Työssä tutkittiin moniarvoisten metalliformiaattien valmistusta ioninvaihto-menetelmällä. Kirjallisuustutkimus käsitteleetunnettuja alumiiniformiaatin ja rautaformiaatin valmistusmenetelmiä, kationinvaihtohartsien ominaisuuksia, ioninvaihtohartsien selektiivisyyttä ja alumiinin, raudan, magnesiumin ja sinkin vesikemiaa. Laboratoriokokeiden avulla tutkittiin sinkki-, magnesium-, rauta(II)- ja alumiiniformiaattien valmistusta ioninvaihdolla. Kokeet suoritettiin kolonnissa, joka oli pakattu makrohuokoisella tai geelimäisellä vahvalla kationin-vaihtohartsilla. Hartsi vaihdettiin natriummuodosta metallimuotoon metallikloridi- tai metallisulfaattiliuoksella.Metalli eluoitiin hartsista natriumformiaatilla. Formiaattien valmistus onnistui makrohuokoista vahvaa kationinvaihtohartsia käyttämällä. Rauta(II)formiaatin valmistus oli vaikeampaa kuin muiden formiaattien, koska rauta(II) hapettui osittain rauta(III):ksi valmistuksen aikana. Alumiiniformiaattia valmistettiin käyttäen sekä makrohuokoista että geelimäistä hartsia. Makrohuokoisen hartsin havaittiin soveltuvan geelimäistä hartsia paremmin alumiiniformiaatin valmistukseen. Kungeelimäistä hartsia käytettiin, noin 30 % alumiinista jäi kiinni hartsiin eikä siten eluoitunut. Ioninvaihdon selektiivisyyskertoimien saamiseksi suoritettiin tasapainokokeita. Selektiivisyyskertoimia käytettiin ioninvaihtokolonnin dynaamisessa simuloinnissa. Ioninvaihdon simuloiminen dynaamisella kolonnimallilla onnistui hyvin.

Salt and heat stresses, which are often combined in nature, induce complementing defense mechanisms. Organisms adapt to high external salinity by accumulating small organic compounds known as osmolytes, which equilibrate cellular osmotic pressure. Osmolytes can also act as "chemical chaperones" by increasing the stability of native proteins and assisting refolding of unfolded polypeptides. Adaptation to heat stress depends on the expression of heat-shock proteins, many of which are molecular chaperones, that prevent protein aggregation, disassemble protein aggregates, and assist protein refolding. We show here that Escherichia coli cells preadapted to high salinity contain increased levels of glycine betaine that prevent protein aggregation under thermal stress. After heat shock, the aggregated proteins, which escaped protection, were disaggregated in salt-adapted cells as efficiently as in low salt. Here we address the effects of four common osmolytes on chaperone activity in vitro. Systematic dose responses of glycine betaine, glycerol, proline, and trehalose revealed a regulatory effect on the folding activities of individual and combinations of chaperones GroEL, DnaK, and ClpB. With the exception of trehalose, low physiological concentrations of proline, glycerol, and especially glycine betaine activated the molecular chaperones, likely by assisting local folding in chaperone-bound polypeptides and stabilizing the native end product of the reaction. High osmolyte concentrations, especially trehalose, strongly inhibited DnaK-dependent chaperone networks, such as DnaK+GroEL and DnaK+ClpB, likely because high viscosity affects dynamic interactions between chaperones and folding substrates and stabilizes protein aggregates. Thus, during combined salt and heat stresses, cells can specifically control protein stability and chaperone-mediated disaggregation and refolding by modulating the intracellular levels of different osmolytes.

A recently developed calculation method to determine stoichiometric dissociation constants of weak acids from potentiometric titration data is described. The titration data from three different weak acids in aqueous salt solutions at 25 °C were used as examples of the use of the method. The salt alone determined the ionic strength of the solutions considered in this study, and salt molalities up to 0,5 mol kg -1 were used.

The AMPK/Snf1 kinase has a central role in carbon metabolism homeostasis in Saccharomyces cerevisiae. In this study, we show that Snf1 activity, which requires phosphorylation of the Thr210 residue, is needed for protection against selenite toxicity. Such protection involves the Elm1 kinase, which acts upstream of Snf1 to activate it. Basal Snf1 activity is sufficient for the defense against selenite, although Snf1 Thr210 phosphorylation levels become increased at advanced treatment times, probably by inhibition of the Snf1 dephosphorylation function of the Reg1 phosphatase. Contrary to glucose deprivation, Snf1 remains cytosolic during selenite treatment, and the protective function of the kinase does not require its known nuclear effectors. Upon selenite treatment, a null snf1 mutant displays higher levels of oxidized versus reduced glutathione compared to wild type cells, and its hypersensitivity to the agent is rescued by overexpression of the glutathione reductase gene GLR1. In the presence of agents such as diethyl maleate or diamide, which cause alterations in glutathione redox homeostasis by increasing the levels of oxidized glutathione, yeast cells also require Snf1 in an Elm1-dependent manner for growth. These observations demonstrate a role of Snf1 to protect yeast cells in situations where glutathione-dependent redox homeostasis is altered to a more oxidant intracellular environment and associates AMPK to responses against oxidative stress.

OBJECTIVE: To assess the iodine status of Swiss population groups and to evaluate the influence of iodized salt as a vector for iodine fortification. DESIGN: The relationship between 24 h urinary iodine and Na excretions was assessed in the general population after correcting for confounders. Single-day intakes were estimated assuming that 92 % of dietary iodine was excreted in 24 h urine. Usual intake distributions were derived for male and female population groups after adjustment for within-subject variability. The estimated average requirement (EAR) cut-point method was applied as guidance to assess the inadequacy of the iodine supply. SETTING: Public health strategies to reduce the dietary salt intake in the general population may affect its iodine supply. SUBJECTS: The study population (1481 volunteers, aged ≥15 years) was randomly selected from three different linguistic regions of Switzerland. RESULTS: The 24 h urine samples from 1420 participants were determined to be properly collected. Mean iodine intakes obtained for men (n 705) and women (n 715) were 179 (sd 68.1) µg/d and 138 (sd 57.8) µg/d, respectively. Urinary Na and Ca, and BMI were significantly and positively associated with higher iodine intake, as were men and non-smokers. Fifty-four per cent of the total iodine intake originated from iodized salt. The prevalence of inadequate iodine intake as estimated by the EAR cut-point method was 2 % for men and 14 % for women. CONCLUSIONS: The estimated prevalence of inadequate iodine intake was within the optimal target range of 2-3 % for men, but not for women.

This study evaluated the tolerance of mango cultivars 'Haden', 'Palmer', 'Tommy Atkins' and 'Uba' grafted on rootstock 'Imbú' to salt stress using chlorophyll fluorescence. Plants were grown in modified Hoagland solution containing 0, 15, 30, and 45 mmol L-1 NaCl. At 97 days the parameters of the chlorophyll fluorescence (F0, Fm, Fv, F0/Fm, Fv/Fm, Fv'/Fm', ΦPSII = [(Fm'-Fs)/(Fm')], D = (1- Fv'/Fm') and ETR = (ΦPSII×PPF×0,84×0,5) were determined. At 100 days, the leaf emission and leaf area, toxicity and leaf abscission indexes were determined. In all cultivars evaluated, in different degree, there were decreases in photochemical efficiency of photosystem II, enhanced concentrations from 15 mmol L-1 NaCl. The decreases in the potential quantum yield of photosystem II (Fv/Fm) were 27.9, 18.7, 20.5, and 27.4%, for cultivars 'Haden', 'Palmer', 'Tommy Atkins', and 'Uba', respectively, when grown in 45 mmol L-1 NaCl. It was found decreases in leaf emission and mean leaf area in all cultivars from 15 mmol L-1 NaCl. There were increases in leaf toxicity of 33.0, 67.5, 41.6 and 80.8% and in leaf abscission of 71.8, 29.2, 32.5, and 67.9% for the cultivars 'Haden', 'Palmer', 'Tommy Atkins', and 'Uba' respectively, when grown in 45 mmol L-1 NaCl. Leaf toxicity and leaf abscission were not observed in 15 mmol L-1 NaCl. The decrease in Fv/Fm ratio were accompanied by decreasing in leaf emission and increased leaf toxicity index, showing, therefore, the potential of chlorophyll fluorescence in the early detection of salt stress in mango tree.

Single-stranded DNA (ssDNA) plays a major role in several biological processes. It is therefore of fundamental interest to understand how the elastic response and the formation of secondary structures are modulated by the interplay between base pairing and electrostatic interactions. Here we measure force-extension curves (FECs) of ssDNA molecules in optical tweezers set up over two orders of magnitude of monovalent and divalent salt conditions, and obtain its elastic parameters by fitting the FECs to semiflexible models of polymers. For both monovalent and divalent salts, we find that the electrostatic contribution to the persistence length is proportional to the Debye screening length, varying as the inverse of the square root of cation concentration. The intrinsic persistence length is equal to 0.7 nm for both types of salts, and the effectivity of divalent cations in screening electrostatic interactions appears to be 100-fold as compared with monovalent salt, in line with what has been recently reported for single-stranded RNA. Finally, we propose an analysis of the FECs using a model that accounts for the effective thickness of the filament at low salt condition and a simple phenomenological description that quantifies the formation of non-specific secondary structure at low forces.

Among the various strategies to reduce the incidence of non-communicable diseases reduction of sodium intake in the general population has been recognized as one of the most cost-effective means because of its potential impact on the development of hypertension and cardiovascular diseases. Yet, this strategic health recommendation of the WHO and many other international organizations is far from being universally accepted. Indeed, there are still several unresolved scientific and epidemiological questions that maintain an ongoing debate. Thus what is the adequate low level of sodium intake to recommend to the general population and whether national strategies should be oriented to the overall population or only to higher risk fractions of the population such as salt-sensitive patients are still discussed. In this paper, we shall review the recent results of the literature regarding salt, blood pressure and cardiovascular risk and we present the recommendations recently proposed by a group of experts of Switzerland. The propositions of the participating medical societies are to encourage national health authorities to continue their discussion with the food industry in order to reduce the sodium intake of food products with a target of mean salt intake of 5-6 grams per day in the population. Moreover, all initiatives to increase the information on the effect of salt on health and on the salt content of food are supported.

BACKGROUND: An inverse correlation between expression of the aldehyde dehydrogenase 1 subfamily A2 (ALDH1A2) and gene promoter methylation has been identified as a common feature of oropharyngeal squamous cell carcinoma (OPSCC). Moreover, low ALDH1A2 expression was associated with an unfavorable prognosis of OPSCC patients, however the causal link between reduced ALDH1A2 function and treatment failure has not been addressed so far. METHODS: Serial sections from tissue microarrays of patients with primary OPSCC (n = 101) were stained by immunohistochemistry for key regulators of retinoic acid (RA) signaling, including ALDH1A2. Survival with respect to these regulators was investigated by univariate Kaplan-Meier analysis and multivariate Cox regression proportional hazard models. The impact of ALDH1A2-RAR signaling on tumor-relevant processes was addressed in established tumor cell lines and in an orthotopic mouse xenograft model. RESULTS: Immunohistochemical analysis showed an improved prognosis of ALDH1A2(high) OPSCC only in the presence of CRABP2, an intracellular RA transporter. Moreover, an ALDH1A2(high)CRABP2(high) staining pattern served as an independent predictor for progression-free (HR: 0.395, p = 0.007) and overall survival (HR: 0.303, p = 0.002), suggesting a critical impact of RA metabolism and signaling on clinical outcome. Functionally, ALDH1A2 expression and activity in tumor cell lines were related to RA levels. While administration of retinoids inhibited clonogenic growth and proliferation, the pharmacological inhibition of ALDH1A2-RAR signaling resulted in loss of cell-cell adhesion and a mesenchymal-like phenotype. Xenograft tumors derived from FaDu cells with stable silencing of ALDH1A2 and primary tumors from OPSCC patients with low ALDH1A2 expression exhibited a mesenchymal-like phenotype characterized by vimentin expression. CONCLUSIONS: This study has unraveled a critical role of ALDH1A2-RAR signaling in the pathogenesis of head and neck cancer and our data implicate that patients with ALDH1A2(low) tumors might benefit from adjuvant treatment with retinoids.
