Objective: To evaluate the factors influencing the results of ulnar nerve neurotization at the motor branch of the brachii biceps muscle, aiming at the restoration of elbow flexion in patients with brachial plexus injury. Methods: 19 patients, with 18 men and 1 woman, mean age 28.7 years. Eight patients had injury to roots C5-C6 and 11, to roots C5-C6-C7. The average time interval between injury and surgery was 7.5 months. Four patients had cervical fractures associated with brachial plexus injury. The postoperative follow-up was 15.7 months. Results: Eight patients recovered elbow flexion strength MRC grade 4; two, MRC grade 3 and nine, MRC <3. There was no impairment of the previous ulnar nerve function. Conclusion: The surgical results of ulnar nerve neurotization at the motor branch of brachii biceps muscle are dependent on the interval between brachial plexus injury and surgical treatment, the presence of associated fractures of the cervical spine and occipital condyle, residual function of the C8-T1 roots after the injury and the involvement of the C7 root. Signs of reinnervation manifested up to 3 months after surgery showed better results in the long term. Level of Evidence: IV, Case Series.

BACKGROUND: Nerve transfers or graft repairs in upper brachial plexus palsies are 2 available options for elbow flexion recovery. OBJECTIVE: To assess outcomes of biceps muscle strength when treated either by grafts or nerve transfer. METHODS: A standard supraclavicular approach was performed in all patients. When roots were available, grafts were used directed to proximal targets. Otherwise, a distal ulnar nerve fascicle was transferred to the biceps branch. Elbow flexion strength was measured with a dynamometer, and an index comparing the healthy arm and the operated-on side was developed. Statistical analysis to compare both techniques was performed. RESULTS: Thirty-five patients (34 men) were included in this series. Mean age was 28.7 years (standard deviation, 8.7). Twenty-two patients (62.8%) presented with a C5-C6 injury, whereas 13 patients (37.2%) had a C5-C6-C7 lesion. Seventeen patients received reconstruction with grafts, and 18 patients were treated with a nerve transfer from the ulnar nerve to the biceps. The trauma to surgery interval (mean, 7.6 months in both groups), strength in the healthy arm, and follow-up duration were not statistically different. On the British Medical Research Council muscle strength scale, 8 of 17 (47%) patients with a graft achieved >= M3 biceps flexion postoperatively, vs 16 of 18 (88%) post nerve transfers (P = .024). This difference persisted when a muscle strength index assessing improvement relative to the healthy limb was used (P = .031). CONCLUSION: The results obtained from ulnar nerve fascicle transfer to the biceps branch were superior to those achieved through reconstruction with grafts.

Elevated levels of copper have been detected in various types of human cancer cells, such as breast cancer cells, and a number of mechanisms have been proposed to explain the action and influence of copper on tumor progress. In this work, we found that stimulating the proliferation of mammary epithelial MCF7 cells with the high-redox-potential copper complex Cu (GlyGlyHis) is associated with the copper-induced intracellular generation of reactive oxygen species (ROS) that induces lipid peroxidation and causes increased roughness of external cell membranes, which leads to the formation of larger cell domes. The results presented herein provide new insights into the molecular link between copper and the proliferation of breast cancer cells and, consequently, into the mechanism by which changes in redox balance and ROS accumulation regulates cell membrane roughness. Self-assembly of poly(4-vynil-N-alkyl)pyridinium bromide with alkyl side chains of 2, 5, 7, 10, or 16 carbons from ethanolic solutions onto flat silica surfaces was studied by means of ellipsometry, atomic force microscopy (AFM), contact angle measurements, and sum-frequency generation (SFG) vibrational spectroscopy in the CH3 and CH2 stretch region. Ab initio quantum-chemical calculations on the N-alkylpyridinium side-group with restricted Hartree-Fock (RHF) method and 6-311G (d,p) basis set were C one to estimate the charge distribution along the pyridinium ring and the alkyl side-chain. SFG results showed that longer side chains promote the disorientation of the alkyl groups at the surface, corroborating with the contact angle values. AFM images revealed film homogeneity, regardless the alkyl side group. However, after 24 h contact with water, ringlike structures appeared on the film surfaces, when the polycation alkyl side chain had 7 or less carbons, and as the alkyl chain increased to 10 or 16 carbons, the films dewetted because the hydrophobic interactions prevailed over the electrostatic interactions between the pyridinium charged groups and the negatively charged SiO2 surface. Under acid conditions (HCl 0.1 mol.L-1), the film mean thickness values decreased up to 50% of original values when the alkyl side chains were ethyl or pentyl groups due to ion-pair disruption, but for longer groups they remained unchanged. Quantum-chemical optimization and Mulliken electron population showed that (i) from C2 to C15 the positive charge at the headgroup (HG) decreased 0.025, while the charge at combined HG + alpha-CH2 increased 0.037; and (ii) for C6 or longer, the alkyl side group presents a tilt in the geometry, moving away from the plane. Such effects summed up over the whole polymer chain give support to suggest that when the side chains are longer than 7 carbons, the hydrophobic interaction decreases film stability and increases acid resistance.

OBJETIVO: Avaliar de forma crítica os fatores que influenciam os resultados da neurotizaão do nervo ulnar no ramo motor do músculo bíceps braquial, visando a restauraão da flexão do cotovelo em pacientes com lesão do plexo braquial. MÉTODOS: 19 pacientes, 18 homens e uma mulher, com idade média de 28,7 anos foram avaliados entre fevereiro de 2003 e maio de 2007. Oito pacientes apresentavam lesão das razes C5-C6 e 11, das razes C5-C6-C7. O intervalo de tempo médio entre a injúria e o tratamento cirúrgico foi 7,5 meses. Quatro pacientes apresentavam fraturas cervicais associadas à lesão do plexo braquial. O seguimento pós-operatório foi de 15,7 meses. RESULTADO: Oito pacientes recuperaram força de flexão do cotovelo MRC grau 4; dois, MRC grau 3 e nove, MRC < 3. Não houve prejuízo da função prévia do nervo ulnar. CONCLUSÃO: Os resultados da cirurgia de neurotizaão do nervo ulnar no ramo motor no músculo bíceps braquial são dependentes: do intervalo decorrido entre a lesão do plexo e o tratamento cirúrgico, da presença de fraturas associadas da coluna cervical e côndilo occipital, da função residual das razes de C8-T1 após o trauma e do acometimento da raiz de C7. Sinais de reinervaão manifestados até 3 meses no pós cirúrgico mostram melhores resultados ao longo prazo. Nível de Evidência: IV, Série de Casos.

The role of mitochondrial dysfunction in cancer has long been a subject of great interest. In this study, such dysfunction has been examined with regards to thyroid oncocytoma, a rare form of cancer, accounting for less than 5% of all thyroid cancers. A peculiar characteristic of thyroid oncocytic cells is the presence of an abnormally large number of mitochondria in the cytoplasm. Such mitochondrial hyperplasia has also been observed in cells derived from patients suffering from mitochondrial encephalomyopathies, where mutations in the mitochondrial DNA(mtDNA) encoding the respiratory complexes result in oxidative phosphorylation dysfunction. An increase in the number of mitochondria occurs in the latter in order to compensate for the respiratory deficiency. This fact spurred the investigation into the presence of analogous mutations in thyroid oncocytic cells. In this study, the only available cell model of thyroid oncocytoma was utilised, the XTC-1 cell line, established from an oncocytic thyroid metastasis to the breast. In order to assess the energetic efficiency of these cells, they were incubated in a medium lacking glucose and supplemented instead with galactose. When subjected to such conditions, glycolysis is effectively inhibited and the cells are forced to use the mitochondria for energy production. Cell viability experiments revealed that XTC-1 cells were unable to survive in galactose medium. This was in marked contrast to the TPC-1 control cell line, a thyroid tumour cell line which does not display the oncocytic phenotype. In agreement with these findings, subsequent experiments assessing the levels of cellular ATP over incubation time in galactose medium, showed a drastic and continual decrease in ATP levels only in the XTC-1 cell line. Furthermore, experiments on digitonin-permeabilised cells revealed that the respiratory dysfunction in the latter was due to a defect in complex I of the respiratory chain. Subsequent experiments using cybrids demonstrated that this defect could be attributed to the mitochondrially-encoded subunits of complex I as opposed to the nuclearencoded subunits. Confirmation came with mtDNA sequencing, which detected the presence of a novel mutation in the ND1 subunit of complex I. In addition, a mutation in the cytochrome b subunit of complex III of the respiratory chain was detected. The fact that XTC-1 cells are unable to survive when incubated in galactose medium is consistent with the fact that many cancers are largely dependent on glycolysis for energy production. Indeed, numerous studies have shown that glycolytic inhibitors are able to induce apoptosis in various cancer cell lines. Subsequent experiments were therefore performed in order to identify the mode of XTC-1 cell death when subjected to the metabolic stress imposed by the forced use of the mitochondria for energy production. Cell shrinkage and mitochondrial fragmentation were observed in the dying cells, which would indicate an apoptotic type of cell death. Analysis of additional parameters however revealed a lack of both DNA fragmentation and caspase activation, thus excluding a classical apoptotic type of cell death. Interestingly, cleavage of the actin component of the cytoskeleton was observed, implicating the action of proteases in this mode of cell demise. However, experiments employing protease inhibitors failed to identify the specific protease involved. It has been reported in the literature that overexpression of Bcl-2 is able to rescue cells presenting a respiratory deficiency. As the XTC-1 cell line is not only respiration-deficient but also exhibits a marked decrease in Bcl-2 expression, it is a perfect model with which to study the relationship between Bcl-2 and oxidative phosphorylation in respiratory-deficient cells. Contrary to the reported literature studies on various cell lines harbouring defects in the respiratory chain, Bcl-2 overexpression was not shown to increase cell survival or rescue the energetic dysfunction in XTC-1 cells. Interestingly however, it had a noticeable impact on cell adhesion and morphology. Whereas XTC-1 cells shrank and detached from the growth surface under conditions of metabolic stress, Bcl-2-overexpressing XTC-1 cells appeared much healthier and were up to 45% more adherent. The target of Bcl-2 in this setting appeared to be the actin cytoskeleton, as the cleavage observed in XTC-1 cells expressing only endogenous levels of Bcl-2, was inhibited in Bcl-2-overexpressing cells. Thus, although unable to rescue XTC-1 cells in terms of cell viability, Bcl-2 is somehow able to stabilise the cytoskeleton, resulting in modifications in cell morphology and adhesion. The mitochondrial respiratory deficiency observed in cancer cells is thought not only to cause an increased dependency on glycolysis but it is also thought to blunt cellular responses to anticancer agents. The effects of several therapeutic agents were thus assessed for their death-inducing ability in XTC-1 cells. Cell viability experiments clearly showed that the cells were more resistant to stimuli which generate reactive oxygen species (tert-butylhydroperoxide) and to mitochondrial calcium-mediated apoptotic stimuli (C6-ceramide), as opposed to stimuli inflicting DNA damage (cisplatin) and damage to protein kinases(staurosporine). Various studies in the literature have reported that the peroxisome proliferator-activated receptor-coactivator 1(PGC-1α), which plays a fundamental role in mitochondrial biogenesis, is also involved in protecting cells against apoptosis caused by the former two types of stimuli. In accordance with these observations, real-time PCR experiments showed that XTC-1 cells express higher mRNA levels of this coactivator than do the control cells, implicating its importance in drug resistance. In conclusion, this study has revealed that XTC-1 cells, like many cancer cell lines, are characterised by a reduced energetic efficiency due to mitochondrial dysfunction. Said dysfunction has been attributed to mutations in respiratory genes encoded by the mitochondrial genome. Although the mechanism of cell demise in conditions of metabolic stress is unclear, the potential of targeting thyroid oncocytic cancers using glycolytic inhibitors has been illustrated. In addition, the discovery of mtDNA mutations in XTC-1 cells has enabled the use of this cell line as a model with which to study the relationship between Bcl-2 overexpression and oxidative phosphorylation in cells harbouring mtDNA mutations and also to investigate the significance of such mutations in establishing resistance to apoptotic stimuli.

ZusammenfassungIn der vorliegenden Arbeit ist eine Enzymimmunoelektrode zur Bestimmung von Atrazin in Wasser entwickelt worden. Die Motivation war, einen Immunoassay zu entwickeln, der ohne die speziellen Geräte, wie einen EIA-Reader, durchgeführt werden konnte. Dafür müssen drei Änderungen vorgenommen werden. Es muß das Detektorsystem EIA-Reader zur Meßwerterfassung ersetzt werden, und damit muß die nur im EIA-Reader verwendbare Mikrotiterplatte ausgetauscht werden. Als drittes muß der Immunoassay dem neuen Detektorsystem angepat werden. Eine pH-Elektrode wurde anstelle des EIA-Readers benutzt. Als Enzym, das eine pH-Änderung induziert, wurde Lactamase ausgewählt. Als Festphase wurden anstelle der Mikrotiterplatte Polystyrolmikropartikel (PSMP) verwendet. Die Entwicklung der Enzymimmunoelektrode erfolgte in drei Schritten: Entwicklung des Immunoassays für Atrazin unter Verwendung von Lactamase, Übertragung auf die Festphase PSMP und Einsatz der pH-Elektrode als Detektorsystem. Zuerst wurden Tracer mit dem Enzym Lactamase hergestellt. Als Haptene wurden 2-Chlor-4-(isopropylamino)-6-[(1-carboxypent-5-yl)amino]-s-Triazin (iPr/Cl/C6), Di-Chloratrazin und Di-Chlorsimazin verwendet. Es wurden unterschiedliche Testmittelpunkte im Immunoassay erreicht, (iPr/Cl/C6 I50 = 1.22µg/L; Dichloratrazin I50 = 0.27µg/L; Di-Chlorsimazin I50 = 0.12µg/L). Aufgrund der nur mäßigen Stabilität der Tracer unter Verwendung der Di-Chlorderivate wurde auf deren Verwendung bei der Entwicklung der Immunoelektrode verzichtet.Im zweiten Schritt erfolgte die Übertragung auf PSMP. Die Verwendung der PSMP hatte außer einer Verbesserung des Testmittelpunktes auf 1.00µg/L noch den Vorteil, da die benötigten Mengen an Antikörper verringert werden konnten.Danach wurde die pH-Elektrode als Signalwandler zur Bestimmung des Atrazins eingesetzt. Unter Verwendung der pH-Elektrode konnte der bisher niedrigste Testmittelpunkt (I50 = 0.005µg/L) zur Bestimmung von Atrazin erreicht werden.

In this study, the use of the discotic liquid crystalline HBCs and conjugated polymers based on 2,7-carbazole were investigated in detail as donor materials in organic bulk-heterojunction solar cells. It has been shown that they perform efficiently in photovoltaic devices in combination with suitable acceptors. The efficiency was found to depend strongly dependent on the morphology of the film. By investigation of a series of donor materials with similar molecular structures based on both discotic molecules and conjugated polymers, a structure-performance relation was established, which is not only instructive for these materials but also serves as a guideline for improved molecular design. For the series of HBCs used in this study, it is found that the device efficiency decreases with increasing length of the alkyl substituents in the HBC. Thus, the derivative with the smallest alkyl mantle, being more crystalline compared to the HBCs with longer alkyl chains, gave the highest EQE of 12%. A large interfacial separation was found in the blend of HBC-C6,2 and PDI, since the crystallization of the acceptor occurred in a solid matrix of HBC. This led to small dispersed organized domains and benefited the charge transport. In contrast, blends of HBC-C10,6/PDI or HBC-C14,10/PDI revealed a rather homogeneous film limiting the percolation pathways due to a mixed phase. For the first time, poly(2,7-carbazole) was incorporated as a donor material in solar cells using PDI as an electron acceptor. The good fit in orbital energy levels and absorption spectra led to high efficiency. This result indicates that conjugated polymers with high band-gap can also be applied as materials to build efficient solar cells if appropriate electron acceptors are chosen. In order to enhance the light absorption ability, new ladder-type polymers based on pentaphenylene and hexaphenylene with one and three nitrogen bridges per repeat unit have been synthesized and characterized. The polymer 2 with three nitrogen bridges showed more red-shifted absorbance and emission and better packing in the solid-state than the analogous polymer 3 with only one nitrogen bridge per monomer unit. An overall efficiency as high as 1.3% under solar light was obtained for the device based on 1 and PDI, compared with 0.7% for the PCz based device. Therefore, the device performance correlates to a large extent with the solar light absorption ability and the lateral distance between conjugated polymer chains. Since the lateral distance is determined by the length and number of attached alkyl side chains, it is possible to assume that these substituents insulate the charge carrier pathways and decrease the device performance. As an additional consequence, the active semiconductor is diluted in the insulating matrix leading to a lower light absorption. This work suggests ways to improve device performance by molecular design, viz. maintaining the HOMO level while bathochromically shifting the absorption by adopting a more rigid ladder-type structure. Also, a high ratio of nitrogen bridges with small alkyl substituents was a desirable feature both in terms of adjusting the absorption and maintaining a low lateral inter-chain separation, which was necessary for obtaining high current and efficiency values.

Das Glaukom stellt eine heterogene Gruppe von okularen Erkrankungen dar, deren Pathogenese sich durch einen langsamen, progradienten Untergang von retinalen Ganglienzellen und ihren Axonen auszeichnet. rnIn den letzten Jahren wurde im Kontext der Glaukompathogenese verstärkt die Beteiligung autoreaktiver Antikörper diskutiert. Ein Schwerpunkt dieser Arbeit bestand in dem Vergleich solcher Autoantikörper-Reaktionen in den Serum- und Kammerwasserproben einzelner Glaukompatienten. Hierdurch sollte geklärt werden, inwieweit die Immunreaktivitäten dieser beiden Körperflüssigkeiten miteinander übereinstimmen und ob sich Hinweise auf eine lokale Antikörperproduktion im immunprivilegierten Auge finden lassen. Mittels eines etablierten Protein-Microarray-Verfahrens wurden die Immunreaktionen gegen 40 verschiedene Antigene, wie z.B. Hitzeschock-Proteine oder neuronale Strukturproteine, untersucht. Die Ergebnisse zeigten, dass die detektierten Autoantikörper-Reaktionen gegen mehr als 80% der untersuchten Antigene in beiden Körperflüssigkeiten miteinander übereinstimmen. Verdeutlicht wird hierdurch, dass die Antikörper-basierenden immunologischen Vorgänge im Auge bzw. Kammerwasser, trotz dessen Abschottung vom Blutkreislauf durch die Blut-Retina-Schranke, denen des Serums stark ähneln. Nur vereinzelt lassen sich Hinweise auf eine lokale Antikörperproduktion im Auge finden, wodurch die Bedeutung der detektierten Serumantikörper-Reaktionen für die Glaukomerkrankung belegt wird. rnEin weiterer Schwerpunkt der Arbeit lag auf der Detektion möglicher veränderter Proteinexpressionen in den Retinae und Serumproben von Glaukompatienten, die potentiell zu den neurodegenerativen Prozessen der Glaukompathogenese beitragen. Um die Analyse spezifischer Proteinexpressionen zu ermöglichen, wurde das Verfahren des Antikörper-Microarrays etabliert und auf die Fragestellung angewendet. Untersucht wurden hierbei vor allem die Abundanzen von Komplementproteinen, Zytokinen und Hitzeschock-Proteinen, aber auch die von verschiedenen neuronalen Strukturproteinen. Als Probenmaterial dienten Serum- und Retinaproben von Glaukompatienten, die vergleichend denen von gesunden Probanden gegenübergestellt wurden. Die Analyse erbrachte die Erkenntnis, dass neben der verstärkten Expression von Komplementproteinen in der Retina (z.B. C3, C6) auch im Serum der Glaukompatienten eine erhöhte Konzentration dieser Proteine vorliegt, die im Rahmen der Glaukomerkrankung möglicherweise ebenfalls eine Rolle spielen. Ähnliches konnte für verschiedene Zytokine, wie z.B. TNF-α, IFN-γ oder IL1-β beobachtet werden, die in den untersuchten Retinae von Glaukomprobanden, teilweise auch in den Serumproben der Patienten, in verstärktem Mae detektiert werden konnten. Die erhöhte Produktion von Zytokinen in der Retina ist wahrscheinlich auf die Aktivierung von Gliazellen zurückzuführen, ein Ereignis für das in dieser Arbeit zahlreiche Hinweise gefunden werden konnten. Die Gliaaktivierung wird vermutlich durch apoptotische Prozesse in der Retina ausgelöst, eventuell aber auch durch eine erfolgte Komplementaktivierung. Darüber hinaus konnten mittels eines massenspektrometrischen Verfahrens weitere Expressionsunterschiede verschiedener retinaler Proteine bei Glaukompatienten festgestellt werden. Diese Veränderungen, wie z.B. geminderte Mengen von ROS-eliminierenden Proteinen, wie der Superoxid Dismutase und Peroxiredoxin-2, begünstigen bzw. verstärken sehr wahrscheinlich die neurodegenerativen Prozesse in der Retina von GlaukompatientenrnInwieweit die untersuchten Faktoren kausativ an den neurodegenerativen Prozessen beteiligt sind, bleibt ungeklärt, jedoch untermauert deren Vielzahl die Notwendigkeit, die Ursache der Glaukomerkrankung als komplexe Interaktion und Wechselwirkung verschiedener Komponenten zu betrachten und nicht als einen einzelnen fehlgesteuerten Mechanismus.rn

Das Enterobakterium Escherichia coli sowie das Bodenbakterium Bacillus subtilis können C4-Dicarbonsäuren als aerobe Kohlenstoffquelle zur Energiekonservierung nutzen. Die Regulation des C4-Dicarboxylatstoffwechsels erfolgt in E. coli und B. subtilis durch das Zweikomponentensystem DcuSREc bzw. DctSRBs, bestehend aus einer Sensorkinase und einem Responseregulator. Diese kontrollieren die Expression des C4-Dicarboxylat-Transporters DctA. Der Sensor DcuSEc benötigt für seine Funktion im aeroben Stoffwechsel den Transporter DctA als Cosensor. Für das DctSRBs-System gibt es Hinweise aus genetischen Untersuchungen, dass DctSBs das Bindeprotein DctBBs und möglicherweise auch DctABs als Cosensoren für seine Funktion benötigt. In dieser Arbeit sollte ein direkter Nachweis geführt werden, ob DctBBs und DctABs gemeinsam oder nur jeweils eine der Komponenten als Cosensoren für DctSBs fungieren. Sowohl für DctBBs als auch für DctABs wurde eine direkte Protein-Protein-Interaktion mit DctSBs durch zwei in vivo Interaktionsmethoden nachgewiesen. Beide Methoden beruhen auf der Co-Reinigung der Interaktionspartner mittels Affinitätschromatographie und werden je nach Affinitätssäule als mSPINE oder mHPINE (Membrane Strep/His-Protein INteraction Experiment) bezeichnet. Die Interaktion von DctSBs mit DctBBs wurde zusätzlich über ein bakterielles Two-Hybrid System nachgewiesen. Nach Coexpression mit DctSBs interagieren DctABs und DctBBs in mSPINE-Tests gleichzeitig mit der Sensorkinase. DctSBs bildet somit eine sensorische DctS/DctA/DctB-Einheit in B. subtilis und das Bindeprotein DctBBs agiert nur als Cosensor, nicht aber als Transport-Bindeprotein. Eine direkte Interaktion zwischen dem Transporter DctABs und dem Bindeprotein DctBBs besteht nicht. Transportmessungen belegen, dass der DctA-vermittelte Transport von [14C]-Succinat unabhängig ist von DctBBs. Außerdem wurde untersucht, ob Zweikomponentensysteme aus anderen Bakteriengruppen nach einem ähnlichen Schema wie DcuSREc bzw. DctSRBs aufgebaut sind. Das thermophile Bakterium Geobacillus kaustophilus verfügt über ein DctSR-System, welches auf genetischer Ebene mit einem Transporter des DctA-Typs und einem DctB-Bindeprotein geclustert vorliegt. Die Sensorkinase DctSGk wurde in E. coli heterolog exprimiert und gereinigt. Diese zeigt in einer E. coli DcuS-Insertionsmutanten Komplementation der DcuS-Funktion und besitzt dabei Spezifität für die C4-Dicarbonsäuren Malat, Fumarat, L-Tartrat und Succinat sowie für die C6-Tricarbonsäure Citrat. In Liposomen rekonstituiertes DctSGk zeigt Autokinase-Aktivität nach Zugabe von [γ-33P]-ATP. Der KD-Wert für [γ-33P]-ATP der Kinasedomäne von DctSGk liegt bei 43 μM, die Affinität für ATP ist damit etwa 10-fach höher als in DcuSEc.

Eisbohrkerne stellen wertvolle Klimaarchive dar, da sie atmosphärisches Aerosol konservieren. Die Analyse chemischer Verbindungen als Bestandteil atmosphärischer Aerosole in Eisbohrkernen liefert wichtige Informationen über Umweltbedingungen und Klima der Vergangenheit. Zur Untersuchung der α-Dicarbonyle Glyoxal und Methylglyoxal in Eis- und Schneeproben wurde eine neue, sensitive Methode entwickelt, die die Stir Bar Sorptive Extraction (SBSE) mit der Hochleistungsflüssigchromatographie-Massenspektrometrie (HPLC-MS) kombiniert. Zur Analyse von Dicarbonsäuren in Eisbohrkernen wurde eine weitere Methode entwickelt, bei der die Festphasenextraktion mit starkem Anionenaustauscher zum Einsatz kommt. Die Methode erlaubt die Quantifizierung aliphatischer Dicarbonsäuren (≥ C6), einschließlich Pinsäure, sowie aromatischer Carbonsäuren (wie Phthalsäure und Vanillinsäure), wodurch die Bestimmung wichtiger Markerverbindungen für biogene und anthropogene Quellen ermöglicht wurde. Mit Hilfe der entwickelten Methoden wurde ein Eisbohrkern aus den Schweizer Alpen analysiert. Die ermittelten Konzentrationsverläufe der Analyten umfassen die Zeitspanne von 1942 bis 1993. Mittels einer Korrelations- und Hauptkomponentenanalyse konnte gezeigt werden, dass die organischen Verbindungen im Eis hauptsächlich durch Waldbrände und durch vom Menschen verursachte Schadstoffemissionen beeinflusst werden. Im Gegensatz dazu sind die Konzentrationsverläufe einiger Analyten auf den Mineralstaubtransport auf den Gletscher zurückzuführen. Zusätzlich wurde ein Screening der Eisbohrkernproben mittels ultrahochauflösender Massenspektrometrie durchgeführt. Zum ersten Mal wurden in diesem Rahmen auch Organosulfate und Nitrooxyorganosulfate in einem Eisbohrkern identifiziert.

Before signals of the visual environment are transferred to higher brain areas via the optic nerve, they are processed and filtered in parallel pathways within the retina. In the past a plethora of functionally distinct ganglion cell types responding to certain aspects of the environment, such as direction of movement, contrast and colour have been described. Aim of this thesis was the anatomical investigation of the selectivity in retinal circuits underlying this diversity. For this purpose, mouse and macaque retinae were analysed. OFF-ganglion cells in the mouse retina received their excitatory drive unselectively from all bipolar cell types stratifying within the area of their dendritic trees. Only the input to direction-selective C6 ganglion cells and bistratified D2 ganglion cells appeared to be weighted. In primates the highly specialised midget-system forms a 1:1 connection from red- and green-sensitive cones onto midget bipolar- and ganglion cells, building the substrate for red/green colour vision. Here it was demonstrated that blue-sensitive (S-) cones also contact OFF-midget bipolars and are, thus, potential candidates to transfer blue-OFF signals to M1 intrinsically photosensitive ganglion cells (ipRGCs). M1 cells received glycinergic input from A8 amacrine cells and express GABAA receptors containing subunit alpha 3. M2 cells, in contrast, received less inhibitory input.

In questo lavoro di tesi ci proponiamo di determinare lo spessore degli isotopi ^157Gd e ^155Gd in vista della misura della sezione d’urto di cattura neutronica presso la facility n_TOF del CERN. La principale motivazione dell’esperimento è legata alla necessità di ottenere misure più accurate per le applicazioni ai reattori nucleari. Inoltre, i nuovi risultati, potranno essere sfruttati anche per applicazioni ai recenti sviluppi nella Terapia di Cattura Neutronica e per costruire nuovi rivelatori nell’ambito della ricerca del neutrino. La misura sarà effettuata nella prima area sperimentale EAR-1 di n_TOF, equipaggiata con rivelatori, come per esempio gli scintillatori liquidi al benzene deuterato (C6 D6) particolarmente adatti per questi tipi di misura. La sezione d’urto di questi due isotopi cambia di molti ordini di grandezza al variare dell’energia dei neutroni incidenti. Per questo motivo, lo studio effettuato in questa tesi ha mostrato che sono necessari due campioni altamente arricchiti per ogni isotopo da misurare: un campione estremamente sottile per energie del neutrone fino a 100 meV, e uno più spesso per energie maggiori. Inoltre per questi campioni sono stati determinati le densità areali necessarie per lo svolgimento dell’esperimento affinchè avvenga il fenomeno di trasmissione dei neutroni.

The most common techniques to perform stellate ganglion blocks (SGBs) are the blind C6 approach and the fluoroscopic-controlled paratracheal C7 approach, both after manual dislocation of the large vessels. Complications due to vascular or esophageal puncture have been reported. The goal of this ultrasound imaging study was to determine how frequently hazardous structures are located along the needle path of conventional SGB and to determine the influence of the dislocation maneuver on their position.

INTRODUCTION: The ultrastructure of venous valves and walls in chronic venous disease was investigated. METHODS: Consecutive patients were categorised into one of three groups (group A: patients with C1 venous disease in accordance with CEAP (Clinical severity, Etiology, Anatomy, Pathophysiology); group B: C2 and C3; group C: C4, C5 and C6). The terminal or preterminal valve and adjacent vessel wall was harvested from the great saphenous vein. Sections were examined with a transmission electron microscope. The volumes of elastin and of collagen per unit surface area of valve were assessed, as well as the surface endothelium of valve and vessel wall. RESULTS: The study population consisted of 17 patients. The elastin ratio was analysed by means of stereology. Mean values were: in group A, 0.45 μm3/m2; in group B, 0.67 μm3/m2; in group C, 0.97 μm3/m2. The ratio was similar for collagen (A, 15.7 μm3/m2; B, 26.8 μm3/m2; C, 30.1 μm3/m2). Surface analysis of the valve endothelium and the adjacent vessel wall endothelium showed a trend towards increasing damage with more severe disease. CONCLUSIONS: With progression of venous disease, the valve elastin content, assessed morphologically, seems to increase, and the endothelium of the venous valve and the vein wall tend to show more damage.
