Investigación producida a partir de una estancia en el Departamento de Agricultura de los Estados Unidos (United States Department of Agricultura - USDA), California, USA, entre el 1 de Julio y el 30 de Septiembre del 2005. Las coberturas comestibles están siendo consideradas en frutas frescas cortadas, como una estrategia para reducir los efectos perjudiciales que inflige el proceso mínimo en los tejidos vegetales. El empleo de puré de frutas constituye una excelente opción para la formación de recubrimientos o películas comestibles, ya que es una matriz constituida por polisacáridos primarios (sustancias pépticas y celulósicas) con propiedades de permeabilidad a la película y plastificantes gracias a los azúcares presentes en las frutas. Estas películas son buenas barreras contra el oxígeno en sistemas alimenticios de baja humedad, mejorando la calidad de los productos y extendiendo su vida útil. Se evalúa películas a base de puré de manzana como soporte de agentes antioxidantes y de aceites esenciales como antimicrobianos. Se estudia su permeabilidad al vapor de agua y al oxígeno, además de las propiedades de tensión, color y antimicrobianas.

Introduction. Critically ill patients suffer from oxidative stress caused by reactive oxygen species (ROS) and reactive nitrogen species (RNS). Although ROS/RNS are constantly produced under normal circumstances, critical illness can drastically increase their production. These patients have reduced plasma and intracellular levels of antioxidants and free electron scavengers or cofactors, and decreased activity of the enzymatic system involved in ROS detoxification. The pro-oxidant/antioxidant balance is of functional relevance during critical illness because it is involved in the pathogenesis of multiple organ failure. In this study the objective was to evaluate the relation between oxidative stress in critically ill patients and antioxidant vitamin intake and severity of illness. Methods. Spectrophotometry was used to measure in plasma the total antioxidant capacity and levels of lipid peroxide, carbonyl group, total protein, bilirubin and uric acid at two time points: at intensive care unit (ICU) admission and on day seven. Daily diet records were kept and compliance with recommended dietary allowance (RDA) of antioxidant vitamins (A, C and E) was assessed. Results. Between admission and day seven in the ICU, significant increases in lipid peroxide and carbonyl group were associated with decreased antioxidant capacity and greater deterioration in Sequential Organ Failure Assessment score. There was significantly greater worsening in oxidative stress parameters in patients who received antioxidant vitamins at below 66% of RDA than in those who received antioxidant vitamins at above 66% of RDA. An antioxidant vitamin intake from 66% to 100% of RDA reduced the risk for worsening oxidative stress by 94% (ods ratio 0.06, 95% confidence interval 0.010 to 0.39), regardless of change in severity of illness (Sequential Organ Failure Assessment score). Conclusion. The critical condition of patients admitted to the ICU is associated with worsening oxidative stress. Intake of antioxidant vitamins below 66% of RDA and alteration in endogenous levels of substances with antioxidant capacity are related to redox imbalance in critical ill patients. Therefore, intake of antioxidant vitamins should be carefully monitored so that it is as close as possible to RDA.

Reduction of the antioxidant capacity of plasma has been linked with the impairment of an effective immune response and so we hypothesized that the carriage rate of Neisseria meningitidis in asymptomatic subjects might correlate with the levels of antioxidants in plasma. To this end we took pharyngeal swabs from 339 children in Marquesado Basic Health Zone, Granada, Spain and in addition determined the total antioxidant capacity (TAC) in plasma samples from these subjects. The overall prevalence of N. meningitidis carriage was 5.9% (mean age 7.1 years) with rates of 10.3% in children aged 3 < or =years, 3.9% between 4 and 7 years and 2.4% in older subjects. Plasma TAC for the < or =3-year-olds was 0.13 for carriers and 1.10 for non-carrier controls (P=0.04), 0.13 for carriers aged 4-7 years (controls 0.63) and 0.28 for carriers aged >7 years (controls 0.52). We analysed the association between TAC in plasma (<0.37 - 2 S.D.) and the carrier state of N. meningitidis. In the carrier state, the odds ratio for this association (TAC in plasma <0.25) was 8.44 (95% CI 1.5-48.9). These findings may suggest a reduced immune response in the host favourable to nasopharyngeal persistence of meningococci.

High postprandial levels of TAG may further induce endothelial dysfunction and inflammation in subjects with high fasting levels of TAG, an effect that seems to be related to oxidative stress. The present study investigated whether minor compounds of olive oil with antioxidant activity decrease postprandial levels of soluble isoforms of intercellular adhesion molecule 1 (sICAM-1) and vascular cell adhesion molecule 1 (sVCAM-1), as surrogate markers of vascular inflammation, after a high-fat meal. A randomized crossover and blind trial on fourteen healthy and fourteen hypertriacylglycerolaemic subjects was performed. The study involved a 1-week adaptation lead-in period on a National Cholesterol Education Program Step I diet supplemented with extra-virgin olive oil (EVOO) containing 1125 mg polyphenols/kg and 350 mg tocopherols/kg, or refined olive oil (ROO) with no polyphenols or tocopherols. After a 12 h fast, the participants ate a high-fat meal enriched in EVOO or ROO (50 g/m2 body surface area), which on average provided 3700 kJ energy with a macronutrient profile of 72% fat, 22% carbohydrate and 6% protein. Blood samples drawn hourly over the following 8 h demonstrated a similar postprandial TAG response for both EVOO and ROO meals. However, in both healthy and hypertriacylglycerolaemic subjects the net incremental area under the curve for sICAM-1 and sVCAM-1 were significantly lower after the EVOO meal. In conclusion,the consumption of EVOO with a high content of minor antioxidant compounds may have postprandial anti-inflammatory protective effects.

The aim of this study was to examine the responses of uric acid, antioxidant defences and pro-oxidant variables after a high-fat meal. Twenty-five healthy persons without criteria for the metabolic syndrome, underwent a high-fat meal with Supracal (60 g fat). Measurements were made at baseline and 3 h after the meal of TAG, uric acid, HDL-cholesterol, total proteins and oxidative stress. Following the high-fat meal, we detected a significant increase in pro-oxidative variables and a decrease in antioxidative variables. The uric acid concentrations were significantly lower after the high-fat meal and the reduction correlated significantly with the oxidative stress variables. The inverse relation between reduced uric acid and increased carbonylated proteins remained in multiple regression analysis. We conclude that uric acid is a powerful antioxidant and its reduction following a high-fat meal may be related with its acute antioxidative action.

Plant-based whole foods provide thousands of bioactive metabolites to the human diet that reduce the risk of developing chronic diseases. β-Caryophyllene (CAR) is a common constituent of the essential oil of numerous plants, vegetables, fruits and medicinal herbs, and has been used as a flavouring agent since the 1930 s. Here, we report the antioxidant activity of CAR, its protective effect on liver fibrosis and its inhibitory capacity on hepatic stellate cell (HSC) activation. CAR was tested for the inhibition of lipid peroxidation and as a free radical scavenger. CAR had higher inhibitory capacity on lipid peroxidation than probucol, α-humulene and α-tocopherol. Also, CAR showed high scavenging activities against hydroxyl radical and superoxide anion. The activity of 5-lipoxygenase, an enzyme that actively participates in fibrogenesis, was significantly inhibited by CAR. Carbon tetrachloride-treated rats received CAR at 2, 20 and 200 mg/kg. CAR significantly improved liver structure, and reduced fibrosis and the expression of Col1a1, Tgfb1 and Timp1 genes. Oxidative stress was used to establish a model of HSC activation with overproduction of extracellular matrix proteins. CAR (1 and 10 μm) increased cell viability and significantly reduced the expression of fibrotic marker genes. CAR, a sesquiterpene present in numerous plants and foods, is as a natural antioxidant that reduces carbon tetrachloride-mediated liver fibrosis and inhibits hepatic cell activation.

Malnutrition affects 40-50% of patients with ear, nose and throat (ENT) cancer. The aim of this study was to assess changes induced by a specific nutritional supplement enriched with n-3 polyunsaturated fatty acids, fiber and greater amounts of proteins and electrolytes, as compared with a standard nutritional supplement, on markers of inflammation, oxidative stress and metabolic status of ENT cancer patients undergoing radiotherapy (RT). Fourteen days after starting RT, 26 patients were randomly allocated to one of two groups, 13 supplemented with Prosure, an oncologic formula enriched with n-3 polyunsaturated fatty acids, fiber and greater amounts of proteins and electrolytes (specific supplement), and 13 supplemented with Standard-Isosource (standard supplement). Patients were evaluated before RT, and 14, 28 and 90 days after starting RT. The results showed that there were no significant differences between the groups, but greater changes were observed in the standard supplement group, such as a decline in body mass index (BMI), reductions in hematocrit, erythrocyte, eosinophil and albumin levels, and a rise in creatinine and urea levels. We concluded that metabolic, inflammatory and oxidative stress parameters were altered during RT, and began to normalize at the end of the study. Patients supplemented with Prosure showed an earlier normalization of these parameters, with more favorable changes in oxidative stress markers and a more balanced evolution, although the difference was not significant.

Abstract A prospective 1-year follow-up study in ear, nose, and throat (ENT) cancer patients was carried out one year after radiotherapy to assess the effect of varying consumption of ω3 fatty acid according to whether they consumed more or less than the 50th percentile of ω3 fatty acids. Clinical, analytical, inflammatory (CRP and IL-6), and oxidative variables (TAC, GPx, GST, and SOD) were evaluated. The study comprised 31 patients (87.1% men), with a mean age of 61.3 ± 9.1 years. Hematological variables showed significant differences in the patients with a lower consumption of ω3 fatty acids. A lower mortality and longer survival were found in the group with ω3 fatty acid consumption ≥50th percentile but the differences were not significant. No significant difference was reached in toxicity, inflammation, and oxidative stress markers. The group with ω3 fatty acid consumption <50th percentile significantly experienced more hematological and immune changes.

The role of the induction of oxidative stress as the mechanism of action of many antitumor drugs is acquiring an increasing interest. In such cases, the antitumor therapy success may be conditioned by the antioxidants present in our own body, which can be synthesized de novo (endogenous) or incorporated through the diet and nutritional supplements (exogenous). In this paper, we have reviewed different aspects of antioxidants, including their classification, natural sources, importance in diet, consumption of nutritional supplements, and the impact of antioxidants on health. Moreover, we have focused especially on the study of the interaction between antioxidants and antitumor therapy, considering both radiotherapy and chemotherapy. In this regard, we found that the convenience of administration of antioxidants during cancer treatment still remains a very controversial issue. In general terms, antioxidants could promote or suppress the effectiveness of antitumor treatment and even protect healthy tissues against damage induced by oxidative stress. The effects may depend on many factors discussed in the paper. These factors should be taken into consideration in order to achieve precise nutritional recommendations for patients. The evidence at the moment suggests that the supplementation or restriction of exogenous antioxidants during cancer treatment, as appropriate, could contribute to improving its efficiency.

O objetivo deste estudo foi avaliar o efeito renoprotetor do fitoterápico Uncaria tomentosa sobre a lesão renal aguda isquêmica induzida pelo clampeamento dos pedículos renais de ratos. A hipóxia e a hipoperfusão geradas com a isquemia intensificam a produção de espécies reativas já presentes no processo inflamatório. Os resultados mostraram que a função renal avaliada pelo clearance de creatinina, a excreção de peróxidos urinários (FOX) e a excreção urinária de malondealdeído (TBARS) desses animais apresentou renoproteção induzida pela UT, provavelmente relacionada às suas atividades antioxidantes.

RESUMO Objetivo Avaliar ação renoprotetora dos flavonoides diosmina e hesperidina na prevenção da nefrotoxicidade da anfotericina B em modelo experimental com ratos. Método Ratos Wistar, adultos, machos foram distribuídos nos seguintes grupos: Salina; diosmina hesperidina (animais receberam 50 mg/kg de diosmina hesperidina em água de bebedouro por dez dias); Anfotericina B (animais receberam 15 mg/kg/dia de anfotericina B intraperitoneal por cinco dias); Anfotericina B+diosmina hesperidina. Foram avaliados função renal, fração de excreção de sódio, potássio e magnésio e os metabólitos oxidativos. Resultados O tratamento com anfotericina B reduziu a função renal, vista peloclearance de creatinina, elevou os marcadores de função tubular como a fração de excreção de sódio, potássio, magnésio e dos metabólitos oxidativos. O pré-condicionamento com diosmina hesperidina elevou o clearance de creatinina e atenuou da lesão tubular e oxidativa. Conclusão A administração de anfotericina B resultou no declínio da função renal com lesão tubular e a diosmina hesperidina demonstrou efeito renoprotetor antioxidante.

Con el objeto de validar una técnica para determinar la actividad sanguínea de glutatión peroxidasa (GSH-Px; EC 1.11.1.9) en el Laboratorio de Patología Clínica de la Universidad Austral de Chile y establecer la correlación entre su actividad y la concentración sanguínea y plasmática de selenio (Se) en bovinos a pastoreo en rebaños lecheros del sur de Chile, se tomaron 5-10 mL de sangre heparinizada a 112 vacas de ocho rebaños en la provincia de Valdivia. La actividad enzimática se analizó mediante una técnica cinética, y el Se por activación de neutrones. Fueron calculadas la inexactitud e imprecisión de la técnica cinética y se describen el rango, promedio y desviación estándar de la actividad enzimática. La correlación entre la actividad sanguínea de GSH-Px y la concentración de Se fue obtenida mediante el coeficiente de correlación simple. La inexactitud e imprecisión fueron 5,9% y 10%, respectivamente. La actividad de GSH-Px fue 89 ± 45 U/g de hemoglobina (Hb) y la correlación entre las variables señaladas fue r=0,97 (P<0,05). Según estos resultados, es posible recomendar el uso rutinario de la técnica descrita. La correlación señalada permite anotar que en bovinos a pastoreo la actividad de GSH-Px está relacionada con la concentración sanguínea de selenio.

O camu-camu [Myrciaria dubia (Humb., Bonpl. & Kunth) McVaugh], da família Myrtaceae, é encontrado em áreas inundáveis da Região Amazônica e utilizado como conservante em antioxidantes por seu alto teor de ácido ascórbico. O objetivo deste trabalho foi avaliar porta-enxertos desta família, adaptados a terra firme, visando à propagação vegetativa de camu-camu. Selecionaram-se duzentas e quarenta mudas de camu-camu, goiabeira (Psidium guajava L.) e pitangueira (Eugenia uniflora L.), que receberam quatro tipos de enxertia, originando doze tratamentos de sessenta plantas, com cinco repetições. Apenas o porta-enxerto de camu-camu se mostrou compatível. A incompatibilidade entre camu-camu e os porta-enxertos de goiabeira e pitangueira foi demonstrada por análises anatômicas.

El objetivo de este estudio fue determinar el balance metabólico nutricional de Cu, Zn, Fe, Mn y Se, mediante la actividad sanguínea de SOD y GSH-Px y establecer la relación entre la concentración de Cu, Fe, Mn y Zn en el forraje y la actividad de SOD. Se tomaron 10 mL de sangre a 105 novillas seleccionadas en 15 rebaños lecheros de Caldas, Colombia (5º4' N y 75º3' O) y se tomaron muestras de forrajes para analizar Cu, Fe, Mn y Zn. El promedio de la actividad de SOD fue 1.390±1.299 U g-1 Hb, y estaba correlacionada con Cu, Mn y Fe en el forraje. La actividad promedio de GSH-Px fue 389±184 U g-1 Hb y fue observada una mayor frecuencia de valores deficitarios y bajo/marginales de 9%, habiendo sido más afectados los animales de zonas altas (>2.000 msnm). Bajo estas condiciones, estos resultados permiten señalar que la SOD es una enzima que puede emplearse como indicador del balance metabólico nutricional de Cu, Mn y Fe en bovinos a pastoreo, no está clara todavía su relación con Zn. La actividad de GSH-Px indica deficiencias en el balance metabólico nutricional de Se, en bovinos a pastoreo.

O objetivo deste trabalho foi avaliar o crescimento inicial, acúmulo de biomassa, trocas gasosas e defesas antioxidativas de soja 'Tracajá', cultivada na Região Amazônica, exposta ao ozônio sob condições controladas. Sementes germinadas em vasos foram levadas para duas câmaras, uma com ar filtrado (AF) e outra com ar filtrado mais 30 ppb de ozônio (AF + O3). Aos 10 e 20 dias após a semeadura, as trocas gasosas, crescimento em altura e acúmulo de biomassa foram medidos; aos 20 dias após a semeadura, as defesas antioxidativas (ácido ascórbico e superóxido dismutase) foram analisadas. Aos 10 dias após a semeadura, a fotossíntese líquida, condutância estomática, transpiração, altura, área foliar e biomassa foram 16, 27, 11, 22, 29 e 18% menores, respectivamente, no tratamento AF + O3. Aos 20 dias após a semeadura, além dessas variáveis, comprimento da raiz, diâmetro do caule e razão raiz:parte aérea foram 10, 15 e 12% menores, respectivamente, apesar da concentração de ácido ascórbico e da atividade da superóxido dismutase terem aumentado. Plântulas de soja 'Tracajá' apresentam baixa tolerância à concentração de 30 ppb de ozônio.
