The colloidal stability of poly(ethylene glycol)-decorated poly(methyl methacrylate), PMMA/Tween-20, particles was investigated by means of phase separation measurements, in the presence of sodium fluoride (NaF), sodium chloride, sodium bromide, sodium nitrate, or sodium thiocyanate (NaSCN) at 1.0 mol L-1. Following Hofmeister's series, the dispersions of PMMA/Tween-20 destabilized faster in the presence of NaF than with NaSCN. After the phase separation, the systems were homogenized and except for the dispersions in NaF, re-dispersed particles took longer to destabilize, indicating that anions adsorbed on the particles, creating a new surface. Except for F- ions, the adsorption of anions on the polar outmost shell was evidenced by means of tensiometry and small-angle X-ray scattering measurements. Fluoride ions induced the dehydration of the polar shell, without affecting the polar shell electron density, and the formation of very large aggregates. A model was proposed to explain the colloidal behavior in the presence of Hofmeister ions.

In gene-banking, primordial germ cells (PGCs), which are embryonic precursor cells of germ cells, are useful for cryopreservation because PGCs have a potential to differentiate into both eggs and sperm via germ-line chimera. Here, we have established vitrification methods for PGCs cryopreservation using 12- to 17-somite stage embryos in loach, Misgurnus anguillicaudatus, which were dechorionated, removed their yolk and injected with green fluorescent protein (GFP) -nos1 3'UTR mRNA to visualize their PGCs. In order to optimize cryopreservation medium for vitrification, the toxicity of cryoprotectants was analyzed. Different concentrations (2, 3, 4, 5 m) of dimethyl sulfoxide (DMSO), methanol (MeOH), ethylene glycol (EG) and propylene glycol (PG) as cryoprotectants were tested. Then, 5 m DMSO showed significantly-high toxicity. Based on this information, combinations called DMP (2 m (14.2% [v/v]) DMSO, 2 m (8.1% [v/v]) MeOH and 2 m (14.4% [v/v]) PG), DP (2 m (14.2% [v/v]) DMSO and 4 m (28.7% [v/v]) PG) and DE (2.1 m (15% [v/v]) DMSO and 2.7 m (15% [v/v]) EG) were evaluated for their toxicities and efficacy of PGCs cryopreservation using two types of equilibration step: direct immersion of cryopreservation media (one-step) and serial exposure to half and full concentration of cryopreservation media (two-step). Viable PGCs were obtained from post-thaw embryos which were cryopreserved by DP and DE with both 1- and 2-step equilibrations. Despite DP showing the highest toxicity, it gave the highest survival rate of embryonic cells after cryopreservation. When PGCs recovered from vitrified embryos were transplanted into host embryos at the blastula stage, the transplanted PGCs were able to migrate to a host genital ridge similarly as endogenous PGCs. It suggests that our methods could be useful to create a germ-line chimera for the production of gametes from PGCs of cryopreserved embryos.

Poly(ethylene tereftalate) (PET) is a polymer highly susceptible to the hydrolytic reactions that occur during applications and mainly in thermomechanical processing. These reactions lead to the decrease of molecular weight of the polymer, limiting the recycling number of the material. The reactive extrusion of the PET in presence of chain extenders is an alternative to recover mechanical and rheological properties that were depreciated by the polymer degradation. In this study, PET wastes from nonwoven fabrics production were extruded in presence of the secondary stabilizer Irgafos 126 (IRG) on variable concentrations. The results showed that Irgafos 126 increased molecular weight, decreased crystallinity and changed processing behavior of the PET, similarly to the effects produced by the well-known chain extender pyromellitic dianhydride (PMDA), showing that the secondary stabilizer Irgafos 126 can also act as a chain extender for the PET.

In this study, Cross-Polarization Magic-angle Spinning CP/MAS, 2D Exchange, Centerband-Only Detection of Exchange (CODEX), and Separated-Local-Field (SLF) NMR experiments were used to study the molecular dynamics of poly(ethylene glycol) (PEG) inside Hectorite/PEG intercalation compounds in both single- and double-layer configurations. The results revealed that the overall amplitude of the motions of the PEG chain in the single-layer configuration is considerably smaller than that observed for the double-layer intercalation compound. This result indicates that the effect of having the polymer chain interacting with both clay platelets is to produce a substantial decrease in the motional amplitudes of those chains. The presence of these dynamically restricted segments might be explained by the presence of anchoring points between the clay platelets and the PEG oxygen atoms, which was induced by the Na+ cations. By comparing the PEG motional amplitudes of the double-layered nanocomposites composed of polymers with different molecular weights, a decrease in the motional amplitude for the smaller PEG chain was observed, which might also be understood using the presence of anchoring points.

In der vorliegenden Arbeit erfolgten Oberflchenmodifizierungen durch Polymere nach zwei Anstzen. Dies war zum einen ein Ansatz, bei dem die Oberflchen mit Diblockcopolymeren versehen wurden. Diese bestanden aus einem Ankerblock, der starke Wechselwirkungen mit der Oberflche zeigt, und einem Bojenblock, der gezielte Eigenschaften trgt. Zum anderen erfolgten Modifizierungen durch auf Plasmaschichten verankerte Homopolymere. Beide Anstze erfolgten auf zwei Substraten von unterschiedlichen Eigenschaften. Diese waren das Siliciumoxid, fr das Modifizierungen durch radikalische in-situ Oberflchenpolymerisation, und das Poly(ethylen-stat-norbornen), fr das Modifizierungen durch ex-situ dargestellte Polymere gewhlt wurden. Beim ersten Ansatz zur Modifizierung der Siliciumoxidoberflche ermglichte ein adsorbierter Poly(e-caprolacton)-Makroinitiator die Oberflchenpolymerisation hin zu oberflchenverankertem Poly(e-caprolacton)-block-poly(alkyl(meth)acrylat). Beim zweiten Ansatz erfolgte die Abscheidung von plasmapolymerisiertem Allylamin, die Immobilisierung des Azoinitiators 4,4-Azobis(4-cyanopentansurechlorid) und die nachfolgende Oberflchenpolymerisation von Methylmethacrylat oder Styrol. Beim ersten Modifizierungsansatz der Poly(ethylen-stat-norbornen)-Oberflche sollte diese mit thermisch interdiffundierten Poly(ethylen-alt-propylen)-block-poly(dimethylsiloxan) versehen werden. Trotz erfolgreicher Synthese wurde gezeigt, da keine Interdiffusion stattfand. Im zweiten Modifizierungsansatz wurde die Oberflche mit aus einem Hexamethyldisiloxan/Sauerstoff-Plasma abgeschiedenem reinem Siliciumoxid beschichtet, woran sich die Adsorption von Poly(dimethylsiloxan) anschlo. Damit konnten die hohen Haftreibungskrfte gegenber Halogenbutylgummi erfolgreich beseitigt werden.

Das Ziel der vorgelegten Arbeit war die Synthese von definierten, sphrischen Polystyrolpartikeln im Grenbreichen von Nanometern, die als Trger fr die Immobilisierung von Metallocenkatalysatoren verwendet werden sollten. Ein wichtiger Anspruch an das System war dabei die Mglichkeit einer homogene Verteilung des Metallocenes auf dem Trger and eine homogene Fragmentierung des getrgerten Katalysators whrend der Polymerisation im Polymerprodukt. Fr diese Zielsetzung wurden unterschiedliche Polystyrolnanopartikel hergestellt. Die Polystyrolnanopartikel waren mit unterschiedlichen funktionellen Gruppen wie Polyethylenoxid- und Polypropylenoxidketten oder Hydroxygruppen auf der Oberflche versehen, um den Metallocenkatalysator und den Cokatalysator MAO immobilisieren zu knnen. In verschiedenen Experimenten wurde der Einfluss dieser Polystyrolnanopartikel als Trger auf die Katalysatoreigenschaften wie Aktivitt oder Produktivitt und die Eigenschaften des produzierten Polyolefins wie z.B. Molekulargewicht und Morphologie untersucht. Im Vergleich zu den PS- Nanopartikeln wurden auerdem PS-Mikropartikel, Silica und Dendrimere als Trger in der heterogenen Olefinpolymerisation eingesetzt. Von all diesen Trgersystemen wurde das Fragmentierungsverhalten durch konfocale Fluoreszenzmikroskopie untersucht. Aus den erhaltenen Ergebnissen kann geschlossen werden, dass die hergestellten Polystyrolnanopartikel neuartige und leistungsfhige Trger fr heterogene Polymerisationsprozesse darstellen. Die hergestellten Polystyrolnanopartikel besaen eine wohldefinierte sphrische Struktur, die eine homogene Verteilung des immobilisierten Metallocenkatalysators und somit auch eine vollstndige Fragmentierung des getrgerten Katalysators im hergestellten Polyolefin ermglichte. Die Katalysatorsysteme, die aus den PS- Nanopartikeln und dem Metallocenkatalysator zusammengesetzt waren, wurden in verschiedenen Polymerisationen wie der Ethylen- oder Propylenhomopolymersation und der Copolymerisation von Ethen mit - Olefinen getestet. Die Oberflchen- funktionalisierten PS Nanopartikel immobilisierten den Metallocenkatalysator ausreichend gut, so dass kein Leachen (Ablsen) des Katalysators von der Trgeroberflche festgestellt werden konnte und deshalb Polymer von sehr guter Morphologie erhalten wurde. Um die Fragmentierung des Katalysators und den inneren Aufbau des Polymers nher untersuchen zu knnen, wurde die konfocale Fluoreszenzmikroskopie fr das PS- Nanopartikeltrgersystem angewendet. Durch farbstoffmarkierte Trgerpartikel konnte die Verteilung des fragmentierten Katalysators innerhalb des Polymers sichtbar gemacht und analysiert werden. Dabei wurde festgestellt, dass sich PS- Nanopartikel und auch Dendrimere als Trger hnlich verhalten wie Ziegler- Natta- Katalysatoren, die auf MgCl2 immobilisiert fr die heterogene Olefinpolymerisation verwendet werden. Das Fragmentierungsverhalten der Silica oder PS- Mirkopartikel getrgerten Systeme entsprach dagegen dem schichtweisen Fragmentierungsverhalten wie es bereits von Fink und Mitarbeitern beschrieben wurde.

The rheological properties of block co-polymers in water solution at different pH have been investigated. The block copolymers are based on different architectures containing poly(ethylene glycol), poly(propylene glycol) and different blocks of polymer that change their hydrophobic/hydrophilic behavior as a function of pH. The polymer chains of the starting material were extended at their functional ends with the pH-sensitive units using ATRP; this mechanism of controlled radical polymerization was chosen because of the need to minimize polydispersity and avoid transfer reactions possibly leading to homopolymeric inpurities. The starting material were modified in order to use them as macroinitiator for ATRP. The kinetic of each ATRP reaction has been investigated, in order to be able to synthesize polymers with different degree of polymerization, stopping the reaction when the desired polymers chain length has been reached. We will use polymer chains with different basicity and degree of polymerization to link any possible effect of their presence to the conditions under which they become hydrophobic. It has been shown that the rate of polymerization changes changing the type of macroinitiator and the type of monomer synthesized. The slowest rate of polymerization is the one with the most hindered monomer synthesized using the macroinitiator with the highest molecular weight. The water solubility of the synthesized polymers changes depending on the pH of the solution and on the structure of the polymers. It has been shown using 1H-NMR that some of the synthesized polymers are capable to self-aggregation in water solution. The self-aggregation and the type of aggregation is influenced from the structure of the polymer and from the pH of the solution. Changing the structure of the polymers and the pH it is possible to obtain different type of aggregates in solution. This aggregates differ for the volume occupied from them, and for their hardness. Rheological measurements have been demonstrated that the synthesized polymers are capable to form gel phases. The gelation temperature changes changing the structure of the aggregates in solution and it is possible to correlate the changing in the gelation temperature with the changing in the structure of the polymer.

Grape berry is considered a non climacteric fruit, but there are some evidences that ethylene plays a role in the control of berry ripening. This PhD thesis aimed to give insights in the role of ethylene and ethylene-related genes in the regulation of grape berry ripening. During this study a small increase in ethylene concentration one week before vraison has been measured in Vitis vinifera L. Pinot Noir grapes confirming previous findings in Cabernet Sauvignon. In addition, ethylene-related genes have been identified in the grapevine genome sequence. Similarly to other species, biosynthesis and ethylene receptor genes are present in grapevine as multi-gene families and their expression appeared tissue or developmental specific. All the other elements of the ethylene signal transduction cascade were also identified in the grape genome. Among them, there were ethylene response factors (ERF) which modulate the transcription of many effector genes in response to ethylene. In this study seven grapevine ERFs have been characterized and they showed tissue and berry development specific expression profiles. Two sequences, VvERF045 and VvERF063, seemed likely involved in berry ripening control due to their expression profiles and their sequence annotation. VvERF045 was induced before vraison and was specific of the ripe berry, by sequence similarity it was likely a transcription activator. VvERF063 displayed high sequence similarity to repressors of transcription and its expression, very high in green berries, was lowest at vraison and during ripening. To functionally characterize VvERF045 and VvERF063, a stable transformation strategy was chosen. Both sequences were cloned in vectors for over-expression and silencing and transferred in grape by Agrobacterium-mediated or biolistic-mediated gene transfer. In vitro, transgenic VvERF045 over-expressing plants displayed an epinastic phenotype whose extent was correlated to the transgene expression level. Four pathogen stress response genes were significantly induced in the transgenic plants, suggesting a putative function of VvERF045 in biotic stress defense during berry ripening. Further molecular analysis on the transgenic plants will help in identifying the actual VvERF045 target genes and together with the phenotypic characterization of the adult transgenic plants, will allow to extensively define the role of VvERF045 in berry ripening.

This thesis is based on three main studies, all dealing with structure-property investigation of semicrystalline polyolefin-based composites. Low density poly(ethylene) (LDPE) and isotactic poly(propylene) (iPP) were chosen as parts of the composites materials and they were investigated either separately (as homoploymers), either in blend systems with the composition LDPE/iPP 80/20 or as filled matrix with layered silicate (montmorillonite). The beneficial influence of adding ethylene-co-propylene polymer of amorphous nature, to low density poly(ethylene)/isotactic poly(propylene) (80/20) blend is demonstrated. This effect is expressed by the major improvement of mechanical properties of ternary blends as examined at a macroscopic size scale by means of tensile measurements. The structure investigation also reveals a clear dependence of the morphology on adding ethylene-copropylene polymer. Both the nature and the content of ethylene-co-propylene polymer affect structure and properties. It is further demonstrated that the extent of improvement in mechanical properties is to be related to the molecular details of the compatibilizer. Combination of high molecular weight and high ethylene content is appropriate for the studied system where the poly(ethylene) plays the role of matrix. A new way to characterize semicrystalline systems by means of Brillouin spectroscopy is presented in this study. By this method based on inelastic light scattering, we were able to measure the high frequency elastic constant (c11) of the two microphases in the case where the spherulites size is exhibit size larger than the size of the probing phonon wavelength. In this considered case, the sample film is inhomogeneous over the relevant length scales and there is an access to the transverse phonon in the crystalline phase yielding the elastic constant c44 as well. Isotactic poly(propylene) is well suited for this type of investigation since its morphology can be tailored through different thermal treatment from the melt. Two distinctly different types of films were used; quenched (low crystallinity) and annealed (high crystallinity). The Brillouin scattering data are discussed with respect to the spherulites size, lamellae thickness, long period, crystallinity degree and well documented by AFM images. The structure and the properties of isotactic poly(propylene) matrix modified by inorganic layered silicate, montmorillonite, are discussed with respect to the clay content. Isotactic poly(propylene)-graft-maleic anhydride was used as compatibilizer. It is clearly demonstrated that the property enhancement is largely due to the ability of layered silicate to exfoliate. The intimate dispersion of the nanometer-thick silicate result from a delicate balance of the content ratio between the isotactic poly(propylene)-graft-maleic anhydride compatibilizer and the inorganic clay.

Ethylene plays an important role in apple fruit development. Its biosynthesis is catalyzed by two enzymes ACS and ACO. The first is considered to catalyzes the rate-limiting step of ethylene production and in apple two different alleles (MdACS1-1 and MdACS1-2) of this gene have been identified. The presence in the promoter region of MdACS1-2 allele of a SINE insertion is considered to be responsible for a low transcription level and a pronounced reduction in ethylene production in apple cultivar homozygous for this allele. However, the specific expression of each MdACS1 allele has never been reported as well as any in vivo analysis of its 5-flanking region. With the present study we addressed these issues by developing a set of qPCR allele specific primers for MdACS1 and by a functional characterization of the MdACS1 promoters by transient expression analysis. qPCR analysis on different apple tissues and stages of development demonstrated that MdACS1-2 allele is never express and that MdACS1-1 allele is ripening-related and expresses predominantly but not exclusively in apple fruit. To test MdACS1 promoter in fruit the only protocol available in literature for transient transformation of apple fruit was evaluated and optimized. Twenty chimeric promoter::reporter constructs were generated and analyzed by Agrobacterium-transient transformation. The in vivo analysis allowed to identify an enhancer-like region of 261 bp in MdACS1 promoter and a region of 57 bp in MdACS1-2 responsible, also if not alone, in the inactivation of the MdACS1-2 allele. Through the assessment of ethylene production in a segregating progeny derived from the cross between Fuji and Mondial Gala (homozygous for MdACS1-2 allele) we demonstrated that at least two other genes may be involved in apple ethylene production. An hypothesis that could explain the difference between Fuji and Mondial Gala have been proposed.

In der hier vorliegenden Arbeit wurden neue eisenhaltige Spincrossover-Komplexerndargestellt und deren magnetische Eigenschaften untersucht. Ausgehend von frheren Ergebnissen wurden verschiedene Strategien zur Optimierung der Eisen-Spincrossover Verbindungen verfolgt. Wie schon frher dokumentiert finden sich bei Eisen-bergangsmetall Komplexen eine Vielzahl von Spincrossover Phnomenen. Ebenso gut dokumentiert sind die Mglichen nderungen des Spin Zustandes durch uere Einflsse wie Temperatur, Druck oder Licht. Darauf aufbauend wurden nunrnverschiedene Eisenkomplexe synthetisiert und auf das Spincrossover Phnomen hin untersucht. Dazu wurden zum Einen Fe(II) Komplexe vom Typ [FeL1(NCS)2] (L1 = pmea, pmap, tepa and tmpa) betrachtet und zum anderen Sternfrmige Fe(III) Komplexe vom Typ [M{(CN-FeIIIL2}x]y+. (M=Fe(II), Co(III), Mo(IV), Ru(II)) undrndodecanukleare Komplexe vom Typ [(L2Fe(III)NC)5Fe(II)CNCo(III)(CNFe(III)L2)5]4+. L2= Bis(R2,3,4 -salicylidenaminoalkyl-R1-amin. Thermischen Spincrossover und LIESST zeigen [3,3/N-H/Sal-H/Fe/Co]; [3,3/N-H/Sal-H/Fe/Ru]; [3,3/N-H/sal-H/Fe/Mo]; [3,3/N-H/Sal-H/Fe/W]; FeII(pmea)(SCN)2; thermischen Spinbergang zeigt [2,3/N-H/Sal-H/Fe/Fe-Co]. Die Ethylen gebrckten Komplexe zeigen weniger guten oder gar keinen Schalteffekt im Gegensatz zum Propylen gebrckten Komplexe. Fe(II)(PMEA)(SCN)2 zeigt vollstndigen thermischen Spinbergang von High Spin nach Low Spin ein LIESST und einen LiPTH Effekt.

Poly(ethylene glycol) (PEG) is used in a broad range of applications due to its unique combination of properties and is approved use in formulations for body-care products, edibles and medicine. This thesis aims at the synthesis and characterization of novel heterofunctional PEG structures and the establishment of diethyl squarate as a suitable linker for the covalent attachment to proteins. Chapter 1 is an introduction on the properties and applications of PEG as well as the fascinating chemistry of squaric acid derivatives. In Chapter 1.1, the synthesis and properties of PEG are described, and the versatile applications of PEG derivatives in everyday products are emphasized with a focus on PEG-based pharmaceuticals and nonionic surfactants. This chapter is written in German, as it was published in the German Journal Chemie in unserer Zeit. Chapter 1.2 deals with PEGs major drawbacks, its non-biodegradability, which impedes parenteral administration of PEG conjugates with polyethers exceeding the renal excretion limit, although these would improve blood circulation times and passive tumor targeting. This section gives a comprehensive overview of the cleavable groups that have been implemented in the polyether backbone to tackle this issue as well as the synthetic strategies employed to accomplish this task. Chapter 1.3 briefly summarizes the chemical properties of alkyl squarates and the advantages in protein conjugation chemistry that can be taken from its use as a coupling agent. In Chapter 2, the application of diethyl squarate as a coupling agent in the PEGylation of proteins is illustrated. Chapter 2.1 describes the straightforward synthesis and characterization of squaric acid ethyl ester amido PEGs with terminal hydroxyl functions or methoxy groups. The reactivity and selectivity of theses activated PEGs are explored in kinetic studies on the reactions with different lysine and other amino acid derivatives, followed by 1H NMR spectroscopy. Further, the efficient attachment of the novel PEGs to a model protein, i.e., bovine serum albumin (BSA), demonstrates the usefulness of the new linker for the PEGylation with heterofunctional PEGs. In Chapter 2.3 initial studies on the biocompatibility of polyether/BSA conjugates synthesized by the squaric acid mediated PEGylation are presented. No cytotoxic effects on human umbilical vein endothelial cells exposed to various concentrations of the conjugates were observed in a WST-1 assay. A cell adhesion molecule - enzyme immunosorbent assay did not reveal the expression of E-selectin or ICAM-1, cell adhesion molecules involved in inflammation processes. The focus of Chapter 3 lies on the syntheses of novel heterofunctional PEG structures which are suitable candidates for the squaric acid mediated PEGylation and exhibit superior features compared to established PEGs applied in bioconjugation. Chapter 3.1 describes the synthetic route to well-defined, linear heterobifunctional PEGs carrying a single acid-sensitive moiety either at the initiation site or at a tunable position in the polyether backbone. A universal concept for the implementation of acetal moieties into initiators for the anionic ring-opening polymerization (AROP) of epoxides is presented and proven to grant access to the degradable PEG structures aimed at. The hydrolysis of the heterofunctional PEG with the acetal moiety at the initiating site is followed by 1H NMR spectroscopy in deuterium oxide at different pH. In an exploratory study, the same polymer is attached to BSA via the squarate acid coupling and subsequently cleaved from the conjugate under acidic conditions. Furthermore, the concept for the generation of acetal-modified AROP initiators is demonstrated to be suitable for cholesterol, and the respective amphiphilic cholesteryl-PEG is cleaved at lowered pH. In Chapter 3.2, the straightforward synthesis of -amino 2-dihydroxyl star-shaped three-arm PEGs is described. To assure a symmetric length of the hydroxyl-terminated PEG arms, a novel AROP initiator is presented, whos primary and secondary hydroxyl groups are separated by an acetal moiety. Upon polymerization of ethylene oxide for these functionalities and subsequent cleavage of the acid-labile unit no difference in the degree of polymerization is seen for both polyether fragments.

Poly(ethylenglykol) (PEG) ist eines der wichtigsten Polymere fr pharmazeutische und biomedizinische Zwecke. Dies lsst sich vor allen Dingen auf seine ausgezeichnete Biokompatibilitt, seine hohe chemische Stabilitt sowie seine sehr gute Wasserlslichkeit zurckfhren. Neben seiner Anwendung in Produkten wie Lebensmitteln und Kosmetika ist PEG vor allem im pharmazeutischen Bereich unersetzlich geworden. Hier dient PEG als Grundlage fr Salben, es kommt aber auch in der sogenannten PEGylierung zum Einsatz. Unter PEGylierung versteht man die kovalente Verknpfung von PEG mit Wirkstoffmoleklen, beispielsweise Proteinen oder niedermolekularen Medikamenten. In der akademischen Forschung sind aber auch PEGylierte Nanopartikel oder durch PEG stablisierte Liposomen fr die Applikation im Bereich der Medizin von hohem Interesse. Trotz seiner breiten Verwendung hat PEG zwei entscheidende Nachteile: Zum einen bentigt man gerade im Hinblick auf PEGylierungen viele funktionelle Gruppe, jedoch trgt PEG maximal zwei Hydroxyl-Gruppen (die Endgruppen), die fr kovalente Verknpfungen genutzt werden knnen. Zum anderen ist PEG nicht in physiologischer Umgebung abbaubar und kann daher in vivo oberhalb eines Molekulargewichts von 40 000 g/mol nicht eingesetzt werden, da sonst eine Ausscheidung ber die Niere nicht mglich ist und eine ungewollte Anreicherung im Krper stattfindet.rnDie durch die geringe Anzahl an Endgruppen limitierte Beladungsdichte kann durch das Design neuer Epoxid-Derivate und deren statistischen Einbau in das PEG Rckgrat deutlich verbessert werden. Im ersten Teil dieser Arbeit werden drei neuartige funktionelle Oxirane vorgestellt, die systematisch mit Ethylenoxid copolymerisiert wurden, was die selektive Einfhrung verschiedener funktioneller Gruppen am Polymerrckgrat ermglicht. Im Vordergrund der Betrachtungen standen die Eigenschaften der neuartigen multifunktionellen (mf)-PEG Copolymere im Hinblick auf ihr thermisches Verhalten sowie die Verteilung der funktionellen Gruppen (Mikrostruktur) innerhalb des PEG-Rckgrats. Die gezielte Adressierbarkeit der funktionellen Gruppen konnte durch verschiedene Modellreaktionen besttigt werden. Darber hinaus konnte gezeigt werden, dass sich mit der vorgestellten Synthesestrategie komplexe Hybridmaterialien, beispielsweise metallhaltige Polyether, darstellen lassen. Mit Hinblick auf die biomedizinischen Anwendungen und die Konkurrenz zu etablierten PEG-Hompolymeren, standen die Wasserlslichkeit und die Toxizitt der synthetisierten Materialien im Zentrum weiterer Untersuchungen. Alle dargestellten Polymere zeigten einen Trbungspunkt in Wasser, der sich in Abhngigkeit der Zusammensetzung und Hydrophobizitt der Comonomere ber ein weites Temperaturspektrum variieren und somit systematisch einstellen lie. Die Toxizitt der statistischen mf-PEGs lag im Bereich von PEG, was die mf-PEGs interessant fr biomedizinische Anwendung macht.rnIm zweiten Teil der Arbeit wurden Copolymerisationen verwendet, um ber erstmals hergestellte Epoxid-Inimere sauer spaltbare Einheiten in das Polyetherrckgrat einzufhren. Die neuen, verzweigten Strukturen wurden auf die Zersetzung in physiologisch relevantem Milieu untersucht. Die erzielte pH-abhngige Spaltbarkeit, kann fr potenzielle Anwendungen beispielsweise in der Krebstherapie, von Vorteil sein.rn

Different concepts for the synthesis of sulfur-containing polymers as well as their adsorption onto gold surfaces were studied. The present work is divided into three parts. The main part focuses on the synthesis of poly(1,2-alkylene sulfides) (polysulfides) with complex architectures on the basis of polyether-based macroinitiators by the anionic ring-opening polymerization of ethylene sulfide and propylene sulfide. This synthetic tool kit allowed the synthesis of star-shaped, brush-like, comb-like and pom-pom-like polysulfides, the latter two with an additional poly(ethylene glycol) chain. Additionally, the number of polysulfide arms as well as the monomer composition could be varied over a wide range to obtain copolymers with multiple thioether functionalities.rnThe second section deals with the synthesis of a novel lipoic acid-based initiator for ring-opening polymerizations for lactones and epoxides. A straightforward approach was selected to accomplish the ability to obtain tailored polymers with a common used disulfide-anchoring group, without the drawbacks of post-polymerization functionalization. rnIn the third part, a new class of block-copolymers consisting of polysulfides and polyesters were investigated. For the first time this approach enabled the use of hydroxyl-terminated poly(propylene sulfide) as macroinitiator for the synthesis of a second block.rnThe adsorption efficiency of those different polymer classes onto gold nanoparticles as well as gold rnsupports was studied via different methods.rn

This work covers the synthesis of second-generation, ethylene glycol dendrons covalently linked to a surface anchor that contains two, three, or four catechol groups, the molecular assembly in aqueous buffer on titanium oxide surfaces, and the evaluation of the resistance of the monomolecular adlayers against nonspecific protein adsorption in contact with full blood serum. The results were compared to those of a linear poly(ethylene glycol) (PEG) analogue with the same molecular weight. The adsorption kinetics as well as resulting surface coverages were monitored by ex situ spectroscopic ellipsometry (VASE), in situ optical waveguide lightmode spectroscopy (OWLS), and quartz crystal microbalance with dissipation (QCM-D) investigations. The expected compositions of the macromolecular films were verified by X-ray photoelectron spectroscopy (XPS). The results of the adsorption study, performed in a high ionic strength ("cloud-point") buffer at room temperature, demonstrate that the adsorption kinetics increase with increasing number of catechol binding moieties and exceed the values found for the linear PEG analogue. This is attributed to the comparatively smaller and more confined molecular volume of the dendritic macromolecules in solution, the improved presentation of the catechol anchor, and/or their much lower cloud-point in the chosen buffer (close to room temperature). Interestingly, in terms of mechanistic aspects of "nonfouling" surface properties, the dendron films were found to be much stiffer and considerably less hydrated in comparison to the linear PEG brush surface, closer in their physicochemical properties to oligo(ethylene glycol) alkanethiol self-assembled monolayers than to conventional brush surfaces. Despite these differences, both types of polymer architectures at saturation coverage proved to be highly resistant toward protein adsorption. Although associated with higher synthesis costs, dendritic macromolecules are considered to be an attractive alternative to linear polymers for surface (bio)functionalization in view of their spontaneous formation of ultrathin, confluent, and nonfouling monolayers at room temperature and their outstanding ability to present functional ligands (coupled to the termini of the dendritic structure) at high surface densities.
