Fever is considered an important component of the acute phase response of the body in defence against invading organisms such as bacteria. Quercetin, an important representative of the flavonoid class, has been extensively studied as an anti-inflammatory agent. In the present study, we investigated the effect of quercetin, administered orally (5, 25 and 50 mg kg(-1)) or intraperitoneally (50 mg kg(-1)), on the febrile response induced by either intraperitoneally (50 mu g kg(-1)) or intravenously (5 mu g kg(-1)) injected lipopolysaccharide (LPS from Escherichia coli) in rats. In contrast with the well known anti-inflammatory activity of quercetin, the results demonstrate that quercetin, at the doses used, did not alter the fever induced by LPS, regardless of the route of administration.

Objective: In this study, we determined the protective effect of isoflavones from Glycine max on human umbilical vein endothelial cell (ECV304) damage induced by hydrogen peroxide (H(2)O(2)) and on nitric oxide (NO) production. Methods: We studied the regulation of NO synthesis in cultured human endothelial cells by phytoestrogens contained in soy extracts in the presence or absence of ICI 182,780 or N(omega)-nitro-L-arginine methyl esther and determined the protective effect of these isoflavones on ECV304 damage induced by H(2)O(2). Results: We show that soy extracts activate NO synthesis in endothelial cells and protect against cell damage. Conclusions: In conclusion, soy isoflavones markedly protect ECV304 cells against H(2)O(2) damage and promote NO synthesizing. Therefore, these isoflavones call potentially act as an NO promoter and as an antioxidant.

Objectives: Collagen disorganization is one of the main degradation patterns found in unsuccessful adhesive restorations. The hypothesis of this study was that pretreatment using natural collagen cross-linking agents rich in proanthocyanidin (PA) would improve mechanical properties and stability over time of the dentin collagen and, thus, confer a more resistant and lasting substrate for adhesive restorations. Methods: PA-based extracts, from grape seed (GSE), cocoa seed (CSE), cranberry (CRE), cinnamon (CNE) and acai berry (ACE) were applied over the demineralized dentin. The apparent elastic modulus (E) of the treated dentin collagen was analyzed over a 12 month period. Specimens were immersed in the respective solution and E values were obtained by a micro-flexural test at baseline, 10, 30, 60, 120 and 240 min. Samples were stored in artificial saliva and re-tested after 3, 6 and 12 months. Data was analyzed using ANOVA and Tukey test. Results: GSE and CSE extracts showed a time-dependent effect and were able to improve [240 min (MPa): GSE = 108.96 +/- 56.08: CSE = 59.21 +/- 24.87] and stabilize the E of the organic matrix [12 months (MPa): GSE = 40.91 +/- 19.69; CSE = 42.11 +/- 13.46]. CRE and CNE extracts were able to maintain the E of collagen matrices constant over 12 months [CRE = 11.17 +/- 7.22; CNE = 9.96 +/- 6.11; MPa]. ACE (2.64 +/- 1.22 MPa) and control groups immersed in neat distilled water (1.37 +/- 0.69 MPa) and ethanol-water (0.95 +/- 0.33 MPa) showed no effect over dentin organic matrix and enable their degradation and reduction of mechanical properties. Significance: Some PA-based extracts were capable of improving and stabilizing collagen matrices through exogenous cross-links induction. (C) 2011 Elsevier Ltd. All rights reserved.

Green tea, a product from Camellia sinensis, consists of over 200 componentes. The most known are the catechins, ou polyphenolic compounds, or flavonoids, but it contains also polysacharide conjugates, amino acids, caffeine and vitamins. Studies have suggested that the regular consumption of green tea reduces the risk of cancer, protecting agains initial events and progression of the disease, may act as antioxidant, has bactericidal properties, and that green tea consumption may be an adjuvant in the treatment of diabetes mellitus, hypertension and hypercholesterolemia. Our experimental study was carried out with male Wistar rats, separated in two groups, experimental and control. The animals in control group were given water ad libitum, and the animals in the experimental group received green tea instead of water. After 2 weeks of experimental period, all the animals, suffered surgical wounds on the dorsum. The animals were sacrificed on I, 3 and 7 days after surgery. Skin samples were used for light microscopy evaluation. Our results allowed us to observe that the experimental group (green tea drinking) presented accelerated epithelial neformation on day 3 after surgery, when epithelial neoformation is in the initial phases. The surgical areas presented 48.20% X 27.32% epithelial neoformation for green tea group and their controls respectivelly at this time. The results of the present study, along with other information about green tea in the literature, strongly suggests that regular use of green tea may be beneficial in surgery situations, when fast epithelial neoformation is desirable.

Este trabalho descreve a investigação química e biológica do extrato bruto e das partições hexano e acetato de etila, das folhas de Pyrostegia venusta (Ker Gawl.) Miers, popularmente conhecida como “cipó de São João”. P. venusta é classificada botanicamente como uma liana de porte mediano, tendo como característica uma exuberante floração vermelha, e por isso, sendo utilizada como planta ornamental. Essa planta possui uma larga utilização na medicina popular, sendo utilizada no tratamento de vitiligo, diarreia, bronquite, resfriado, icterícia e infecções. Os objetivos deste trabalho foram identificar as classes de metabólitos secundários presentes, avaliar o potencial antioxidante das amostras de P. vesnuta (extrato bruto, frações acetato de etila e hexano), quantificar o teor de flavonoides no extrato bruto, verificar a segurança do uso dessa planta, em termos de viabilidade celular (VC) frente à macrófagos murinos (RAW 264.7) (ensaio de imunotoxicidade). Adicionalmente os resultados de viabilidade celular foram comparados com quatro compostos anti-inflamatórios comerciais (ácido acetilsalicílico, indometacina, betametasona e piroxicam), e testar o extrato bruto quanto à inibição de catepsinas K e V. Os testes de identificação fitoquímica confirmaram a presença de flavonoides, cumarinas e esteroides nas amostras. A metodologia cromatográfica associada à análises por espectrometria de massas, levou a identificação dos compostos: fitol (1), sitosterol (2), estigmasterol (3) e campesterol (4). O extrato bruto demonstrou ter atividade inibitória frente as duas catepsinas testadas (K e V). A fração acetato de etila foi a que apresentou maior atividade antioxidante nas metodologias de inibição do radical DPPH (IC50 38,62 μg/mL) e radical ABTS (IC50 27,58 μg/mL). O teor de flavonoides total para o extrato bruto foi de 148,5±7,65 μg/mg (14,85 % (m/m)), o que justifica a observada atividade antioxidante, já que estes possuem atividade antioxidante. As amostras de P. venusta obtiveram valores de VC maiores do que os anti-inflamatórios comerciais, estes apresentaram VC abaixo do controle negativo, assim como o extrato bruto e a fração acetato de etila, a fração hexano obteve valores acima do controle negativo, sendo estes os maiores resultados de VC entre as amostras de P. venusta.

The objective of this study was to evaluate the influence of the color and phenolic compounds of strawberry jam on acceptance during storage. Jams were processed, stored for 120 days and evaluated monthly for chromatic characteristics, total phenolic compounds, total anthocyanins (ANT), total ellagic acid (TEA), flavonoids and free ellagic acid (FEA), and sensory acceptance as well. Data were submitted to analysis of variance (ANOVA) and the means were compared by the Least Significant Difference (LSD). Cluster Analysis and Partial Least Square Regression (PLS) were performed to investigate the relationships between instrumental data and acceptance. Contents of ANT, TEA and redness decreased during storage. Other chemical characteristics and sensory acceptance showed a nonlinear behavior. Higher acceptance was observed after 60 days, suggesting a trend of quality improvement followed by decline to the initial levels. The same trend was observed for lightness, non-pigment flavonoids and FEA. According to PLS map, for consumers in cluster 2, acceptance was associated to jams at 60 days and to luminosity, FEA, and non-pigment flavonoids. For cluster 1, a positive association between flavor liking, jam at initial storage, and the contents of TEA and ANT was indicated. Jams at 120 days were positively associated to hue and negatively associated to color liking, for cluster 1. Color and texture were positively correlated to overall liking for cluster 2, whereas for cluster 1, overall acceptance seemed to be more associated to flavor liking. Changes in color and phenolic compounds slightly influenced the acceptance of strawberry jams, but in different ways for consumers clusters.

Nowadays, new emerging products claiming antioxidant properties are becoming more frequent. However, information about this topic in their labels is usually scarce. In this paper, we analyzed total phenolics, total flavonoids and ascorbic acid contents, as well as DPPH scavenging activity of several commercial samples, namely green tea and other herbal infusions, dietary supplements, and fruit juices, available in the Portuguese market. In general, beverages containing green tea and hibiscus showed higher phenolics contents (including flavonoids) and antioxidant activity than those without these ingredients. A borututu infusion presented the lowest concentrations of bioactive compounds and scavenging activity, due to the low recommended amount of plant to prepare the beverage. Some juices without antioxidant claims in the label presented similar values to those with it.

Os polifenóis constituem um grupo biologicamente relevante de compostos naturais, que têm gerado um crescente interesse por parte dos consumidores e das indústrias alimentar, farmacêutica e de cosméticos. Os compostos fenólicos exibem uma vasta gama de propriedades fisiológicas, tais como anti-alérgica, anti-aterogénica, anti-inflamatória, anti-microbiana, antioxidante, anti-trombótica, possuindo efeitos cardio-protetores e vasodilatadores. Por conseguinte, o interesse na obtenção deste compostos ativos de uma forma fácil, barata e rápida tem vindo a aumentar. Uma das fontes de polifenóis são os subprodutos agrícolas, tais como os produtos derivados do vinho e/ou processamento de uvas. As folhas, os caules e os resíduos obtidos durante o processo de produção do vinho têm sido subutilizados, apesar de representarem uma boa fonte de compostos antioxidantes e bioativos. Portugal é um dos mais importantes países produtores de vinho, onde todos os anos são geradas várias toneladas de subprodutos associados a esta indústria. A extração, caracterização, quantificação e avaliação da atividade antioxidante de compostos bioativos extraídos destes subprodutos são, pois, tarefas importantes a fim de avaliar o potencial de utilização destes produtos como novas fontes de compostos antioxidantes. O objetivo deste trabalho foi quantificar e avaliar os teores de polifenóis de extratos preparados a partir de folhas e caules de videira, e bagaço, engaço e grainha de uva. Os parâmetros experimentais da extração foram otimizados, incluindo a proporção de solvente de extração (água/etanol), a temperatura e o tempo de extração. A quantificação dos polifenóis (como fenóis totais e flavonoides) foi efetuada por espetrofotometria de UV/Vis. Verificou-se que a melhor razão de solventes extratores foi de 50/50 (v/v) em água/etanol para todos os subprodutos estudados. No caso dos subprodutos folhas e caules, as condições ideais de extração foram obtidas quando se usou uma temperatura de 40ºC e um tempo de extração de 30 min. Para o bagaço, a temperatura ideal foi de 55ºC durante 5 min. Para o engaço e grainha a utilização de uma temperatura de 40ºC durante 5 min e 30 min, respetivamente, revelou-se o ideal para se obter os compostos. Com este trabalho conclui-se que os subprodutos da vinha e do vinho efetivamente possuem compostos de elevado valor e com capacidade antioxidante.

The antioxidant profiles of 39 water samples (29 flavored waters based on 10 natural waters) and 6 flavors used in their formulation (furnished by producers) were determined. Total phenol and flavonoid contents, reducing power, and DPPH radical scavenging activity were the optical techniques implemented and included in the referred profile. Flavor extracts were analyzed by HS-SPME/GC-MS to obtain the qualitative and quantitative profiles of the volatile fraction of essential oils. Results pointed out a higher reducing power (0.14 11.8 mg of gallic acid/L) and radical scavenging activity (0.29 211.5 mg Trolox/L) of flavored waters compared with the corresponding natural ones, an interesting fact concerning human health. Bioactive compounds, such as polyphenols, were present in all samples (0.5 359 mg of gallic acid/L), whereas flavonoids were not present either in flavored waters or in flavors. The major components of flavor extracts were monoterpenes, such as citral, R-limonene, carveol, and R-terpineol.

Folk medicine is a relevant and effective part of indigenous healthcare systems which are, in practice, totally dependent on traditional healers. An outstanding coincidence between indigenous medicinal plant uses and scientifically proved pharmacological properties of several phytochemicals has been observed along the years. This work focused on the leaves of a medicinal plant traditionally used for therapeutic benefits (Angolan Cymbopogon citratus), in order to evaluate their nutritional value. The bioactive phytochemical composition and antioxidant activity of leaf extracts prepared with different solvents (water, methanol and ethanol) were also evaluated. The plant leaves contained ~60% of carbohydrates, protein (~20%), fat (~5%), ash (~4%) and moisture (~9%). The phytochemicals screening revealed the presence of tannins, flavonoids, and terpenoids in all extracts. Methanolic extracts also contained alkaloids and steroids. Several methods were used to evaluate total antioxidant capacity of the different extracts (DPPH; NO; and H2O2 scavenging assays, reducing power, and FRAP). Ethanolic extracts presented a significantly higher antioxidant activity (p < 0.05) except for FRAP, in which the best results were achieved by the aqueous extracts. Methanolic extracts showed the lowest radical scavenging activities for both DPPH; and NO; radicals.

Fresh-cut vegetables are a successful convenient healthy food. Nowadays, the presence of new varieties of minimally processed vegetables in the market is common in response to the consumers demand for new flavours and high quality products. Within the most recent fresh-cut products are the aromatic herbs. In this work, the objective was to evaluate the nutritional quality and stability of four fresh-cut aromatic herbs. Several physicochemical quality characteristics (colour, pH, total soluble solids, and total titratable acidity) were monitored in fresh-cut chives, coriander, spearmint and parsley leaves, stored under refrigeration (3 ± 1 ºC) during 10 days. Their nutritional composition was determined, including mineral composition (phosphorous, potassium, sodium, calcium, magnesium, iron, zinc, manganese and copper) and fat- and water-soluble vitamin contents. Total soluble phenolics, flavonoids and the antioxidant capacity were determined by spectrophotometric methods. The aromatic herbs kept their fresh appearance during the storage, maintaining their colour throughout shelf-life. Their macronutrient composition and mineral content were stable during storage. Coriander had the highest mineral and fatsoluble vitamin content, while spearmint showed the best scores in the phenolic, flavonoid and antioxidant capacity assays. Vitamins and antioxidant capacity showed some variation during storage, with a differential behaviour of each compound according to the sample.

The effect of organic and conventional agricultural systems on the physicochemical parameters, bioactive compounds content, and sensorial attributes of tomatoes (‘‘Redondo’’ cultivar) was studied. The influence on phytochemicals distribution among peel, pulp and seeds was also accessed. Organic tomatoes were richer in lycopene (+20%), vitamin C (+30%), total phenolics (+24%) and flavonoids (+21%) and had higher (+6%) in vitro antioxidant activity. In the conventional fruits, lycopene was mainly concentrated in the pulp, whereas in the organic ones, the peel and seeds contained high levels of bioactive compounds. Only the phenolic compounds had a similar distribution among the different fractions of both types of tomatoes. Furthermore, a sensorial analysis indicated that organic farming improved the gustative properties of this tomato cultivar.
