The larvicidal activity of the neotropical "matico" Piper tuberculatum was evaluated. The secondary compounds were extracted of leaves, stems and mature spikes with fruits and seeds from wild plants and in vitro plants of Piper tuberculatum. The acute toxicities to the fall armyworm, Spodoptera frugiperda (Smith) (Lepidoptera: Noctuidae), of extracts of spikes with fruits and seeds and in vitro plants of P tuberculatum were evaluated by means of contact bioassays. Only CH2Cl2:MeOH (2:1) and EtOH extracts of mature spikes and CH2C12:MeOH (2:1) extract from in vitro plants showed significant levels of larval mortality. The CH2Cl2:MeOH (2:1) and EtOH extracts of mature spikes caused 90% mortality when doses of 0.1850 mg/mu L were applied to the S. frugiperda in 24 and 48 h of exposure, respectively. The CH2Cl2:MeOH (2:1) extract from in vitro plants caused 95% mortality when doses of 0.1850 mg/mu L were too applied in 48 h of exposure. The mature spikes test best results were: LD50 0.001 mg/mu L with EtOH and 0.007 mg/mu L with CH2Cl2:MeOH (2:1) and LD90 0.027 mg/mu L with EtOH and 0.103 mg/mu L with CH2Cl2:MeOH (2:1); and, in the case of in vitro plants, only CH2Cl2:MeOH (2:1) extract was: LD50 0.003 mg/mu L and LD90 0.060 mg/mu L. The potential value of extracts derived from P. tuberculatum as efficient insecticides against S.frugiperda is discussed.

Psychiatric co-morbidities in epilepsy are common in patients with temporal lobe epilepsy (TLE). Pathological alterations in TLE are well characterised; however, neuropathologic data are relatively scale regarding the association between psychiatric diseases and epilepsy. Our objective was to evaluate the clinical data of 46 adult TLE patients with and without psychiatric co-morbidities and to correlate the data with hippocampal neuronal density and mossy fiber sprouting. Accordingly, patients were grouped as follows: TLE patients without history of psychiatric disorder (TLE, n = 16), TLE patients with interictal psychosis (TLE + P, n = 14), and TLE patients with major depression (TLE + D, n = 16). Hippocampi from autopsies served as non-epileptic controls (n = 10). TLE + P exhibited significantly diminished mossy fiber sprouting and decreased neuronal density in the entorhinal cortex when compared with TLE. TLE + P showed significantly poorer results in verbal memory tasks. TLE + D exhibited significantly increased mossy fiber sprouting length when compared with TLE and TLE + P. Further, a higher proportion of TLE + D and TLE + P presented secondarily generalised seizures than did TLE. Our results indicate that TLE patients with psychiatric disorders have distinct features when compared with TLE patients without psychiatric co-morbidities and that these changes may be involved in either the manifestation or the maintenance of psychiatric co-morbidities in epilepsy. Stress is the most commonly reported precipitating factor for seizures in patients with epilepsy. Despite compelling anecdotal evidence for stress-induced seizures, animal models of the phenomena are sparse and possible mechanisms are unclear. Here, we tested the hypothesis that increased levels of the stress-associated hormone corticosterone ( CORT) would increase epileptiform activity and spontaneous seizure frequency in mice rendered epileptic following pilocarpine-induced status epilepticus. We monitored video-EEG activity in pilocarpine-treated mice 24/7 for a period of four or more weeks, during which animals were serially treated with CORT or vehicle. CORT increased the frequency and duration of epileptiform events within the first 24 hours of treatment, and this effect persisted for up to two weeks following termination of CORT injections. Interestingly, vehicle injection produced a transient spike in CORT levels - presumably due to the stress of injection - and a modest but significant increase in epileptiform activity. Neither CORT nor vehicle treatment significantly altered seizure frequency; although a small subset of animals did appear responsive. Taken together, our findings indicate that treatment of epileptic animals with exogenous CORT designed to mimic chronic stress can induce a persistent increase in interictal epileptiform activity.

Objective: There is accumulating evidence that the limbic system is pathologically involved in cases of psychiatric comorbidities in temporal lobe epilepsy (TLE) patients. Our objective was to develop a conceptual framework describing how neuropathological, neurochemical and electrophysiological aspects might contribute to the development of psychiatric symptoms in TLE and the putative neurobiological mechanisms that cause mood disorders in this patient subgroup. Methods: In this review, clinical, experimental and neuropathological findings, as well as neurochemical features of the limbic system were examined together to enhance our understanding of the association between TLE and psychiatric comorbidities. Finally, the value of animal models in epilepsy and mood disorders was discussed. Conclusions: TLE and psychiatric symptoms coexist more frequently than chance would predict. Alterations and neurotransmission disturbance among critical anatomical networks, and impaired or aberrant plastic changes might predispose patients with TLE to mood disorders. Clinical and experimental studies of the effects of seizures on behavior and electrophysiological patterns may offer a model of how limbic seizures increase the vulnerability of TLE patients to precipitants of psychiatric symptoms.

It has been revealed that the network of excitable neurons via attractive coupling can generate spikes under stimuli of subthreshold signals with disordered phases. In this paper, we explore the firing activity induced by phase disorder in excitable neuronal networks consisting of both attractive and repulsive coupling. By increasing the fraction of repulsive coupling, we find that, in the weak coupling strength case, the firing threshold of phase disorder is increased and the system response to subthreshold signals is decreased, indicating that the effect of inducing neuron firing by phase disorder is weakened with repulsive coupling. Interestingly, in the large coupling strength case, we see an opposite situation, where the coupled neurons show a rather large response to the subthreshold signals even with small phase disorder. The latter case implies that the effect of phase disorder is enhanced by repulsive coupling. A system of two-coupled excitable neurons is used to explain the role of repulsive coupling on phase-disorder-induced firing activity.

Background: Kinins, with bradykinin and des-Arg(9)-bradykinin being the most important ones, are pro-inflammatory peptides released after tissue injury including stroke. Although the actions of bradykinin are in general well characterized; it remains controversial whether the effects of bradykinin are beneficial or not. Kinin-B2 receptor activation participates in various physiological processes including hypotension, neurotransmission and neuronal differentiation. The bradykinin metabolite des-Arg(9)-bradykinin as well as Lys-des-Arg(9)-bradykinin activates the kinin-B1 receptor known to be expressed under inflammatory conditions. We have investigated the effects of kinin-B1 and B2 receptor activation on N-methyl-Daspartate (NMDA)-induced excitotoxicity measured as decreased capacity to produce synaptically evoked population spikes in the CA1 area of rat hippocampal slices. Principal Findings: Bradykinin at 10 nM and 1 mu M concentrations triggered a neuroprotective cascade via kinin-B2 receptor activation which conferred protection against NMDA-induced excitotoxicity. Recovery of population spikes induced by 10 nM bradykinin was completely abolished when the peptide was co-applied with the selective kinin-B2 receptor antagonist HOE-140. Kinin-B2 receptor activation promoted survival of hippocampal neurons via phosphatidylinositol 3-kinase, while MEK/MAPK signaling was not involved in protection against NMDA-evoked excitotoxic effects. However, 100 nM Lys-des-Arg(9)-bradykinin, a potent kinin-B1 receptor agonist, reversed bradykinin-induced population spike recovery. The inhibition of population spikes recovery was reversed by PD98059,showing that MEK/MAPK was involved in the induction of apoptosis mediated by the B1 receptor. Conclusions: Bradykinin exerted protection against NMDA-induced excitotoxicity which is reversed in the presence of a kinin-B1 receptor agonist. As bradykinin is converted to the kinin-B1 receptor metabolite des-Arg(9)-bradykinin by carboxypeptidases, present in different areas including in brain, our results provide a mechanism for the neuroprotective effect in vitro despite of the deleterious effect observed in vivo.

Purinergic receptors participate, in almost every cell type, in controlling metabolic activities and many physiological functions including signal transmission, proliferation and differentiation. While most of P2Y receptors induce transient elevations of intracellular calcium concentration by activation of intracellular calcium pools and forward these signals as waves which can also be transmitted into neighboring cells, P2X receptors produce calcium spikes which also include activation of voltage-operating calcium channels. P2Y and P2X receptors induce calcium transients that activate transcription factors responsible for the progress of differentiation through mediators including calmodulin and calcineurin. Expression of P2X2 as well as of P2X7 receptors increases in differentiating neurons and glial cells, respectively. Gene expression silencing assays indicate that these receptors are important for the progress of differentiation and neuronal or glial fate determination. Metabotropic receptors, mostly P2Y1 and P2Y2 subtypes, act on embryonic cells or cells at the neural progenitor stage by inducing proliferation as well as by regulation of neural differentiation through NFAT translocation. The scope of this review is to discuss the roles of purinergic receptor-induced calcium spike and wave activity and its codification in neurodevelopmental and neurodifferentiation processes.

The first part of my thesis presents an overview of the different approaches used in the past two decades in the attempt to forecast epileptic seizure on the basis of intracranial and scalp EEG. Past research could reveal some value of linear and nonlinear algorithms to detect EEG features changing over different phases of the epileptic cycle. However, their exact value for seizure prediction, in terms of sensitivity and specificity, is still discussed and has to be evaluated. In particular, the monitored EEG features may fluctuate with the vigilance state and lead to false alarms. Recently, such a dependency on vigilance states has been reported for some seizure prediction methods, suggesting a reduced reliability. An additional factor limiting application and validation of most seizure-prediction techniques is their computational load. For the first time, the reliability of permutation entropy [PE] was verified in seizure prediction on scalp EEG data, contemporarily controlling for its dependency on different vigilance states. PE was recently introduced as an extremely fast and robust complexity measure for chaotic time series and thus suitable for online application even in portable systems. The capability of PE to distinguish between preictal and interictal state has been demonstrated using Receiver Operating Characteristics (ROC) analysis. Correlation analysis was used to assess dependency of PE on vigilance states. Scalp EEG-Data from two right temporal epileptic lobe (RTLE) patients and from one patient with right frontal lobe epilepsy were analysed. The last patient was included only in the correlation analysis, since no datasets including seizures have been available for him. The ROC analysis showed a good separability of interictal and preictal phases for both RTLE patients, suggesting that PE could be sensitive to EEG modifications, not visible on visual inspection, that might occur well in advance respect to the EEG and clinical onset of seizures. However, the simultaneous assessment of the changes in vigilance showed that: a) all seizures occurred in association with the transition of vigilance states; b) PE was sensitive in detecting different vigilance states, independently of seizure occurrences. Due to the limitations of the datasets, these results cannot rule out the capability of PE to detect preictal states. However, the good separability between pre- and interictal phases might depend exclusively on the coincidence of epileptic seizure onset with a transition from a state of low vigilance to a state of increased vigilance. The finding of a dependency of PE on vigilance state is an original finding, not reported in literature, and suggesting the possibility to classify vigilance states by means of PE in an authomatic and objectic way. The second part of my thesis provides the description of a novel behavioral task based on motor imagery skills, firstly introduced (Bruzzo et al. 2007), in order to study mental simulation of biological and non-biological movement in paranoid schizophrenics (PS). Immediately after the presentation of a real movement, participants had to imagine or re-enact the very same movement. By key release and key press respectively, participants had to indicate when they started and ended the mental simulation or the re-enactment, making it feasible to measure the duration of the simulated or re-enacted movements. The proportional error between duration of the re-enacted/simulated movement and the template movement were compared between different conditions, as well as between PS and healthy subjects. Results revealed a double dissociation between the mechanisms of mental simulation involved in biological and non-biologial movement simulation. While for PS were found large errors for simulation of biological movements, while being more acurate than healthy subjects during simulation of non-biological movements. Healthy subjects showed the opposite relationship, making errors during simulation of non-biological movements, but being most accurate during simulation of non-biological movements. However, the good timing precision during re-enactment of the movements in all conditions and in both groups of participants suggests that perception, memory and attention, as well as motor control processes were not affected. Based upon a long history of literature reporting the existence of psychotic episodes in epileptic patients, a longitudinal study, using a slightly modified behavioral paradigm, was carried out with two RTLE patients, one patient with idiopathic generalized epilepsy and one patient with extratemporal lobe epilepsy. Results provide strong evidence for a possibility to predict upcoming seizures in RTLE patients behaviorally. In the last part of the thesis it has been validated a behavioural strategy based on neurobiofeedback training, to voluntarily control seizures and to reduce there frequency. Three epileptic patients were included in this study. The biofeedback was based on monitoring of slow cortical potentials (SCPs) extracted online from scalp EEG. Patients were trained to produce positive shifts of SCPs. After a training phase patients were monitored for 6 months in order to validate the ability of the learned strategy to reduce seizure frequency. Two of the three refractory epileptic patients recruited for this study showed improvements in self-management and reduction of ictal episodes, even six months after the last training session.

Zusammenfassung Es wurden im Rahmen dieser Arbeit elementspezifische Wechselwirkungen von Humin-stoffen (HS) untersucht. Grundlage war hierbei die Kopplung eines ICP-MS mit einer HPLC unter Verwendung der Size Exclusion Chromatography (SEC). Durch die Kopplung des empfindlichen und elementspezifischen Massenspektrometers als Detektor mit einer HPLC als chromatographisches Trennverfahren von HS-Spezies konnten Gehalte im natürlichen Konzentrationsbereich und Wechselwirkungen einzelner HS-Fraktionen be-stimmt werden.Zur Gehaltsbestimmung in Echtzeit wurde die online MSIVA (massenspektrometrische Isotopenverdünnungsanalyse) für C angewendet und für die simultane Bestimmung von S und Hg modifiziert. Hierzu wurde ein auf 34S bzw. 201Hg angereicherter Spike verwendet. Mit Hilfe der entwickelten online MSIVA für die simultane Detektion von C, S und Hg konnten Gehalte in chromatographischen Fraktionen mit einer Nachweisgrenze von 3 ng C/s, 0.1 ng S/s und 2 pg Hg/s bestimmt werden.Um die Nachweisstärke und Richtigkeit der Methode zu überprüfen, wurden die C- und S-Gesamtgehalte von HS-Proben bestimmt und mit den elementar-analytischen Daten anderer Arbeitgruppen verglichen. Weiterhin wurde an einer internationalen Laborstudie (IMEP-9) als zertifizierendes Labor teilgenommen. Die in dieser Arbeit erhaltenen Werte für Cr, Pb und U wurden zur Ermittlung der zertifizierten Werte herangezogen und zeigten eine große Übereinstimmung mit den Werten anderer Arbeitgruppen.Nach der Analyse von Gesamtgehalten wurden auch die C- und S-Gehalte in einzelnen Fraktionen von HS nach SEC ermittelt. Hierdurch konnte im Vergleich zur bisher ange-wendeten Elementaranalyse der Gesamtprobe eine Verbesserung bei der Bestimmung des S-Anteils in HS erreicht werden. So ergeben sich Fehler bei der Bestimmung des C/S-Verhältnisses durch anorganisches Sulfat (S-Überbestimmung) sowie durch nieder-molekulare Anteile, die nicht zu den HS gerechnet werden (C-Überbestimmung).Durch die simultane Detektion von Hg konnten die Wechselwirkungen mit HS aufgezeigt werden. Als Bindungsstellen wurden in erster Linie schwefelhaltige Fraktionen identifiziert, wobei S in reduzierter Form (-2) eine weitaus höhere Affinität zu Hg hat. Jedoch konnte auch die Bindung an schwefelfreie Fraktionen nachgewiesen werden. Dies ist für die Untersuchung der Mobilität und Bioverfügbarkeit von Hg in der Umwelt eine wichtige Erkenntnis.Die entwickelte online MSIVA für Schwefel wurde in Zusammenarbeit mit A. Prange und D. Schaumlöffel auf die CE/ICP-MS-Kopplung übertragen. Hierbei musste ein neues Konzept für die Zuführung des isotopenangereicherten Spikes und der Kalibration des Systems entwickelt werden. Mit Hilfe der online MSIVA konnten erstmalig mit CE fraktionierte Metallothioneine aus Leber-Cytosol quantifiziert werden, indem über den S-Gehalt und den Cysteinanteil der Proteingehalt ermittelt wurde.Weiterhin wurde durch Isotopenaustauschversuche erstmalig die kinetische Stabilität von Cr(III)- und Co(III)-Komplexen in mit SEC getrennten Fraktionen nachgewiesen. Analoge Cu(II)- und Co(II)-Komplexe sind hingegen kinetisch labil. Dies ist mit Hilfe der Ligandenfeldstabilisierungsenergie des Übergangszustandes zu er-klären. Bei d3- und d6-Spinsystemen, wie den Cr(III)- und Co(III)-Komplexen, liegt diese Energie deutlich höher als die der d7- und d9-Spinsysteme, wie Cu(II)- und Co(II)-Komplexe. Die höhere Energie-barriere des Übergangzustandes verlangsamt daher die Austausch-geschwin-digkeit, wodurch kinetisch stabile Komplexe entstehen.

Zur Untersuchung der speziesspezifischen Transformationsprozesse des Quecksilbers in der Umwelt wurden erstmalig Mikrokosmosexperimente unter Verwendung von isotopenangereicherten Verbindungen durchgeführt. Es wurden naturrelevante Bedingungen simuliert, um eine spätere Übertragbarkeit der Ergebnisse auf den biogeochemischen Kreislauf des Quecksilbers zu gewährleisten. Die aufgebauten Mikrokosmen bestanden aus Boden/Pflanzen/Luft-Kompartimenten. Der Boden der Mikrokosmen wurde mit isotopenangereicherten Quecksilberspezies dotiert. Durch die Verwendung von isotopenangereicherten Verbindungen, verbunden mit gleichzeitiger ICP/MS-Detektion, konnten auftretende Transformationsprozesse beobachtet werden. Die Messung der Quecksilberspezies erfolgte mittels GC-ICP/MS nach vorheriger Derivatisierung mit Natriumtetraethylborat und Anreicherung per 'purge and trap'.Die Massenspuren der einzelnen Quecksilberisotope wurden für alle Quecksilberspezies gemessen und daraus dann für jede Spezies die Isotopenverhältnisse gebildet. Bei einer Änderung des Isotopenverhältnisses kann von einer Speziestransformation ausgegangen werden.Aus den Mikrokosmosexperimenten, denen Methylquecksilber als angereicherte Isotopenverbindung zugegeben wurde, konnte gefolgert werden, dass Methylquecksilber im Boden zunächst zu anorganischem Quecksilber demethyliert wurde. Im Anschluss daran erfolgte eine Reduktion zu elementarem Quecksilber. Dieses gebildete elementare Quecksilber verflüchtigte sich nahezu vollständig (ca. 90-100%) vom Boden in die Atmosphäre.Bei der Zugabe von anorganischem Quecksilber als angereicherte Isotopenverbindung in den Boden wurde vorwiegend eine Reduktion zu elementarem Quecksilber beobachtet, das dann in die Atmosphäre emittiert. Es konnte aber auch eine geringe Methylierung (ca. 5%) zu Methylquecksilber beobachtet werden. Daraus kann gefolgert werden, dass die methylierten Quecksilberverbindungen eine wesentliche Rolle im natürlichen Kreislauf des Quecksilbers spielen.Parallel zu den Mikrokosmosexperimenten wurden Feldversuche in einem flussnahen Feuchtgebiet, aus dem auch der Boden für die Mikrokosmosexperimente entnommen worden war, durchgeführt. In den Feldversuchen wurden Quecksilberkonzentrationen und der Quecksilberfluss zwischen dem Boden und der Atmosphäre mit Hilfe von Flusskammerexperimenten bestimmt. Es konnten mit Hilfe von Mikrokosmen, die natürliche Verhältnisse simulieren sollten, und mit Hilfe verschieden angereicherter Isotopenspikes erstmals Speziestransformationen des Quecksilbers direkt beobachtet werden. Die GC-ICP/MS-Methode ermöglichte eine eindeutige Identifikation von Edukt und Produkt der jeweiligen Umwandlung. Allerdings wurde die Untersuchung eingeschränkt durch die irreversiblen biologischen Veränderungen in den eingesetzten Mikrokosmen nach über einer Woche und durch die Notwendigkeit, vergleichsweise hohe Konzentrationen der Spikes einzusetzen, um eine statistisch signifikante Auswertung der Veränderung der Isotopenverhältnisse zu erreichen. Somit sind Mikrokosmosexperimente nur eingeschränkt für Untersuchungen des Quecksilberverhaltens geeignet.Im Rahmen dieser Arbeit ist es aber gelungen, die Mikrokosmenexperimente unter Verwendung von isotopenangereicherten Verbindungen und der GC-ICP/MS-Methode als leistungsstarkes Verfahren zur Beobachtung von Speziestransformationsprozessen des Quecksilbers zu etablieren.

AII Amakrinzellen sind Interneurone in der Retina und ein wichtiges Element der Stäbchenbahn von Säugetieren. Bei ihren Antworten auf Lichtreize generieren sie Aktionspotentiale, obwohl die ihnen vor- und nachgeschalteten Bipolarzellen graduierte Membranpotentiale aufweisen. Um die Verarbeitung der Lichtsignale in der Stäbchenbahn der Säuger besser zu verstehen wurden in der vorliegenden Arbeit Membranströme von AII Amakrinzellen und Veränderungen der intrazellulären Kalziumkonzentration mittels Indikatorfarbstoffe bei Mäusen simultan gemessen.Die spannungsabhängigen Kalziumkanäle waren durch eine negative Aktivierungsschwelle und eine sehr langsame Inaktivierung gekennzeichnet¸ ausserdem wurden sie von Dihydropyridinen (Agonisten und Antagonisten) moduliert. Sie fanden sich vor allem auf den keulenförmigen Fortsätzen von AII Amakrinzellen. Lokale Applikationen von Glutamat, AMPA oder Kainat lösten einwärtsgerichtete Ströme aus. Diese Ströme gingen einher mit einer Erhöhung der Fluoreszenz und zwar vor allem in den distalen Dendriten. NMDA löste keine Veränderung der Kalziumkonzentration aus und nur in wenigen Fällen Ströme (7 von 23).Diese Befunde deuten darauf hin, dass es sich bei den ionotropen Glutamat-Rezeptoren auf AII Amakrinzellen um solche vom AMPA Typ handelt. Diese befinden sich, sofern sie kalziumpermeabel sind (oder durch andere Mechanismen zu einer Erhöhung der [Ca2+]i führen) auf den distalen Dendriten nahe der Ganglienzellschicht.

An accurate and sensitive species-specific GC-ICP-IDMS (gas chromatography inductively coupled plasma isotope dilution mass spectrometry) method for the determination of trimethyllead and a multi-species-specific GC-ICP-IDMS method for the simultaneous determination of trimethyllead, methylmercury, and butyltins in biological and environmental samples were developed. They allow the determination of corresponding elemental species down to the low ng g-1 range. The developed synthesis scheme for the formation of isotopically labeled Me3206Pb+ can be used for future production of this spike. The novel extraction technique, stir bar sorptive extraction (SBSE), was applied for the first time in connection with species-specific isotope dilution GC-ICP-MS for the determination of trimethyllead, methylmercury and butyltins. The results were compared with liquid-liquid extraction. The developed methods were validated by the analysis of certified reference materials. The liquid-liquid extraction GC-ICP-IDMS method was applied to seafood samples purchased from a supermarket. The methylated lead fraction in these samples, correlated to total lead, varied in a broad range of 0.01-7.6 %. On the contrary, the fraction of methylmercury is much higher, normally in the range of 80-98 %. The highest methylmercury content of up to 12 µg g-1 has been determined in shark samples, an animal which is at the end of the marine food chain, whereas in other seafood samples a MeHg+ content of less than 0.2 µg g-1 was found. Butyltin species could only be determined in samples, where anthropogenic contaminations must be assumed. This explains the observed broad variation of the butylated tin fraction in the range of <0.3-49 % in different seafood samples. Because all isotope-labelled spike compounds, except trimethyllead, are commercially available, the developed multi-species-specific GC-ICP-IDMS method has a high potential in future for routine analysis.
