiple Output (MIMO), is predicted by using the developed physical model. Based on Subcarrier Multiplexing (SCM) techniques, four-channel signals with 100 MHz bandwidth per channel are generated and used to drive the RSOA modulator. The transmission performance of the RSOA modulator under the broadband multi channels is depicted with the figure of merit, EVM under di erent adrature Amplitude Modulation (QAM) level of 64 and 254 for various number of Orthogonal Frequency Division Multiplexing (OFDM) subcarriers of 64, 512, 1024 and 2048.

The mechanisms of secretory granule biogenesis and regulated secretion of digestive enzymes in pancreatic acinar cells are still not well understood. To shed light on these processes, which are of biological and clinical importance (e.g., pancreatitis), a better molecular understanding of the components of the granule membrane, their functions and interactions is required. The application of proteomics has largely contributed to the identification of novel zymogen granule (ZG) proteins but was not yet accompanied by a better characterization of their functions. In this study we aimed at a) isolation and identification of novel membrane-associated ZG proteins; b) characterization of the biochemical properties and function of the secretory lectin ZG16p, a membrane-associated protein; c) exploring the potential of ZG16p as a new tool to label the endolysosomal compartment. First, we have performed a suborganellar proteomics approach by combining protein analysis by 2D-PAGE and identification by mass spectrometry, which has led to the identification of novel peripheral ZGM proteins with proteoglycan-binding properties (e.g., chymase, PpiB). Then, we have unveiled new molecular properties and (multiple) functions of the secretory lectin ZG16p. ZG16p is a unique mammalian lectin with glycan and proteoglycan binding properties. Here, I revealed for the first time that ZG16p is highly protease resistant by developing an enterokinase-digestion assay. In addition I revealed that ZG16p binds to a high molecular weight complex at the ZGM (which is also protease resistant) and forms highly stable dimers. In light of these findings I suggest that ZG16p is a key component of a predicted submembranous granule matrix attached to the luminal side of the ZGM that fulfils important functions during sorting and packaging of zymogens. ZG16p, may act as a linker between the matrix and aggregated zymogens due to dimer formation. Furthermore, ZG16p protease resistance might be of higher importance after secretion since it is known that ZG16p binds to pathogenic fungi in the gut. I have further investigated the role of ZG16p binding motifs in its targeting to ZG in AR42J cells, a pancreatic model system. Point mutations of the glycan and the proteoglycan binding motifs did not inhibit the targeting of ZG16p to ZG in AR42J cells. I have also demonstrated that when ZG16p is present in the cytoplasm it interacts with and modulates the endo-lysosomal compartment. Since it is known that impaired autophagy due to lysosomal malfunction is involved in the course of pancreatitis, a potential role of ZG16p in pancreatitis is discussed.

Systems equipped with multiple antennas at the transmitter and at the receiver, known as MIMO (Multiple Input Multiple Output) systems, offer higher capacities, allowing an efficient exploitation of the available spectrum and/or the employment of more demanding applications. It is well known that the radio channel is characterized by multipath propagation, a phenomenon deemed problematic and whose mitigation has been achieved through techniques such as diversity, beamforming or adaptive antennas. By exploring conveniently the spatial domain MIMO systems turn the characteristics of the multipath channel into an advantage and allow creating multiple parallel and independent virtual channels. However, the achievable benefits are constrained by the propagation channel’s characteristics, which may not always be ideal. This work focuses on the characterization of the MIMO radio channel. It begins with the presentation of the fundamental results from information theory that triggered the interest on these systems, including the discussion of some of their potential benefits and a review of the existing channel models for MIMO systems. The characterization of the MIMO channel developed in this work is based on experimental measurements of the double-directional channel. The measurement system is based on a vector network analyzer and a two-dimensional positioning platform, both controlled by a computer, allowing the measurement of the channel’s frequency response at the locations of a synthetic array. Data is then processed using the SAGE (Space-Alternating Expectation-Maximization) algorithm to obtain the parameters (delay, direction of arrival and complex amplitude) of the channel’s most relevant multipath components. Afterwards, using a clustering algorithm these data are grouped into clusters. Finally, statistical information is extracted allowing the characterization of the channel’s multipath components. The information about the multipath characteristics of the channel, induced by existing scatterers in the propagation scenario, enables the characterization of MIMO channel and thus to evaluate its performance. The method was finally validated using MIMO measurements.

O objetivo do estudo foi saber como em Portugal mulheres e homens distribuem a atividade familiar na variável do Trabalho Não Pago, nomeadamente no que concerne ao tempo que dispõem para as Tarefas Domésticas e Prestação de Cuidados. Para (Scott, 1990) o termo 'género' constitui um dos aspetos daquilo que se poderia chamar de busca de legitimidade académica para os estudos feministas, nos anos 80. A interpretação de género enfoca a diferença sexual como determinante na forma como homens e mulheres se comunicam, pois, segundo essa visão, " (...) homens e mulheres têm essências diferentes, coisa que refletiria na sua forma diferente de utilizar a linguagem (os homens tenderiam a se expressar de forma mais direta e autoritária e as mulheres dominariam uma linguagem mais cheia de nuances" (Costa, 2000). O trabalho, têm diferentes formas — designadamente trabalho remunerado em contexto profissional e trabalho não pago no contexto dos agregados domésticos (Perista, 2002); O trabalho não pago constitui-se como um dos domínios da atividade humana nos quais a valorização social dos tempos se espelha de modo mais assimétrico (Perista, 2002); Igualmente pretendemos saber como evoluiu desde 1999, em relação ao estudo nacional realizado pelo Instituto Nacional de Estatística (INE) e Comissão para a Igualdade no Trabalho e Emprego (CITE) designado por ―Inquérito à Ocupação do Tempo 1999‖, tendo em linha de conta e emergência significativa de novos perfis de relação familiar, como as uniões de facto e os casamentos entre pessoas do mesmo sexo, e o eventual paradigma comportamental das novas gerações. A opção por estas variáveis teve em linha de conta que foram as que, naquele estudo, atingiram valores mais elevados entre homens e mulheres, com maior prestação destas.

O aumento significativo de patologias relacionadas com a alimentação, designadamente a obesidade, considerada pela Organização Mundial de Saúde como a “epidemia do século XXI”, remete o profissional da área da nutrição para a necessidade de uma intervenção cada vez mais eficiente. O objectivo deste estudo é avaliar a necessidade do desenvolvimento de novas tecnologias que possibilitem o auxílio aos processos de apoio nutricional, assim como medir as preferências no desenvolvimento de software. Para isso, utilizou-se a Análise Conjunta, na qual os resultados agregados foram hierarquizados mediante uma Análise Categórica de Componentes Principais, para posterior segmentação dos individuos, mediante uma Análise de Clusters Hierárquicos. Neste estudo, pode verificar-se que não existe preferências no desenvolvimento de software genérico, havendo uma grande receptividade dos profissionais de saúde ao desenvolvimento da Telemedicina na área da nutrição. Por fim, existem diferenças, estatisticamente significativas, entre os profissionais da área da nutrição, o que permite a sua caracterização.
