Selected parts of ten frozen core samples from Holes 482A, 482B, 483A, and 485A, Leg 65 of the Deep Sea Drilling Project (DSDP), were analyzed for residual carbohydrates in order to determine the provenance and history of the organic material in the sediments. The samples, which represented silty-clay, shale, and nannofossil- chalk sediments, were analyzed for water-soluble monosaccharides, acid-soluble monosaccharides, and for starch and cellulose. Most samples yielded positive results for acid-extractable (polymeric) arabinose, fucose, xylose, mannose, galactose, and glucose. Amylose was detected in seven of the samples, whereas cellulose was found in only one. Possible explanations for the relatively high levels of free sugars are suggested in the conclusions to this chapter.

Fours sets of PM10 samples were collected in three sites in SEQ from December 2002 to August 2004. Three of these sets of samples were collected by QLD EPA as a part of their regular air monitoring program at Woolloongabba, Rocklea and Eagle Farm. Half of the samples were used in this study for the analysis of water-soluble ions, which are Na+, K+, Mg2+, Ca2+, NH4 +, Cl-, NO3 -, SO4 2-, F-, Br-, NO2 -, PO4 -3 and the other half was retained by QLD EPA. The fourth set of samples was collected at Rocklea, specifically for this study. A quarter of the samples obtained from this set of samples were used to analyse water-soluble ions; a quarter of the sample was used to analyse Pb, Cu, Al, Fe, Mn and Zn; and the rests were used to analyse US EPA 16 priority PAHs. The water-soluble ions were extracted ultrasonically with water and the major watersoluble anions as well as NH4 + were analysed using IC. Na+, K+, Mg2+, Ca2+ Pb, Cu, Al, Fe, Mn and Zn were analysed using ICP-AES while PAHs were extracted by acetonitrile and analysed using HPLC. Of the analysed water-soluble ions, Cl-, NO3 -, SO4 2-, Na+, K+, Mg2+ and Ca2+ were high in concentration and determined in all the samples. F-, Br-, NO2 -, PO4 -3 and NH4 + ions were lower in concentration and determined only in some samples. Na+ and Cl- were high in all samples indicating the importance of a marine source. Principal Component Analysis (PCA) was used to examine the temporal variations of the water-soluble ions at the three sites. The results indicated that there was no major difference between the three sites. However, comparing the average concentrations of ions and Cl-/Na+ it was concluded that Woolloongabba had more marine influence than the other sites. Al, Fe and Zn were detected in all samples. Al and Fe were high in all samples indicating the significance of a source of crustal matter. Cu, Mn and Pb were in low concentrations and were determined only in some samples. The lower Pb concentrations observed in the study than in previous studies indicate that the phasing-out of leaded petrol had an appreciable impact on Pb levels in SEQ. This study reports for the first time, simultaneous data on the water-soluble, metal ion and PAH levels of PM10 aerosols in Brisbane, and provides information on the most likely sources of these chemical species. Such information can be used alongside those that already exist to formulate PM10 pollution reduction strategies for SEQ in order to protect the community from the adverse effects of PM pollution.

Fully atomistic molecular dynamics simulations have been carried out to investigate the correlation of biological activity with dynamics of water molecules in an aqueous protein solution of the toxic domain of enterotoxin (PDB ID: 1ETN). This is a small protein of 13 amino acid residues. Our study of this water soluble protein clearly reveals that water dynamics slows down in the hydration layer. Despite this general slowing down, water molecules in the vicinity of the second beta turn of this protein exhibit faster dynamics than those near other regions of the protein. Since this beta turn is believed to play a critical role in the receptor binding of this protein, the faster dynamics of water near the beta turn m ay have biological significance. The collective orientational dynamics of the water molecules in the protein solution exhibits a characteristic long time component of 27 ps, which agrees well with dielectric relaxation experiments.

Solar distillation can be used to produce potable water from contaminated water. However, studies show that ions such as F(-) and NO(3)(-) occur in distillates from solar stills. In order to understand the reasons for this behavior, imaging and distillation experiments were conducted. White dots were seen in the vapor space above the interface of hot water poured into containers. The concentrations of various ions such as F(-) and SO(4)(2-) in the distillates from thermal and solar distillation experiments were roughly comparable when the feed consisted of deionized water and also solutions having fluoride concentrations of 100 and 10 000 mg/L. These observations suggest that aerosols enter the distillation setup through leaks and provide nuclei for the condensation of water vapor. The water-soluble component of aerosols dissolves in the drops formed, and some of the drops are transferred to the distillate by buoyancy-driven convection.

A new carbazole-based tetraimidazole ligand 1,3,6,8-tetra(1H-imidazol-1-yl)-9-methyl-9H-carbazole (L) has been synthesized. The unsymmetrical nature of L as well as the rotational freedom of imidazole donor moieties around C-N bond make it a special building unit, which upon treatment with cis-(tmeda)Pd(NO3)(2) produced an unprecedented single linkage-isomeric Pd-8 tetrafacial molecular nanobarrel (PSMBR-1) tmeda N,N,N',N'-tetramethylethane-1,2-diamine]. Unlike closed architectures, open barrel architecture of water-soluble PSMBR-1 makes it an ideal host for some water insoluble polyaromatic hydrocarbons in aqueous medium; one such inclusion complex coroneneCPSMBR-1 was characterized by X-ray diffraction study. Moreover, the potential application of PSMER-1 as carrier in aqueous medium for the transportation of water insoluble fluorophore (perylene) for live cell imaging is explored.

A study on self-assembly of anisotropically substituted penta-aryl fullerenes in water has been reported. The penta-phenol-substituted amphiphilic fullerene derivative C60Ph5(OH)(5)],exhibited self-assembled vesicular nanostructures in water under the experimental conditions. The size of the vesicles Was observed to depend upon the kinetics of self-assembly and could be varied from similar to 300 to similar to 70 nm. Our mechanistic study indicated that the self-assembly of C60Ph5(OH)(5) is driven by extensive intermolecular as well as water-mediated hydrogen :bonding along with fullerene-fullerene hydrophobic interaction in water. The cumulative effect of these interactions is responsible for the stability of vesicular structures even on the removal of solvent. The substitution of phenol with anisole resulted in different packing and interaction of the fullerene derivative, as Indicated in the molecular dynamics studies, thus resulting in different self-assembled nanostructures. The hollow vesicles were further encapsulated with a hydrophobic conjugated polymer and water-soluble dye as guest molecules. Such confinement of pi-conjugated polymers in fullerene has significance in bulk heterojunction devices for efficient exciton diffusion.

A water soluble third generation poly(alkyl aryl ether) dendrimer was examined for its ability to solubilize hydrophobic polyaromatic molecules in water and facilitate non-radiative resonance energy transfer between them. One to two orders of magnitude higher aqueous solubilities of pyrene (PY), perylene (PE), acridine yellow (AY) and acridine orange (AO) were observed in presence of a defined concentration of the dendrimer. A reduction in the quantum yield of the donor PY* emission and a partial decrease in lifetime of the donor excited state revealed the occurrence of energy transfer from dendrimer solubilized excited PY to ground state PE molecules, both present within a dendrimer. The energy transfer efficiency was estimated to be similar to 61%. A cascade resonance energy transfer in a three component system, PY*-to-PE-to-AY and PY*-to-PE-to-AO, was demonstrated through incorporation of AY or AO in the two component PY-PE system. In the three-component system, excitation of PY resulted in emission from AY or AO via a cascade energy transfer process. Careful choice of dye molecules with good spectral overlap and the employment of dendrimer as the medium enabled us to expand absorption-emission wavelengths, from similar to 330 nm to similar to 600 nm in aqueous solution. As matas inundáveis e brejos presentes nas restingas desencadeiam uma série de processos que influenciam as características físico-químicas e biológicas do solo, levando as plantas a apresentarem mecanismos de aclimatação ou adaptação ao estresse da inundação, como alterações morfológicas e fisiológicas de forma a minimizar os efeitos da falta de oxigênio. Dentre as espécies vegetais de samambaias ocorrentes em ambientes inundáveis nas restingas, se destacam três espécies: Acrostichum danaeifolium Langsd. & Fisch., Blechnum serrulatum Rich. e Thelypteris interrupta (Willd.) K.Iwats. O objetivo deste trabalho é caracterizar os aspectos ecofisiológicos que os esporófitos dessas samambaias apresentam para sobreviver em ambientes de inundação na restinga de Maricá, estado do Rio de Janeiro. Neste sentido, foi determinada a caracterização física e química dos sítios de ocorrências destas samambaias, as variações foliares entre elas, espessura, densidade, massa por unidade de folha, teor de clorofilas e atributos quantitativos das células epidérmicas, além da quantificação e determinação à distribuição dos carboidratos. Para as variáveis dos vegetais foram feitas coletas na estação chuvosa e seca e para variáveis do solo na estação seca. Os sítios analisados se mostraram extremamente ácidos, de baixa fertilidade e com toxidez por macro e micro nutrientes, indicando que as samambaias apresentam tolerância a estes fatores. Na época chuvosa (inundação), as samambaias apresentaram queda na densidade foliar, acompanhada de um aumento de massa por unidade de folha. Esta habilidade de conseguir ganhar massa seca por área classifica todas as samambaias analisadas como tolerantes à inundação. Os altos valores de carboidratos solúveis nas folhas indicam aumento da degradação do amido foliar e o menor teor de carboidrato solúvel encontrado nos caules explicita a redução na respiração das raízes destas plantas sob anoxia/ hipoxia, para evitar a oxidação e o incremento do estoque de amido de reserva, elucidando estratégia de tolerância à inundação. A menor disponibilidade de água na estação seca afeta diretamente os atributos foliares diminuindo o índice estomático, a suculência e a massa por unidade de folha, no qual reflete na queda das concentrações de clorofilas. Os menores valores nas concentrações de clorofila têm influencia direta na presença de amidos foliar que são estocado e, alterando toda a dinâmica dos carboidratos nestas espécies. A análise do sítio onde cresce Acrostichum danaeifolium indica níveis críticos de Na no solo e provavelmente, a produção de mucilagem no caule e no pecíolo é uma estratégia de tolerância ao ambiente salino e inundado. O elevado índice de cobertura de Blechnum serrulatum em ambientes inundados indica que esta espécie possui adaptações a solos hidromórficos, entre elas, grande capacidade de estocagem de amido no caule. A maior sinuosidade das células epidérmicas em T. interrupta permite uma alta suculência mantendo o status hidrológico da folha em ambas as estações. Os resultados apresentados, além de agregar informações sobre a biologia das samambaias nos neotrópicos, irão contribuir para a compreensão da dinâmica de ocupação de espécies herbáceas em ambientes alagáveis nas restingas brasileiras

This report is the second in a series from a project to assess land-based sources of pollution (LBSP) and effects in the St. Thomas East End Reserves (STEER) in St. Thomas, USVI, and is the result of a collaborative effort between NOAA’s National Centers for Coastal Ocean Science, the USVI Department of Planning and Natural Resources, the University of the Virgin Islands, and The Nature Conservancy. Passive water samplers (POCIS) were deployed in the STEER in February 2012. Developed by the US Geological Survey (USGS) as a tool to detect the presence of water soluble contaminants in the environment, POCIS samplers were deployed in the STEER at five locations. In addition to the February 2012 deployment, the results from an earlier POCIS deployment in May 2010 in Turpentine Gut, a perennial freshwater stream which drains to the STEER, are also reported. A total of 26 stormwater contaminants were detected at least once during the February 2012 deployment in the STEER. Detections were high enough to estimate ambient water concentrations for nine contaminants using USGS sampling rate values. From the May 2010 deployment in Turpentine Gut, 31 stormwater contaminants were detected, and ambient water concentrations could be estimated for 17 compounds. Ambient water concentrations were estimated for a number of contaminants including the detergent/surfactant metabolite 4-tert-octylphenol, phthalate ester plasticizers DEHP and DEP, bromoform, personal care products including menthol, indole, n,n-diethyltoluamide (DEET), along with the animal/plant sterol cholesterol, and the plant sterol beta-sitosterol. Only DEHP appeared to have exceeded a water quality guideline for the protection of aquatic organisms.

Centrifuge tests were carried out to determine the effect of 5 different water-soluble chemicals on a thin consolidated disc of clay. The evolution of changes in the clay permeability with time was investigated and other structural changes due to chemical attack were monitored. The findings presented here demonstrate that the permeability of the clay appear to be generally related to the polarity of the chemicals and the dielectric constant, with the exception of Butanol. In the case of Butanol at low flow rate and low stress level, the action of the chemical caused the clay to crack, with a consequent large increase in flow.

A method has been evolved to enhance the production of natural feed in brackish water fish farms by providing substrates for bio-growth ('aquafeed' production) which is a biomass complex consisting of sedentary and associated organisms of plant and animal species. The seasonal fluctuations of the aquafeed production over different substrates ranged as: 787-1830g/coconut leaf (6m²)/45 days, 16.0-072.9g/glass panel (2x10x10cm²)/30 days, 52-230g/nylon mat (2x25x25cm²)/30 days and 18.6-123.1g/wooden block (6x10x10cm²)/30 days. The average dry weight composition of the major components of aquafeed obtained in the present study was sand-silt-clay 40%, protein 22%, carbohydrate (water soluble) 1.8% and fat 3.35% (water content 85%). Mugil cephalus of 1.85 cm reared in a 0.01ha pond and fed on aquafeed attained a size of 23 cm length and 146.73g weight during one year. Survival rate was 54% at a density of 1000/ha. Salinity and temperature of the pond during the culture period ranged between 1.4 and 32.8‰ and 28.1 and 36.5°C respectively.

Quality of analog fishery products invariably depends on the gel characteristics and nutritional status of minced meat. With an objective to find out the effect of water washing on kamaboko gel, the minced meat from Croaker fish was washed for four times (5 minutes each) using chilled water at a temperature of 8-10°C. Results reflected noticeable improvement in folding test and SSN% of kamaboko with essential decrease in fat content, water soluble proteins, expressible water and quality parameters like NPN, VBN, TMA, FFA and PV denoting superior gel quality than control sample after repeated washing. The results indicated that there was a definite improvement in functional properties such as gel forming ability, expressible water content of the croaker minced meat essential decrease in fat content, water soluble proteins, expressible water and quality parameters after each wash, but two washes of 5 minutes duration each was necessary to achieve satisfactory results.

Microcoleus vaginatus isolated from a desert algal crust of Shapotou was cultured in BG-11 medium containing 0.2mol l(-1) NaCl or 0.2mol l(-1) NaCl plus 100mg l(-1) sucrose, extracellular polymeric substances (EPS) or hot water-soluble polysaccharides (HWP), respectively. Photosynthetic oxygen evolution rates, photosystem 11 activity (Fv/Fm) and dark respiration of NaCl-stressed cells were enhanced significantly by the added sucrose or EPS under salt stress conditions (0.2mol l(-1) NaCl). Compared with cells treated with salt alone, sodium contents in cells reduced significantly; the content of cellular total carbohydrate did not change, and intracellular sucrose, water-soluble sugar increased significantly following the addition of exogenous carbohydrates. Sucrose synthase (SS) activity of NaCl-stressed cells increased following the addition of sucrose, and sucrose phosphate synthase (SPS) activity of NaCl-stressed cells increased following the addition of exogenous sucrose, EPS or HWP compared with cells stressed with NaCl only. The results suggested that the extruded EPS might be re-absorbed by cells of M. vaginatus as carbon source, they could increase salt tolerance of M. vaginatus through the changes of carbohydrate metabolism and the selective uptake of sodium ions.
