Ultimamente si stanno sviluppando tecnologie per rendere più efficiente la virtualizzazione a livello di sistema operativo, tra cui si cita la suite Docker, che permette di gestire processi come se fossero macchine virtuali. Inoltre i meccanismi di clustering, come Kubernetes, permettono di collegare macchine multiple, farle comunicare tra loro e renderle assimilabili ad un server monolitico per l'utente esterno. Il connubio tra virtualizzazione a livello di sistema operativo e clustering permette di costruire server potenti quanto quelli monolitici ma più economici e possono adattarsi meglio alle richieste esterne. Data l'enorme mole di dati e di potenza di calcolo necessaria per gestire le comunicazioni e le interazioni tra utenti e servizi web, molte imprese non possono permettersi investimenti su un server proprietario e la sua manutenzione, perciò affittano le risorse necessarie che costituiscono il cosiddetto "cloud", cioè l'insieme di server che le aziende mettono a disposizione dei propri clienti. Il trasferimento dei servizi da macchina fisica a cloud ha modificato la visione che si ha dei servizi stessi, infatti non sono più visti come software monolitici ma come microservizi che interagiscono tra di loro. L'infrastruttura di comunicazione che permette ai microservizi di comunicare è chiamata service mesh e la sua suddivisione richiama la tecnologia SDN. È stato studiato il comportamento del software di service mesh Istio installato in un cluster Kubernetes. Sono state raccolte metriche su memoria occupata, CPU utilizzata, pacchetti trasmessi ed eventuali errori e infine latenza per confrontarle a quelle ottenute da un cluster su cui non è stato installato Istio. Lo studio dimostra che, in un cluster rivolto all'uso in produzione, la service mesh offerta da Istio fornisce molti strumenti per il controllo della rete a scapito di una richiesta leggermente più alta di risorse hardware.

Negli ultimi anni si è sviluppato un crescente interesse per lo studio e il monitoraggio di fenomeni meteorologici estremi. In particolare, stanno aumentando osservazioni e testimonianze relative a manifestazioni temporalesche molto violente, spesso caratterizzate da grandine di notevole dimensione che rappresenta la causa principale di pericolo per le persone e di danni anche ingenti al comparto agricolo, infrastrutturale e alle automobili. Per questi motivi, in questo lavoro di tesi è stata presentata una nuova tecnica meteorologica denominata “Multi-sensor Approach for Satellite Hail Advection” (MASHA) che si basa sulla combinazione delle misure fornite dai satelliti geostazionari per trasportare l’informazione e da quelli in orbita polare per riconoscere le nubi grandinigene. I risultati della tecnica MASHA forniscono ogni cinque minuti su tutto il bacino del Mediterraneo la probabilità che la nube osservata abbia formato della grandine in un intervallo compreso tra 0 e 1. Le potenzialità del metodo MASHA riguardano soprattutto l’elevata risoluzione spaziale e temporale con la quale vengono riprodotti i sistemi grandinigeni, la capacità del modello di ricostruire i campi di grandine e l’evoluzione di questi sistemi anche in assenza di passaggi recenti dei satelliti polari. Per migliorare le performance della tecnica, è stato sviluppato un nuovo indice di severità chiamato Hail Severity Index (HSI) in grado di migliorare la localizzazione e la stima dell’intensità dei sistemi grandinigeni attraverso l’integrazione di nuove variabili: l’overshooting index, i fulmini e i dati radar. Le performance della tecnica MASHA e le potenzialità dell’indice HSI mostrate in questo lavoro di tesi, suggeriscono implementazioni operative a supporto di sistemi per il nowcasting di eventi temporaleschi particolarmente severi. Questo approccio, potrebbe essere molto efficace per fornire informazioni in tempo reale ai servizi meteorologici e nel contesto della protezione civile.

L’importanza sempre crescente dell’elettronica è accompagnata da una crescente necessità di compattezza ed efficienza energetica. Una parte sostanziale del costo e delle dimensioni dei moderni dispositivi elettronici è legata ai sistemi di conversione di potenza, il cui volume è dominato dai passivi. Per poter affrontare la miniaturizzazione dei circuiti elettronici di potenza sono dunque necessari metodi di design e tecnologie che permettano di ridurre i requisiti di immagazzinamento di energia. Un possibile approccio è aumentare la frequenza di commutazione nel range delle decine di MHz facendo in modo che l’efficienza non venga penalizzata dall’aumento delle perdite in commutazione. Ciò è reso possibile dall’impiego di topologie di convertitori risonanti che implementano la condizione ZVS. Oltre all’impiego di convertitori risonanti, l’aumento della frequenza operativa, mantenendo elevata l’efficienza, è abilitato dall’impiego di dispositivi a semiconduttore a largo band-gap come il nitruro di gallio (GaN), i quali mostrano performance superiori al silicio in termini di temperature operative, frequenze di funzionamento e densità di potenza. Inoltre, ad elevate frequenze di commutazione, l’utilizzo di magnetici coreless diventa una valida alterativa ai magnetici tradizionali, con vantaggi in termini di costo, ingombro e di efficienza. Il focus di questa tesi è il progetto di un convertitore DC-DC risonante con isolamento coreless ad alta efficienza e ad alta frequenza in tecnologia GaN a 650 V pensato per applicazioni wall-adapter. A seguito dello studio di alcune topologie di inverter risonanti e dei rispettivi rettificatori, si è scelta la topologia phi2 per il design del convertitore DC-DC double phi2 isolato (simulato con LTspice). È stato poi effettuato il design di un trasformatore coreless su PCB tramite simulatore elettromagnetico (ADS Keysight Momentum). Il convertitore complessivo presenta un’efficienza del 95,8% con una efficienza del link del 98%.

Nel presente progetto di tesi sono state preparate e studiate delle miscele polimeriche di poli(ottilene 2,5-furanoato) (POF) e il poli(trietilene 2,5-furanoato) (PTEF) a diversa composizione allo scopo di ottenere materiali adatti al packaging alimentare flessibile e sostenibile. I due omopolimeri sono stati sintetizzati mediante policondensazione in massa, ed in seguito sono state preparate le miscele fisiche per solvent casting, processate in forma di film per pressofusione. Dopo una completa caratterizzazione molecolare, i materiali realizzati sono stati caratterizzati morfologicamente (SEM), termicamente (TGA e DSC), strutturalmente (WAXS), meccanicamente (prove a trazione), e ne sono studiate le proprietà barriera. Le miscele sono risultate immiscibili ma comunque caratterizzate da buona compatibilità e stabilità termica. Considerando che il POF è un materiale gommoso e semicristallino mentre il PTEF è gommoso ma amorfo, le miscele presentano caratteristiche meccaniche coerenti con la loro composizione: in particolare, al diminuire della porzione cristallizabile POF e all’aumentare della porzione gommosa PTEF, si registra un progressivo aumento degli allungamenti a rottura e una diminuzione del modulo elastico. Inoltre, nelle blend contenenti il 50 e il 60% di PTEF, le performance barriera risultano migliori di quasi due ordini di grandezza rispetto a quelle dell’omopolimero POF. Per quanto riguarda le prove di compostaggio, le miscele con una quantità di POF superiore al 50% non subiscono degradazione, mentre si registra una totale compostabilità per le due miscele più ricche in PTEF.

The current dominance of African runners in long-distance running is an intriguing phenomenon that highlights the close relationship between genetics and physical performance. Many factors in the interesting interaction between genotype and phenotype (eg, high cardiorespiratory fitness, higher hemoglobin concentration, good metabolic efficiency, muscle fiber composition, enzyme profile, diet, altitude training, and psychological aspects) have been proposed in the attempt to explain the extraordinary success of these runners. Increasing evidence shows that genetics may be a determining factor in physical and athletic performance. But, could this also be true for African long-distance runners? Based on this question, this brief review proposed the role of genetic factors (mitochondrial deoxyribonucleic acid, the Y chromosome, and the angiotensin-converting enzyme and the alpha-actinin-3 genes) in the amazing athletic performance observed in African runners, especially the Kenyans and Ethiopians, despite their environmental constraints.

A rapid, sensitive and specific method for quantifying propylthiouracil in human plasma using methylthiouracil as the internal standard (IS) is described. The analyte and the IS were extracted from plasma by liquid-liquid extraction using an organic solvent (ethyl acetate). The extracts were analyzed by high performance liquid chromatography coupled with electrospray tandem mass spectrometry (HPLC-MS/MS) in negative mode (ES-). Chromatography was performed using a Phenomenex Gemini C18 5μm analytical column (4.6mm×150mm i.d.) and a mobile phase consisting of methanol/water/acetonitrile (40/40/20, v/v/v)+0.1% of formic acid. For propylthiouracil and I.S., the optimized parameters of the declustering potential, collision energy and collision exit potential were -60 (V), -26 (eV) and -5 (V), respectively. The method had a chromatographic run time of 2.5min and a linear calibration curve over the range 20-5000ng/mL. The limit of quantification was 20ng/mL. The stability tests indicated no significant degradation. This HPLC-MS/MS procedure was used to assess the bioequivalence of two propylthiouracil 100mg tablet formulations in healthy volunteers of both sexes in fasted and fed state. The geometric mean and 90% confidence interval CI of Test/Reference percent ratios were, without and with food, respectively: 109.28% (103.63-115.25%) and 115.60% (109.03-122.58%) for Cmax, 103.31% (100.74-105.96%) and 103.40% (101.03-105.84) for AUClast. This method offers advantages over those previously reported, in terms of both a simple liquid-liquid extraction without clean-up procedures, as well as a faster run time (2.5min). The LOQ of 20ng/mL is well suited for pharmacokinetic studies. The assay performance results indicate that the method is precise and accurate enough for the routine determination of the propylthiouracil in human plasma. The test formulation with and without food was bioequivalent to reference formulation. Food administration increased the Tmax and decreased the bioavailability (Cmax and AUC).

The aim of this study was to evaluate the performance of the Centers for Dental Specialties (CDS) in the country and associations with sociodemographic indicators of the municipalities, structural variables of services and primary health care organization in the years 2004-2009. The study used secondary data from procedures performed in the CDS to the specialties of periodontics, endodontics, surgery and primary care. Bivariate analysis by χ2 test was used to test the association between the dependent variable (performance of the CDS) with the independents. Then, Poisson regression analysis was performed. With regard to the overall achievement of targets, it was observed that the majority of CDS (69.25%) performance was considered poor/regular. The independent factors associated with poor/regular performance of CDS were: municipalities belonging to the Northeast, South and Southeast regions, with lower Human Development Index (HDI), lower population density, and reduced time to deployment. HDI and population density are important for the performance of the CDS in Brazil. Similarly, the peculiarities related to less populated areas as well as regional location and time of service implementation CDS should be taken into account in the planning of these services.

To examine the influence of l-arginine supplementation in combination with physical training on mitochondrial biomarkers from gastrocnemius muscle and its relationship with physical performance. Male Wistar rats were divided into four groups: control sedentary (SD), sedentary supplemented with l-arginine (SDLA), trained (TR) and trained supplemented with l-arginine (TRLA). Supplementation of l-arginine was administered by gavage (62.5mg/ml/day/rat). Physical training consisted of 60min/day, 5days/week, 0% grade, speed of 1.2km/h. The study lasted 8weeks. Skeletal muscle mitochondrial enriched fraction as well as cytoplasmic fractions were obtained for Western blotting and biochemical analyses. Protein expressions of transcriptor coactivator (PGC-1α), transcriptor factors (mtTFA), ATP synthase subunit c, cytochrome oxidase (COXIV), constitutive nitric oxide synthases (eNOS and nNOS), Cu/Zn-superoxide dismutase (SOD) and manganese-SOD (Mn-SOD) were evaluated. We also assessed in plasma: lipid profile, glycemia and malondialdehyde (MDA) levels. The nitrite/nitrate (NOx(-)) levels were measured in both plasma and cytosol fraction of the gastrocnemius muscle. 8-week l-arginine supplementation associated with physical training was effective in promoting greater tolerance to exercise that was accompanied by up-regulation of the protein expressions of mtTFA, PGC-1α, ATP synthase subunit c, COXIV, Cu/Zn-SOD and Mn-SOD. The upstream pathway was associated with improvement of NO bioavailability, but not in NO production since no changes in nNOS or eNOS protein expressions were observed. This combination would be an alternative approach for preventing cardiometabolic diseases given that in overt diseases a profound impairment in the physical performance of the patients is observed.

Objective Adapt the 6 minutes walking test (6MWT) to artificial gait in complete spinal cord injured (SCI) patients aided by neuromuscular electrical stimulation. Method Nine male individuals with paraplegia (AIS A) participated in this study. Lesion levels varied between T4 and T12 and time post injured from 4 to 13 years. Patients performed 6MWT 1 and 6MWT 2. They used neuromuscular electrical stimulation, and were aided by a walker. The differences between two 6MWT were assessed by using a paired t test. Multiple r-squared was also calculated. Results The 6MWT 1 and 6MWT 2 were not statistically different for heart rate, distance, mean speed and blood pressure. Multiple r-squared (r2 = 0.96) explained 96% of the variation in the distance walked. Conclusion The use of 6MWT in artificial gait towards assessing exercise walking capacity is reproducible and easy to apply. It can be used to assess SCI artificial gait clinical performance.

The pathological mechanisms underlying cognitive dysfunction in multiple sclerosis (MS) are not yet fully understood and, in addition to demyelinating lesions and gray-matter atrophy, subclinical disease activity may play a role. To evaluate the contribution of asymptomatic gadolinium-enhancing lesions to cognitive dysfunction along with gray-matter damage and callosal atrophy in relapsing-remitting MS (RRMS) patients. Forty-two treated RRMS and 30 controls were evaluated. MRI (3T) variables of interest were brain white-matter and cortical lesion load, cortical and deep gray-matter volumes, corpus callosum volume and presence of gadolinium-enhancing lesions. Outcome variables included EDSS, MS Functional Composite (MSFC) subtests and the Brief Repeatable Battery of Neuropsychological tests. Cognitive dysfunction was classified as deficits in two or more cognitive subtests. Multivariate regression analyses assessed the contribution of MRI metrics to outcomes. Patients with cognitive impairment (45.2%) had more cortical lesions and lower gray-matter and callosal volumes. Patients with subclinical MRI activity (15%) had worse cognitive performance. Clinical disability on MSFC was mainly associated with putaminal atrophy. The main independent predictors for cognitive deficits were high burden of cortical lesions and number of gadolinium-enhancing lesions. Cognitive dysfunction was especially related to high burden of cortical lesions and subclinical disease activity. Cognitive studies in MS should look over subclinical disease activity as a potential contributor to cognitive impairment.

Synthetic dyes are much used in processed foods. HPLC was applied to different types of snacks, such as colored cereals, chocolate confetti, chewing gums and candies for the determination of those additives. In the case of artificially colored breakfast cereals, 71% of the samples exceeded the allowed limits. Regarding the portions recommended for consumption by the makers of two of the samples, the amounts exceeded those allowed by the Brazilian legislation. In the case of chocolate confetti and candies none of the samples showed higher amounts than those allowed. However 37% of the chewing gum samples presented larger contents than the authorized ones, and one sample contained five times more synthetic dyes than allowed.

The high cost of sensitivity commercial calorimeters may represent an obstacle for many calorimetric research groups. This work describes the construction and calibration of a batch differential heat conduction calorimeter with sample cells volumes of about 400 μL. The calorimeter was built using two small high sensibility square Peltier thermoelectric sensors and the total cost was estimated to be about US$ 500. The calorimeter was used to study the excess enthalpy of solution of binary mixtures of liquids, as a function of composition, for the following binary systems of solvents: water + 1,4-dioxane or + dimethylsulfoxide at 298,2 ± 0,5 K.

This paper presents the behavior of three bored piles conducted in diabasic soil submitted to uplift forces. The piles were built at the site for Experimental Studies in Soil Mechanics and Foundations of UNICAMP, located in the city of Campinas, Brazil. Field tests have already been conducted at the site (SPT, CPT, DMT and PMT), as well as laboratory tests by using sample soils taken from a well up to 17 m deep. The water table is not checked until a depth of 17 m. In order to check the behavior of the piles when submitted to uplift forces, slow static load tests were carried out as the recommendations of NBR 12131. The carrying capacity of these piles was also provided by means of theoretical methods, appropriate for uplift forces, and through semi-empirical methods appropriate for compression forces, considering only the portion of lateral resistance. The values estimated by using the considered methods were compared to those obtained by means of load tests. One of the tested piles was extracted from the soil to be the subject of a study on its geometry.

Remote sensing data are each time more available and can be used to monitor the vegetal development of main agricultural crops, such as the Arabic coffee in Brazil, since that the relationship between spectral and agronomical data be well known. Therefore, this work had the main objective to assess the use of Quickbird satellite images to estimate biophysical parameters of coffee crop. Test area was composed by 25 coffee fields located between the cities of Ribeirão Corrente, Franca and Cristais Paulista (SP), Brazil, and the biophysical parameters used were row and between plants spacing, plant height, LAI, canopy diameter, percentage of vegetation cover, roughness and biomass. Spectral data were the reflectance of four bands of QUICKBIRD and values of four vegetations indexes (NDVI, GVI, SAVI and RVI) based on the same satellite. All these data were analyzed using linear and nonlinear regression methods to generate estimation models of biophysical parameters. The use of regression models based on nonlinear equations was more appropriate to estimate parameters such as the LAI and the percentage of biomass, important to indicate the productivity of coffee crop.

cDNA arrays are a powerful tool for discovering gene expression patterns. Nylon arrays have the advantage that they can be re-used several times. A key issue in high throughput gene expression analysis is sensitivity. In the case of nylon arrays, signal detection can be affected by the plastic bags used to keep membranes humid. In this study, we evaluated the effect of five types of plastics on the radioactive transmittance, number of genes with a signal above the background, and data variability. A polyethylene plastic bag 69 μm thick had a strong shielding effect that blocked 68.7% of the radioactive signal. The shielding effect on transmittance decreased the number of detected genes and increased the data variability. Other plastics which were thinner gave better results. Although plastics made from polyvinylidene chloride, polyvinyl chloride (both 13 μm thick) and polyethylene (29 and 7 μm thick) showed different levels of transmittance, they all gave similarly good performances. Polyvinylidene chloride and polyethylene 29 mm thick were the plastics of choice because of their easy handling. For other types of plastics, it is advisable to run a simple check on their performance in order to obtain the maximum information from nylon cDNA arrays.
