Au Mali, le monde rural est un système complexe dont le développement implique son appréhension dans sa complexité. Cependant, la réalité malienne montre un manque de formation, d'organisation et une exploitation irrationnelle des ressources entravant les efforts du développement. Dans cette situation, il est difficile au crédit rural de jouer son rôle dans le développement. Les paysans du Mali, et plus précisément ceux dans la zone Office de la Haute Vallée du Niger (OHVN), tirent annuellement de leurs cultures, des revenus assez faibles. Malheureusement ces sommes ne restent pas dans le secteur rural pour faciliter la promotion de l'agriculture. Elles sont, pour la plupart, utilisées pour payer les arriérés de crédit. Ainsi, au cours d'un stage que nous avons eu à effectuer à Bancoumana (un secteur tabacole encadré par l'Office), il a été constaté que les paysans sont confrontés à plusieurs problèmes, qu'ils soient d'origine économique, sociologique ou autre. Entre autres problèmes: la faiblesse des revenus d'où incapacité d'investir et d'épargner; - l'insuffisance de sensibilisation sur le crédit rural; - l'insuffisance de formation en gestion et organisation. Dans cette problématique, nous soulevons des points d'ordre économique et social. En fait, dans la politique de crédit pratiquée par de nombreux organismes de développement (expérience vécue en Madagascar), on ne tient pas compte des habitudes et de la tradition en matière de crédit. Les politiques de crédit restent incomprises et inappliquées par les paysans. L'élaboration de ces politiques ne s'appuie pas solidement sur le vécu des participants et des bénéficiaires-cibles, encore moins sur les valeurs. En se basant sur nos investigations antérieures, nous avons jugé nécessaire de voir comment évolue la politique de crédit dans la zone de l'Office de la Haute Vallée du Niger. La présente étude a pour but de comprendre le fonctionnement du crédit rural dans notre zone d'étude, à l'aide d'une analyse descriptive. Nous proposons des recommandations et des suggestions s'il y a lieu.

Au Niger, le taux de mortalité maternelle est estimé à 535 décès pour 100 000 naissances vivantes (INS, 2013) et la probabilité pour un nouveau-né de mourir avant l’âge d’un mois est de 33 ‰. Depuis 2006, le Niger a mis en place une politique de gratuité des soins pour les femmes enceintes et les enfants de 0 à 5 ans, ce qui a contribué à une amélioration significative de la fréquentation des centres de santé. En mars 2012, un processus délibératif fut organisé pendant une conférence de trois jours pour échanger sur les acquis, limites et perspectives de cette nouvelle politique avec 160 participants dont des chercheurs, des humanitaires, des décideurs politiques et des intervenants sur le terrain. L’objectif de cette recherche est de comprendre les effets de cette conférence ainsi que d’explorer les activités du comité de suivi de la feuille de route. La recherche a été réalisée durant deux mois en été 2014 à Niamey et à N’guiguimi. Elle a reposé sur l’utilisation du cadre conceptuel de Boyko et al., (2012) qui permet de décrire les principales caractéristiques et les effets attendus des dialogues délibératifs et comprendre comment les dialogues délibératifs peuvent contribuer à l’élaboration de politiques sur la base de données probantes. Nous avons mis un accent particulier sur les trois formes d’utilisation des connaissances présentées par Dagenais et al., (2013) : instrumentale, conceptuelle et persuasive. Des entretiens semi-directifs ont été effectués avec 22 acteurs impliqués dans la mise en oeuvre des recommandations. Ils ont été enregistrés, retranscrits intégralement et traités avec le logiciel QDA Miner. Les résultats de l’analyse des discours recueillis révèlent une utilisation instrumentale des recommandations et plus visible chez les humanitaires que les décideurs et les acteurs de la société civile. Il ressort aussi de cette analyse une utilisation conceptuelle et persuasive des recommandations à un degré plus faible parmi tous les acteurs. Le comité de suivi de la feuille route de la conférence n’a pratiquement pas fonctionné, par conséquent, le processus n’a pas eu l’impact souhaité. Les principales raisons de cet échec sont liées au contexte de mise en oeuvre des recommandations (arrestation de plusieurs agents du ministère de la Santé publique qui sont des membres clés du comité de suivi à cause du détournement des fonds GAVI, manque de volonté technique et politique) et/ou aux conditions financières (absence de primes pour les membres du comité et de budget de fonctionnement.). Les iv résultats obtenus ont permis de comprendre les énormes défis (contextuels, financiers notamment) qui restent à relever en matière de transfert de connaissance dans le secteur de santé publique au Niger. En ce qui concerne la suite de la conférence, il faudrait accélérer la redynamisation du comité de suivi en le dotant d’un fonds de fonctionnement et en créant une agence autonome de gestion de la gratuité des soins; et renforcer le soutien politique autour de l’Initiative Santé Solidarité Sahel.

The injectivity, containment and storage capacity of sandstone reservoirs in a field in the Coastal Swamp depobelt of the onshore eastern Niger Delta were evaluated using wireline logs and seismic data to assess their potentials for carbon dioxide storage and geosequestration. The reservoir formation consists of multilayered alternating beds of sandstone and shale cap rocks. Active seismicity and fracturing intensity are low and growth faults provide the reservoir sealing mechanisms. Three reservoirs were delineated at depths between 3319 m and 3539 m which will keep injected CO2 in a supercritical state. The reservoir depth of at least 800 m, porosity and permeability of more than 10 percent and 20 mD, and a caprock thickness of at least 10 m, in addition to geothermal gradients of 13.46 to 33.66 ºC /km are the ideal conditions for the efficacy of storage. Comparison of the derived reservoir and seal properties such as porosity, permeability, thickness and depth with the minimum recommended site selection criteria shows that the reservoirs are potential candidates for carbon geosequestration with a total theoretical storage capacity of 147MM tons.

The work focuses on the screening of Polyalthia longifolia and Aloe vera for their phytoextractability of heavy metal in soil of the Niger Delta. 5kg of soil was polluted with 100 mg of Zn, Fe and Pb each replicated 9 times. Each set was separated into 3 subgroups. The subgroups were phytoextracted with Polyalthia longifolia and Aloe vera alongside a control (no phytoextraction) respectively. After 12 weeks, the concentration of Lead, Iron and Zinc in soils, roots and shoots was determined. Results showed that the two plants have phytoextraction ability with reductions in Zn, Fe, and Pb in the phytoextraction soil . Metal transfer factor was PB: Aloe vera (0.881) > P. longifolia (0.315); Zn: P. longifolia (0.614) > Aloe vera (0.606); Fe: Aloe vera (0.812) > P. longifolia (0.774). Translocation factors for the two plants were in the order: Zn: P. longifolia (0.79) > Aloe vera (0.36); Fe: P. longifolia (0.63) > Aloe vera (0.05); Pb: P. longifolia (0.57) > Aloe vera (0.23). Since the translocation factors were < 1, the plants can be classified as non- hyperaccumulators for these metals.

Diversos trabalhos têm procurado aumentar a eficiência da hidrólise enzimática da biomassa lignocelulósica. Nesse contexto, o melhoramento de cepas produtoras de enzimas celulolíticas e hemicelulolíticas pode resultar em misturas enzimáticas mais eficientes. A linhagem parental de Aspergillus niger 3T5B8, referenciada como produtora de poligalacturonase, foi utilizada para o melhoramento genético visando aumentar a produção de celulases e hemicelulases. A produção das enzimas CMCase, xilanase, beta-glicosidase e poligalacturonase por fermentação submersa usando duas linhagens mutantes P49 e P83 foi avaliada e comparada com a linhagem parental. Os resultados mostraram um destaque para a cepa P83 com um aumento na produção de 56% de CMCase, 76% de beta-glicosidase, 23% de xilanase e 216% na poligalacturonase.

As lipases são enzimas que catalisam a hidrólise parcial ou total de triacilglicerois produzindo ácidos graxos livres, diacilglicerol, monoacilglicerol e glicerol. Este trabalho caracterizou a especificidade, a temperatura ótima e o pH ótimo das lipases obtidas por duas cepas de Aspergillus niger, sendo uma selvagem C e outra mutante 11T53A14. Os resultados mostram que os extratos enzimáticos apresentam especificidade diferentes, sendo a cepa selvagem mais específica para ácidos graxos de 8 carbonos e a cepa mutante inespecífica em relação ao tamanho do ácido graxo. As duas cepas apresentaram atividade em uma ampla faixa de pH, porém observou-se uma redução da atividade de mais de 50% em pH acima de 9,0. Em relação à temperatura, as lipases das duas cepas se mostraram mais ativas a 35°C.

The basidiomycete fungus Gloeophyllum trabeum causes a typical brown rot and is known to use reactive oxygen species in the degradation of cellulose. The extracellular Cel12A is one of the few endo-1,4-β-glucanase produced by G. trabeum. Here we cloned cel12A and heterologously expressed it in Aspergillus niger. The identity of the resulting recombinant protein was confirmed by mass spectrometry. We used the purified GtCel12A to determine its substrate specificity and basic biochemical properties. The G. trabeum Cel12A showed highest activity on β-glucan, followed by lichenan, carboxymethylcellulose, phosphoric acid swollen cellulose, microcrystalline cellulose, and filter paper. The optimal pH and temperature for enzymatic activity were, respectively, 4.5 and 50 °C on β-glucan. Under these conditions specific activity was 239.2 ± 9.1 U mg(-1) and the half-life of the enzyme was 84.6 ± 3.5 hours. Thermofluor studies revealed that the enzyme was most thermal stable at pH 3. Using β-glucan as a substrate, the Km was 3.2 ± 0.5 mg mL(-1) and the Vmax was 0.41 ± 0.02 µmol min(-1). Analysis of the effects of GtCel12A on oat spelt and filter paper by scanning electron microscopy revealed the morphological changes taking place during the process.

In this work, different reactions in vitro between an environmental bacterial isolate and fungal species were related. The Gram-positive bacteria had terminal and subterminal endospores, presented metabolic characteristics of mesophilic and acidophilic growth, halotolerance, positive to nitrate reduction and enzyme production, as caseinase and catalase. The analysis of partial sequences containing 400 to 700 bases of the 16S ribosomal RNA gene showed identity with the genus Bacillus. However, its identity as B. subtilis was confirmed after analyses of the rpoB, gyrA, and 16S rRNA near-full-length sequences. Strong inhibitory activity of environmental microorganisms, such as Penicillium sp, Aspergillus flavus, A. niger, and phytopathogens, such as Colletotrichum sp, Alternaria alternata, Fusarium solani and F. oxysporum f.sp vasinfectum, was shown on co-cultures with B. subtilis strain, particularly on Sabouraud dextrose agar (SDA) and DNase media. Red and red-ochre color pigments, probably phaeomelanins, were secreted by A. alternata and A. niger respectively after seven days of co-culture.

Background: In spite of its advantageous physiological properties for bioprocess applications, the use of the yeast Kluyveromyces marxianus as a host for heterologous protein production has been very limited, in constrast to its close relative Kluyveromyces lactis. In the present work, the model protein glucose oxidase (GOX) from Aspergillus niger was cloned into K. marxianus CBS 6556 and into K. lactis CBS 2359 using three different expression systems. We aimed at verifying how each expression system would affect protein expression, secretion/localization, post-translational modification, and biochemical properties. Results: The highest GOX expression levels (1552 units of secreted protein per gram dry cell weight) were achieved using an episomal system, in which the INU1 promoter and terminator were used to drive heterologous gene expression, together with the INU1 prepro sequence, which was employed to drive secretion of the enzyme. In all cases, GOX was mainly secreted, remaining either in the periplasmic space or in the culture supernatant. Whereas the use of genetic elements from Saccharomyces cerevisiae to drive heterologous protein expression led to higher expression levels in K. lactis than in K. marxianus, the use of INU1 genetic elements clearly led to the opposite result. The biochemical characterization of GOX confirmed the correct expression of the protein and showed that K. marxianus has a tendency to hyperglycosylate the protein, in a similar way as already observed for other yeasts, although this tendency seems to be smaller than the one of e. g. K. lactis and S. cerevisiae. Hyperglycosylation of GOX does not seem to affect its affinity for the substrate, nor its activity. Conclusions: Taken together, our results indicate that K. marxianus is indeed a good host for the expression of heterologous proteins, not only for its physiological properties, but also because it correctly secretes and folds these proteins.

The trails formed by many ant species between nest and food source are two-way roads on which outgoing and returning workers meet and touch each other all along. The way to get back home, after grasping a food load, is to take the same route on which they have arrived from the nest. In many species such trails are chemically marked by pheromones providing orientation cues for the ants to find their way. Other species rely on their vision and use landmarks as cues. We have developed a method to stop foraging ants from shuttling on two-way trails. The only way to forage is to take two separate roads, as they cannot go back on their steps after arriving at the food or at the nest. The condition qualifies as a problem because all their orientation cues-chemical, visual or any other - are disrupted, as all of them cannot but lead the ants back to the route on which they arrived. We have found that workers of the leaf-cutting ant Atta sexdens rubropilosa can solve the problem. They could not only find the alternative way, but also used the unidirectional traffic system to forage effectively. We suggest that their ability is an evolutionary consequence of the need to deal with environmental irregularities that cannot be negotiated by means of excessively stereotyped behavior, and that it is but an example of a widespread phenomenon. We also suggest that our method can be adapted to other species, invertebrate and vertebrate, in the study of orientation, memory, perception, learning and communication.

Milkfat-soybean oil blends were enzymatically interesterified (EIE) by Aspergillus niger lipase immobilized on SiO(2)-PVA hybrid composite in a solvent free system. An experimental mixture design was used to study the effects of binary blends of milkfat-soybean oil (MF:SBO) at different proportions (0:100; 25:75; 33:67; 50:50; 67:33; 75:25; 100:0) on the compositional and textural properties of the EIE products, considering, as response variables, the interesterification yield (IY), consistency and hardness. Lipase-catalysed interesterification reactions increased the relative proportion of TAGs` C(46)-C(52) and decreased the TAGs` C(40)-C(42) and C(54) concentrations. The highest IY was attained (10.8%) for EIE blend of MF:SBO 67:33 resulting in a more spreadable material at refrigerator temperature in comparison with butter, milkfat or non-interesterified (NIE) blend. In this case, consistency and hardness values were at least 32% lower than values measured for butter. Thus, using A. niger lipase immobilized on SiO(2)-PVA improves the textural properties of milkfat and has potential for development of a product incorporating unsaturated and essential fatty acids from soybean oil. This study aimed to correlate the efficiency of enzymatic hydrolysis of the cellulose contained in a sugarcane bagasse sample pretreated with dilute H(2)SO(4) with the levels of independent variables such as initial content of solids and loadings of enzymes and surfactant (Tween 20), for two cellulolytic commercial preparations. The preparations, designated cellulase I and cellulase II, were characterized regarding the activities of total cellulases, endoglucanase, cellobiohydrolase, cellobiase, beta-glucosidase, xylanase, and phenoloxidases (laccase, manganese and lignin peroxidases), as well as protein contents. Both extracts showed complete cellulolytic complexes and considerable activities of xylanases, without activities of phenoloxidases. For the enzymatic hydrolyses, two 2(3) central composite full factorial designs were employed to evaluate the effects caused by the initial content of solids (1.19-4.81%, w/w) and loadings of enzymes (1.9-38.1 FPU/g bagasse) and Tween 20 (0.0-0.1 g/g bagasse) on the cellulose digestibility. Within 24 h of enzymatic hydrolysis, all three independent variables influenced the conversion of cellulose by cellulase I. Using cellulase II, only enzyme and surfactant loadings showed significant effects on cellulose conversion. An additional experiment demonstrated the possibility of increasing the initial content of solids to values much higher than 4.81% (w/w) without compromising the efficiency of cellulose conversion, consequently improving the glucose concentration in the hydrolysate.

Penicillium chrysogenum is widely used as an industrial antibiotic producer, in particular in the synthesis of g-lactam antibiotics such as penicillins and cephalosporins. In industrial processes, oxalic acid formation leads to reduced product yields. Moreover, precipitation of calcium oxalate complicates product recovery. We observed oxalate production in glucose-limited chemostat cultures of P. chrysogenum grown with or without addition of adipic acid, side-chain of the cephalosporin precursor adipoyl-6-aminopenicillinic acid (ad-6-APA). Oxalate accounted for up to 5% of the consumed carbon source. In filamentous fungi, oxaloacetate hydrolase (OAH; EC3.7.1.1) is generally responsible for oxalate production. The P. chrysogenum genome harbours four orthologs of the A. niger oahA gene. Chemostat-based transcriptome analyses revealed a significant correlation between extracellular oxalate titers and expression level of the genes Pc18g05100 and Pc22g24830. To assess their possible involvement in oxalate production, both genes were cloned in Saccharomyces cerevisiae, yeast that does not produce oxalate. Only the expression of Pc22g24830 led to production of oxalic acid in S. cerevisiae. Subsequent deletion of Pc22g28430 in P. chrysogenum led to complete elimination of oxalate production, whilst improving yields of the cephalosporin precursor ad-6-APA. (C) 2011 Elsevier Inc.
