Tobacco consumption is a global epidemic responsible for a vast burden of disease. With pharmacological properties sought-after by consumers and responsible for addiction issues, nicotine is the main reason of this phenomenon. Accordingly, smokeless tobacco products are of growing popularity in sport owing to potential performance enhancing properties and absence of adverse effects on the respiratory system. Nevertheless, nicotine does not appear on the 2011 World Anti-Doping Agency (WADA) Prohibited List or Monitoring Program by lack of a comprehensive large-scale prevalence survey. Thus, this work describes a one-year monitoring study on urine specimens from professional athletes of different disciplines covering 2010 and 2011. A method for the detection and quantification of nicotine, its major metabolites (cotinine, trans-3-hydroxycotinine, nicotine-N′-oxide and cotinine-N-oxide) and minor tobacco alkaloids (anabasine, anatabine and nornicotine) was developed, relying on ultra-high pressure liquid chromatography coupled to triple quadrupole mass spectrometry (UHPLC-TQ-MS/MS). A simple and fast dilute-and-shoot sample treatment was performed, followed by hydrophilic interaction chromatography-tandem mass spectrometry (HILIC-MS/MS) operated in positive electrospray ionization (ESI) mode with multiple reaction monitoring (MRM) data acquisition. After method validation, assessing the prevalence of nicotine consumption in sport involved analysis of 2185 urine samples, accounting for 43 different sports. Concentrations distribution of major nicotine metabolites, minor nicotine metabolites and tobacco alkaloids ranged from 10 (LLOQ) to 32,223, 6670 and 538 ng/mL, respectively. Compounds of interest were detected in trace levels in 23.0% of urine specimens, with concentration levels corresponding to an exposure within the last three days for 18.3% of samples. Likewise, hypothesizing conservative concentration limits for active nicotine consumption prior and/or during sport practice (50 ng/mL for nicotine, cotinine and trans-3-hydroxycotinine and 25 ng/mL for nicotine-N′-oxide, cotinine-N-oxide, anabasine, anatabine and nornicotine) revealed a prevalence of 15.3% amongst athletes. While this number may appear lower than the worldwide smoking prevalence of around 25%, focusing the study on selected sports highlighted more alarming findings. Indeed, active nicotine consumption in ice hockey, skiing, biathlon, bobsleigh, skating, football, basketball, volleyball, rugby, American football, wrestling and gymnastics was found to range between 19.0 and 55.6%. Therefore, considering the adverse effects of smoking on the respiratory tract and numerous health threats detrimental to sport practice at top level, likelihood of smokeless tobacco consumption for performance enhancement is greatly supported.

We set out to determine the seroprevalence of hepatitis B and C among human immunodeficiency virus type-1 (HIV-1) infected individuals in North-Central Nigeria to define the influence of these infections on CD4+ lymphocytes cells among our patients as access to antiretroviral therapy improves across the Nigerian nation. The CD4+ values of 180 confirmed HIV-1 infected individuals were enumerated using a superior fluorescence-activated cell sorter system. These patients were tested for the presence of hepatitis B surface antigen and anti-hepatitis C virus (HCV) using third generation enzyme-linked immunosorbent assays. Fifty (27.8%) patients had active hepatitis B virus (HBV) infection while 33 (18.3%) tested positive for anti-HCV antibody. Of these infections, 110 (61.1%), 37 (20.6%), and 20 (11.1%) had HIV only, HBV/HIV-only, and HCV/HIV-only respectively. A HBV/HCV/HIV coinfection prevalence of 7.2% (13 patients) was recorded. Patients coinfected with HIV/HBV/HCV appeared to have lower CD4+ counts (mean = 107 cells/µl; AIDS defining) when compared to HBV/HIV-only (mean = 377 cells/µl), HCV/HIV-only (mean = 373 cells/µl) and patients with mono HIV infection (mean = 478 cells/µl). Coinfection with HBV or HCV is relatively common among HIV-infected patients in Nigeria and should be a big consideration in the initiation and choice of therapy.

A sensitive and specific ultra performance liquid chromatography-tandem mass spectrometry method for the simultaneous quantification of nicotine, its metabolites cotinine and trans-3'-hydroxycotinine and varenicline in human plasma was developed and validated. Sample preparation was realized by solid phase extraction of the target compounds and of the internal standards (nicotine-d4, cotinine-d3, trans-3'-hydroxycotinine-d3 and CP-533,633, a structural analog of varenicline) from 0.5mL of plasma, using a mixed-mode cation exchange support. Chromatographic separations were performed on a hydrophilic interaction liquid chromatography column (HILIC BEH 2.1×100mm, 1.7μm). A gradient program was used, with a 10mM ammonium formate buffer pH 3/acetonitrile mobile phase at a flow of 0.4mL/min. The compounds were detected on a triple quadrupole mass spectrometer, operated with an electrospray interface in positive ionization mode and quantification was performed using multiple reaction monitoring. Matrix effects were quantitatively evaluated with success, with coefficients of variation inferior to 8%. The procedure was fully validated according to Food and Drug Administration guidelines and to Société Française des Sciences et Techniques Pharmaceutiques. The concentration range was 2-500ng/mL for nicotine, 1-1000ng/mL for cotinine, 2-1000ng/mL for trans-3'-hydroxycotinine and 1-500ng/mL for varenicline, according to levels usually measured in plasma. Trueness (86.2-113.6%), repeatability (1.9-12.3%) and intermediate precision (4.4-15.9%) were found to be satisfactory, as well as stability in plasma. The procedure was successfully used to quantify nicotine, its metabolites and varenicline in more than 400 plasma samples from participants in a clinical study on smoking cessation.

The change over time in the fecundity and weight of queens was investigated in three monogynous, independent colony founding species,Lasius niger, Camponotus ligniperda andC. herculaneus, and two polygynous dependent colony founding species,Plagiolepis pygmaea andIridomyrmex humilis. Queens of the three species founding independently exhibited a similar pattern with a significant loss of weight between mating and the emergence of the first workers. In contrast, weights of queens of the species employing dependent colony founding remained more stable. Fecundity of queens founding independently increased slowly with time whereas fecundity of queens founding dependently reached the maximum level some weeks after the beginning of the first reproductive season. These results are discussed in relation to some differences in the life history (e.g., life-span) between queens utilizing independent and dependent colony founding.

BACKGROUND Mutational analysis of the KRAS gene has recently been established as a complementary in vitro diagnostic tool for the identification of patients with colorectal cancer who will not benefit from anti-epidermal growth factor receptor (EGFR) therapies. Assessment of the mutation status of KRAS might also be of potential relevance in other EGFR-overexpressing tumors, such as those occurring in breast cancer. Although KRAS is mutated in only a minor fraction of breast tumors (5%), about 60% of the basal-like subtype express EGFR and, therefore could be targeted by EGFR inhibitors. We aimed to study the mutation frequency of KRAS in that subtype of breast tumors to provide a molecular basis for the evaluation of anti-EGFR therapies. METHODS Total, genomic DNA was obtained from a group of 35 formalin-fixed paraffin-embedded, triple-negative breast tumor samples. Among these, 77.1% (27/35) were defined as basal-like by immunostaining specific for the established surrogate markers cytokeratin (CK) 5/6 and/or EGFR. KRAS mutational status was determined in the purified DNA samples by Real Time (RT)-PCR using primers specific for the detection of wild-type KRAS or the following seven oncogenic somatic mutations: Gly12Ala, Gly12Asp, Gly12Arg, Gly12Cys, Gly12Ser, Gly12Val and Gly13Asp. RESULTS We found no evidence of KRAS oncogenic mutations in all analyzed tumors. CONCLUSIONS This study indicates that KRAS mutations are very infrequent in triple-negative breast tumors and that EGFR inhibitors may be of potential benefit in the treatment of basal-like breast tumors, which overexpress EGFR in about 60% of all cases.

Trois constats majeurs sont à l'origine de notre étude. Premièrement, les processus d'acquisition mobilisés par l'enseignement bilingue ont fait l'objet de très peu de recherches, alors que les résultats issus de ce type d'enseignement ont été largement explorés et ont globalement reçu une évaluation positive. Deuxièmement, l'apprentissage de la langue seconde (L2) et le bénéfice qu'il peut tirer d'un enseignement bilingue reste l'objet privilégié pour la plupart des études, alors que les particularités et les enjeux de l'apprentissage des disciplines dites "non linguistiques" (DNL) (telles l'histoire, la physique ou les mathématiques) restent largement inexplorées. Troisièmement, l'intégration a été souvent étudiée au niveau du curriculum scolaire pour dégager les interrelations entre enseignement de la L2 et enseignement des DNL, mais l'intégration demandait encore à être décrit de manière détaillée en tant qu'incorporation de l'apprentissage des savoirs disciplinaires et linguistiques et en tant que travail intégré des savoirs linguistiques et disciplinaires.Notre analyse s'inscrit alors dans une triple perspective prenant en compte ces trois axes de recherche : une perspective interactionniste et située de l'apprentissage des langues permettant d'envisager les processus d'acquisition mobilisés par l'enseignement bilingue in situ ; une perspective bi-plurilingue qui invite à étudier les spécificités de l'enseignement bilingue et s'affilie à une didactique du plurilinguisme valorisant un travail bilingue et intégré en classe ; une perspective des DNL permettant de considérer les enjeux de l'enseignement bilingue pour l'apprentissage des DNL, et non seulement pour l'apprentissage de la L2, et de considérer le travail intégré des savoirs linguistiques et disciplinaires.Ainsi, notre étude vise la description des pratiques effectives en classe bilingue s'appuyant sur l'analyse d'un important corpus d'interactions enregistrées dans diverses classes du secondaire II, orientée vers une méthodologie à caractère ethnographique et basée sur une démarche empirique et descriptive, ainsi que sur les outils de l'analyse conversationnelle appliquée à l'apprentissage des L2.Dans ce contexte, notre travail repose sur l'hypothèse que l'enseignement bilingue profite non seulement à l'apprentissage des savoirs linguistiques, mais également à celui des savoirs disciplinaires, lorsqu'il se base sur un travail intégré de la dimension linguistique et disciplinaire de cet apprentissage, et qu'un travail dans les deux langues en présence contribue à une intégration forte des savoirs linguistiques et disciplinaires. Il vise alors l'examen de l'articulation entre travail disciplinaire et travail linguistique dans différentes DNL et activités, ainsi que l'élaboration bilingue des concepts disciplinaires.D'abord, nos analyses montrent deux manières d'aborder l'élaboration des concepts disciplinaires dans les classes bilingues observées, le mode unilingue et le mode bilingue, et mettent en lumière l'apport du mode bilingue ou travail bilingue à l'élaboration des savoirs disciplinaires. Puis, la description détaillée de séquences-types permet de dégager les fonctionnements spécifiques à un enseignement bilingue et intégré, ainsi que les degrés et les procédés d'intégration des savoirs linguistiques et disciplinaires dans l'apprentissage en classe bilingue.En résumé, nos observations montrent que le bénéfice de l'enseignement bilingue réside dans l'attention particulière portée aux enjeux linguistiques des DNL et dans le travail linguistique, lorsqu'il est orienté vers un but disciplinaire. Aussi, cette orientation s'affirme dans la dynamique entre travail sur la dimension disciplinaire et travail sur la dimension linguistique.

El peso del servicio de televisión dentro de la oferta de triple play (Internet, telefonía, televisión) de los operadores de telecomunicación por cable en España está siendo uno de los ejes de análisis del mercado comunicológico, gracias a la tendencia del mercado digital hacia la convergencia y la irrupción de nuevos contenidos audiovisuales a través de Internet. El análisis de datos estadísticos aportados por los operadores de cable refleja, salvo en el caso de TeleCable, que el servicio televisivo en España está lejos de la realidad de otros países europeos, donde el contenido televisivo es líder sólido.

BACKGROUND In this study, we evaluated the ability of gene expression profiles to predict chemotherapy response and survival in triple-negative breast cancer (TNBC). METHODS Gene expression and clinical-pathological data were evaluated in five independent cohorts, including three randomised clinical trials for a total of 1055 patients with TNBC, basal-like disease (BLBC) or both. Previously defined intrinsic molecular subtype and a proliferation signature were determined and tested. Each signature was tested using multivariable logistic regression models (for pCR (pathological complete response)) and Cox models (for survival). Within TNBC, interactions between each signature and the basal-like subtype (vs other subtypes) for predicting either pCR or survival were investigated. RESULTS Within TNBC, all intrinsic subtypes were identified but BLBC predominated (55-81%). Significant associations between genomic signatures and response and survival after chemotherapy were only identified within BLBC and not within TNBC as a whole. In particular, high expression of a previously identified proliferation signature, or low expression of the luminal A signature, was found independently associated with pCR and improved survival following chemotherapy across different cohorts. Significant interaction tests were only obtained between each signature and the BLBC subtype for prediction of chemotherapy response or survival. CONCLUSIONS The proliferation signature predicts response and improved survival after chemotherapy, but only within BLBC. This highlights the clinical implications of TNBC heterogeneity, and suggests that future clinical trials focused on this phenotypic subtype should consider stratifying patients as having BLBC or not.
