Introduction: This study aimed to investigate the effects of the two peptide NOP partial agonists (UFP-113 and [F/G]N/OFQ(1-13)NH2) and the non peptide NOP partial agonist (AT-090) in the mouse emotional behavior as well as in the intracellular transduction pathways following the receptor binding. Methods: Male Swiss or CD-1 mice were used in this study together with NOP(+/+) and NOP(-/-) mice. The elevated plus maze (EPM) was used to evaluate the effects of compounds on anxiety-like behaviors. Diazepam and the NOP agonists, N/OFQ and Ro 65-6570, were used as positive controls in the EPM. NOP(+/+) and NOP(-/-) mice were used to evaluate the selectivity of those compounds that induced anxiolytic-like behaviors. The forced swim test (FST) was used to evaluate the effects of compounds on depressive-like behaviors. Nortriptyline and the NOP antagonists, UFP-101 and SB-612111, were used as positive controls in the FST. The effects of N/OFQ, UFP-101, SB-612111, UFP-113, [F/G]N/OFQ(1-13)NH2, and AT-090 were assessed in the methylphenidate-induced hyperlocomotion (MIH) test; in this assay valproate was used as positive control. The G protein and β-arrestin 2 transduction pathways of NOP receptor agonists (N/OFQ and Ro 65-6570), antagonist (UFP-101), and partial agonists (UFP-113, [F/G]N/OFQ(1-13)NH2, and AT-090) were also evaluated using an innovative assay that measures a bioluminescence resonance energy transfer process. For this, cell lines permanently co-expressing the NOP receptor coupled to luciferase (energy donor), and green fluorescent protein (energy acceptor) coupled to one of the effector proteins (G protein or β-arrestin 2) were used. Results: Diazepam (1 mg/kg), N/OFQ (1 nmol), Ro 65-6570 (0.1 mg/kg), and AT-090 (0.01 mg/kg) induced anxiolytic-like effect in mice in the EPM. The effects of Ro 65-6570 and AT-090 were selective to NOP receptor. UFP-113 (0.01-1 nmol) and [F/G]N/OFQ(1-13)NH2 (0.1-3 nmol) were inactive in the EPM. In the FST, nortriptyline (30 mg/kg), UFP-101 (10 nmol), SB-612111 (10 mg/kg), UFP-113 (0.01 and 0.1 nmol), and [F/G]N/OFQ(1-13)NH2 (0.3 and 1 nmol) induced antidepressant-like effects, while AT-090 (0.001-0.1 mg/kg) was inactive in this assay. The effects of UFP-113 and [F/G]N/OFQ(1-13)NH2 were selective to NOP receptor. Valproate (400 mg/kg) counteracted methylphenidate (MPH, 10 mg/kg)-induced hyperlocomotion in mice in the open field. N/OFQ (1 nmol), UFP-113 (0.01-0.1 nmol), and [F/G]N/OFQ(1-13)NH2 (1 nmol) were also able to reduce the MPH-induced hyperlocomotion, without changing the locomotor activity per se. The effect of UFP-113 was selective to NOP receptor. The UFP-101 (10 nmol), SB-612111 (10 mg/kg), and AT-090 (0.001-0.03 mg/kg) did not change the hyperlocomotor effect of methylphenidate. In vitro, N/OFQ and Ro 65-6570 behaved as NOP full agonists for G-protein and β-arrestin 2 pathways. AT-090 behaved as NOP receptor partial agonist for both transduction pathways, while UFP-113 and [F/G]N/OFQ(1-13)NH2 behaved as partial agonists and antagonists of NOP receptor for NOP/G protein and NOP/β-arrestin 2, respectively. UFP-101 behaved as NOP receptor antagonist for both transduction pathways. Conclusion: NOP ligands producing same effects on NOP/G protein interaction (partial agonism), but with opposite effects on β-arrestin 2 recruitment (partial agonism vs antagonism), can promote different in vivo effects on anxiety and mood as it was observed in the behavioral tests. This work corroborates the potential of NOP receptor as an innovative pharmacological target for the treatment of emotional disorders.

The medicinal plants constitute a rich source of biologically active compounds used for the treatment of many psychiatric disorders, such as anxiety disorders and depression. Generalized anxiety disorder has increased significantly, being the second most prevalent disorder in care facilities to public health. Depression is considered a chronic and common psychiatric disorder that affects 350 million people of all ages around the world. In this context, the pharmacological intervention conduits have been employed, effective, although leave to be desired when observed adverse effects. The genus Passiflora is commonly commercially known by its fruit, but is also widely used in traditional Brazilian medicine. Passiflora edulis displays considerable morphological variability. This plant produces two types of fruit: Purple (Passiflora edulis Sims fo. edulis) and yellow (Passiflora edulis fo. flavicarpa Degener). This study investigated the central effects of aqueous extract of the leaves of the two varieties of the species Passiflora edulis in tests used to assess behavior related to anxiety and depression, as well as investigating the potential effect of the antidepressant-like fractions of edulis fo. edulis and neuropharmacological mechanisms responsible for this action. To conduct this study used male Swiss mice (2 months old, weighing 30-35 g). The animals received the aqueous extract of the leaves of the two species of Passiflora: edulis fo. edulis (100, 300, 1000 mg / kg) and fractions ethyl acetate, butanol and aqueous waste (25, 50, 75, 100 mg / kg) and edulis fo. flavicarpa (30, 100, 300, 1000 mg / kg) or saline by gavage 60 minutes prior to the maze tests at high cross the open field test, test forced swim test and sedation induced by thiopental. To investigate the mechanism of action of the activity of antidepressant type of fractions the following drugs were used: PCPA (inhibitor of 5-HT synthesis) AMPT (inhibitor of catecholamine synthesis), DSP-4 (noradrenergic neurotoxin) and Sulpiride (antagonist selective dopamine D2 receptor). They were used as a standard positive control, fluoxetine and nortriptyline. The results of the phytochemical profile show very different characteristics to the aqueous extract of the varieties of Passiflora edulis "flavicarpa" and "edulis". The aqueous extracts of both varieties of Passiflora edulis share anxiolytic activity type (edulis fo. edulis 300 mg/kg; edulis fo. flavicarpa 300 and 1000 mg/kg) and antidepressant (edulis fo. edulis 300 mg/kg; edulis fo flavicarpa 1000 mg/kg), while the effect hipolocomotor/sedative was only seen for edulis fo. edulis (1000 mg/kg). Both fractions ethyl acetate, butanol aqueous extract edulis fo. edulis showed activity type antidepressant at a dose of 50 mg/kg in the forced swim test. The data suggest that the effect of antidepressant-like fractions edulis fo. edulis involves catecholaminergic and serotonergic neurotransmission, particularly dopaminergic, there is seen that pre-treatment DSP-4 is not affected antidepressant action of fractions as was dependent activation of dopamine D2 receptors.

CHAPTER II - This study evaluated the effects of two different types of acute aerobic exercise on the osmotic stability of human erythrocyte membrane and on different hematological and biochemical variables that are associated with this membrane property. The study population consisted of 20 healthy and active men. Participants performed single sessions of two types of exercise. The first session consisted of 60 min of moderate-intensity continuous exercise (MICE). The second session, executed a week later, consisted of high-intensity interval exercise (HIIE) until exhaustion. The osmotic stability of the erythrocyte membrane was represented by the inverse of the salt concentration (1/H50) at the midpoint of the sigmoidal curve of dependence between the absorbance of hemoglobin and the NaCl concentration. The values of 1/H50 changed from 2.29 ± 0.1 to 2.33 ± 0.09 after MICE and from 2.30 ± 0.08 to 2.23 ± 0.12 after HIIE. In MICE has occurred an increase in the mean corpuscular volume, probably due to in vivo lysis of older erythrocytes, with preservation of cells that were larger and more resistant to in vitro lysis. The study showed that a single bout of acute exercise affected the erythrocyte osmotic stability, which increased after MICE and decreased after HIIE.

The aim of this study was to test the sediment preference of L. vannamei shrimp. It was observed shrimp visit frequency, swimming and burying behaviour at different sediment compositions for 24h. Juvenile (0.93 ± 0.29g) and sub-adult shrimps (10.0 ± 1.18g) were obtained from the aquaculture station at Universidade Federal Rural do Semi-Árido UFERSA, and held in a plastic tank (water volume 500 L) supplied with aerated water and kept at constant temperature, pH, and salinity. Shrimp was fed by commercial shrimp dry food. The experimental substrates were composed by A: medium sand + thick sand + very thick sand + gravel; B: very fine sand + fine sand; and C: silt + clay. Thus, six different substrate combinations were tested: A, B, C, A+B, A+C, B+C. To test preference, it was used a cylindrical tank (40 l) divided into six differently substrate compartments. A single shrimp was introduced each tank and the frequency at which this shrimp visited each compartment was recorded over a 24h study period. It was tested 54 shrimp (18 sub-adult males, 18 subadult females and 18 juveniles). For each trial, sediment and water were changed to avoid pheromones and residues influence. Shrimp were weighted and sub-adults were divided by sex: males present petasma and females present thelycum. Data were collected on the experimental day at 19:30; 20:30; 00:30; 1:30; 05:30; 06:30; 13:30 and 14:30 h. At each time point, shrimp were observed for 20-min periods, in which we noted down which compartment the shrimp was occupying at 2-min intervals. Thus, for each period we had eleven observations (88 observations per day). For observations at night, it was used dim red light that did not affect shrimp behaviour. At each 20-min period, it was observed visit frequency in each substrate, if shrimp was burred or not or if it was swimming. There was not significant difference between light and dark burry activity for females. Swimming activity was significantly higher at night, mainly at 00:30 and 01:30 h. All L. vannamei shrimp showed preference for sediment B. This animal presents cyclic activity, spends the day light period buried and swims at night

Objective: To verify the sociodemographic profile and the frequency of musculoskeletal injuries in elderly people who practice competitive and noncompetitive sports activities in the city of Pelotas, RS. Methods: Descriptive study, including 29 male subjects, aged 65 years or older, who practiced sports modalities in a competitive and noncompetitive way, in the city of Pelotas, in 2015. Data collection was performed through the application of a questionnaire with questions addressing economic, sociodemographic, nutritional and behavioral issues, and injuries sustained in sports activities. Statistical analysis was performed by calculating measures of central tendency for continuous variables and proportions for categorical variables. Results: The current practice of competitive sports was described by 58.6% (n=17) of the subjects, and the most practiced sports were 7-a-side football (53.0%, n=9), tennis (23.5%, n=4) and swimming (23.5%, n=4). Noncompetitive sports were practiced by 44.8% (n=13), and tennis was the most popular sport played (92.3%, n=12). The frequency of injuries among individuals practicing competitive and noncompetitive sports was, respectively, 35.3% (n=6), and 38.5% (n=5). The most frequent injuries were epicondylitis (50.0%, n=3) and meniscus injuries (60.0%, n=3), and the most affected body regions were knee (27.3%, n=3), elbow (27.3%, n=3), and shoulder (18.2%, n=2). The injuries occurred during the sports activities, and the dominant side was the one affected in 63.6% (n=7). Conclusion: This study verified that the occurrence of injuries in elderly individuals who practice sports is relevant, even among those who practice them noncompetitively, with epicondylitis as the most frequent among them, whereas meniscus injuries are the most frequent among those who practice competitive sports.

Introdução – A frequência de dislipidemia em crianças e adolescentes tem vindo a aumentar rapidamente nos últimos anos, estando o seu aparecimento associado a fatores genéticos e ambientais. Este indicador, associado a outras doenças como a obesidade, constituem a síndrome metabólica e são considerados fatores de risco para doença cardiovascular e diabetes, que quando presentes na infância podem conduzir precocemente ao aparecimento destas doenças. Objetivos – Avaliar o perfil de indicadores de saúde de crianças no início de um Programa de Educação Contínua; Verificar os efeitos do Programa de Educação Contínua aplicado durante 36 meses (ginástica funcional, brincadeiras de rua e das aulas de natação), sobre os resultados dos exames iniciais de colesterol, triglicerídeos e do IMC. Métodos - Estudo quantitativo, exploratório e descritivo de corte transversal, realizado entre 2012 e 2015 numa amostra não probabilística por conveniência de 165 crianças, 70,0% da população-alvo. As crianças tinham entre 7 a 12 anos de idade, 40,59% entre 11 e 12 anos e 54% eram do sexo feminino. Eram beneficiárias de um plano de saúde que integrava um programa de intervenção (educação contínua e atividade física), designado Crescendo com Saúde e foram selecionadas através de critérios, como: dislipidemia (colesterol total e/ou triglicerídeos elevados), sobrepeso e/ou obesidade, e história familiar de HAS e DM. Os dados clínicos foram obtidos na consulta de enfermagem, utilizando-se a recolha de sangue para dosemanento do perfil lipídico no início do programa e a cada 6 meses e avaliação do IMC na fase inicial do programa e semanalmente. Para classificação do estado nutricional foram utilizados os pontos de corte da OMS (2007). Resultados – no início do programa 69% das crianças apresentavam hipercolesterolémia, 32% aumento do colesterol, 45% tinham excesso de peso (sobrepeso e obesidade) e 12% risco de sobrepeso. Após o programa de intervenção, das 53% crianças que participaram de forma regular, 29% apresentaram redução do colesterol, 16% dos triglicerídeos e o excesso de peso reduziu em 9%, aumentando contudo o risco de sobrepeso para 22%. Das que participaram de forma irregular, apenas 3% reduziram o perfil lipídico e 1% o estado nutricional. Conclusões – O programa de intervenção permitiu uma redução dos fatores de risco de doença metabólica e permitiu melhorar os hábitos das crianças estudadas e suas famílias. Estes resultados comprovam a eficácia a curto e médio prazo dos programas de intervenção na comunidade, e realçam o papel das intervenções preventivas de educação contínua e de atividade física regular, para o processo de redução dos indicadores de risco metabólico desde a infância. Atualmente o exercício físico vem sendo utilizado com intuito de redução de massa corporal, em especial de massa gorda. Entretanto cresce o número de indivíduos que, associado ao exercício físico, utiliza substâncias com característica lipolítica, como é o caso do ácido lipóico. Tanto o exercício físico, quanto a utilização de suplementação com ácido lipóico são responsáveis por remodelagem vascular (devido à interferência no processo de angiogênese) e modificação de fatores de risco cardiovascular (como hipertensão e lipídeos sanguíneos elevados). Desta forma, o presente estudo buscou analisar a influência do exercício físico de moderada intensidade e da suplementação com ácido lipóico sobre a frequência cardíaca, pressão arterial, bioquímica sanguínea e angiogênese no músculo cardíaco e no músculo esquelético de ratos Wistar. Foram utilizados 80 ratos Wistar, divididos em quatro grupos: controle, ácido lipóico (LA), exercício (E) e associação (ELA). Os animais foram submetidos a um programa de adaptação e treinamento de natação (9 e 17 semanas) com um aumento progressivo no tempo natação (até 1h/dia) e intensidade de carga (até 5% do peso corporal). Os animais receberam ácido lipóico 5 vezes por semana (da 10ª à 17ª semana), 60 mg / Kg / dia. O exercício crônico de intensidade moderada promoveu bradicardia, mas sua associação com a suplementação de ácido lipóico interrompeu este benefício. A suplementação com LA mostrou-se eficaz em melhorar o perfil lipídico, mas associado ao exercício não apresentou redução. A angiogênese foi aumentada no coração e gastrocnêmio dos animais exercitados, a largura da fibra de E, LA e ELA foi reduzida no coração, enquanto no gastrocnêmio apresentaram um aumento na largura das fibras apenas por LA e ELA. A espessura do ventrículo esquerdo diminuiu no grupo E, enquanto que a área da câmara do ventrículo esquerdo, e os níveis de VEGF circulantes, não mostraram nenhuma diferença significativa. Foi observada uma interação negativa entre o exercício físico e a suplementação com ácido lipóico (supressão da bradicardia do exercício de um lado, e a perda da adaptação do perfil lipídico induzido por suplementação de AL por outro lado). Este estudo mostra pela primeira vez, a interação entre o exercício crônico de intensidade moderada e a suplementação com ácido lipóico sobre a remodelação cardíaca e angiogênese, confirmando os benefícios da prática física em melhorar o fornecimento de sangue do músculo, que não foi afetado pelo consumo de ácido lipóico. O ácido lipóico em animais não treinados não foi capaz de estimular a 9 angiogênese cardíaca e ao contrário mostram uma tendência para a redução dos novos vasos.

O estudo pretende comparar o efeito de diferentes tipos de exercício físico na composição corporal e força em jovens desportistas do sexo feminino. A amostra foi constituída por vinte e seis desportistas femininas com idades compreendidas entre os 16 e 21 anos. Previamente ao início do estudo foram sujeitas ao 1º momento de avaliação, e após vinte e quatro semanas realizaram o segundo e ultimo momento de avaliação. Os parâmetros avaliados foram a composição corporal através da técnica de absorciometria radiológica de dupla energia; a força isocinética, nomeadamente os momentos de força (peak-torques) e rácios nos músculos extensores e flexores do joelho; a potência muscular nos membros inferiores através dos saltos verticais Squat jump e Countermovement jump. Posteriormente foram separadas em quatro grupos, grupo natação e exercício vibratório (NAT EV; n= 6), grupo natação (NAT; n=6), grupo futsal (FUTS; n= 6) e o grupo de controlo (CONT; n= 8). O grupo (NAT/EV), participou num programa de exercício vibratório (EV), três sessões por semana complementar ao treino de natação. O grupo de (NAT) cumpriu o plano de treino correspondente à disciplina, o grupo de (FUTS) cumpriu o treino referente à respetiva modalidade, o grupo controlo (CONT) realizou somente os exercícios físicos inerentes às aulas de educação física na escola. Resultados: Nas comparações inter grupos, verificou-se no grupo FUTS um aumento de 0,1 (g/m2) ± 0,0 nos valores da DMO-PE, quando comparado com o grupo de CONT, nas comparações intra grupo o grupo de NAT EV registou um acréscimo de 0,1 (g/m2) ± 0,1 nos valores da DMO-TC. Em conclusão, os resultados obtidos sugerem que a modalidade de FUTS promoveu mais alterações na composição corporal, nomeadamente na DMO-PE, no entanto dados conseguidos pelo grupo de NAT EV sugerem que o exercício vibratório poderá influenciar positivamente o incremento da DMO; ABSTRACT: The objective of this study was to compare the effect of different types of exercise on body composition and strength in young female athletes. The sample consisted of twenty-six female athletes aged between 16 and 21. Before the study there was a 1st evaluation point, after twenty-four weeks there was the 2nd and final evaluation. We evaluated body composition through x-ray absorptiometry technique of dual energy, isokinetic strength, including the peak-torques and ratios in the extensor and flexor muscles of the knee; the muscle power in the lower limbs was evaluatated through the vertical jumps Squat jump and countermovement jump. During the study they were separated into four groups, swimming exercise group and vibration (NAT EV; n = 6), swimming group (NAT; n= 6), footsal group (FUTS; n= 6) and control group (CONT; n= 8). The NAT/EV group, participated in a vibrating exercise program (EV), complementary to swimming training, with three sessions per week. The NAT group fulfilled the corresponding swimming workout plan, FUTS group fulfilled the training related to futsal, the CONT group only performed the usual exercises in physical education classes at school. Results: In intergroup comparisons, there was an increase in FUTS group of 0.1 (g / m2) ± 0.0 in the values of MBD-PE, when compared with the CONT group comparisons in intra-group group NAT EV increased by 0.1 (g / m2) ± 0.1 in BMD-TC values. In conclusion, the results suggest that FUTS group promoted more changes in body composition, particularly in BMD-PE, but data obtained by NAT EV group suggest that vibration exercise can positively influence the increase in BMD.

O presente relatório de estágio enquadra-se no âmbito do mestrado em Exercício e Saúde da Universidade de Évora. Estágio que decorreu no Eugénios Health Club & SPA Club. Com este relatório pretende-se essencialmente, fazer uma ponte entre a componente académica e prática comum na área do desporto. Este estágio final de ano, teve como principal objetivo aplicar os conhecimentos e capacidades adquiridas ao longo de todo o percurso académico, assim como desenvolver as competências pessoais e profissionais, de forma a que o ponto de partida para um currículo profissional seja o mais real possível para integração do mercado de trabalho. Este relatório, encontra-se estruturado de forma a descrever as atividades planeadas e desenvolvidas ao longo do estágio. Desta forma estão relatadas as tarefas inicialmente definidas (responsável por acompanhamento na sala de Cardio-Musculação, aulas de natação e realização de aulas de Hidroginástica, bem como as atividades complementares); ABSTRACT: The present internship report fills in the ambit of the master’s degree in Health and Exercise at the Universidade de Évora. This internship occurred at the Eugénios Health & SPA Club. This report aims essentially to make a bridge between the academic and practice components in the sport area. This final internship had the main goal of applying the knowledge and acquired skills throughout the academic path as well the development of the personal and professional abilities, so that the starting point of the professional resume could be as real as possible for a future integration in the employment market. This report finds itself structured to describe the planned and developed activities over the year, making a passage through the theory learned. This way the initial defined tasks are reported – monitoring the cardio/bodybuilding room; organization and execution of different levels of swimming classes; executing water aerobics classes as other complementary activities.
