The aim of this study was to determine whether image artifacts caused by orthodontic metal accessories interfere with the accuracy of 3D CBCT model superimposition. A human dry skull was subjected three times to a CBCT scan: at first without orthodontic brackets (T1), then with stainless steel brackets bonded without (T2) and with orthodontic arch wires (T3) inserted into the brackets' slots. The registration of image surfaces and the superimposition of 3D models were performed. Within-subject surface distances between T1-T2, T1-T3 and T2-T3 were computed and calculated for comparison among the three data sets. The minimum and maximum Hausdorff Distance units (HDu) computed between the corresponding data points of the T1 and T2 CBCT 3D surface images were 0.000000 and 0.049280 HDu, respectively, and the mean distance was 0.002497 HDu. The minimum and maximum Hausdorff Distances between T1 and T3 were 0.000000 and 0.047440 HDu, respectively, with a mean distance of 0.002585 HDu. In the comparison between T2 and T3, the minimum, maximum and mean Hausdorff Distances were 0.000000, 0.025616 and 0.000347 HDu, respectively. In the current study, the image artifacts caused by metal orthodontic accessories did not compromise the accuracy of the 3D model superimposition. Color-coded maps of overlaid structures complemented the computed Hausdorff Distances and demonstrated a precise fusion between the data sets.

Long-distance correlations (LDCs) of plasma potential fluctuations in the plasma edge have been investigated in the TCABR tokamak in the regime of edge biasing H-mode using an array of multi-pin Langmuir probes. This activity was carried out as part of the scientific programme of the 4th IAEA Joint Experiment (2009). The experimental data confirm the effect of amplification of LDCs in potential fluctuations during biasing recently observed in stellarators and tokamaks. For long toroidal distances between probes, the cross-spectrum is concentrated at low frequencies f < 60 kHz with peaks at f < 5 kHz, f = 13-15 kHz and f similar to 40 kHz and low wave numbers with a maximum at k = 0. The effects of MHD activity on the LDCs in potential fluctuation are investigated.

Aedes aegypti is the most important vector of dengue viruses in tropical and subtropical regions. Because vaccines are still under development, dengue prevention depends primarily on vector control. Population genetics is a common approach in research involving Ae. aegypti. In the context of medical entomology, wing morphometric analysis has been proposed as a strong and low-cost complementary tool for investigating population structure. Therefore, we comparatively evaluated the genetic and phenotypic variability of population samples of Ae. aegypti from four sampling sites in the metropolitan area of Sao Paulo city, Brazil. The distances between the sites ranged from 7.1 to 50 km. This area, where knowledge on the population genetics of this mosquito is incipient, was chosen due to the thousands of dengue cases registered yearly. The analysed loci were polymorphic, and they revealed population structure (global F-ST = 0.062; p < 0.05) and low levels of gene flow (Nm = 0.47) between the four locations. Principal component and discriminant analyses of wing shape variables (18 landmarks) demonstrated that wing polymorphisms were only slightly more common between populations than within populations. Whereas microsatellites allowed for geographic differentiation, wing geometry failed to distinguish the samples. These data suggest that microevolution in this species may affect genetic and morphological characters to different degrees. In this case, wing shape was not validated as a marker for assessing population structure. According to the interpretation of a previous report, the wing shape of Ae. aegypti does not vary significantly because it is stabilised by selective pressure. (C) 2011 Elsevier B.V. The dispersal of plant-feeding mites can occur involuntarily, through transportation of infested plant parts, or voluntarily, by walking to new plant parts or to suitable spots where biotic (phoresis) or abiotic (wind, agricultural tools, etc.) factors carry them over long distances. Elucidating the dispersal mechanisms of the coconut mite, Aceria guerreronis Keifer, is important for understanding the process of colonization of new fruits of a same or different plants, essential for the improvement of control strategies of this serious coconut pest. Thus, the objective of this work was to investigate the voluntary dispersal mechanisms of this mite. The hypothesis that the coconut mite disperses by walking, phoresis or wind were tested. The coconut mite was shown to be able to walk short distances between fruits of the same bunch or between bunches of the same plant. Phoresis on insects of the orders Hymenoptera (Apidae), Coleoptera (Curculionidae) and Lepidoptera (Phycitidae) was evaluated in the laboratory and in the field. Although in the laboratory mites were shown to be able to climb onto honeybees, field investigations failed to show these insects as important carriers of the pest, corroborating findings of previous works; however, both laboratory and field investigations suggested the curculionid Parisoschoenus obesulus Casey to be able to transport the coconut mite between plants. Similarly, laboratory and field investigations suggested wind to be important in the dispersal of the coconut mite between plants.

PURPOSE. We previously demonstrated that most eyes have regionally variable extensions of Bruch's membrane (BM) inside the clinically identified disc margin (DM) that are clinically and photographically invisible. We studied the impact of these findings on DM- and BM opening (BMO)-derived neuroretinal rim parameters. METHODS. Disc stereo-photography and spectral domain optical coherence tomography (SD-OCT, 24 radial B-scans centered on the optic nerve head) were performed on 30 glaucoma patients and 10 age-matched controls. Photographs were colocalized to SD-OCT data such that the DM and BMO could be visualized in each B-scan. Three parameters were computed: (1) DM-horizontal rim width (HRW), the distance between the DM and internal limiting membrane (ILM) along the DM reference plane; (2) BMO-HRW, the distance between BMO and ILM along the BMO reference plane; and (3) BMO-minimum rim width (MRW), the minimum distance between BMO and ILM. Rank-order correlations of sectors ranked by rim width and spatial concordance measured as angular distances between equivalently ranked sectors were derived. RESULTS. The average DM position was external to BMO in all quadrants, except inferotemporally. There were significant sectoral differences among all three rim parameters. DM- HRW and BMO-HRW sector ranks were better correlated (median rho = 0.84) than DM- HRW and BMO-MRW (median rho = 0.55), or BMO-HRW and BMO-MRW (median rho = 0.60) ranks. Sectors with the narrowest BMO-MRW were infrequently the same as those with the narrowest DM-HRW or BMO-HRW. CONCLUSIONS. BMO-MRW quantifies the neuroretinal rim from a true anatomical outer border and accounts for its variable trajectory at the point of measurement. Introduction: This study aimed to describe the anatomy of mandibular premolars with type IX canal configuration by using micro–computed tomography. Methods: Mandibular premolars with radicular grooves (n = 105) were scanned, and 16 teeth with type IX configuration were selected. Number and location of canals, distances between anatomic landmarks, occurrence of apical delta, root canal fusion, and furcation canals, as well as 2-dimensional (area, perimeter, roundness, major and minor diameters) and 3-dimensional (volume, surface area, and structuremodel index) analysis were performed. Data were statistically compared by using analysis of variance and Kruskal-Wallis tests (a = 0.05). Results: Overall, specimens had 1 root with a main canal that divided into mesiobuccal, distobuccal, and lingual canals at the furcation level. Mean length of the teeth was 22.9 2.06 mm, and the configuration of the pulp chamber was mostly triangle-shaped. Mean distances from the furcation to the apex and cementoenamel junction were 9.14 2.07 and 5.59 2.19 mm, respectively. Apical delta, root canal fusion, and furcation canals were present in 4, 5, and 10 specimens, respectively. No statistical differences were found in the 2-dimensional and 3-dimensional analyses between root canals (P > .05). Conclusions: Type IX configuration of the root canal system was found in 16 of 105 mandibular premolars with radicular grooves. Most of the specimens had a triangle-shaped pulp chamber. Within this anatomic configuration, complexities of the root canal systems such as the presence of furcation canals, fusion of canals, oval-shaped canals in the apical third, small orifices at the pulp chamber level, and apical delta were also observed

This study aimed to evaluate the spatial variability of leaf content of macro and micronutrients. The citrus plants orchard with 5 years of age, planted at regular intervals of 8 x 7 m, was managed under drip irrigation. Leaf samples were collected from each plant to be analyzed in the laboratory. Data were analyzed using the software R, version 2.5.1 Copyright (C) 2007, along with geostatistics package GeoR. All contents of macro and micronutrients studied were adjusted to normal distribution and showed spatial dependence.The best-fit models, based on the likelihood, for the macro and micronutrients were the spherical and matern. It is suggest for the macronutrients nitrogen, phosphorus, potassium, calcium, magnesium and sulfur the minimum distances between samples of 37; 58; 29; 63; 46 and 15 m respectively, while for the micronutrients boron, copper, iron, manganese and zinc, the distances suggests are 29; 9; 113; 35 and 14 m, respectively.

Das Zweikomponentensystem DcuSR reguliert die Expression der Gene der anaeroben Fumaratatmung in E. coli in Abhängigkeit von externen C4-Dicarbonsäuren. Die membranständige Histidinkinase DcuS detektiert den Reiz und leitet ihn über die Membran an den Responseregulaor DcuR weiter, der die Aktivität der Zielgene reguliert. Das Substratspektrum von DcuS wurde näher untersucht und strukturelle Eigenschaften der Substrate sowie ihre Affinität zu DcuS bestimmt. Es wird vermutet, dass Histidinkinasen im aktiven Zustand als Dimere oder höhere Oligomere vorliegen. Der Oligomerisierungszustand von DcuS in der Membran wurde mittels EPR-Spektroskopie untersucht. Es wurden funktionelle Cysteinmutanten von DcuS hergestellt, die nur an bestimmten Positionen der periplasmatischen Domäne Cysteinreste, aber sonst keine weiteren Cysteinreste, enthielten. Die Proteine wurden isoliert, über die Cysteinreste mit Nitroxiden markiert und in Liposomen rekonstituiert. Erste EPR-Messungen zeigten, dass rekonstituiertes DcuS in einem geordneten Zustand in der Membran vorliegt, der diskrete Abstände zwischen den Monomeren aufweist. Die Struktur von rekonstituiertem DcuS in der Membran soll durch Festkörper-NMR aufgeklärt werden. Ein geeignetes C-terminal verkürztes Konstrukt, DcuS-PD/PAS wurde zu diesem Zweck hergestellt. Das Protein ließ sich in hoher Reinheit isolieren und konnte wieder in Liposomen rekonstituiert werden. Vorbereitende NMR-Messungen zeigten, dass eine Strukturaufklärung an diesem Protein möglich ist. Weitere Strukturuntersuchungen werden zur Zeit durchgeführt.

Se le trasformazioni sociali in atto tendono a esasperare il senso di incertezza, sradicamento ed individualismo, sussistono pratiche che si contrappongono alle tendenze dominanti, finalizzate a ricucire i legami sociali su scala locale. La progettazione urbano-architettonica interiorizza il nuovo bisogno di comunità originando soluzioni abitative tese a favorire gli scambi informali fra vicini, facendo leva sul concetto di capitale sociale, attaccamento al quartiere, identità del luogo e partecipazione. La casa, simbolo di stabilità e sicurezza ma anche di privacy, privatismo familiare, diventa sempre più oggetto di studi, domanda sociale e intervento politico. Soprattutto è sempre più intesa come un nodo di relazioni familiari in una rete di relazioni sociali più ampie. Casa e quartiere incidono nella esperienza di benessere e socialità familiare? In che modo gli spazi urbani e architettonici influenzano la coesione sociale? Quale il ruolo degli abitanti nello sviluppare socialità e integrazione? Sono queste le domande che ci siamo posti per rilevare le dinamiche sociali e culturali dell’abitare attraverso uno studio di caso condotto in due quartieri simili. Dalla ricerca emerge come il significato della casa non sia univoco ma cambi rispetto al ciclo di vita familiare e a quello economico e ciò incide nella partecipazione alle attività di quartiere. Mostriamo inoltre come lo spazio fisico costruito crea importanti opportunità per gli scambi informali e per il benessere familiare e individuale dei bambini ma che, il contesto sociale sia una discriminate fondamentale. Nel quartiere dove è presente una organizzazione di abitanti il numero delle relazioni di vicinato aumenta, cambiano anche la qualità delle relazioni e le distanze fisiche fra i vicini. Emerge inoltre che la reciprocità è il principale strumento di costruzione della coesione comunitaria interna e crea un atteggiamento di apertura e fiducia che va al di là dei confini di quartiere.

Der Haupt-Lichtsammelkomplex (LHCII) des Photosyntheseapparates höherer Pflanzen gehört zu den häufigsten Membranproteinen der Erde. Seine Kristallstruktur ist bekannt. Das Apoprotein kann rekombinant in Escherichia coli überexprimiert und somit molekularbiologisch vielfältig verändert werden. In Detergenzlösung besitzt das denaturierte Protein die erstaunliche Fähigkeit, sich spontan zu funktionalen Protein-Pigment-Komplexen zu organisieren, welche strukturell nahezu identisch sind mit nativem LHCII. Der Faltungsprozess findet in vitro im Zeitbereich von Sekunden bis Minuten statt und ist abhängig von der Bindung der Cofaktoren Chlorophyll a und b sowie verschiedenen Carotinoiden.rn Diese Eigenschaften machen LHCII besonders geeignet für Strukturuntersuchungen mittels der elektronenparamagnetischen Resonanz (EPR)-Spektrokopie. Diese setzt eine punktspezifische Spinmarkierung des LHCII voraus, die in dieser Arbeit zunächst optimiert wurde. Einschließlich der Beiträge Anderer stand eine breite Auswahl von über 40 spinmarkierten Mutanten des LHCII bereit, einen N-terminalen „Cys walk“ eingeschlossen. Weder der hierfür notwendige Austausch einzelner Aminosäuren noch die Anknüpfung des Spinmarkers beeinträchtigten die Funktion des LHCII. Zudem konnte ein Protokoll zur Präparation heterogen spinmarkierter LHCII-Trimere entwickelt werden, also von Trimeren, die jeweils nur ein Monomer mit einer Spinmarkierung enthalten.rn Spinmarkierte Proben des Detergenz-solubilisierten LHCII wurden unter Verwendung verschiedener EPR-Techniken strukturell analysiert. Als besonders aussagekräftig erwies sich die Messung der Wasserzugänglichkeit einzelner Aminosäurepositionen anhand der Electron Spin Echo Envelope Modulation (ESEEM). In Kombination mit der etablierten Double Electron-Electron Resonance (DEER)-Technik zur Detektion von Abständen zwischen zwei Spinmarkern wurde der membranständige Kernbereich des LHCII in Lösung eingehend untersucht und strukturell der Kristallstruktur für sehr ähnlich befunden. Die Vermessung kristallographisch nicht erfasster Bereiche nahe dem N-Terminus offenbarte die schon früher detektierte Strukturdynamik der Domäne in Abhängigkeit des Oligomerisierungsgrades. Der neue, noch zu vervollständigende Datensatz aus Abstandsverteilungen und ESEEM-Wasserzugänglichkeiten monomerer wie trimerer Proben sollte in naher Zukunft die sehr genaue Modellierung der N-terminalen Domäne des LHCII ermöglichen.rn In einem weiteren Abschnitt der Arbeit wurde die Faltung des LHCII-Apoproteins bei der LHCII-Assemblierung in vitro untersucht. Vorausgegangene fluoreszenzspektroskopi-sche Arbeiten hatten gezeigt, dass die Bindung von Chlorophyll a und b in aufeinanderfolgenden Schritten im Zeitbereich von weniger als einer Minute bzw. mehreren Minuten erfolgten. Sowohl die Wasserzugänglichkeit einzelner Aminosäurepositionen als auch Spin-Spin-Abstände änderten sich in ähnlichen Zeitbereichen. Die Daten deuten darauf hin, dass die Ausbildung der mittleren Transmembran-Helix mit der schnelleren Chlorophyll-a-Bindung einhergeht, während sich die Superhelix aus den beiden anderen Transmembranhelices erst im langsameren Schritt, zusammen mit der Chlorophyll-b-Bindung, ausbildet.rn

In many plant species, the genetic template of early life-stages is formed by animal-mediated pollination and seed dispersal and has profound impact on further recruitment and population dynamics. Understanding the impact of pollination and seed dispersal on genetic patterns is a central issue in plant population biology. In my thesis, I investigated (i) contemporary dispersal and gene flow distances as well as (ii) genetic diversity and spatial genetic structure (SGS) across subsequent recruitment stages in a population of the animal-pollinated and dispersed tree Prunus africana in Kakamega Forest, West Kenya. Using microsatellite markers and parentage analyses, I inferred distances of pollen dispersal (father-to-mother), seed dispersal/maternal gene flow (mother-to-offspring) as well as paternal gene flow (father-to-offspring) for four early life stages of the species (seeds and fruits, current year seedlings, seedlings ≤ 3yr, seedlings > 3yr). Distances of pollen and seed dispersal as well as paternal gene flow were significantly shorter than expected from the spatial arrangement of trees and sampling plots. They were not affected by the density of conspecific trees in the surrounding. At the propagule stage, mean pollen dispersal distances were considerably (23-fold) longer than seed dispersal distances, and paternal gene flow distances exceeded maternal gene flow by a factor of 25. Seed dispersal distances were remarkably restricted, potentially leading to a strong initial SGS. The initial genetic template created by pollination and seed dispersal was extensively altered during later recruitment stages. Potential Janzen-Connell effects led to markedly increasing distances between offspring and both parental trees in older life stages. This showed that distance and density-dependent mortality factors are not exclusively related to the mother tree, but also to the father. Across subsequent recruitment stages, the pollen to seed dispersal ratio and the paternal to maternal gene flow ratio dropped to 2.1 and 3.4, respectively, in seedlings > 3yr. The relative changes in effective pollen dispersal, seed dispersal, and paternal gene flow distances across recruitment stages elucidate the mechanisms affecting the contribution of the two processes pollen and seed dispersal to overall gene flow. Using the same six microsatellite loci, I analyzed genetic diversity and SGS across five life stages, from seed rain to adults. Levels of genetic diversity within the studied P. africana population were comparable to other Prunus species and did not vary across life stages. In congruence with the short seed dispersal distances, I found significant SGS in all life stages. SGS decreased from seed and early seedling stages to older juvenile stages, and it was higher in adults than in late juveniles of the next generation. A comparison of the data with direct assessments of contemporary gene flow patterns indicate that distance- or density-dependent mortality, potentially due to Janzen-Connell effects, led to the initial decrease in SGS. Intergeneration variation in SGS could have been driven by variation in demographic processes, the effect of overlapping generations, and local selection processes. Overall, my study showed that complex sequential processes during recruitment contribute to the spatial genetic structure of tree populations. It highlights the importance of a multistage perspective for a comprehensive understanding of the impact of animal-mediated pollen and seed dispersal on spatial population dynamics and genetic patterns of trees.

Im Rahmen dieser Arbeit wurden neuartige funktionale Nanographene synthetisiert und hinsichtlich ihrer strukturellen und elektronischen Eigenschaften charakterisiert. Basierend auf dem Strukturmotiv des Graphens konnten anellierte polyzyklische aromatische Kohlenwasserstoffe (PAKs) mit unterschiedlichen Seitenverhältnissen strukturdefiniert erhalten und gezielt in der Peripherie funktionalisiert werden. Basierend auf dem Synthesekonzept einer „Vorplanarisierung“ konnten Nanographen-Scheiben mit einem Durchmesser von bis zu 3 nm in hoher Reinheit erhalten werden. Durch die Entwicklung von Polyphenylen-Vorläufern mit einem gewinkelten Rückgrat konnten erstmals defektfreie und lösliche Nanographen-Streifen (GNRs) mit Breiten von 1,0 - 2,1 nm und Längen von über 40 nm synthetisiert werden.rnrnAm Hexa-peri-hexabenzocoronen (HBC) war es möglich, durch die Einführung kurzer linearer Alkylreste in der Peripherie den inter- und intrakolumnaren Abstand nach Selbstorganisation zu reduzieren. In Mischungen mit Perylentetracarboxydiimid (PDI) als Akzeptor konnte durch eine erhöhte Dichte und eine verbesserte Ladungsträgermobilität eine relative Steigerung der Effizienz von Donor-Akzeptor-Heteroübergangs-Solarzellen um 9 % erreicht werden. Eine kovalente Verknüpfung von HBC und PDI erlaubte hier die vollständige Kontrolle der supramolekularen Organisation, des Phasenverhaltens sowie des Abstandes zwischen Donor und Akzeptor.rnrnBasierend auf den im Rahmen dieser Arbeit entwickelten Synthesekonzepten, eröffnen sich nun zahlreiche Möglichkeiten zur Entwicklung weiterer Nanographene, die entsprechend der gewünschten Anwendung funktionalisiert werden können und ein besseres Verständnis der Eigenschaften graphenartiger Materialien erlauben werden.
