Lo scopo della seguente tesi è di presentare una proposta e analisi di sottotitolaggio dell’episodio Blackwater della seconda stagione della serie Game of Thrones. A spingermi a scegliere questo tema è stata la mia passione per la serie e la saga di libri da cui è stata adattata e il desiderio di redigerne i sottotitoli. È stata un’esperienza utile e stimolante perché mi ha dato la possibilità di mettere in pratica le nozioni teoriche acquisite durante questi anni di studio. Il mio lavoro si sviluppa in tre capitoli e un’appendice. Il primo capitolo è dedicato alla contestualizzazione dell’episodio. Nel secondo capitolo sono analizzati gli aspetti tecnici della traduzione multimediale e del sottotitolaggio. Il terzo capitolo contiene l’analisi delle principali difficoltà incontrate nel processo traduttivo, mentre l’appendice contiene la tabella dei sottotitoli sia in inglese sia in italiano.

Some inducible yeast genes relocate to nuclear pores upon activation, but the general relevance of this phenomenon has remained largely unexplored. Here we show that the bidirectional hsp-16.2/41 promoter interacts with the nuclear pore complex upon activation by heat shock in the nematode Caenorhabditis elegans. Direct pore association was confirmed by both super-resolution microscopy and chromatin immunoprecipitation. The hsp-16.2 promoter was sufficient to mediate perinuclear positioning under basal level conditions of expression, both in integrated transgenes carrying from 1 to 74 copies of the promoter and in a single-copy genomic insertion. Perinuclear localization of the uninduced gene depended on promoter elements essential for induction and required the heat-shock transcription factor HSF-1, RNA polymerase II, and ENY-2, a factor that binds both SAGA and the THO/TREX mRNA export complex. After induction, colocalization with nuclear pores increased significantly at the promoter and along the coding sequence, dependent on the same promoter-associated factors, including active RNA polymerase II, and correlated with nascent transcripts.

Attention has recently been drawn to Enterococcus faecium because of an increasing number of nosocomial infections caused by this species and its resistance to multiple antibacterial agents. However, relatively little is known about the pathogenic determinants of this organism. We have previously identified a cell-wall-anchored collagen adhesin, Acm, produced by some isolates of E. faecium, and a secreted antigen, SagA, exhibiting broad-spectrum binding to extracellular matrix proteins. Here, we analysed the draft genome of strain TX0016 for potential microbial surface components recognizing adhesive matrix molecules (MSCRAMMs). Genome-based bioinformatics identified 22 predicted cell-wall-anchored E. faecium surface proteins (Fms), of which 15 (including Acm) had characteristics typical of MSCRAMMs, including predicted folding into a modular architecture with multiple immunoglobulin-like domains. Functional characterization of one [Fms10; redesignated second collagen adhesin of E. faecium (Scm)] revealed that recombinant Scm(65) (A- and B-domains) and Scm(36) (A-domain) bound to collagen type V efficiently in a concentration-dependent manner, bound considerably less to collagen type I and fibrinogen, and differed from Acm in their binding specificities to collagen types IV and V. Results from far-UV circular dichroism measurements of recombinant Scm(36) and of Acm(37) indicated that these proteins were rich in beta-sheets, supporting our folding predictions. Whole-cell ELISA and FACS analyses unambiguously demonstrated surface expression of Scm in most E. faecium isolates. Strikingly, 11 of the 15 predicted MSCRAMMs clustered in four loci, each with a class C sortase gene; nine of these showed similarity to Enterococcus faecalis Ebp pilus subunits and also contained motifs essential for pilus assembly. Antibodies against one of the predicted major pilus proteins, Fms9 (redesignated EbpC(fm)), detected a 'ladder' pattern of high-molecular-mass protein bands in a Western blot analysis of cell surface extracts from E. faecium, suggesting that EbpC(fm) is polymerized into a pilus structure. Further analysis of the transcripts of the corresponding gene cluster indicated that fms1 (ebpA(fm)), fms5 (ebpB(fm)) and ebpC(fm) are co-transcribed, a result consistent with those for pilus-encoding gene clusters of other Gram-positive bacteria. All 15 genes occurred frequently in 30 clinically derived diverse E. faecium isolates tested. The common occurrence of MSCRAMM- and pilus-encoding genes and the presence of a second collagen-binding protein may have important implications for our understanding of this emerging pathogen.

Spinocerebellar Ataxia type 7 (SCA7) is a neurodegenerative disease caused by expansion of a CAG repeat encoding a polyglutamine tract in ATXN7, a component of the SAGA histone acetyltransferase (HAT) complex. Previous studies provided conflicting evidence regarding the effects of polyQ-ATXN7 on the activity of Gcn5, the HAT catalytic subunit of SAGA. Here I showed that reducing Gcn5 expression accelerates both cerebellar and retinal degeneration in a mouse model of SCA7. Deletion of Gcn5 in Purkinje cells in mice expressing wild type Atxn7, however, causes only mild ataxia and does not lead to the early lethality observed in SCA7 mice. Reduced Gcn5 expression strongly enhances retinopathy in SCA7 mice, but does not affect the transcriptional targets of Atxn7, as expression of these genes is not further altered by Gcn5 depletion. These findings demonstrate that loss of Gcn5 functions can contribute to the time of onset and severity of SCA7 phenotypes, but suggest that non-transcriptional functions of SAGA may play a role in neurodegeneration in this disease.

The objective of this study was to determine if area measurements of pleural fluid on computed tomography (CT) reflect the actual pleural fluid volume (PEvol) as measured at autopsy, to establish a formula to estimate the volume of pleural effusion (PEest), and to test the accuracy and observer reliability of PEest.132 human cadavers, with pleural effusion were divided into phase 1 (n = 32) and phase 2 (n = 100). In phase 1, PEvol was compared to area measurements on axial (axA), sagittal (sagA), and coronal (corA) CT images. Linear regression analysis was used to create a formula to calculate PEest. In phase 2, intra-class correlation (ICC) was used to assess inter-reader reliability and determine the agreement between PEest and PEvol. PEvol correlated to a higher degree to axA (r s mean = 0.738; p < 0.001) than to sagA (r s mean = 0.679, p < 0.001) and corA (r s mean = 0.709; p < 0.001). PEest can be established with the following formula: axA × 0.1 = PEest. Mean difference between PEest and PEvol was less than 40 mL (ICC = 0.837-0.874; p < 0.001). Inter-reader reliability was higher between two experienced readers (ICC = 0.984-0.987; p < 0.001) than between an inexperienced reader and both experienced readers (ICC = 0.660-0.698; p < 0.001). Pleural effusions may be quantified in a rapid, reliable, and reasonably accurate fashion using single area measurements on CT.

Enterococci are normal flora in the human intestinal tract, and also one of the leading causes of nosocomial infections, with most of the clinical isolates being Enterococcus faecalis and Enterococcus faecium. Despite extensive studies on the antibiotic resistance, the pathogenicity of enterococci is not well understood, especially for E. faecium. To identify potential virulence factors based on their antigenicity during infection, E. faecium genomic libraries were constructed and screened using sera from patients with E. faecium endocarditis. ^ As one of my projects, total polysaccharides were extracted from E. faecalis OG1RF and from two epa mutants constructed previously, TX5179 and TX5180, and western blots with patient sera showed that an immuno-reactive polysaccharide present in wild type OG1RF was not produced by either of the two epa mutants. The epa mutants were more sensitive to ethanol stress, neutrophil killing and neutrophil phagocytosis than the wild type OG1RF. ^ Expression of virulence factors is commonly regulated by two component systems. A BLAST search was performed to identify potential two component systems in the E. faecalis V583 genome database using PhoP/PhoS as query sequences, and 11 gene pairs were identified, seven of which were disrupted in E. faecalis OGIRF. ^ Finally, an in vitro translocation model was established for enterococci. E. faecalis strain OG1RF and E. faecium strain DO were shown to be able to translocate across a T84 monolayer, while E. coli strain DH5α and E. faecalis strain E1 could not. ^ In conclusion, several E. faecium antigens expressed in infection (whose antibodies present in sera from patients with E. faecium endocarditis) were identified, two of which, SagA and GlyA, were characterized and suggested to be involved in cell wall metabolism. E. faecalis epa gene cluster (involving in polysaccharide biosynthesis and known to be involved in virulence of E. faecalis in mice) was shown to be involved in hindering neutrophil killing. Several two-component systems were identified in E. faecalis and two of which, EtaRS and EtbRS, were involved in E. faecalis virulence in a mouse peritonitis model.^

Objective: Colorectal cancer (CRC) can be largely prevented or effectively treated in its early stages, yet disparities exist in timely screening. The aim of this study was to explore the disparities in CRC screening on the basis of health insurance status including private, Medicare, Medicaid, and State Administered General Assistance (SAGA). Methods: A retrospective chart review for the period January 2000 to May 2007 (95 records) was conducted at two clinic sites; a private clinic and a university hospital clinic. All individuals at these sites who met study criteria (>50 years old with screening colonoscopy) were included. Age, gender, date of first clinic visit when screening referral was made, and date of completed procedure (screening colonoscopy) were recorded. Groups were dichotomized between individuals with private health insurance and individuals with public health insurance. Individuals with any history of CRC, known pre-cancerous conditions as well as family history of CRC requiring frequent colonoscopy were excluded from the study. Linear model analysis was performed to compare the average waiting time to receiving screening colonoscopy between the groups. T-test was performed to analyze age or gender related differences between the two groups as well as within each group. Results: The average waiting time (33 days) for screening colonoscopy in privately insured individuals was significantly lower than publicly insured individuals (200 days). The time difference between the first clinic visit and the procedure was statistically significant (p < 0.0001) between the two groups. There was no statistical difference (p=0.089) in gender between these groups (public vs. private). There were also no statistically significant gender or age related differences found within each group. Conclusions: Disparities exist in timely screening for CRC and one of the barriers leading to delayed CRC screening includes health insurance status of an individual. Even within the insured group, type of insurance plays major role. There is a negative correlation between public health insurance status and timely screening. Differences in access to medical care and delivery of care experienced by patients who are publicly insured through Medicaid, Medicare, and SAGA, suggests that the State of Connecticut needs to implement changes in health care policies that would provide timely screening colonoscopy. It is evident that health insurance coverage facilitates timely access to healthcare. Therefore, there is a need for increased efforts in advocacy for policy, payment and physician participation in public insurance programs. A state-wide comprehensive program involving multiple components targeting different levels of change such as provider, patients and the community should help reduce some of the observed causes of healthcare disparities based on the insurance status.

A partir de los aportes críticos de Enrique Anderson Imbert y de Umberto Eco acerca de las posibles relaciones existentes entre los escritos de Borges y Chesterton es que, en el presente trabajo, me propongo estudiar el motivo del cuarto cerrado y su relación con el detective en Seis problemas para don Isidro Parodi de Honorio Bustos Domecq (seudónimo de Adolfo Bioy Casares y Jorge Luis Borges). En tal sentido, intento fundamentar la relación paródica que a nivel de procedimientos se establece con la saga del padre Brown de Gilbert R. Chesterton. Con tal finalidad, reseño el itinerario diacrónico que los tres elementos fundamentales de este tipo de relatos -el cuarto cerrado, el detective y la víctima-, han tenido a lo largo de la historia de la literatura de índole policial con el objetivo preciso de señalar la peculiar textualización de los mismos en la obra analizada. La originalidad del tratamiento de estos motivos típicos, tanto por parte de Biorges como por parte de Chesterton, ha evitado su posible petrificación en el tiempo como tópico literario.

Ya desde su primera novela publicada (Barbarie, 1927), el novelista mendocino Carlos Alberto Arroyo se inscribe en una línea de especial relevancia dentro de nuestras letras provinciales: la que responde a una intencionalidad política, nutrida por las complejas alternativas de la historia lugareña, a tal punto que un vasto sector de su obra podría ser denominada "la saga de la Mendoza lencinista". Lo sucesos narrados en La furia de los vencidos giran alrededor de la Revolución del '30 y su repercusión en el ambiente político mendocino, en particular dentro del círculo de una tradicional familia mendocina, los Ansay, cuya casa es el escenario de gran parte de la acción. Sucesivas polaridades espaciales inscritas en los pasajes descriptivos tienden a resaltar el valor simbólico de la casa como ámbito de acción en La furia de los vencidos: la verticalidad que se establece a través del concepto de "clase" como estrato dirigente de la sociedad, respaldada en el ejercicio del poder político; la dialéctica entre el adentro y el afuera, vale decir, la intimidad familiar opuesta al espacio de actuación pública y la idea de lo cerrado, manifiesta en la construcción de un espacio doméstico, asociado a lo femenino -hortus conclusus descripto con una tópica consagrada por la tradición- son otras tantas equivalencias del actuar de los personajes, sometidos a una coyuntura de transformación social que influye de manera decisiva en su destino.

La cultura clásica, en especial la griega, a menudo y sostenidamente ha atraído a escritores mendocinos de cuna o por adopción. En ese marco, el presente trabajo explora primero rasgos de la épica helénica y luego analiza elementos de la cultura americana precolombina, en el tratamiento de personajes, intervención divina, sucesos, espacio y tiempo en Los días del venado, obra reconocida con el primer premio de la Fundación "El Libro" a la mejor publicación juvenil de 2000. Hipótesis de trabajo: En Los días del venado se concentran dos vertientes ancestrales en tensión, la helena y la americanista indígena. Esta última predomina en la toma de posición ideológica de la historia narrada. Método: Análisis de contenido sin categorías fijadas a priori, por cuanto se trata de una metodología fenomenológíca y cualitativa. Resultados previstos: aporte para la comprensión crítica, con una lectura reflexiva de doble vía de la novela: el reconocimiento de la herencia helena (helenos, en efectos, son los nombres de los vasallos de Misáianes que invaden las Tierras Fértiles) más una importante cosmovisión americanista -en especial maya y araucana-, que la diferencia notablemente de las cualidades eurocéntricas de una saga al estilo de J. R. R. Tolkien.

A partir del análisis del genotexto de la emigración italiana a la Argentina en "Emigrat;" de A. Marazzi (1880) y en la saga "Il Roscaldi" de Nella Pasini (1924,1930), el trabajo se propone reconstruir las estructuraciones y las modelaciones ideológicas inscriptas, especialmente aquellas vinculadas con dos posiciones políticas contrapuestas, es decir, la "antiemigración" y la "proemigración" desde Italia.
