El artículo que a continuación se presenta describe un estudio de casos evaluativo realizado en un proyecto de Aprendizaje y Servicio Solidario (APS) que se desarrolla en la villa marginada Los Piletones, de la ciudad de Buenos Aires, Argentina. En concreto, con el estudio, se identifican y analizan nueve competencias transversales o generales (responsabilidad social y compromiso ciudadano, capacidad de aplicar los conocimientos en la práctica, capacidad de comunicarse con personas no expertas en la materia, etc.) que el proyecto logra desarrollar en los estudiantes universitarios de veterinaria que participan y se implican en él.

In cork oak (Quercus suber L.), recurrent embryogenesis is produced in vitro through autoembryony without exogenous plant growth regulators (PGRs); secondary embryos appear on the embryo axis but seldom on cotyledons. Focusing mainly on the histological origin of neoformations, we investigated the influence of the embryo axis and exogenous PGRs on the embryogenic potential of somatic embryo cotyledons. Isolated cotyledons of somatic embryos became necrotic when cultured on PGR-free medium but gave secondary embryos when cultured on media containing benzyladenine and naphthaleneacetic acid. Cotyledons of cork oak somatic embryos are competent to give embryogenic responses. Isolated cotyledons without a petiole showed a lower percentage of embryogenic response than did those with a petiole. In petioles, somatic embryos arose from inner parenchyma tissues following a multicellular budding pattern. Joined to the embryo axis, cotyledons did not show morphogenic responses when cultured on PGR-free medium but revealed budlike and phylloid formations when cultured on medium with PGRs. The different morphogenic behavior displayed by somatic cotyledons indicates an influence of the embryo axis and indicates a relationship between organogenic and embryogenic regeneration pathways

The in vivo accessibility of the chick embryo makes it a favoured model system for experimental developmental biology. Although the range of available techniques now extends to miss-expression of genes through in ovo electroporation, it remains difficult to knock out individual gene expression. Recently, the possibility of silencing gene expression by RNAi in chick embryos has been reported. However, published studies show only discrete quantitative differences in the expression of the endogenous targeted genes and unclear morphological alterations. To elucidate whether the tools currently available are adequate to silence gene expression sufficiently to produce a clear and specific null-like mutant phenotype, we have performed several experiments with different molecules that trigger RNAi: dsRNA, siRNA, and shRNA produced from a plasmid coexpressing green fluorescent protein as an internal marker. Focussing on fgf8 expression in the developing isthmus, we show that no morphological defects are observed, and that fgf8 expression is neither silenced in embryos microinjected with dsRNA nor in embryos microinjected and electroporated with a pool of siRNAs. Moreover, fgf8 expression was not significantly silenced in most isthmic cells transformed with a plasmid producing engineered shRNAs to fgf8. We also show that siRNA molecules do not spread significantly from cell to cell as reported for invertebrates, suggesting the existence of molecular differences between different model systems that may explain the different responses to RNAi. Although our results are basically in agreement with previously reported studies, we suggest, in contrast to them, that with currently available tools and techniques the number of cells in which fgf8 gene expression is decreased, if any, is not sufficient to generate a detectable mutant phenotype, thus making RNAi useless as a routine method for functional gene analysis in chick embryos.

Background: Nolz1 is a zinc finger transcription factor whose expression is enriched in the lateral ganglionic eminence (LGE), although its function is still unknown. Results: Here we analyze the role of Nolz1 during LGE development. We show that Nolz1 expression is high in proliferating neural progenitor cells (NPCs) of the LGE subventricular zone. In addition, low levels of Nolz1 are detected in the mantle zone, as well as in the adult striatum. Similarly, Nolz1 is highly expressed in proliferating LGE-derived NPC cultures, but its levels rapidly decrease upon cell differentiation, pointing to a role of Nolz1 in the control of NPC proliferation and/or differentiation. In agreement with this hypothesis, we find that Nolz1 over-expression promotes cell cycle exit of NPCs in neurosphere cultures and negatively regulates proliferation in telencephalic organotypic cultures. Within LGE primary cultures, Nolz1 over-expression promotes the acquisition of a neuronal phenotype, since it increases the number of β-III tubulin (Tuj1)- and microtubule-associated protein (MAP)2-positive neurons, and inhibits astrocyte generation and/or differentiation. Retinoic acid (RA) is one of the most important morphogens involved in striatal neurogenesis, and regulates Nolz1 expression in different systems. Here we show that Nolz1 also responds to this morphogen in E12.5 LGE-derived cell cultures. However, Nolz1 expression is not regulated by RA in E14.5 LGE-derived cell cultures, nor is it affected during LGE development in mouse models that present decreased RA levels. Interestingly, we find that Gsx2, which is necessary for normal RA signaling during LGE development, is also required for Nolz1 expression, which is lost in Gsx2 knockout mice. These findings suggest that Nolz1 might act downstream of Gsx2 to regulate RA-induced neurogenesis. Keeping with this hypothesis, we show that Nolz1 induces the selective expression of the RA receptor (RAR)β without altering RARα or RARγ. In addition, Nozl1 over-expression increases RA signaling since it stimulates the RA response element. This RA signaling is essential for Nolz1-induced neurogenesis, which is impaired in a RA-free environment or in the presence of a RAR inverse agonist. It has been proposed that Drosophila Gsx2 and Nolz1 homologues could cooperate with the transcriptional co-repressors Groucho-TLE to regulate cell proliferation. In agreement with this view, we show that Nolz1 could act in collaboration with TLE-4, as they are expressed at the same time in NPC cultures and during mouse development. Conclusions: Nolz1 promotes RA signaling in the LGE, contributing to the striatal neurogenesis during development.

These organizers are located at the interface of the expression domains of key patterning genes (Fezf-Irx and Otx-Gbx, respectively). To gain insights into the evolutionary origin of the ZLI, we studied Fezf in bilaterians. Results: In this paper, we identified a conserved sequence motif (Fezf box) in all bilaterians. We report the expression pattern of Fezf in amphioxus and Drosophila and compare it with those of Gbx, Otx and Irx. We found that the relative expression patterns of these genes in vertebrates are fully conserved in amphioxus and flies, indicating that the genetic subdivisions defining the location of both secondary organizers in early vertebrate brain development were probably present in the last common ancestor of extant bilaterians. However, in contrast to vertebrates, we found that Irx-defective flies do not show an affected Fezf expression pattern. Conclusions: The absence of expression of the corresponding morphogens from cells at these conserved genetic boundaries in invertebrates suggests that the organizing properties might have evolved specifically in the vertebrate lineage by the recruitment of key morphogens to these conserved genetic locations.

The Snail zinc-finger transcription factors trigger epithelial-mesenchymal transitions (EMTs), endowing epithelial cells with migratory and invasive properties during both embryonic development and tumor progression. During EMT, Snail provokes the loss of epithelial markers, as well as changes in cell shape and the expression of mesenchymal markers. Here, we show that in addition to inducing dramatic phenotypic alterations, Snail attenuates the cell cycle and confers resistance to cell death induced by the withdrawal of survival factors and by pro-apoptotic signals. Hence, Snail favors changes in cell shape versus cell division, indicating that with respect to oncogenesis, although a deregulation/increase in proliferation is crucial for tumor formation and growth, this may not be so for tumor malignization. Finally, the resistance to cell death conferred by Snail provides a selective advantage to embryonic cells to migrate and colonize distant territories, and to malignant cells to separate from the primary tumor, invade, and form metastasis.

Notch signaling is involved in cell fate choices during the embryonic development of Metazoa. Commonly, Notch signaling arises from the binding of the Notch receptor to its ligands in adjacent cells driving cell-to-cell communication. Yet, cell-autonomous control of Notch signaling through both ligand-dependent and ligand-independent mechanisms is known to occur as well. Examples include Notch signaling arising in the absence of ligand binding, and cis-inhibition of Notch signaling by titration of the Notch receptor upon binding to its ligands within a single cell. Increasing experimental evidences support that the binding of the Notch receptor with its ligands within a cell (cis-interactions) can also trigger a cell-autonomous Notch signal (cis-signaling), whose potential effects on cell fate decisions and patterning remain poorly understood. To address this question, herein we mathematically and computationally investigate the cell states arising from the combination of cis-signaling with additional Notch signaling sources, which are either cell-autonomous or involve cell-to-cell communication. Our study shows that cis-signaling can switch from driving cis-activation to effectively perform cis-inhibition and identifies under which conditions this switch occurs. This switch relies on the competition between Notch signaling sources, which share the same receptor but differ in their signaling efficiency. We propose that the role of cis-interactions and their signaling on fine-grained patterning and cell fate decisions is dependent on whether they drive cis-inhibition or cis-activation, which could be controlled during development. Specifically, cis-inhibition and not cis-activation facilitates patterning and enriches it by modulating the ratio of cells in the high-ligand expression state, by enabling additional periodic patterns like stripes and by allowing localized patterning highly sensitive to the precursor state and cell-autonomous bistability. Our study exemplifies the complexity of regulations when multiple signalng sources share the same receptor and provides the tools for their characterization.

A colaboração docente on-line permite a criação de comunidades virtuais. As ferramentas virtuais de uma equipe em rede permitem criar aulas virtuais em diferentes localizações geográficas. Estes ambientes permitem na educação a transferência do conhecimento entre alunos e docentes, incluindo atividades síncronas e assíncronas. Neste trabalho descrevemos nossa experiência em colaboração docente on-line, utilizando ferramentas virtuais na educação universitária. Os softwares utilizados foram 1-Skype: Software de aplicação para chamadas na Internet (VoIP). 2- LAN: Aula virtual digital disponibilizado em http://www.cevap.unesp.br/abertura.htm. Estas mídias foram utilizadas em dias e locais de transmissão e recepção diferentes. Os temas foram: 1-"Como preparar uma videoconferência", 2-"Importância das TICs na docência Universitária", 3-"Plataformas virtuais com bases de dados automatizadas na busca bibliográfica", 4-"Uso do laboratório virtual no ensino de Biologia Celular, Histologia e Embriologia". As ferramentas virtuais permitem a colaboração on-line de docentes localizados em diferentes localizações geográficas, além de formar recursos humanos que as usarão para melhorar o desempenho da educação universitária.

OBJETIVOS: analisar a contagem dos corpos lamelares em comparação com o teste de Clements na avaliação da maturidade pulmonar fetal em gestantes diabéticas. MÉTODOS: estudo prospectivo envolvendo 62 gestantes submetidas a amniocentese entre a 26ª e a 39ª semana. O líquido amniótico foi imediatamente submetido ao teste de Clements e à contagem de corpos lamelares. Os partos ocorreram até três dias após a amniocentese. A ocorrência de síndrome de angústia respiratória, indicativa de imaturidade pulmonar, foi confrontada com os resultados de imaturidade da amniocentese (ausência de anel completo no 3º tubo e menos de 50.000 corpos lamelares). O teste do chi2 foi utilizado para comparar o desempenho dos dois métodos e p<0,05 foi considerado significante. RESULTADOS: sete neonatos apresentaram síndrome de angustia respiratória (11,3%). A contagem de corpos lamelares e o teste de Clements tiveram desempenhos semelhantes quanto à sensibilidade (100 vs 71,4%, respectivamente) e valor preditivo negativo (100 vs 93,5%). A contagem de corpos lamelares teve melhor especificidade (87,3% vs 52,7; p<0,001), valor preditivo positivo (50 vs 16,1%; p=0,017) e acurácia (88,7% versus 54,8%; p<0,001) que o teste de Clements. CONCLUSÕES: a contagem de corpos lamelares revelou ser método simples e preciso na avaliação da maturidade pulmonar fetal. Comparado ao teste de Clements, tem a vantagem de não necessitar de manipulações e reagentes e de ter melhor desempenho quanto à especificidade, valor preditivo positivo e acurácia. Assim como o teste de Clements, a contagem de corpos lamelares tem alto valor preditivo negativo, assegurando que diante de um resultado maduro (50.000 ou mais), não ocorrerá síndrome de angustia respiratória.

OBJETIVO: avaliar a distância das fissuras cerebrais fetais à borda interna da calota craniana por meio da ultrassonografia tridimensional (US3D). MÉTODOS: realizou-se um estudo de corte transversal em 80 gestantes normais entre a 21ª e 34ª semanas de gestação. Avaliou-se a distância entre a tábua óssea interna da calota craniana fetal e as fissuras de Sylvius, parieto-occipital, hipocampo e calcarina. Para a obtenção desta distância para as três primeiras fissuras, realizou-se uma varredura tridimensional através do plano axial (nível dos ventrículos laterais). Para a obtenção da distância da fissura calcarina utilizou-se uma varredura coronal (nível dos lobos occiptais). Para avaliar a correlação entre as fissuras e a idade gestacional foram realizadas regressões de primeiro grau, sendo os ajustes calculados pelo coeficiente de determinação (R²). Foram determinados percentis 5, 50 e 95 para cada fissura. Avaliou-se ainda a correlação entre a distância destas fissuras com os diâmetros biparietal (DBP) e circunferência craniana (CC) utilizando o coeficiente de correlação de Pearson (r). RESULTADOS: todas as medidas das fissuras apresentaram correlação linear com a idade gestacional (Sylvius: R²=0,5; parieto-occipital: R²=0,7; hipocampo: R²=0,3 e calcarina: R²=0,3). A média da distância das fissuras variou de 7,0 a 14,0 mm, 15,9 a 28,7 mm, 15,4 a 25,4 mm e 15,7 a 24,8 mm para as fissuras de Sylvius, parieto-occipital, hipocampo e calcarina, respectivamente. As fissuras de Sylvius e parieto-occipital apresentaram as maiores correlações com o DBP (r=0,8 e 0,7, respectivamente) e a CC (r=0,7 e 0,8, respectivamente). CONCLUSÕES: a distância das fissuras cerebrais fetais à borda interna da calota craniana por meio da US3D apresentou correlação positiva com a idade gestacional.

O gato doméstico (Felis catus) foi nomeado por Carolus Linnaeus em seu livro Systema Naturae, em 1798. A família Felidea apresenta muita semelhança morfológica com os felinos selvagens. O estudo da embriologia do gato doméstico é de grande valia, uma vez que, é considerado um importante modelo animal quando comparado aos gatos selvagem em extinção, especialmente relacionado às pesquisas sobre biologia reprodutiva. Este trabalho objetivou análisar e comparar as fases embrionárias de quatro embriões e um feto de felinos domésticos. Nos embriões com idade gestacional estimada em 17 dias (0,5cm CR) podemos observar pela análise macroscópica a presença de dilatação rostral correspondente ao prosencéfalo, o local placóide do cristalino, a flexura cervical, os quatro arcos faríngeos com os sulcos que o dividem, a proeminência cardíaca, o indício do brotamento do membro pélvico, além da presença de somitos. Na região caudal do embrião, visualizamos a curvatura cranio-caudal, permitindo ao mesmo uma posição em formato de "C". Nos embriões com idade gestacional estimada em 22 dias (1,2cm CR), na análise macroscópica foi visualizado o prosencéfalo, vesícula óptica com pigmentação da retina, vesícula ótica, quarto ventrículo, fígado, membros torácicos e pélvicos com discreta distinção dos dígitos e vascularização superficial. Nos embriões com idade gestacional estimada em 25 dias (1,5cm CR) notamos a presença do prosencéfalo e mesencéfalo, a curvatura cervical pronunciada, vesícula óptica com forte pigmentação da retina, vesícula ótica, membros pélvicos e torácicos bem desenvolvidos, com distinção dos dígitos e fígado bem pronunciado. Os fetos com idade gestacional estimada em 52 dias (10cm CR) possuem estruturas internas e externas facilmente identificadas em animais adultos. Com relação às estruturas ósseas notamos que as mesmas não apresentam nenhuma epífise óssea formada, sendo visíveis somente as diáfises ósseas. Na análise microscópica, o embrião de idade gestacional de 19 dias (0,9cm CR) revelou a presença do rostro, cavidade oral com lábio superior e inferior, cavidade nasal, olho e a abertura do 4º ventrículo encefálico, esôfago, coração com átrio e ventrículo, pulmão, fígado, crista mesonéfrica, gônada primitiva, estômago, broto do membro torácico, coluna vertebral e a medula espinhal em formação. Esse trabalho é de grande importância para o estudo da morfologia externa e interna de gatos domésticos, principalmente no que diz respeito ao desenvolvimento ósseo e articular, considerando as alterações que podem ou não ser promovidas pelo uso de terapias medicamentosas ou celulares durante o desenvolvimento embrionário e fetal.

Resumo O desenvolvimento embrionário dos peixes é de grande importância para a piscicultura e na reintrodução de espécies ameaçadas de extinção em seus ambientes, e seu conhecimento constitui uma importante maneira para minimizar doenças e mortalidades dessas espécies. Com o auxílio de técnicas como a Microscopia Eletrônica de Varredura (MEV) e a imuno-histoquimica para identificar proteínas ósseas, foi possível avaliar as fases de desenvolvimento com mais riqueza de detalhes, facilitando a compreensão de hábitos e da biologia da espécie. Neste trabalho pudemos observar a ontogenia e osteogênese da Piapara (Leporinus elongatus), desde a fecundação até a fase juvenil, sendo evidenciadas estruturas importantes como o tamanho do vitelo, essencial para a nutrição do embrião; o fechamento do blastóporo, evento principal da embriogênese, que indica as taxas de fertilização; a metamorfose, que indica a formação dos primeiros e principais órgãos do animal e a formação de sua estrutura óssea. As Proteínas Ósseas Morfogenéticas (BMP-2 e BMP-4), moléculas essenciais reguladoras no desenvolvimento embrionário e na formação óssea, foram observadas apenas no estádio larval até o período juvenil, não sendo evidenciadas nos estágios anteriores. Os resultados desse trabalho trouxeram novas informações quanto à biologia do desenvolvimento dessa espécie, que certamente poderão auxiliar no aprimoramento de técnicas reprodutivas visando uma melhora na sua produção seja para fins comerciais ou de repovoamento.
