To investigate the effect of the N-terminal Slit2 protein on neuronal survival and development, recombinant human N-terminal Slit2 (N-Slit2) was assayed against isolated embryonic chick dorsal root ganglion sensory, ciliary ganglion and paravertebral sympathetic neurons. N-Slit2 promoted significant levels of neuronal survival and neurite extension in all of these populations. The protein was also assayed against postnatal mouse dorsal root ganglion neurons and found to promote neuronal survival in a similar manner. These findings suggest the Slit proteins may play an important role during development of the nervous system, mediating cellular survival in addition to the well documented role these proteins play in axonal and neuronal chemorepulsion.

Calcium-binding proteins (CBPs) such as calbindin, parvalbumin and calretinin are used as immunohistochemical markers for discrete neuronal subpopulations. They are particularly useful in identifying the various subpopulations of GABAergic interneurons that control output from prefrontal and cingulate cortices as well as from the hippocampus. The strategic role these interneurons play in regulating output from these three crucial brain regions has made them a focus for neuropathological investigation in schizophrenia. The number of pathological reports detailing subtle changes in these CBP-containing interneurons in patients with schizophrenia is rapidly growing. These proteins however are more than convenient neuronal markers. They confer survival advantages to neurons and can increase the neuron's ability to sustain firing. These properties may be important in the subtle pathophysiology of nondegenerative phenomena such as schizophrenia. The aim of this review is to introduce the reader to the functional properties of CBPs and to examine the emerging literature reporting alterations in these proteins in schizophrenia as well as draw some conclusions about the significance of these findings. Members of the GATA transcription factor gene family have been implicated in a variety of developmental processes, including that of the vertebrate central nervous system. However, the role of GATA proteins in spinal cord development remains unresolved. In this study, we investigated the expression and function of two GATA proteins, GATA2 and GATA3, in the developing chick spinal cord. We show that both proteins are expressed by a distinct subpopulation of ventral interneurons that share the same dorsoventral position as CHX10-positive V2 interneurons. However, no coexpression is observed between the two GATA proteins and CHX10. By in vivo notochord grafting and cyclopamine treatment, we demonstrate that the spatially restricted pattern of GATA3 expression is regulated, at least in part, by the signaling molecule Sonic hedgehog. In addition, we further show that Sonic hedgehog induces GATA3 expression in a dose-dependent manner. Using in ovo electroporations, we also demonstrate that GATA2 is upstream of GATA3 in the same epigenetic cascade and that GATA3 is capable of inducing GATA2 expression in vivo. Furthermore, the ectopically expressed GATA proteins can repress differentiation of other ventral cell fates, but not the development of progenitor populations identified by PAX protein expression. Taken together, our findings strongly suggest an important role for GATA2 and GATA3 proteins in the establishment of a distinct ventral interneuron subpopulation in the developing chick spinal cord. We review investigations that have lead to a model of how the ventral spinal cord of higher vertebrate embryos is patterned during development. Central to this model is the secreted morphogen protein, Sonic hedgehog. There is now considerable evidence that this molecule acts in a concentration-dependent manner to direct the development of the spinal cord. Recent studies have suggested that two classes of homeodomain proteins are induced by threshold concentrations of Sonic hedgehog. Reciprocal inhibition between the two classes acts to convert the continuous gradient of Sonic hedgehog into defined domains of transcription factor expression. However, a number of aspects of ventral spinal cord patterning remain to be elucidated. Some issues currently under investigation involve temporal aspects of Shh-signalling, the role of other signals in ventral patterning and the characterisation of ventral interneurons. In this review, we discuss the current state of knowledge of these issues and present some preliminary studies aimed at furthering understanding of these processes in spinal cord patterning.

Although the polyunsaturated fatty acids arachidonic acid (AA) and docosahexaenoic acid (DHA) are enriched in the olfactory mucosa, their possible contribution to olfactory transduction has not been investigated. This study characterized their effects on voltage-gated K+ and Na+ channels of rat olfactory receptor neurons. Physiological (3-10 mum) concentrations of AA and DHA potently and irreversibly inhibited the voltage-gated K+ current in a voltage-independent manner. In addition, both compounds significantly reduced the inhibitory potency of the odorants acetophenone and amyl acetate at these channels. By comparison, the steady-state effects of both AA and DHA on the voltage-gated Na+ channel were relatively weak, with half-maximal inhibition requiring approximate to 35 mum of either compound. However, a surprising finding was that the initial application of 3 mum AA to a naive neuron caused a strong but transient inhibition of the Na+ current. The channels became almost completely resistant to this inhibition within 1 min, and a 2-min wash in control solution was insufficient to restore the strong inhibitory effect. These observations suggest that polyunsaturated fatty acids have the potential to strongly influence the coding of odorant information by olfactory receptor neurons.

Approximately half of the motoneurons generated during normal embryonic development undergo programmed cell death. Most of this death occurs during the time when synaptic connections are being formed between motoneurons and their target, skeletal muscle. Subsequent muscle activity stemming from this connection helps determine the final number of surviving motoneurons. These observations have given rise to the idea that motoneuron survival is dependent upon access to muscle derived trophic factors, presumably through intact neuromuscular synapses. However, it is not yet understood how the muscle regulates the supply of such trophic factors, or if there are additional mechanisms operating to control the fate of the innervating motoneuron. Recent observations have highlighted target independent mechanisms that also operate to support the survival of motoneurons, such as early trophic-independent periods of motoneuron death, trophic factors derived from Schwann cells and selection of motoneurons during pathfinding. Here we review recent investigations into motoneuron cell death when the molecular signalling between motoneurons and muscle has been genetically disrupted. From these studies, we suggest that in addition to trophic factors from muscle and/or Schwann cells, specific adhesive interactions between motoneurons and muscle are needed to regulate motoneuron survival. Such interactions, along with intact synaptic basal lamina, may help to regulate the supply and presentation of trophic factors to motoneurons.

The synaptic conductance of the On-Off direction-selective ganglion cells was measured during visual stimulation to determine whether the direction selectivity is a property of the circuitry presynaptic to the ganglion cells or is generated by postsynaptic interaction of excitatory and inhibitory inputs. Three synaptic asymmetries were identified that contribute to the generation of direction-selective responses: (1) a presynaptic mechanism producing stronger excitation in the preferred direction, (2) a presynaptic mechanism producing stronger inhibition in the opposite direction, and (3) postsynaptic interaction of excitation with spatially offset inhibition. Although the on- and off-responses showed the same directional tuning, the off-response was generated by all three mechanisms, whereas the on- response was generated primarily by the two presynaptic mechanisms. The results indicate that, within a single neuron, different strategies are used within distinct dendritic arbors to accomplish the same neural computation.

Whole body studies of Plotosus tandanus revealed that ampullary pores occur over the entire body of the fish, but are in higher concentrations in the head region. These pores give rise to a short canal (50-60 mum) produced by columnar epithelial cells bound together by tight junctions and desmosomes. At the junction. of the canal and the ampulla, cuboidal epithelial cells make up the wall. The ampulla consists of layers of collagen fibers that surround flattened epithelial cells in the lateral regions and give rise to supportive cells-that encase a small number of receptor cells (10-15). The ampullary wall comprises several types of cells that are adjoined via tight junctions and desmosomes between cell types. The ovoid receptor cells possess microvilli along the luminar apical area. Beneath this area, the cells are rich in mitochondria and rough endoplasmic reticulum. An unmyelinated neuron adjoins with each receptor cell opposite multiple presynaptic bodies. This form of microampulla has not been previously described within the Family Plotosidae. (C) 2002 Wiley-Liss, Inc.

Atm gene-disrupted mice recapitulate the majority of characteristics observed in patients with the genetic disorder ataxia-telangiectasia (A-T). However, although they exhibit defects in neuromotor function and a distinct neurological phenotype, they do not show the progressive neurodegeneration seen in human patients, but there is evidence that ataxia-telangiectasia mutated ( Atm)-deficient animals have elevated levels of oxidized macromolecules and some neuropathology. We report here that in vitro survival of cerebellar Purkinje cells from both Atm knock-out and Atm knock-in mice was significantly reduced compared with their wild-type littermates. Although most of the Purkinje neurons from wild-type mice exhibited extensive dendritic elongation and branching under these conditions, most neurons from Atm-deficient mice had dramatically reduced dendritic branching. An antioxidant ( isoindoline nitroxide) prevented Purkinje cell death in Atm-deficient mice and enhanced dendritogenesis to wild-type levels. Furthermore, administration of the antioxidant throughout pregnancy had a small enhancing effect on Purkinje neuron survival in Atm gene-disrupted animals and protected against oxidative stress in older animals. These data provide strong evidence for a defect in the cerebellum of Atm-deficient mice and suggest that oxidative stress contributes to this phenotype.

In the rodent central nervous system (CNS) during the five days prior to birth, both growth hormone (GH) and its receptor (GHR) undergo transient increases in expression to levels considerably higher than those found postnatally. This increase in expression coincides with the period of neuronal programmed cell death (PCD) in the developing CNS. To evaluate the involvement of growth hormone in the process of PCD, we have quantified the number of motoneurons in the spinal cord and brain stem of wild type and littermate GHR-deficient mice at the beginning and end of the neuronal PCD period. We found no change in motoneuron survival in either the brachial or lumbar lateral motor columns of the spinal cord or in the trochlear, trigeminal, facial or hypoglossal nuclei in the brain stem. We also found no significant differences in spinal cord volume, muscle fiber diameter, or body weight of GHR-deficient fetal mice when compared to their littermate controls. Therefore, despite considerable in vitro evidence for GH action on neurons and glia, genetic disruption of GHR signalling has no effect on prenatal motoneuron number in the mouse, under normal physiological conditions. This may be a result of compensation by the signalling of other neurotrophic cytokines.

A Internet é um meio privilegiado de comunicação, no qual a informação não conhece fronteiras, e a sua divulgação toma uma dimensão universal. No mundo empresarial a comunicação é hoje tida, como o elemento fulcral dos processos de negócio, em que interação com as partes interessadas, pode ser traduzida em termos do seu expoente máximo pelo website da empresa. Grande parte da informação hoje divulgada à sociedade pela empresa, tem por objetivo demonstrar a «transparência» dos processos da organização, promovendo assim o seu escrutínio público. A investigação visou a análise exploratória dos websites de 422 empresas que se encontravam certificadas no ano 2011, no âmbito dos sistemas de gestão da qualidade (ISO 9001), ambiente (ISO 14001), segurança e saúde do trabalho (OHSAS 18001/NP 4397). O objeto de estudo visou determinar a percentagem de empresas que recorre ao website, para disponibilizar a Política da Segurança e Saúde do Trabalho (PSST), às partes interessadas (stakeholders), no cumprimento da alínea g) do requisito normativo 4.2 dos referenciais OHSAS 18001/NP 4397, bem como compreender a extensão ao website do requisito 4.4.3.1 (comunicação) no âmbito da divulgação da PSST.

A Internet é um meio priviligiado de comunicação, no qual a informação não conhece fronteiras, e a sua divulgação toma uma dimensão universal. No mundo empresarial a comunicação é hoje tida, como o elemneto fulcral dos processos de negócio , em que interação com as partes interessadas, pode ser traduzida em termos do seu exposente máximo pelo website da empresa. Muita da informação hoje divulgada à sociedade pela empresa, visa o seu próprio escrutínio público, na strês dimensões (económica, ambiental e social) do Desenvolvimento Sustentável (DS). A presente investigação visou uma análise exploratória dos websites de 523 empresas que se encontravam certificadas, no fianl do ano de 2011, no âmbito dos sistemas de gestão da qualidade (ISO 9001), ambiente (ISO 14001), segurança e saúde do trabalho (OHSAS 18001). O obketo de estudo proposto visou conhecer quais os conteúdos que são frequentemente mais divulgados, e que perfil tem a empresa portuguesa de Gestão (SIG), Sustentabilidade Empresarial (SE) e Responsabilidade Social Empresarial (RSE). O método de investigação baseou-se na técnica da Análise de Conteúdo, a qual permitiu quantificar por categoria de análise (indicador) e subcategorias de análise (itens), os conteúdos da informação divulgada no website da empresa. A visão holística sobre a dimensão dos contéudos disponibilizados no website da empresa, foir determinada (medida) com auxílio de divulgação no website das empresas, são: a Missão, a Política da Qualidade, Ambiente e Segurança, o Código de Ética & Conduta Empresarial, o Relatório de Sustentabilidade, o Relatório e Contas, os Indicadores & Índices Financeiros, os certificados e marcas do Sistema de Gestão da Qualidade (SGQ), do trabalho (SGSST) e, por fim, os Projetos de Envolvimento com a Comunidade no âmbito da RSE. Em termos de perfil, as empresas portugueas de maior dimensão (volume de vendas), e as sociedades anónimas (S.A:), divulgam mais informação no website, que as outras. Não tendo sido porém, encontrada evidência estatística significativa no estudo da relação das empresas localizadas no distrito de Lisboa & Setúbal, das empresas do setor de atividade secundário (2.º), e das empresas com exposição aos media (rankings nacionais), com a maior divulgação de informação no website da empresa.

São mostrados os resultados da avaliação da eficiência do programa CEAPE (Centro de Educação e Alimentação do Pré-Escolar), eficiência esta estimada por meio de sistema simplificado compatível com as infra-estruturas locais. A freqüência média do pré-escolar foi considerada alta, uma vez que as médias anuais de 56 Centros de Educação e Alimentação do Pré-Escolar (CEAPE) em andamento, esteve acima de 85% (87,4% em 1978, 89,7% em 1979). A freqüência média das "mães" dos pré-escolares também foi tida como alta, desde que 75% das unidades alcançaram mais de 90% de participação. A merenda servida não foi padronizada uma vez que ela foi assumida espontaneamente e extra-oficialmente pelas autoridades municipais; nestas condições seu valor calórico unitário variou de acordo com os recursos locais, entre 250 e 350 calorias e a média protéica esteve em torno de 13 gramas diárias. As atividades lúdicas sociais e cognitivas foram selecionadas e distribuídas equitativamente entre as diferentes áreas do desenvolvimento infantil o que foi conseguido com a aplicação de um "Plano Rotativo de Recreação Orientada". O programa CEAPE, mesmo sendo de modesta sofisticação, conseguiu alcançar desejável nível de eficiência em sua operacionalização.

O Corpo enquanto matéria, transformado em tela sobre a qual se inscrevem mensagens gritantes, é muito mais que um cenário que se constrói para enquadrar mensagens ou facilitar a comunicação com o receptor. O Corpo passa a ser a própria mensagem. É a imagem física de uma identidade que se busca hoje com sofreguidão para que possamos perceber quem somos. As remodelações desse espelho de identidades são constantes, são projectos sempre inacabados, sempre imperfeitos, com as possibilidades sempre em aberto e onde a essência do Ser não é tida como estática e absoluta. Cada vez mais, de forma mais flagrante, essa necessidade de Ser do indivíduo moderno se entrelaça com a necessidade de possuir bens de consumo que significam algo acerca da sua essência. Estes, são bens particularmente significantes, escolhidos com criteriosa atenção para compor o quadro que a tela do corpo socialmente mostra. Culturalmente, cada vez se esbatem mais as fronteiras entre Ser e Ter. O Corpo eleito como suporte privilegiado das mensagens que se pretendem construir é palco tanto das propostas artísticas, como das imagens que invadem os media e que apresentam elas próprias propostas de identidade. São deco-identidades onde os bens de consumo funcionam como adereços e o corpo é tido como espaço transformável e estilizado.
