A simple device for multiplex ELISA made from melt-extruded plastic microcapillary film

We present a simple device for multiplex quantitative enzyme-linked immunosorbant assays (ELISA) made from a novel melt-extruded microcapillary film (MCF) containing a parallel array of 200µm capillaries along its length. To make ELISA devices different protein antigens or antibodies were immobilised inside individual microcapillaries within long reels of MCF extruded from fluorinated ethylene propylene (FEP). Short pieces of coated film were cut and interfaced with a pipette, allowing sequential uptake of samples and detection solutions into all capillaries from a reagent well. As well as being simple to produce, these FEP MCF devices have excellent light transmittance allowing direct optical interrogation of the capillaries for simple signal quantification. Proof of concept experiments demonstrate both quantitative and multiplex assays in FEP MCF devices using a standard direct ELISA procedure and read using a flatbed scanner. This new multiplex immunoassay platform should find applications ranging from lab detection to point-of-care and field diagnostics.

The integration of proteomics and systems approaches to map regulatory mechanisms underpinning platelet function.

Platelets in the circulation are triggered by vascular damage to activate, aggregate and form a thrombus that prevents excessive blood loss. Platelet activation is stringently regulated by intracellular signalling cascades, which when activated inappropriately lead to myocardial infarction and stroke. Strategies to address platelet dysfunction have included proteomics approaches which have lead to the discovery of a number of novel regulatory proteins of potential therapeutic value. Global analysis of platelet proteomes may enhance the outcome of these studies by arranging this information in a contextual manner that recapitulates established signalling complexes and predicts novel regulatory processes. Platelet signalling networks have already begun to be exploited with interrogation of protein datasets using in silico methodologies that locate functionally feasible protein clusters for subsequent biochemical validation. Characterization of these biological systems through analysis of spatial and temporal organization of component proteins is developing alongside advances in the proteomics field. This focused review highlights advances in platelet proteomics data mining approaches that complement the emerging systems biology field. We have also highlighted nucleated cell types as key examples that can inform platelet research. Therapeutic translation of these modern approaches to understanding platelet regulatory mechanisms will enable the development of novel anti-thrombotic strategies.

Some implications of a sample of practical Turing tests

A series of imitation games involving 3-participant (simultaneous comparison of two hidden entities) and 2-participant (direct interrogation of a hidden entity) were conducted at Bletchley Park on the 100th anniversary of Alan Turing’s birth: 23 June 2012. From the ongoing analysis of over 150 games involving (expert and non-expert, males and females, adults and child) judges, machines and hidden humans (foils for the machines), we present six particular conversations that took place between human judges and a hidden entity that produced unexpected results. From this sample we focus on features of Turing’s machine intelligence test that the mathematician/code breaker did not consider in his examination for machine thinking: the subjective nature of attributing intelligence to another mind.

Negotiating the British landscape

According to many accounts, a key paradigm for understanding art in Post WWII Britain is one of Englishness versus internationalism or abstraction versus realism . These terms have a rich inflection of meanings that have been subject to interrogation over the last few decades. Anwar Shemza came to Britain and practiced his art at a time when these competing claims were at their height. In a postcolonial reading entitled “Black Diaspora Artists in Britain: Three ‘Moments’ in Post-War Britain” Stuart Hall recently used David Scott’s framework of a ‘problem space’, that is discursively defined through questions, tensions and conjunctures, that couched the entry of what he describes as first waive British commonwealth artists into critical visibility in Britain. This can be characterized in part by the reviews of WG Archer and GM Butcher, both supporters of Shemza and prominent critics of the period. Hall includes Shemza in this framework that defines the work and his aspirations as constituted through the tensions of what was perceived to be anti-colonialist aims of modernism through universalism and the ‘nativist’ current in anti-colonial nationalism . This text will focus particularly on the problematic of Landscape as a ‘problem space’ of vernacular and modernism, over here and over there. The aim is not to define Shemza within the tradition of English landscape nor to exclude him but to position him within a discursive field of landscape and modernism in mid twentieth Century art.

Capillary assay device with internal hydrophilic coating

The present invention provides assay devices having a unitary body with an exterior surface, the unitary body being substantially transparent to visible light and formed from a material having a refractive index in the range 1.26 to 1.40, the refractive index being measured at 20 °C with light of wavelength 589 nm, and wherein the unitary body is formed from a hydrophobic material, and at least two capillary bores extending internally along the unitary body, wherein at least a portion of the surface of each capillary bore includes a hydrophilic layer for retaining an assay reagent, and wherein the hydrophilic layer is also substantially transparent to visible light to allow optical interrogation of the capillary bores through the capillary wall. The present invention also provides assay systems including such assay devices, methods of performing an assay using such assay devices and method of method for manufacturing such assay devices.

Euphemism: Sweet Talking or Deception?

The aim of this paper is to shed light on euphemism in two different senses: sweet talking and deception. I shall treat euphemism from two different perspectives: the usual use of euphemism, sweet talking, in which it is used to maintain one's face and the orthophemistic sense, deception, where 'torture' is referred to as "enhanced interrogation techniques". I shall analyze examples, taken from religious, cultural, political backgrounds, on each case. Moreover, I shall talk about taboo since it is usually associated with euphemism. I shall talk about the referential (semantic) and expletive (pragmatic) aspects of swearing expressions. In this essay, I shall show that euphemism can be used in two different senses: sweet talking and deception.

Equidade e descentralização: os mitos do SUS

O Sistema Único de Saúde (SUS) foi configurado para ser um instrumento de justiça social, utilizando a estratégia de descentralização político-administrativa, com vistas à equidade, universalização e integridade das ações de saúde. No entanto, após oito anos de aprovação e legitimação constitucional, o seu processo de implantação tem sofrido desvios na sua trajetória idealizada pela Reforma Sanitária. Nesta dissertação serão enfocadas as possíveis causas dos obstáculos desencaminhadores do SUS. Pontualizando duas questões conceituais: a equidade e a descentralização dos serviços de saúde. A grande interrogação que permeia todo o trabalho está no questionamento sobre a factibilidade do SUS, enquanto um modelo teórico, realizado em moldes utópicos, passível de transformar-se em mais um mito do imaginário social brasileiro, do que uma efetiva política social, voltada para a obtenção de uma sociedade democrática e participativa.

As encruzilhadas da autogestão: imaginário e simbólico nas empresas assumidas por trabalhadores: uma análise de empreendimentos da economia solidária

Nesta pesquisa a economia solidária (ES) é o pano de fundo. O enfoque pretendido é uma aproximação pela perspectiva organizacional. O problema de pesquisa é definido pela ausência da questão organizacional, na construção teórica da ES. O objetivo do trabalho é compreender os fenômenos organizacionais gestados por trabalhadores, no âmbito da ES. A pesquisa está delimitada às empresas assumidas por trabalhadores (EAT), são empresas convencionais que entraram em processo de crise ou falência, sendo assumidas por ex-empregados. A questão de pesquisa recai sobre a maneira com que os trabalhadores reinventam a gestão dos empreendimentos assumidos, quais as transformações operadas e as mudanças realizadas nas práticas de gestão herdadas. O imaginário e o simbólico são as categorias analíticas utilizadas para acessar a realidade dessas organizações. Articuladas à aproximação pela ótica dos grupos minoritários permitem apreender a dinâmica das transformações gestadas, o sentido das novas práticas e mapear a trajetória percorrida pelo grupo de trabalhadores ao construí-las. Dois estudos de caso são utilizados para ilustrar a dinâmica dessas transformações.

Configuração interferómetrica diferencial para medição de deformação e temperatura

Este documento descreve a implementação de um sistema interferométrico diferencial para a interrogação de redes de Bragg com o objectivo da medição diferencial de deformação e temperatura. Foram implementados dois sistemas de interrogação, um utiliza uma configuração implementada através de instrumentação física, e outro, uma configuração implementada através de instrumentação virtual, desenvolvida com recurso ao ambiente LabVIEW®. O esquema diferencial permite diminuir os efeitos de sensibilidade cruzada indissociáveis a estes sistemas. Foram realizados testes de deformação e temperatura de forma a validar o funcionamento do sistema virtual. Em ambos os casos, os resultados foram próximos dos obtidos com o sistema convencional, verificando-se que a instrumentação virtual é uma forma de aproveitar as vantagens que este esquema de interrogação apresenta: grande sensibilidade e elevada gama dinâmica, e permite reduzir uma de suas desvantagens, o elevado custo. Verificou-se ainda a viabilidade da aplicação da configuração estudada num sistema de análise térmica diferencial para identificação de compostos em amostras desconhecidas. O sistema foi demonstrado utilizando uma mistura de acetona e metanol. Do trabalho realizado nesta dissertação foi possível efectuar algumas publicações que foram submetidas em Revistas e Conferências Científicas. Vitrine das letras: o discurso jornalístico e a modernidade pedagógica em Sergipe/Brasil e Portugal - (1910-1920)

Dans cette étude, j'ai eu comme objectif d'établir les connexions et/ou les non-connexions dans les discours journalistiques sur la modernité pédagogique, dans les journaux O Correio de Aracaju (Le Courrier d'Aracaju, Sergipe, Brésil) et O Século (Le Siècle, Portugal), dans la période 1910-1920. J'ai tout particulièrement cherché le dit dans les journaux, dans le sens foucaldien du terme, en continu dialogue avec d'autres auteurs, dans une perspective interdisciplinaire. J'ai tenté de répondre à mon interrogation initiale : quelles sont les connexions et non connexions que l'on pourrait établir entre l'École Moderne du Portugal et celle de Sergipe, au Brésil, en prenant comme source d'analyse les discours journalistiques dans des journaux des deux pays ? Á partir de ce questionnement, à travers un abordage historico-comparatif interculturel, j'ai soutenu la THÈSE que les discours journalistiques du Século et du Correio de Aracaju, bien qu'ils relèvent de genres journalistiques différents quant au niveau de développement de la presse, ont traduit une perception et une assimilation de la modernité pédagogique articulées à une même epistémè. Ainsi, ces discours ont émergé comme objets métis, dont la création s'est faite dans des interlocutions, échanges, impositions et débats dans un processus continu de communication interculturelle. La nature messianique de ces discours, dans le système d'idées républicain, traduisait le sens de la modernité pédagogique prétendue et diffusée par les textes qui exprimaient l'aspiration de rupture et de progrès, tant au Portugal que dans l'état brésilien de Sergipe. Il a également été possible de faire des approximations/comparaisons entre la dispute (article d'opinion), dans le Correio de Aracaju et l'enquête (interview et reportage), dans O Século et de souligner ainsi les connexions et/ou non-connexions présentes sur le thème de la modernité pédagogique dans les discours journalistiques

A credibilidade do testemunho da criança vítima de abuso sexual no contexto judiciário

Violência sexual contra crianças não é um evento incomum; no entanto, há a dificuldade de denúncia, pois, além do estabelecimento da relação de dominação que o agressor exerce sobre a vítima, a maneira como tal fato é recebido pela sociedade e como é encaminhado pelas instituições judiciárias responsáveis também é determinante para as omissões. Inserida no universo dos interrogatórios, muitas vezes, a criança causa confusão ao desmentir o que havia falado antes, reforçando possíveis preconceitos em relação a si mesma. O presente trabalho traz a análise das relações entre a infância e a instituição judiciária, com principal enfoque no sistema de comunicação e de notificação dos crimes sexuais contra a criança e as consequentes intervenções profissionais que buscam a validação, ou não, de seu testemunho. Para tanto, foram pesquisados 51 processos judiciais, dos quais foram selecionados dois casos exemplares. Este trabalho evidencia a possibilidade de preservar a criança da revitimização causada pela multiplicidade de interrogatórios, sem deixar de cumprir as normas jurídicas necessárias. A fragilidade da palavra da criança está na forma como é acolhida pelos adultos, desde a revelação na família até a denúncia aos órgãos oficiais, revelando a urgência de alterações nos procedimentos judiciais relacionados a essa problemática

Da acusação às professoras pelo aprisionamento do sujeito em um diagnóstico a uma interrogação acerca da posição do analista

This study arises with two questionings: what is the usefulness of a diagnosis in school? And what moves that demand for diagnosis? Such questions were drawn up in answer to a diagnostic demand produced in the context of our internship in Scholar/Educational Psychology. On the perspective of working these issues, we conducted a literature research on diagnosis, with regard to its history, as a review of the psychoanalytic literature about the subject. This venture led us to a new interrogation: what are the elaborations that teachers produce from the child diagnosis, which place her as having special educational needs? The need of deciding the method that would lead us to answer such question, taking as reference the psychoanalytic theory, led us to an incursion to the subject research in psychoanalysis. This tracking points us that, according to Freud, on what comes to psychoanalysis, theory and research go together and that psychoanalysis is not a totalitarian world vision. On Lacan, the research is from the analysand, research that always implies the analyst and its praxis. Such path forced us to position a change to question the positions we occupy, in this experience, guided by an analytical listening. To discuss our position, we started from two cases and submitted them to construction and analysis. As a result, we found out that there is no way to know in advance what will be done from a diagnosis, which will be its uses. Point we used to considerate devastating to a child. So, to us, all children that received a diagnosis would be destined to a tragedy and what the research has shown us is that not always, not all of them. Thus, more than knowing what moves the demand, the important is the subject uses and our position towards it so they can generate a work

Eletromanometria do esfíncter superior do esôfago antes e após perfusão esofágica com ácido clorídrico 0,1N: estudo experimental no cão

Racional — A resposta do esfíncter esofágico superior ao refluxo gastroesofágico é controvertida. A perfusão esofágica com ácido clorídrico é modelo de estudo da ação do ácido o componente mais agressivo do refluxo sobre o esfíncter. Objetivos - Estudar o efeito da acidificação esofágica sobre o esfíncter esofágico superior através da eletromanometria esofágica. - Material e Métodos - em 30 cães adultos de ambos os sexos foram registrados estudos eletromanométricos do esôfago. A técnica utilizada foi a de puxada intermitente da sonda e perfusão contínua dos cateteres com água destilada. Estes exames permitiram as medidas da amplitude da pressão (mm Hg) e do comprimento (cm) do esfíncter superior do esôfago em condições basais (momento 1). Após esta fase, os animais foram submetidos a perfusão esofágica e divididos em 3 grupos de 10, na dependência da solução utilizada na perfusão e do momento do estudo: Grupo 1: perfusão esofágica com água destilada e estudos eletromanométricos realizados aos 15 minutos (momento 2) e 30 minutos (momento 3) do término da perfusão; Grupo 2: perfusão esofágica com HCl 0,1 N e estudos eletromanométricos realizados 15 minutos após o término da perfusão (momento 2); Grupo 3: perfusão esofágica com HCl 0,1 N e estudos eletromanométricos realizados 30 minutos após o término da perfusão (momento 3). Resultados/Conclusão - Os resultados observados permitiram concluir que a acidificação do esôfago não provocou alterações significativas sobre a amplitude de pressão e comprimento do esfíncter superior do esôfago.
