We report herein the successful long term engraftment of highly purified hematopoietic stem cells (HSCs) without any facilitating cells in fully allogeneic recipient mice across the entire major histocompatibility complex (MHC) transplantation barrier. This finding challenges the assumption that highly purified marrow HSCs alone cannot produce long-lived allogeneic bone marrow chimeras across the MHC barrier. In the present experiments, 1 × 105 HSCs from 5-fluorouracil (5-FU)-treated donors, without any facilitating cells, have been found to repopulate lethally irradiated fully allogeneic recipients. Low density, lineage-negative (CD4−, CD8−, B220−, Mac-1−, Gr-1−), CD71-negative, class I highly positive, FACS-sorted cells from 5-FU-treated C57BL/6 (B6) donor mice were transplanted into lethally irradiated BALB/c recipients. (BALB/c → BALB/c) → BALB/c T cell-depleted marrow cells used as compromised cells were also transplanted into the recipients to permit experiments to be pursued over a long period of time. Cells of donor origin in all recognized lineages of hematopoietic cells developed in these allogeneic chimeras. One thousand HSCs were sufficient to repopulate hemiallogeneic recipients, but 1 × 104 HSCs alone from 5-FU-treated donors failed to repopulate the fully allogeneic recipients. Transplantation of primary marrow stromal cells or bones of the donor strain into recipient, together with 1 × 104 HSCs, also failed to reconstitute fully allogeneic recipients. Suppression of resistance of recipients by thymectomy or injections of granulocyte colony-stimulating factor before stem cell transplantation enhanced the engraftment of allogeneic HSCs. Our experiments show that reconstitution of all lymphohematopoietic lineages across the entire MHC transplantation barriers may be achieved by transplanting allogeneic HSCs alone, without any facilitating cells, as long as a sufficient number of HSCs is transplanted.

We are developing quantitative assays to repeatedly and noninvasively image expression of reporter genes in living animals, using positron emission tomography (PET). We synthesized positron-emitting 8-[18F]fluoroganciclovir (FGCV) and demonstrated that this compound is a substrate for the herpes simplex virus 1 thymidine kinase enzyme (HSV1-TK). Using positron-emitting FGCV as a PET reporter probe, we imaged adenovirus-directed hepatic expression of the HSV1-tk reporter gene in living mice. There is a significant positive correlation between the percent injected dose of FGCV retained per gram of liver and the levels of hepatic HSV1-tk reporter gene expression (r2 > 0.80). Over a similar range of HSV1-tk expression in vivo, the percent injected dose retained per gram of liver was 0–23% for ganciclovir and 0–3% for FGCV. Repeated, noninvasive, and quantitative imaging of PET reporter gene expression should be a valuable tool for studies of human gene therapy, of organ/cell transplantation, and of both environmental and behavioral modulation of gene expression in transgenic mice.

V(D)J recombination is thought to be regulated by changes in the accessibility of target sites, such as modulation of methylation. To test whether demethylation of the kappa locus can activate recombination, we generated two recombinationally active B cell lines in which the gene for maintenance of genomic DNA methylation, Dnmt1, was flanked with loxP sites. Transduction with a retrovirus expressing both the cre recombinase and green fluorescent protein allowed us to purify recombinationally active cells devoid of methylation. Loss of methylation of the kappa locus was not sufficient to activate recombination, although transcription was activated in one line. It appears that demethylation of the kappa locus is not the rate-limiting step for altering accessibility and thus regulated demethylation does not generate specificity of recombination.

Upon the completion of the Saccharomyces cerevisiae genomic sequence in 1996 [Goffeau,A. et al. (1997) Nature, 387, 5], several creative and ambitious projects have been initiated to explore the functions of gene products or gene expression on a genome-wide scale. To help researchers take advantage of these projects, the Saccharomyces Genome Database (SGD) has created two new tools, Function Junction and Expression Connection. Together, the tools form a central resource for querying multiple large-scale analysis projects for data about individual genes. Function Junction provides information from diverse projects that shed light on the role a gene product plays in the cell, while Expression Connection delivers information produced by the ever-increasing number of microarray projects. The Stanford Microarray Database (SMD) stores raw and normalized data from microarray experiments, and provides web interfaces for researchers to retrieve, analyze and visualize their data. The two immediate goals for SMD are to serve as a storage site for microarray data from ongoing research at Stanford University, and to facilitate the public dissemination of that data once published, or released by the researcher. Of paramount importance is the connection of microarray data with the biological data that pertains to the DNA deposited on the microarray (genes, clones etc.). The flavoprotein (R)-(+)-mandelonitrile lyase (MDL; EC 4.1.2.10), which plays a key role in cyanogenesis in rosaceous stone fruits, occurs in black cherry (Prunus serotina Ehrh.) homogenates as several closely related isoforms. Biochemical and molecular biological methods were used to investigate MDL microheterogeneity and function in this species. Three novel MDL cDNAs of high sequence identity (designated MDL2, MDL4, and MDL5) were isolated. Like MDL1 and MDL3 cDNAs (Z. Hu, J.E. Poulton [1997] Plant Physiol 115: 1359–1369), they had open reading frames that predicted a flavin adenine dinucleotide-binding site, multiple N-glycosylation sites, and an N-terminal signal sequence. The N terminus of an MDL isoform purified from seedlings matched the derived amino acid sequence of the MDL4 cDNA. Genomic sequences corresponding to the MDL1, MDL2, and MDL4 cDNAs were obtained by polymerase chain reaction amplification of genomic DNA. Like the previously reported mdl3 gene, these genes are interrupted at identical positions by three short, conserved introns. Given their overall similarity, we conclude that the genes mdl1, mdl2, mdl3, mdl4, and mdl5 are derived from a common ancestral gene and constitute members of a gene family. Genomic Southern-blot analysis showed that this family has approximately eight members. Northern-blot analysis using gene-specific probes revealed differential expression of the genes mdl1, mdl2, mdl3, mdl4, and mdl5.

O auto sombreamento das folhas posicionadas nas porções inferiores do dossel de plantas pode limitar a produtividade em cultivos tutorados. Assim, a produtividade do tomateiro pode ser aumentada por meio da suplementação luminosa posicionada no interior do dossel, técnica conhecida como interlighting. O sistema de condução do tomateiro também interfere na distribuição da radiação solar, além de afetar os tratos culturais, a competição intra e entre plantas e a relação entre as partes vegetativas e reprodutivas. Desta forma, o objetivo deste trabalho foi avaliar a influência do sistema de condução de minitomate cultivar \'Sweet Grape\' em diferentes números de hastes por planta (duas, três e quatro hastes) e da suplementação luminosa com módulos de LED na produtividade e qualidade dos frutos, na eficiência do uso de água e nutrientes, além da morfologia das plantas e fatores relacionados ao manejo cultural, em ambiente protegido nas condições climáticas do sudeste brasileiro. Ao longo do ciclo de cultivo foram avaliados os seguintes parâmetros: volume irrigado, pH, condutividade elétrica, porcentagem e volume da solução drenada pelos vasos. As colheitas foram realizadas semanalmente, a partir de 90 dias após o transplante. Os frutos colhidos foram classificados, contados e pesados para a obtenção do número e massa de frutos grandes, médios, pequenos, comercial, não comercial e total. Amostras de frutos e tecidos foliares foram coletadas em cinco períodos e avaliadas quanto ao teor de sólidos solúveis, pH, acidez titulável e concentração de ácido ascórbico nos frutos e teor de nutrientes nas folhas. Além disso, ao final do ciclo cultural, foram realizadas as medições dos seguintes parâmetros morfológicos nas plantas: comprimento de hastes, número de cachos normais e bifurcados por hastes e diâmetro apical, mediano e basal das hastes. A suplementação luminosa apresentou aumento no número e na massa de frutos grandes e médios, elevando a produtividade total em 12%. Plantas com duas e três hastes apresentaram maior acúmulo de massa total de frutos, porém plantas com três hastes apresentaram maior massa de frutos não comerciais, com redução na massa de frutos comerciais. A maior eficiência no uso de água e nutrientes foi alcançada em plantas cultivadas com duas hastes. Plantas com quatro hastes demandaram mais solução nutritiva comparada às plantas com duas e três hastes. Esta maior demanda de solução, acarretou em aumento da condutividade elétrica da solução drenada. Plantas com quatro hastes apresentaram maiores teores de sólidos solúveis nos frutos. A suplementação luminosa também resultou em aumento do teor de sólidos solúveis e ligeiro aumento no teor de ácido ascórbico nos frutos. A suplementação luminosa favoreceu o acúmulo de nitrogênio, fósforo e potássio nas folhas do tomateiro. Desta forma, conclui-se que a suplementação luminosa é uma estratégia de manejo tecnicamente viável nas condições climática estudada. O sistema de condução de haste afeta a produtividade e qualidade dos frutos do tomateiro. Plantas com duas hastes além de apresentar maior produtividade de frutos comerciais, mostrou-se a estratégia mais eficiente no uso da água e nutrientes.

Discovering a History: The School of Art at the University of Denver explores the early history of art education in Denver, and the significance of visual art education at the University of Denver within that history beginning in 1865, when the first classes in art were offered, and ending in 1929 when the University acquired the Chappell School of Art—an independent art school—and appointed Vance Kirkland as director. This paper also explores competing art institutions, which at times posed great hindrances to the University. Further, it illustrates how the artists who taught at the University of Denver School of Art, such as Ida De Steiguer, Preston Powers, Emma Richardson Cherry, and Henry Read, were amongst the great contributors to Denver’s burgeoning artistic culture.

This layer is a georeferenced raster image of the historic paper map entitled: Map of Philadelphia, Camden and vicinity : compiled from city plans & personal surveys, engraved by Albert Volk. It was published by Elvino V. Smith in 1921. Scale 1:35,000. The image inside the map neatline is georeferenced to the surface of the earth and fit to the Pennsylvania South State Plane Coordinate System NAD83 (in Feet) (Fipszone 3702). All map collar and inset information is also available as part of the raster image, including any inset maps, profiles, statistical tables, directories, text, illustrations, index maps, legends, or other information associated with the principal map. This map shows features such as roads, railroads, drainage, county, township, and city ward boundaries, parks, cemeteries, and more. This layer is part of a selection of digitally scanned and georeferenced historic maps from The Harvard Map Collection as part of the Imaging the Urban Environment project. Maps selected for this project represent major urban areas and cities of the world, at various time periods. These maps typically portray both natural and manmade features at a large scale. The selection represents a range of regions, originators, ground condition dates, scales, and purposes.

Compiled from 1:24,000 scale maps of Sable, Derby, Arvada, Fort Logan, Englewood, Fitzsimons, Parker, Highlands Ranch, and Littleton 1957 7.5 minute quadrangles. The image inside the map neatline is georeferenced to the surface of the earth and fit to the State Plane Colorado Central NAD 1927 coordinate system (in Feet) (Fipszone 0502). All map collar and inset information is also available as part of the raster image, including any inset maps, profiles, statistical tables, directories, text, illustrations, index maps, legends, or other information associated with the principal map. This is a typical topographic map portraying both natural and manmade features. It shows and names works of nature, such as mountains, valleys, lakes, rivers, vegetation, etc. It also identify the principal works of humans, such as roads, railroads, boundaries, transmission lines, major buildings, etc. Relief is shown with standard contour intervals of 10 feet. This layer is part of a selection of digitally scanned and georeferenced historic maps from The Harvard Map Collection as part of the Imaging the Urban Environment project. Surveying the landscape following the Swiss referendum on February 9th, Adam Łazowski observes that once Swiss voters are deprived of the benefits of the EU internal market, they may come to appreciate that their days of cherry-picking from among EU policies are over. This might present the EU with a golden opportunity to press for a comprehensive framework agreement with Switzerland that would simplify the existing regime and provide for a uniform institutional set-up. He concludes, however, that what both sides cannot avoid is a frank discussion about free movement of persons, noting that that dossier will be crucial for any future steps that will be taken by the EU and the Swiss authorities.

Once the West’s ally, Turkey has been an ever more problematic partner in recent years. The Turkish leadership no longer views the alliance with the European Union and membership in NATO as based on shared values; rather, it is now merely a cherry-picked and shaky community of interests. Turkey is also increasingly alienated politically in the Middle East. In the aftermath of the Arab Spring and the regional developments which followed, Ankara has lost much of the influence it had built in region in previous years. Turkey’s growing international isolation is a consequence of the country ever more fully subordinating its foreign policy to the ideology of the ruling AKP. The world vision offered by that ideology does not square with the diagnoses of Turkey’s partners. The objectives it sets for Turkish foreign policy are incompatible with its partners’ expectations. Moreover, a foreign policy rooted in ideology is less flexible and less capable of adjusting to current international dynamics.
